      • 중금속이 nalidixic acid 향균활성에 미치는 효과

        In the present study, the metallodrug formation of nalidixic acid, the first member of the quinolone antibiotics with heavy metal ions such as Cu^2+ and Tb^3+ was studied by UV-spectrophotometric titration method, and the effect of the nalidixic acid-metal complex on the growth of both gram negative (E. coli K1100) and gram positive (B. subtilis KCTC1210) bacteria was investigated. It was found that the complex was more effective in the inhibition of the growth than only nalidixic acid alone without the metal. The antibiotic effect of the metallodrug may result from the inhibition of DNA synthesis of the bacteria.

      • Bacillus subtilis KS1이 생산하는 항진균물질의 정제 및 특성

        인삼포 토양으로부터 강력한 항진균력을 보이는 KS1 균주를 분리하고 Bacillus subtilis로 동정하였다. 온상시험에서 B. subtilis KS1의 배양액은 오이잿빛곰팡이병, 밀붉은녹병 등의 진균성 농작물병에 대하여 탁월한 방제효과를 나타내었다. 또한, B. subtilis KS1 배양액의 butanol 분획은 Botrytis maydis, Chytridium logenarium, Candida albicans 등 식물 또는 사람의 병원성 진균을 비롯한 몇몇 진균류에 대하여 억제효과를 나타내었다. 이 균주가 생성하는 항진균물질을 pep-RPC reverse phase column과 μ Bondapak C_18 reverse phase column을 이용하여 분리 정제 하였다. 정제된 항진균물질의 열안정성 및 pH 안정성을 조사한 결과, -20-121℃와 pH 4.0-10.0의 범위에서 안정하였다. 이 물질의 조성 및 구조적 특징을 HPLC와 ^1H-, ^1H-^1H-, COSY, NOESY, COSY-NOESY 및 HOHAHA NMR spectroscopy를 통하여 각각 분석한 결과, B. subtilis가 생산하는 고리형 peptide 중 iturin A에 속하는 물질로 확인되었다. 그러나, 지금까지 알려진 iturin A계 물질들의 β-아미노산에 붙어 있는 탄화수소 사슬이 곁가지가 없거나 terminus 또는 subterminus에 1개의 -CH_3 곁가지를 갖는 것과는 달리, SW1의 β-아미노산은 2개의 -CH_3 곁가지가 붙어 있는 탄화수소 사슬을 가짐이 확인되었다. A bacterial strain, KS1, possessing strong antifungal activity was isolated from soil samples of ginseng fields and identified as Bacillus subtilis. In greenhouse test, the culture filtrate of B. subtilis KS1 showed strong protective effect against several fungal diseases of agricultural plants such as cucumber gray mold and wheat leaf rust. In addition, the crude butanol fraction of the culture filtrate exhibited antagonistic effect against several fungi including plant or human pathogens, such as Botrytis maydis, Chytridium lagenarium and Candida albicans. The antifungal compound, SW1, produced by B. subtilis KS1 was purified through consecutive chromatographic separations on a pep-RPC column and a μ Bondapak C_18 reverse phase column. Temperature and pH showed little effect on the stability of the compound in the ranges -20-121℃ and pH 4.0-10.0, respectively. The composition and structural characteristics of SW1 were analysed by HPLC and by ^1H-, ^1H-^1H- COSY, NOESY, COSY-NOESY and HOHAHA NMR spectroscopy, respectively, which revealed that the compound belongs to iturin A, a typical cyclic antifungal compound produced by B. subtilis. In contrast to the previously reported iturin A compounds which have one or no -CH, side chain in the hydrophobic hydrocarbon chain of β-amino acids, SW1 was shown to have a β-amino acid containing 12-carbon skeleton with two -CH, side chains.

      • 염기성 폴리펩티드류에 의한 p-Nitrophenyl Acetate의 가수분해

        The hydrolysis of p-nitrophenyl acetate was catalytically promoted by the synthetic basic polypeptides. The first-order rate constants were linearly proportional to the tested concentration range (up to 4 mg/ml) of the basic polypeptides. The 1:1 (molar ratio of basic to hydrophobic residue) copolymer showed higher reactivity than the 4:1 copolymer. A plot of the logarithm of the second-order rate constants (Log K_cat/K_R) versus pK_a values of the basic residues showed linear-free-energy -relationship (LFER), and the slope was 0.4.

        內病外治藥物에 관한 文獻的 硏究

        The external treatment of internal diseases was first recorded in Hwangjenaegyeong(黃帝內經), and two hundred and fifty odd cases were found in the Bonchogangmok(本草綱目) and Oechisusebang(外治壽世方). Bonchogangmok(本草綱目) is a technical book devoted to explaining materia media as well as the external treatment of internal diseases. Oechisusebang(外治壽世方) is also a technical book, which make 36 references to the disease patterns of internal medicine. The medicinals employed as external treatment of internal diseases are grouped into 300 classes. These medicinals do not set limits to trees and plants but also uses birds and beasts, reptiles, grains, feces and urine, soil, stone etc., applied to various disease patterns. This study researched the medical texts in order to treat the disease patterns of internal medicine.

      • 4방향성 탄소/탄소 복합재의 함침거동 특성

        For the preparation of 4-directional carbon fiber reinforced carbon composites(4D CFRCs) which have good thermal stability and high mechanical properties under the high temperature, pressure impregnation, carbonization and graphitization processes were performed after fabrication of 4D preform using the pultruded rod and weaving method. Due to the good compactability of pitch matrix precursor and reinforcement, carbon matrix was infiltrated effectively into the pore and crack of 4D CFRCs during the densification process. Carbon yield of matrix pitch was high level by adapting of the pressure impregnation and carbonization process. Rod network type 4D preform was more easily densified than weaving type 4D preform.

      • 형광을 이용한 미생물 성장의 연속 측정 방법

        연속적 미생물 성장 측정을 위해 배양 세포의 형광을 이용하는 방법에 대하여 가능성과 문제점들을 알아보고 computer를 연결시켜 발효조 내의 미생물 농도를 on-line으로 측정할 수 있는 시스템을 구성하였다. 실험 결과 세포의 형광을 이용한 미생물 성장의 연속 측정 방법은 배지 조성물의 영향을 배제한다면 실용화 가능성이 좋은 방법임을 알 수 있었다. 높은 세포 농도에서도 희석없이 미생물 농도의 측정이 가능하며 특히, 발효조의 기존 sampling line을 이용하여 배양액을 순환시켜 fluorometer로 형광을 측정하는 방법을 간단히 발효조에 적용하여 사용할 수 있는 장점이 있다. Fluorometer에서 나오는 신호를 computer로 처리한 결과 효과적으로 발효조의 미생물 성장을 관찰할 수 있었다. Culture Fluorescence was measured to follow the cell growth in a fermentor. Signals from a fluorometer were connected to an Apple computer via A/D converter and operational amplifier. Medium composition should be considered to avoid a high background fluorescence originated from fluorophores in the medium. Measuring fluorescence has an advantage over the O.D. measurement since no dilution is needed at a high cell concentration. Cell broth circulated through a flow cell in the fluorometer provides us with monitoring cell growth in the fermentor.

        『黃帝內經』中"死"字意味硏究

        Objective : Every language has twofold aspects in meaning of characters such as ``lead`` means guide as a verb and ``lead`` means one of metals. Chinese character has several meanings because it is phonogram, even if those are exactly same in shapes. we have concern how different meaning of the character ``Si(死)`` is in traditional medical classics. Method : we try to find out the meaning of ``Si`` excluding meaning of ``Si``, ``death`` in 『Huangdineijing』. Result : ``Si`` means ``death`` primary and means ``serious disease to death``, ``losing consciousness just like death`` and so on. Conclusion : Someone who have concern traditional medical classics have to give attentions that characters in classics could get different meanings in same character.
