        The effect of Lfcin-B on non-small cell lung cancer H460 cells is mediated by inhibiting VEGF expression and inducing apoptosis

        Lfcin-B, an antimicrobial peptide found invarious exocrine secretions of mammals, showed antitumoreffects. However, the effect and relative mechanism ofLfcin-B on non-small cell lung cancer is unclear. In thisstudy, assay of cell viability, quantitative real-time PCR,Western blot, annexin V/propidium iodide assay, flowcytometry and tumor-xenograft model were applied toelucidate the mechanism of Lfcin-B on non-small cell lungcancer NCI-H460 (H460) cells. Lfcin-B significantly suppressedthe proliferation of H460 cells in vitro. Additionally,the transcription and translation of the VEGF gene inH460 cells were restrained after exposure to Lfcin-B. Moreover, the apoptosis of H460 cells was induced byLfcin-B through stimulating caspase-3, caspase-9 andpreventing survivin expression on both the transcriptionand translation level. Meanwhile, Lfcin-B increased theproduction of reactive oxygen species and suppressed theRNA of antioxidant enzymes (GPX1, GPX2, SOD3 andcatalase) in H460 cells. Finally, Lfcin-B significantly preventedthe tumor growth in the H460-bearing mice model. These results indicated that Lfcin-B could be a potentialcandidate for the treatment of lung cancer.

      • Improved DTCWT-LMS and FastICA Based sEMG Signals Filtering

        A novel design of dual-tree complex wavelet transform (DTCWT) and fastICA was proposed, aiming at the noise interference and aliasing between multi-channels sEMG signals. Firstly, DTCWT was utilized to decompose signals to different frequency band. Secondly, an improved LMS adaptive filter was designed for filtering sub band noise layer by layer. Finally, fastICA algorithm was introduced to separate crosstalk between channels. Some experiments were carried out to compare the proposed method with other algorithms, and the results showed that the algorithm proposed could filter noise effectively, keep better convergence especially in low signal-to-noise ratio and eliminate crosstalk more thoroughly by fastICA.

        ZL-2, a cathelicidin-derived antimicrobial peptide, has a broad antimicrobial activity against gram-positive bacteria and gram-negative bacteria in vitro and in vivo

        Alloferons are a group of naturally occurringpeptides primarily isolated from insects that are capable ofstimulating mouse and human NK cell cytotoxicity towardcancer cells. In this study, we found that a modified antibacterialpeptide had a broad range of action against bothgram-positive and gram-negative bacteria. A time-courseexperiment showed that CFU counts rapidly decreasedafter ZL-2 treatment, with the bacteria nearly eliminatedwithin 4 h. We also examined the synergy between thepeptide and antibiotics. The peptide ZL-2 resulted in asignificant synergistic improvement in the potencies ofampicillin, erythromycin and ceftazidime against methicillin-resistant bacteria. In addition, ZL-2 had no detectablecytotoxicity in mouse spleen cells or a mouse animalmodel. In the mouse model by i.p. inoculation with Escherichiacoli, timely treatment of i.p. injection with ZL-2resulted in 100-fold reduction in bacteria load in blood aswell as 80 % protection from death in the inoculated animals. In conclusion, we successfully identified a modifiedpeptide with maximal bactericidal activity. This study alsoprovides a potential therapeutic for the treatment of E. colisepticemia by increasing the activity of antimicrobials.

        Interaction of Bilobalide and Ginkgolides B with Bovine Serum Albumin: A Fluorescence Quenching Study

        The interaction of bilobalide (BB) and ginkgolides B (GB) with bovine serum albumin (BSA) was investigated by fluorescent technique and UV/vis absorption spectroscopy. The results showed that BB and GB could intensively quench the fluorescence of BSA through a static quenching procedure. The binding constants (Ka) and the average binding distance between the donor (BSA) and the acceptor (ginkgolides) were obtained ($r_{BB}$ = 5.33 nm and $r_{GB}$ = 4.20 nm) by the theory of non-radiation energy transfer, and then the thermodynamic parameters such as ${\Delta}S^0$ (0.17-0.32 kJ/mol), ${\Delta}G^0$ (-20.76 ~ -17.79 kJ/mol) and ${\Delta}H^0$ (32.47-76.52 kJ/mol) could be calculated, respectively. All these results revealed that the interaction of BB and GB with BSA were driven mainly by hydrophobie force. The synchronous fluorescence spectroscopy was applied to examine the effect of two ginkgolides on the configuration of BSA. The configuration alteration of BSA could be induced by the hydrophobicitv environment of tyrosine with the increase of the drug concentration.

      • Vertically constructed monolithic electrodes for sodium ion batteries: toward low tortuosity and high energy density

        <P>Nanomaterials for battery applications are strongly appealing to the industrial community, yet conventional electrode fabrication by employing a slurry paste restricts the potential performances of nanoengineered materials, particularly with commercial-level thickness and high mass loading. Here, physical vapor deposition (PVD) prepared monolithic electrodes composed of 40 μm-thick anisotropic WS2 vertical arrays on Al foil with a mass loading of 17.5 mg cm<SUP>−2</SUP>, and several sparsely inserted nanometre-thick carbon layers are reported. The monolithic anode shows an ultra-fast Na-ion diffusivity of 2.05 × 10<SUP>−10</SUP> cm<SUP>2</SUP> s<SUP>−1</SUP>, which is two orders of magnitude faster than that of the anode prepared from a slurry paste. As a sodium-ion battery (SIB) anode, the monolithic electrode achieves prominent areal and volumetric capacities of 5.57 mA h cm<SUP>−2</SUP> and 1.39 A h cm<SUP>−3</SUP>, respectively, at a current density of 100 mA g<SUP>−1</SUP> with less than 20% decay during 300 cycles. Furthermore, a SIB full cell composed of the monolithic anode and a Na2V3(PO4)3/C cathode can provide a reversible capacity around 125 mA h g<SUP>−1</SUP> at 2C with an output voltage of 1.77 V, which demonstrates its potential for further development.</P>

        Large-eddy simulation of low-swirl multi-nozzle combustion with co- and counter-swirling arrays

        Large-eddy simulation (LES) of CH 4 -air low-swirl flame was carried out in a multi-nozzle combustor with two burner configurations by using a premixed flamelet model. The multi-nozzle burner includes a co-swirling array where all five nozzles act in the same direction and a counter-swirling array where the center nozzle is in the opposite swirling direction to the outer nozzles. LES results are in good agreement with OH-planar laser-induced florescence data in terms of OH concentrations and combustion progress variables. Numerical results show that the flow of each nozzle is constant before merging. The neighboring flows interact with each other and generate a highvelocity zone with intensive turbulence. The kinetic energy in the interacting region for the co-swirling array is larger than that for the counter-swirling array. After neighboring flow combining, the flow develops into a unified swirling motion similar to a single swirling flow for the co-swirling array, whereas the flow maintains the individual swirling structures for the counter-swirling arrangement. However, the swirling array exerts minimal effect on multi-nozzle combustion in terms of the temperature distributions and combustion progress of premixed low-swirl multi-nozzle flames.

        Analysis of heat transfer performance of ORC direct contact heat exchanger by GRA-VMD-LSSVM model using optimization

        The relationship between heat transfer effect and influencing factors in direct contact with evaporator isstudied. It is important to optimize the design and setting of direct contact heat exchangers and further improve theheat transfer process of heat exchangers by understanding the influencing factors of direct contact evaporator. Themethods used are grey correlation analysis, variational mode decomposition and least square support vector machinealgorithm to calculate the experimental heat transfer coefficient. The conclusions are that grey relation analysis can findthe complex relationship between different influencing factors and reduce the amount of data to improve the accuracyof prediction. Hence, the novelty of this paper is to propose a simple, efficient and accurate hybrid prediction model forVHTC prediction. The place where it goes beyond previous efforts in the literature is that different influencing factorson the heat transfer performance in the direct contact heat transfer process are considered. The results show that theprediction accuracy of the heat transfer coefficient can be improved by 7% by optimizing the data only with grey correlationanalysis. The prediction accuracy can be improved up to 53% after using the hybrid model.

        Prevalence and risk factors for incidental prostate cancer in patients after transurethral resection of the prostate with negative results on prostate biopsy: A retrospective study

        Purpose: This study aimed to explore the prevalence and predictors of incidental prostate cancer (IPC) after transurethral resection of the prostate (TURP) with negative results on transperineal magnetic resonance imaging (MRI)/transrectal ultrasonography (TRUS) fusion prostate biopsy or TRUS-guided prostate biopsy. Materials and Methods: Data of 253 patients who underwent TURP with a preliminary diagnosis of benign prostatic hyperplasia (BPH) were evaluated. The prevalence of IPC was calculated. Univariate and multivariate logistic regression analyses were conducted to explore independent predictive factors of IPC. Results: A total of 253 patients were included. IPC was diagnosed in 12 patients (4.7%). The mean age of the patients and the mean prostate volume were 69.8±7.07 years and 89.3±49.29 mL, respectively. The prevalence of IPC was higher in the TRUS guided prostate biopsy group than in the transperineal MRI/TRUS fusion prostate biopsy group (11 of 203 [5.4%] vs. 1 of 50 [2.0%], p=0.47), but the difference was not statistically significant. Our results indicated that older age (≥70 y) (odds ratio [OR], 1.14; 95% confidence interval [CI], 1.02–1.27; p=0.025) and smaller prostate volume (OR, 0.97; 95% CI, 0.938–0.998; p=0.039) were associated with an increased incidence of IPC after TURP. Conclusions: Our findings indicate that the prevalence of IPC may be higher among patients who undergo transrectal prostate biopsy before TURP than among those who undergo transperineal MRI/TRUS fusion prostate biopsy. Older age and smaller prostate volume were independent predictors of increasing the risk for IPC after TURP.

        Acute Effects of Transforming Growth Factor-β1 on Neuronal Excitability and Involvement in the Pain of Rats with Chronic Pancreatitis

        Background/Aims This study was to investigate whether transforming growth factor-β1 (TGF-β1) plays a role in hyperalgesia in chronic pancreatitis (CP) and the underlying mechanisms. Methods CP was induced in male adult rats by intraductal injection of trinitrobenzene sulfonic acid (TNBS). Abdominal hyperalgesia was assessed by referred somatic behaviors to mechanical stimulation of rat abdomen. Dil dye injected into the pancreas was used to label pancreas-specific dorsal root ganglion (DRG) neurons. Whole cell patch clamp recordings and calcium imaging were performed to examine the effect of TGF-β1 on acutely isolated pancreas-specific DRG neurons. Western blot analysis was carried out to measure the expression of TGF-β1 and its receptors. Results TNBS injection significantly upregulated expression of TGF-β1 in the pancreas and DRGs, and TGF-β1 receptors in DRGs (T9-T13) in CP rats. Intrathecal injection of TGF-β receptor I antagonist SB431542 attenuated abdominal hyperalgesia in CP rats. TGF-β1 application depolarized the membrane potential and caused firing activity of DRG neurons. TGF-β1 application also reduced rheobase, hyperpolarized action potential threshold, and increased numbers of action potentials evoked by current injection of pancreas-specific DRG neurons. TGF-β1 application also increased the concentration of intracellular calcium of DRG neurons, which was inhibited by SB431542. Furthermore, intrathecal injection of TGF-β1 produced abdominal hyperalgesia in healthy rats. Conclusions These results suggest that TGF-β1 enhances neuronal excitability and increases the concentration of intracellular calcium. TGF-β1 and its receptors are involved in abdominal hyperalgesia in CP. This and future study might identify a potentially novel target for the treatment of abdominal pain in CP.
