        Repression of Transcriptional Activity of Estrogen Receptor alpha by a Cullin3/SPOP Ubiquitin E3 Ligase Complex

        The role of SPOP in the ubiquitination of ERα by the Cullin3-based E3 ubiquitin ligase complex was investigated. We showed that the N-terminal region of SPOP containing the MATH domain interacts with the AF-2 domain of ERα in cultured human embryonic 293 cells. SPOP was required for coimmunoprecipitation of ERα with Cullin3. This is the first report of the essential role of SPOP in ERα ubiquitination by the Cullin3- based E3 ubiquitin ligase complex.         인삼추출물이 포화지방산 Lard 와 Alcohol 을 먹인 흰쥐 혈청 지질성분에 미치는 영향

        The present study has been undertaken to investigate the effects of crude ginseng extract added to food on the lipid composition in serum of rats fed lard and alcohol. Thirty-five males of Sprague-Dawley strains weighed about 130 g were divided into 7 groups, each group receiving a different diet for 10 weeks ; i.e. basal diet plus 15% lard, basal diet plus 5% alcohol, basal diet plus 0.5% crude ginseng extract, basal diet plus 15% lard and 0.5% crude ginseng extract. Determinations were carried out on the net weight gain, food efficiency ratio, weight of organs, triglyceride, total cholesterol, HDL-cholesterol, free cholesterol. The results obtained were as follows: 1. Rats given feed containing lard and alcohol showed significant decrease in net weight gain, but crude ginseng extract caused an increase in food efficiency ratio. 2. Lard supplementation group showed increase in the weight of liver, kidney, spleen, but another groups did not. 3. The contents ratio of triglyceride, total cholesterol in serum of lard-fed group showed significant increase compared to controls and but the simultaneous supplementation of crude ginseng extract and lard decreased sinificantly than those of lard-fed group. 4. The contents of free-cholesterol and ester cholesterol in serum of the simultaneous supplementation of crude ginseng extract group decreased as compared with the lard single fed group. 5. Crude ginseng extract single-fed group showed increse of plasma HDL-cholesterol compared to the control but not in any other groups. 6. The ratio of contents of VLDL, LDL-cholesterol and HDL-cholesterol in serum was significantly high in the only lard containing group and alcohol group, the crude ginseng extract supplemented group was showed lower tendency than only lard and alcohol group. The above results suggest that crude ginseng extract would prevent the metabolic disease by improving hyperlipidemia.

Lard supplementation caused an increase in the weight of liver, kidney, spleen, but another groups did not. 3. AST, ALT, ALP, LDH of serum were significantly increased in lard and alcohol containing group but garlic feeding decreased enzyme activities compared to lard and alcohol containing group.

      • Stearic acid, Oleic acid, Elaidic acid에 의한 간세포의 성장 및 지방산 조성의 변화

        This study was carried out in order to investigate the effect of stearic acid (18:0,SA), oleic (18:1cis.0A) and elaidic acid (18:1 trans, EA) on the cell growth and fatty acid composition of Hep-G2 cells. The cells were incubated in serum-free medium 25, 50, 100 and 200uM of fatty acid combined with albumin for 2 days. The fatty acid concentration up to lOOuM showed the normal growth, but the cell growth decreased in the presence of 200uM fatty acid. The treatment of cells with lOOuM of a fatty acid for two days significantly (p<0.05) increased the cellular triglyceride (TG) content in all fatty acid groups compared to control, but TG content was not significantly different among all treatment group, but total cholesterol (TC) was the highest level in EA group. Cellular phospholipid(PL) contents were similar between the control and all faty acid groups.

        황금약침의 급성독성에 관한 안전성 연구

        연구 목적: Sprague-Daewey rat에 의한 黃芩 藥針의 安全性을 검토하기 위해 國立 保健 硏究院 急性 毒性試驗 基準을 기본으로 하여 急性 毒性 試驗을 修行하였다. 방법: 食品醫藥品 安全性 고시 제1998-116호의 `醫藥品 등의 毒性 試驗 기준`에 준하여 실시하였다. SD계 rat는 독성 시험에 적당한 실험 동물로서 急性 毒性 試驗에 널리 사용되고 있다. 본 계통의 rat는 풍부한 試驗基礎資料가 蓄積되어 있어서, 評價 時 實驗 結果의 해석이 용이하다. 결과: 對照群을 포함한 모든 實驗群에서 外觀 및 一般 行動에서의 이상 症狀은 觀察되지 않았으며, 實驗군에 藥針을 시술한 직후의 行動 이상 및 投與 직후 사이의 行動 이상도 觀察되지 않았다. 또한 剖檢時 이상 소견이 對照群과 모든 投與群에서 觀察되지 않았다. 본 시험에서의 黃金藥針은 安全性에는 문제가 없는 것으로 사료되었다. 황금 약침 施術群에 대한 血液 生化學적인 檢査에서 glucose의 농도는 증가하는 경향(P<0.05)을 나타냈다. 반면에 GOT, GPT 농도는 黃金 藥針 施術群에서 오히려 감소하는 有意性 있는 흥미로운 結果를 얻을 수 있었다. 결론: 食品醫藥品 安全性 고시 제1998-116호의 `醫藥品 등의 毒性 試驗 기준`에 준하여 黃金藥針의 急性 毒性 試驗을 修行한 結果 본 試驗에서의 黃金 藥針은 安全性에는 문제가 없는 것으로 사료되었다.         길경 분말이 고지방식이로 유도된 비만 흰쥐 혈청 중의 간효소 활성도에 미치는 영향

        N/A Objectives: The present study has been undertaken to investigate the effects of Platycodi Radix powder on the activities of several enzymes in serum of rats fed high fat diet. Methods: Twenty four males of Sprague-Dawley strains weighed about 580g were divided into 4 groups, each group receiving a different diet for 10 weeks ; i.e. control group, control+ 20% Platycodi Radix powder group, high fat diet group, high fat diet + 20% Platycodi Radix powder group. Determinations were carried out on the net weight gain, food efficiency ratio, and AST, ALT, lactate dehydrogenase, alkaline phosphatase activities in serum of rats. Results: The results obtained were as follows : AST, ALT, ALP, LDH of serum were significantly increased in lard containing group but Platycodi Radix powder feeding groups decreased enzyme activities compared to lard group. Conclusions: The above results suggest that Platycodi Radix powder would prevent the metabolic disease of liver by preventing hyperlipemia caused by high fat diet.

        V79 배양 세포에 있어 인삼추출물이 DNA복제 및 회복 합성에 미치는 분자생물학적 연구

        본 연구는 인삼추출물이 DNA 복제 및 회복 합성에 미치는 영향을 조사하기 위해 수행되었다. DNA 복제 및 회복 합성도의 측정은 V79 chinese hamster lung cell의 핵 내 DNA에 [3H]-tymidine이 유입되는 양으로 측정하였다. 그 결과 인삼추출물의 농도(0~1ug/㎖)가 높을수록 점점 더 크게 감소하였다. 이와 같이 인삼추출물에 의하여 DNA 복제 합성도가 감소하는 현상은 농도가 더 높을수록 효과적인 것으로 보아 농도에 의존한다는 것을 알 수 있었다. 한편 DNA 회복 합성도에 인삼추출물이 미치는 영향을 알아보기 위해 우선 DNA를 손상시켰다. DNA를 손상시키는 물질은 여러 가지 있으나, 본 실험에서는 자외선과 methylmethane sulfonate(MMS)를 사용하였다. 이렇게 손상된 DNA가 인삼추출물(0~1ug/㎖)에 의하여 얼마나 회복되었는가를 알아보기 위하여 인삼추출물의 농도를 각각 달리하여 V79 cell에 첨가하였다. 그 결과 인삼추출물의 농도가 높을수록 DNA 회복합성도는 증가하였다. 이와 같이 인삼추출물이 DNA 복제 합성은 억제하고, DNA 회복 합성은 증가시키는 상반된 효과를 보여 주었다. 이러한 상반된 효과는 복제합성과 회복합성의 두 과정에서 서로 다른 효소체계가 관여하기 때문인 것으로 보인다. 인삼추출물이 DNA복제 및 회복합성에 미치는 영향에 대하여 보다 더 구체적인 분자생물학적 메커니즘 연구가 이루어져야 할 것이다.
