      • Identification and Characterization of a RecA-like Protein Induced by DNA Damaging Agents in Fluorescent Pseudomonas sp.

        A RecA-like protein (RecAps) was identified from fluorescent Pseudomonas sp. and the inducible nature of the protein was characterized in detail. It was shown by dose-response and time-course experiments using two DNA damaging agents, nalidixic acid and mitomycin-C, that the cellular level of RecAps protein was increased 3-8 fold compared to that of the control. The most effective doses of nalidixic acid and mitomycin-C for the protein induction were $30{\mu}g/ml$ and $0.3{\mu}g/ml$ at the treatment time point of 150 min, respectively. The enhanced level of RecAps protein was gradually decreased to the control level after 10 hr in normal medium. Interestingly, the cellular level of RecAps protein was increased by the same DNA damaging agents even when cell growth was completely inhibited by treatment with $170{\mu}g/ml$ of chloramphenicol, an inhibitor of protein synthesis, suggesting that new protein synthesis is not required for the induction of RecAps. All these results suggest that a typical S0S repair function driven by RecA-like protein is conserved in Pseudomonas sp. cells as in E, coli.

        The affects of the temperature and light on the germination of the seed of three kinds of the radish(F1 Fine Leaf, Treated Spring Radish and F1 King Kong) were studied at varied temperature(10℃∼28℃) with the light for a period of 12 hours and in the dark. The mean germination rate of all radish at the low temperature was lower than the rate at the high temperature. The significant difference was between the low and high temperature after 24 hours but the non-significant difference was after 72 hours. The temperature of maximum germination rate of F1 Fine Leaf and Treated Spring Radish was 22℃ and F1 King Kong was 28℃. When photoperiod was 12 hours, three kinds of the radish initiated to germinate in 24 hours. In dark condition, F1 Fine Leaf and Treated Spring Radish initiated after 24 hours, and F1 King Kong in 24 hours at the low temperature.

        The effects of salinity on the growth and production of rice were studied at Seosan reclaimed land from July to October, 1995. The plant height, the number of living leaves, dead leaves and total leaves, the number of the grains and the dry weight of the grains per individual, and the dry weight of above the ground in 25cm×25cm quadrat were investigated on 5 plots where were different salinity. The plant height, the number of living leaves and total leaves and the number of grains and the dry weight of grains per individual, and the dry weight of above the ground decreased as the salinity of water increase and the number of dead leaves of rice increased as the salinity of water decrease. The effect of salinity on the reproductive production is severer than the vegetative production. Because of the salinity, the growth and the production of the rice at Seosan reclaimed land are worse than the normal rice field.

        The effects of the temperature and light on the seed germination of three kinds of the chinese cabbage(F1 Glamour, F1 Spring Green and F1 D-R 60 days) were studied at varied temperature(10℃~28℃) with the light and the dark for a period of 12 hours, and with the dark for 24 hours in a day. The mean seed germination rate of three kinds of chinese cabbage at the low temperature was lower than the rate at the high temperature and the significant difference accepted. The temperature of maximum seed germination rate of F1 Glamour and F1 Spring Green was 25℃, and F1 D-R 60 days was not different according to the temperature gradient with the light and the dark for a period of 12 hours. When photoperiod was 12 hours, three kinds of the chinese cabbage initiated to germinate within 24 hours at all temperature. In dark condition, F1 Glamour and F1 D-R 60 days initiated to germinate after 24 hours, and F1 Spring Green initiated after 48 hours at 10℃. The mean seed germination rate of three kinds of chinese cabbage with the light and the dark for a period of 12 hours was higher than the rate in the dark. In the dark condition, the light accelerated the seed germination of the chinese cabbage on the low temperature.

        자동차 판매 비즈니스 모델 연구

        The objective of this study is offering new on line business model for automotive market. In order for the research aim, we conduct direct in-depth interviews with CEO, including analysis of business patterns on eight automotive firms in Gwangju city. As a result, we suggest new business model so called, Online to offline (o2o) that combines advantages in both off-line and on-line business. Techniques that o2o automotive companies may employ include in-store pick up of items purchased online, allowing customers. Purchasing the automotive is not only a car but also services including insurance, warrant services. o2o business model suggested shows the operating of the goods and service sale. When it comes to characteristics of automotive conventional sale patterns, utilization of the online-to-offline commerce has not eliminated the advantages that e-commerce companies have. That model can enhance the new values for customers as well as new market creation. Also, this new business model (o2o) can contribute the emerging industry business model installed based in particular, emergence of Iot (Internet of thing) in terms of sharing knowledge and information between enterprise and customers.

      • Ribonucleotide Reductase Activity of Schizosaccbarornyces pombe Is Inhibited by Escherichia coli RecA Antibody

        분열형 호모인 Schizosaccbarornyces pombe에서 RecA 유사 단백질은 hydroxyurea (HU)나 methotrexate (MTX)와 같은 nucleotide pool 형성에 관여하는 효소의 저해제에 의해 유도발현된다. 본 연구에서는 대장균의 RecA 단백질과 S. pombe의 ribonucleotide reductase가 구조적으로 유사한지를 RecA 항체를 이용한 면역침전법과 [5-$^3$H]CDP를 기질로하는 thin layerchromatography방법을 통하여 알아보고자 하였다. S. pombe의 rinonucleotide reductase활성은 100mM HU에 의해 대조군에 비해 26.3% 저해되었으며, RecA 항체를 이용한 면역침전에 의해서도 43.3% 저해되었다. 이와 같은 결과는 S. pombe의 rinonucleotide reductase가 대장균의 RecA 단백질과 구조적으로 유사함을 시사하는 것이다. We have previoosly demonstrated that the RecA-like protein of Schizosaccharomyces pombe (S. pombe) is immunologically related to Escherichia coil (E. coil) RecA protein and that the cellular level of the protein is significantly increased by inhibitors of nucleotide pool-forming enzymes such as hydroxyurea (HU) and methotrexate (MTX) (lee and Park, 1994; lee et al., 1994). In this study, we report that the ribonudeotide redudase activity of S. pombe is inhibited by E. coil RecA antibody, as determined by thin layer chromatography using [5-$^3$H]CDP as a substrate. The relative activity of ribonucleotide reductase was dramatically inhibited by 100 mM of flu (26.4% reduction) in in vitro assay, compared to that of non-treated control. The ribonucleotide reductase activity was also inhibited by immunoprecipitation with E. coil RecA antibody (43.3% reduction). These results indicate that the strudure of S. pombe ribonucleotide reductase is in part similar to that of E. coil RecA protein.
