Kidney Toxicity Induced by 13 Weeks Exposure to the Fruiting Body of Paecilomyces sinclairii in Rats

Paecilomyces sinclairiis (PS) is known as a functional food or human health supplement. However concerns have been raised about its kidney toxicity. This study was performed to investigate the kidney toxicity of PS by 13 week-oral administration to rats. Blood urea nitrogen (BUN), serum creatinine, and kidney damage biomarkers including beta-2-microglobulin (β2m), glutathione S-transferase alpha (GST-α), kidney injury molecule 1 (KIM-1), tissue inhibitor of matrix metalloproteinase 1 (TIMP-1), vascular endothelial growth factor (VEGF), calbindin, clusterin, cystatin C, neutrophil gelatinase-associated lipocalin (NGAL) and osteopontin were measured during or after the treatment of PS. BUN, creatinine and kidney damage biomarkers in serum were not changed by PS. However, kidney cell karyomegaly and tubular hypertrophy were observed dose-dependently with higher severity in males. KIM-1, TIMP-1 and osteopontin in kidney and urine were increased dose dependently in male or at the highest dose in female rats. Increased urinary osteopontin by PS was not recovered at 2 weeks of post-exposure in both genders. Cystatin C in kidney was decreased at all treatment groups but inversely increased in urine. The changes in kidney damage biomarkers were more remarkable in male than female rats. These data indicate that the PS may provoke renal cell damage and glomerular filtration dysfunction in rats with histopathological lesions and change of kidney damage biomarkers in kidney or urine. Kidney and urinary KIM-1 and cystatin C were the most marked indicators, while kidney weight, BUN and creatinine and kidney damage biomarkers in serum were not influenced.

<P><B>Abstract</B></P><P>Methods using high performance liquid chromatography with diode array detection (HPLC-DAD) and tandem mass spectrometry (HPLC–MS/MS) were developed and validated for the simultaneous determination of 5 chromones and 6 coumarins: prim-<I>O</I>-glucosylcimifugin (1), cimifugin (2), nodakenin (3), 4′-<I>O</I>-β-<SMALL>D</SMALL>-glucosyl-5-<I>O</I>-methylvisamminol (4), sec-<I>O</I>-glucosylhamaudol (5), psoralen (6), bergapten (7), imperatorin (8), phellopterin (9), 3′-<I>O</I>-angeloylhamaudol (10) and anomalin (11), in Radix Saposhnikoviae. The separation conditions for HPLC-DAD were optimized using an Ascentis Express C18 (4.6mm×100mm, 2.7μm particle size) fused-core column. The mobile phase was composed of 10% aqueous acetonitrile (A) and 90% acetonitrile (B) and the elution was performed under a gradient mode at a flow rate of 1.0mL/min. The detection wavelength was set at 300nm. The HPLC-DAD method yielded a base line separation of the 11 components in 50% methanol extract of Radix Saposhnikoviae with no interfering peaks detected. The HPLC-DAD method was validated in terms of linearity, accuracy and precision (intra- and inter-day), limit of quantification (LOQ), recovery, and robustness. Specific determination of the 11 components was also accomplished by a triple quadrupole tandem mass spectrometer equipped with an electrospray ionization (ESI) source. This HPLC–MS/MS method was also validated by determining the linearity, limit of quantification, accuracy, and precision. Quantification of the 11 components in 51 commercial Radix Saposhnikoviae samples was successfully performed using the developed HPLC-DAD method. The identity, batch-to-batch consistency, and authenticity of Radix Saposhnikoviae were successfully monitored by the proposed HPLC-DAD and HPLC–MS/MS methods.</P>

<P> The present study attempted to predict selective exposure to media messages as a function of personality and situation, which has rarely been examined in prior relevant research. Employing a quasi-experimental method, the interplay between prevailing perceived threat from the economic crisis that started in 2008 and each of the two personality types when dealing with threat - repression and sensitization - was examined to predict online news selection behavior, as unobtrusively recorded. A significant interaction between perceived threat and sensitization tendency was obtained for both the selection of financial crisis-related news and avoidance of financial crisis-unrelated news. The implications of this exclusive pattern of online news selection behavior are discussed. </P>

<P>The three-dimensional Petersen-torus network 3PT is based on the Petersen graph and has recently been proposed as an interconnection network. 3PT is better than the well-known 3D torus and 3D honeycomb mesh in terms of diameter and network cost. In this paper, we propose one-to-all and all-to-all broadcasting algorithms for 3PT(l;m;n) under SLA (single-link available) and MLA (multiple-link available) models.</P>
Mineralogical Characteristics of Na- and Ca-bentonite

Buffer materials play an important role in preventing the leakage of radionuclides from the residue. The mineralogical properties of these buffer materials are critical in repository design. This study presents the fundamental properties of Na-type MX80 and a novel Ca-type Bentonil- WRK. The CaO to MgO ratio in Bentonil-WRK was approximately 1:1, and the CaO to Na2O ratio was approximately 2.8:1. These results suggest that Bentonil-WRK demonstrates a lower swelling index compared to Gyeongju bentonite due to its CaO-to-MgO ratio’s proximity to 1:1, despite having a higher montmorillonite content than Gyeongju bentonite. The results of this research can provide useful foundational data for the evaluation of the thermal-hydraulic-mechanical-chemical behavior of buffer materials.

<P>Delivering isolated chondrocytes with matrix is a promising approach to promote the cartilage repair. The present study attempted to combine the advantages of porous scaffold and hydrogel in delivering chondrocytes to partial-thickness cartilage defects. An electrospun, gelatin-incorporated PLCL scaffold mechanically similar to natural cartilage was fabricated, and chondrocytes were seeded using an injectable heparin-based hydrogel for efficient cell seeding. The scaffold/hydrogel composite showed more enhanced expression of chondrogenic genes and production of GAGs than those prepared without hydrogel. In addition, significant cartilage formation showing good integration with surrounding, similar to natural cartilage, was observed by scaffold/hydrogel composite system in partial-thickness defects of rabbit knees while no regeneration was observed in control defects. Although no exogenous chondrogenic factors were added, it was evident that the scaffold/hydrogel composite system was highly effective and better than the scaffold alone system without hydrogel for cartilage regeneration both in vitro and in vivo.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/bomaf6/2012/bomaf6.2012.13.issue-8/bm3005353/production/images/medium/bm-2012-005353_0011.gif'></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/bm3005353'>ACS Electronic Supporting Info</A></P>
