        Development of an immune or autoimmune response involves T-cell activation in lymphoid organs and subsequent migration to peripheral tissues. Here we show that T-cell-specific ablation of the kinase TBK1 promotes T-cell activation but causes retention of effector T cells in the draininglumph node in a neuroinflammatory autoimmunity model,experimental autoimmune encephalomyelitis (EAE).Atolder ager,the T-cell-conditional TBK1-knockout mice also spontaneously accumulate T cells with activated phenotype.TBK1 contrlos the activation of AKT and its downsream kinase m TIORC1 by a mechanism involving TBK1-stimulated AKT ubiquitination and degradation. The deregulated AKT-mTORC1 signalling in turn contributes to enhanced T-cell activation and impaired effector T-cell egress from draining lymph nodes. Treatment of mice with a small-molecule inhibitor fo TBK1 inhibits EAE induction. These results suggest a role for TBK1 in regulating T-cell migration and establish TBK1 as a regulator of the AKT-mTORC1 signalling axis.

        A Novel Virtual Space Vector Modulation Strategy for the Neutral-Point Potential Comprehensive Balance of Neutral-Point-Clamped Converters

        A novel Virtual Space Vector (VSV) modulation strategy for complete control of potential neutral point (NP) issues is proposed in this paper. The neutral point potential balancing problems of multi-level converters, which include elimination of low frequency oscillations and self-balancing for NP dc unbalance, are investigated first. Then a set of improved virtual space vectors with dynamic adjustment factors are introduced and a multi-objective optimization algorithm which aims to optimize these adjustment factors is presented in this paper. The improved virtual space vectors and the multi-objective optimization algorithm constitute the novel Virtual Space Vector modulation. The proposed novel Virtual Space Vector modulation can simultaneously recover NP dc unbalance and eliminate low frequency oscillations of the neutral point. Experiment results show that the proposed strategy has excellent performance, and that both of the neutral point potential issues can be solved.

        First-principles study of transport properties of endohedral Li@C20 metallofullerene

        The transport properties of the endohedral Li@C20 metallofullerene are studied using density functional non-equilibrium Green’s function method. The equilibrium conductance of Li@C20 metallofullerene becomes larger than that of the empty C20 fullerene molecule. The I.V curve under low-bias voltage shows the characteristic of metallic behavior; another, the novel negative differential resistance behavior is also observed. It is found that the doping effect of Li atom significantly changes the transport properties of C20 fullerene.

        Mechanism and Experimental Investigation of Ultra High Pressure Water Jet on Rubber Cutting

        The recycle and reuse of rubber materials has become a global environment problem with the fast increasing amount of waste rubber. Compared to traditional recycle methods, ultra high pressure water jet, as a fast developing tool of cool cutting, can be used inrecycling rubber materials without damaging the internal organization structure of materials. This paper investigates ultra highpressure jet cutting rubber in mechanism and experiment. Different from metallic materials and brittle materials, rubber materialsare cut by the force of shear and tensile under the impinging of ultra high pressure water jet and the erosion of high speed waterflow with rubber powder. The overall structure of kerf is Y shape. The upper section of Y shape is caused by shear, the middle sectionis resulted from both shear and tensile, and the bottom section is a result of the erosion and the tensile. The results of experimentshow: (1) a significant linear correlation between cutting depth and pump pressure;(2) 2.0 mm/s is the optimal transverse velocityunder the current experiment condition;(3) the cutting depth has a decreasing tendency with the stand-off distance increased. It canbe concluded from orthogonal analysis that pump pressure has a signification effect on cutting depth, nozzle transverse velocity isnot remarkable, and the effect of stand-off distance is not obvious.

        Improving the quality of breast ultrasound examination performed by inexperienced ultrasound doctors with synchronous tele-ultrasound: a prospective, parallel controlled trial

        Purpose: This prospective study explored the value of synchronous tele-ultrasound (US) to aid doctors inexperienced in US with breast US examinations.Methods: In total, 99 patients were enrolled. Two trainee doctors who were inexperienced in US (trainee A [TA] and trainee B [TB]) and one doctor who was an expert in US completed the US examinations sequentially. TA completed the US examinations independently, while TB was instructed by the expert using synchronous tele-US. Subsequently, the expert performed on-site US examinations in person. Separately, they selected the most clinically significant nodule as the target nodule. Consistency with the expert and image quality were compared between TA and TB to evaluate tele-US. Furthermore, TB and the patients evaluated tele-US through questionnaires.Results: TB demonstrated higher consistency with the expert in terms of target nodule selection than TA (93.3% vs. 63.3%, P<0.001). TB achieved good inter-observer agreement (ICC, >0.75) with the expert on five US features (5/9, 55.6%), while TA only did so for one (1/9, 11.1%) (P=0.046). TB’s image quality was higher than TA’s in gray value, time gain compensation, depth, color Doppler adjustment, and the visibility of key information (P=0.018, P<0.001, P<0.001, P=0.033, and P=0.006, respectively). The comprehensive assessment score was higher for TB than for TA (3.96±0.82 vs. 3.09±0.87, P<0.001). Tele-US was helpful in 69.7% of US examinations and had a training effect in 68.0%. Furthermore, 63.6% of patients accepted tele-US and 60.6% were willing to pay.Conclusion: Tele-US can help doctors inexperienced in US to perform breast US examinations.

      • ODV-Associated Proteins of the <i>Pieris rapae</i> Granulovirus

        <P><I>Alphabaculovirus</I> (lepidopteran-specific nucleopolyhedroviruses, NPV) and <I>Betabaculovirus</I> (granuloviruses, GV) are two main genera of the family Baculoviridae. The virion proteomes of <I>Alphabaculovirus</I> have been well studied; however, the <I>Betabaculovirus</I> virion compositions remain unclear. <I>Pieris rapae</I> granulovirus (PrGV) can kill larvae of <I>P. rapae</I>, a worldwide and important pest of mustard family crops. In this study, the occlusion-derived virus (ODV)-associated proteins of PrGV were identified using three mass spectrometry (MS) approaches. The MS analyses demonstrated that 47 proteins were present in PrGV-ODV. Of the 47 PrGV-ODV proteins, 33 have homologues identified previously in other baculovirus ODV/BVs, whereas 14 (P10, Pr21, Pr29, Pr35, Pr42, Pr54, P45/48, Pr83, Pr84, Pr89, Pr92, Pr111, Pr114 and FGF3) were newly identified ODV proteins. Seven of the 14 newly identified ODV proteins are specific to <I>Betabaculovirus</I>, including Pr35, Pr42, Pr54, Pr83, Pr84, Pr111 and Pr114. Furthermore, the data derived from these MS approaches were validated by immunoblotting analysis using antisera prepared from 11 randomly selected recombinant PrGV-ODV proteins (including 5 <I>Betabaculovirus</I>-unique proteins). Comparison analyses revealed the similar and different compositions between <I>Betabaculovirus</I> and <I>Alphabaculovirus</I> virions, which deepen our understanding of the baculovirus virion structure and provide helpful information on <I>Betabaculovirus</I>–host interaction studies.</P><P>We used three mass spectrometry (MS) approaches to identify the occlusion-derived virus (ODV)-associated proteins of the <I>Pieris rapae</I> Granulovirus.         IGBT Open-Circuit Fault Diagnosis for 3-Phase 4-Wire 3-Level Active Power Filters based on Voltage Error Correlation

        A novel open-circuit fault diagnosis method for 3-phase 4-wire 3-level active power filters based on voltage error correlation is proposed in this paper. This method is based on observing the output pole voltage error of the active power filter through two kinds of algorithms. One algorithm is a voltage error analytical algorithm, which derives four output voltage error analytic expressions through the pulse state, current value and dc bus voltage, respectively, assuming that all of the IGBTs of a certain phase come to an OC fault. The other algorithm is a current circuit equation algorithm, which calculates the real-time output voltage error through basic circuit theory. A correlation is introduced to measure the similarity of the output voltage errors between the two algorithms, and OC faults are located by the maximum of the correlations. A FPGA has been chosen to implement the proposed method due to its fast prototyping. Simulation and experimental results are presented to show the performance of the proposed OC fault diagnosis method.

      • siRNA-mediated Silencing of Notch-1 Enhances Docetaxel Induced Mitotic Arrest and Apoptosis in PCa Cells

        Purpose: Notch is an important signaling pathway that regulates cell fate, stem cell maintenance and the initiation of differentiation in many tissues. It has been reported that activation of Notch-1 contributes to tumorigenesis. However, whether Notch signaling might have a role in chemoresistance of prostate cancer is unclear. This study aimed to investigate the effects of Notch-1 silencing on the sensitivity of prostate cancer cells to docetaxel treatment. Methods: siRNA against Notch-1 was transfected into PC-3 prostate cancer cells. Proliferation, apoptosis and cell cycle distribution were examined in the presence or absence of docetaxel by MTT and flow cytometry. Expression of $p21^{waf1/cip1}$ and Akt as well as activation of Akt in PC-3 cells were detected by Western blot and Real-time PCR. Results: Silencing of Notch-1 promoted docetaxel induced cell growth inhibition, apoptosis and cell cycle arrest in PC-3 cells. In addition, these effects were associated with increased $p21^{waf1/cip1}$ expression and decreased Akt expression and activation in PC-3 cells. Conclusion: Notch-1 promotes chemoresistance of prostate cancer and could be a potential therapeutic target.
