Anti-septic effects of pelargonidin on HMGB1-induced responses in vitro and in vivo

A certain nucleosomal protein—high mobilitygroup box-1 (HMGB1)—has recently been established as alate mediator of sepsis, with a relatively wide therapeuticwindow for pharmacological intervention. Pelargonidin(PEL) is a well-known red pigment found in plants; it hasimportant biological activities that are potentially beneficialfor human health. In the present study, we investigatedwhether PEL can modulate HMGB1-mediated inflammatoryresponses in human umbilical vein endothelial cells(HUVECs) and in mice. The anti-inflammatory activities ofPEL were determined by measuring permeability, leukocyteadhesion and migration, and activation of pro-inflammatoryproteins in HMGB1-activated HUVECs andmice, as well as the beneficial effects of PEL on survivalrate in the mouse sepsis model. The data showed that PELhad effectively inhibited lipopolysaccharide (LPS)-inducedrelease of HMGB1 and suppressed HMGB1-mediatedseptic responses, such as hyperpermeability, adhesion andmigration of leukocytes, and expression of cell adhesionmolecules. Furthermore, PEL inhibited the HMGB1-mediatedproduction of tumor necrosis factor alpha (TNF-a)and interleukin 6 (IL-6), as well as the activation of nuclearfactor-jB (NF-jB) and extracellular signal-regulatedkinases 1 and 2 (ERK1/2). Collectively, these resultsindicate that PEL could be used to treat various severevascular inflammatory diseases via the inhibition of theHMGB1 signaling pathway.
Suppressive effects of zingerone on TGFBIp-mediated septic responses

Zingerone (ZGR), a phenolic alkanone isolatedfrom ginger, has been reported to possess various pharmacologicalactivities. Transforming growth factor b-inducedprotein (TGFBIp) is an extracellular matrix proteinwhose expression in several cell types is greatly increasedby TGF-b. TGFBIp is released by human umbilical veinendothelial cells and functions as a mediator of experimentalsepsis. We hypothesized that ZGR could reduceTGFBIp-mediated severe inflammatory responses inhuman endothelial cells and mice. Here, we investigatedthe anti-septic effects and underlying mechanisms of ZGRagainst TGFBIp-mediated septic responses. ZGR effectivelyinhibited lipopolysaccharide-induced release ofTGFBIp and suppressed TGFBIp-mediated septic responses. In addition, ZGR suppressed TGFBIp-induced sepsislethality and pulmonary injury. In conclusion, ZGR suppressedTGFBIp-mediated and CLP-induced septicresponses. Therefore, ZGR could be a potential therapeuticagent for treatment of various severe vascularinflammatory diseases via inhibition of the TGFBIp signalingpathway.
도시와 농촌 노인의 골다공증 지식, 자기효능감과 예방적 건강증진행위 비교 연구

Purpose: The purpose of this study was to investigate the osteoporosis knowledge, self-efficacy, and preventive health promoting behaviors of the elderly who reside in urban and rural areas. Methods: Convenience sampling was used (urban elderly=127, rural elderly=112). The Osteoporosis Quiz, Osteoporosis Self-Efficacy scale, and Health Promotion Inventory for the Prevention of Osteoporosis were used to collect data. Data were analyzed using chi-square, t-tests, two-way ANOVAs, and multiple linear regression. Results: Elders residing in rural areas had significantly less knowledge of osteoporosis (t=-4.884, p<.001), self-efficacy (t=-6.153, p<.001), and preventive health-promoting behaviors (t=-5.710, p<.001). Multiple-regression analysis showed osteoporosis self-efficacy (p=.002) and chronic disease (p=.027) were significant predictors of preventive health-promoting behaviors for elders in urban areas. In rural areas, osteoporosis knowledge (p=.002), osteoporosis self-efficacy (p<.001), and regular medical examinations (p=.014) were significant predictors of preventive health-promoting behaviors. Conclusion: Promoting osteoporosis knowledge and self-efficacy may be significant in improving preventive health-promoting behaviors. Also, nursing interventions suitable for rural elders should be developed to increase their level of preventive health-promoting behaviors.
