        인공모델막에 대한 Polyene 계 항생물질의 영향 ( 2 )

        The effects of water soluble derivatives of polyene antibiotics, amphotericin B methyl ester (AME) and nystatin methyl ester, on the osmotic behavior of sterol-containing liposomes were investigated. The activity of polyene methyl esters on different sterol-containing liposomes as a function of sterol contents was observed. Cholesterol, ergosterol and 7-dehydrocholesterol were incorporated to egg phosphatidylcholine/dicetyl phosphate liposomes, which showed a good osmotic stability. When polyene derivatives were added to liposomes containing ergosterol, the osmotic stability was disturbed at final concentration of 6.45×10^(-5)M. Cholesterol-containing liposomes were not affected by these antibiotic solutions of the same concentration. 7-Dehydrocholesterol-containing liposomes have shown the intermediate sensitivity to AME when compared with cholesterol- and ergosterol-containing liposomes. Selectiveactivity on ergosterol-containing liposomes was also observed for nystatin methyl ester at a higher concentration above 2.31×10^(-4)M. These results suggest that AME is more potent than nytatin methyl ester and that the selectivity of polyene antibiotics for fungi is due to a stronger interaction of polyenes with ergosterol than with cholesterol.
