        산말의 항산화 및 멜라닌 생성 억제활성

        This study investigated the functional study in anti-oxidant and anti-melanin activity by Desmarestia dudresnayi subsp. Tabacoides, Desmarestia viridis, Desmarestia ligulate leaf extracts. The DPPH radical scavenging activity of D. viridis extract showed a high antioxidant activity of 68.0 ± 1.9% at the concentration of 2.5 mg / mL, the ABTS and scavenging activity showed a high antioxidant activity of 84.6 ± 1.7% at the concentration of 2.5 mg / mL, and the NO radical scavenging activity was 2.5. It required a high antioxidant capacity of 91.6 ± 1.1% at mg/mL concentration. In addition, in B16F10 cells, at a concentration of 100 ㎍/mL, D. viridis extract is 35.0 ± 1.7% melanin among, which has the lowest amount of melanin among the three species. With these contents, it seems that there is a possibility that D. viridis could be developed as a cosmetic material. These results D. viridis considered that it can be used as a natural cosmetic material such as anti-oxidant and anti-melanin activity. 본 연구는 산말 3종류 Desmarestia dudresnayi subsp. Tabacoides (담배잎산말), Desmarestia viridis (쇠꼬리산말), Desmarestia ligulate (참산말) 잎 추출물의 항산화 및 항 멜라닌 활성에 대한 기능적 연구를위해 수행하였다. 3종류의 산말 중 담배잎산말 추출물이 2.5 mg / mL의 농도에서 DPPH 및 ABTS 라디칼 소거활성이 각각 68.0 % ± 1.9 % 및 84.6 % ± 1.7 %로 강한 항산화력을 나타냈다. 질소 (NO) 라디칼 소거 활성은 91.6 % ± 1.1 %의 높은 항산화능을 보였다 산말 추출물을 이용한 B16F10세포의 세포독성은 100 ㎍/mL 농도에서 3종의 산말 추출물이 모두 85% 이상의 세포 생존률을 보였고, 100 ㎍ / mL농도에서 담배잎산말 및 쇠꼬리산말이 70% 이상의 멜라닌 억제효과를 보였다. 이러한 내용은 산말이 항산화 및 항멜라닌 활성과 같은 천연 화장품 소재로 사용될 수 있을 것으로 사료된다.

        Purpose: This research is to determine the degree of knowledge about cervical cancer, Human Papillomavirus(HPV), HPV vaccination of boy and girl high school students in Republic of Korea, and aimed to provide a basis of the education and policies which are for the reduction of HPV infection risks. Methods: The research subjects consisted of 132 boy students and 142 girl students. The data were collected using self-reported questionnaires composed of 10 questions about cervical cancer, 10 questions about HPV, and 5 questions about HPV vaccination. The collected data were analyzed with frequency, percentage, average, standard deviation, t-test and ANOVA by using SPSS version 19.0. Results: As a result, the degree of knowledge about cervical cancer, HPV, HPV vaccination of the research subjects was very low, and each average was 4.47±2.98, 2.79±2.91, and 1.20±1.46 points. There were statistically significant differences in the degree of knowledge about cervical cancer, HPV, and HPV vaccination depending on the research subjects’ sex (p<0.01), school types (p<0.05), majors (p<0.05), whether they had heard about cervical cancer or not (p<0.01). Conclusion: Concrete educational programs about cervical cancer, HPV, and HPV vaccination for high school students should be developed considering the students’ sex, school types, and majors.

        산말의 기능성 탐색 및 기능성물질 D-Mannuronic acid의 항염활성

        This study was conducted to investigate the anti-inflammatory activity of Desmarestia ligulate (D. ligulate) leaf. The effects on cell viability and anti-inflammatory activities of D. ligulata leaf extract, as well as those of D-mannuronic acid (DMA), on RAW 264.7 cells (macrophages) and peripheral blood mononuclear cells (PBMCs) during inflammation-induced stress were analyzed by determining the levels of NO, IL-17, IL-4, and TNF-α using ELISA and evaluating the expression of iNOS and MPO genes. D. ligulata leaf extract and DMA exhibited low toxicity in RAW 264.7 cells and PBMCs, which secreted lower levels of TNF-α and IL-4. The anti-inflammatory effect of DMA was higher than that of D. ligulata leaf extract as indicated by the levels of the inflammatory cytokine IL-17. Furthermore, treatment with D. ligulata leaf extract and DMA decreased the expression of iNOS and MPO genes after stress induction. These findings suggested that D. ligulata leaf extract and DMA exert anti-inflammatory effects as well as regulate cytokine release activities and inflammatory gene expression. Therefore, D. ligulata leaf extract and DMA may be used as cosmetic materials to control skin inflammation.

        마그놀롤의 HDF세포에서 Nrf2-SOCS3-Jak2-STAT3에 의한 UVB 유래 염증데미지 조절

        본 연구는 magnolol에 의한 UVB 유도 세포 손상의 복구를 조사하였다. 우리는 약물재배치를 위해 STAT3 기작을 분석하였고, magnolol HDF 세포에서 세포 생존력을 향상시키며, STAT3의 억제제인 것을 확인하였다. IL-6, UVB 및 IFNγ로 처리 된 HDF 세포는 Jak2 및 인산화 된 STAT3 (p-STAT3)의 높은 발현을 나타냈다. Magnolol의 처리는 UVB 유도 세포에서 Jak2 및 p-STAT3의 발현을 감소시킬 수 있었다. 또한, UVB- 손상된 세포 성장은 용량 의존적 방식으로 재 활성화 및 magnolol 과의 상관 관계가 상당히 증가되었다. UVB 처리 된 HDF 세포에 대한 AG490 (Jak2 억제제) 처리와 비교하여, 세포 증식이 유의하게 증가 하였다. 우리는 AG490 및 magnolol 이 TNF-α 농도를 감소시키는 것을 확인했다. Western blot (단백질 수준)은 오직 magnolol 처리 된 세포에서만 Jak2 및 p-STAT3 발현의 감소를 나타냈고, Jak2, p-STAT3 및 SOCS3의 발현은 또한 magnolol 처리한 세포에서만 증가하였다. 세포를 magnolol 및 ML385 (NRF2 억제제)로 동시 처리시 세포 증식 및 NRF2 발현을 감소시켰다. MMP9의 양은 magnolol 및 ML385 로의 처리에 의해 증가되었다. 종합적으로, 이들 결과는 NRF2, SOCS3, Jak2 및 STAT3의 발현을 조절함으로써 UVB 손상 후 세포를 회복시키는데 있어 magnolol의 가능성을 입증한다. This study investigated the repair of UVB-induced cell damage by magnolol. We performed a drug-repurposing screen, and, in the STAT3 reporter gene assay, magnolol was identified as a suppressor of STAT3 that improves the cell viability of HDF cells. HDF cells treated with IL-6, UVB, and IFNγ showed the highest expression of Jak2 and phosphorylated STAT3 (p-STAT3), and magnolol was able to decrease the expression of Jak2 and p-STAT3 in UVB-induced cells. Moreover, UVB-damaged cell growth increased significantly in correlation with both reactivation and with magnolol in a dose-dependent manner. Compared with AG490 (a Jak2 inhibitor) treatment of UVB-treated HDF cells, cell proliferation increased significantly. We confirmed that AG490 and magnolol reduced TNF-α concentrations, and Western blotting (protein level) showed decreases in Jak2 and p-STAT3 expression in only the magnolol-treated cells. The expression of Jak2, p-STAT3, and SOCS3 also increased only after treatment with magnolol. Cells were treated with magnolol and ML385 (an NRF2 inhibitor), and these secondary metabolites reduced cell proliferation and NRF2 expression. The amount of MMP9 was also increased by cotreatment with magnolol and ML385. Collectively, these results demonstrate the potential of magnolol for repairing cells after UVB-induced damage by regulating the expression of NRF2, SOCS3, Jak2, and STAT3.

        홍만병초 분획물에 의한 HT-29대장암 세포의 Wnt/β-catenin 신호전달 조절

        본 연구에서는 Rhododendron brachycarpum (RB, 홍만병초)의 80% 메탄올 추출물 및 분획물의 항암 활성을 규명하고자 하였다. RB n-hexane 분획물은 HT-29 세포에서 가장 높은 활성 저해를 보였다(IC50 = 20.2±1.2 μg/ml). 더욱이, 콜로니와 구형 형성은 수와 크기는 유의적으로 감소시켰다. RB의 n-hexane 분획물에서(0.22±0.02 fold change) TOP / FOP 플래시 리포터 억제 활성은 추출물 및 다른 분획물 보다 낮게 나타났다. n-hexane 및 ethyl acetate 분획물은 세포 내 β-catenin의 발현을 조절하였다. 2 차 대사 산물이 β-catenin 분해를 감소시킬 수 있는지 여부를 조사하기 위해 Western blot을 실시한 결과 n-hexane 분획물에서 p-GSK3β를 조절하였으며, 세포내 β-catenin은 핵에서 정량적인 변화를 가져왔다. 이러한 결과는 RB의 n-hexane 분획물로부터 천연 항암 물질을 포함하고 있음을 보여줍니다. The aim of this study was to determine the anticancer activities of an 80% methanol extract and various fractions of Rhododendron brachycarpum (RB). The n-hexane fraction of RB showed the highest inhibitory activity (Inhibit concentration 50% = 20.2±1.2 μg/ml) in HT-29 cells. Colony- and sphere-forming abilities were significantly correlated with a decrease in the cell count and size. A TOP/FOP flash reporter assay revealed that the inhibitory activity of the n-hexane fraction of RB (0.22±0.02 fold change) was lower than that of the 80% methanol extract and that of other fractions. The n-hexane and ethyl acetate fractions of RB were predominantly dependent on the expression levels of intracellular β-catenin. Western blotting using p-GSK3β with only the n-hexane fraction of RB was conducted to examine whether these secondary metabolites reduced β-catenin degradation. Intracellular β-catenin regulation resulted in quantitative changes in the nucleus. In summary, these results demonstrate the potential of the n-hexane fraction of RB as a natural anticancer agent.
