Abstract

A rapid and multiplexed immunosensor was developed based on a quantum dot (QD)-reverse assaying strategy (RAS) and immuno-magnetic beads (IMBs) for one-step and simultaneous detection of Escherichia coli O157: H7 and Salmonella. In a conventional QD-based immunosensor, the fluorescence signal of the “IMBs-target-QD” immunoconjugate is directly used as the assaying readout. However, the fluorescence signal is affected by IMBs due to light scattering and the “IMBs-target-QD” immunoconjugate needs multiple washing and re-suspension steps. To address these problems, we use the surplus QD-antibody conjugate as signal readout in the RAS, which prevents interference from the IMBs, increases the fluorescence signal, and avoids complex operations. Compared with conventional QD-based immunosensor, the sensitivity of QD-RSA immunosensor for detection of Escherichia coli O157: H7 has been improved fifty-fold, and whole analysis procedure can be finished within 1h. Therefore, this RSA strategy is promising for improving the performance of QD-based immunosensors and could greatly broaden their applications.

Highlights

The immunosensor based on quantum dot-reverse assaying strategy (QD-RAS) was developed.

The sensitivity of QD-RAS improved 50 times compared to conventional QDs-immunosensor.

The whole analysis in QD-RAS can be finished in one-step within 1h.

Objective: FAS/FASL gene promoter polymorphisms have been repeatedly associated with gastric cancer risk, but findings are inconclusive across studies. To address a more precise estimation of the relationship, a meta-analysis was performed. Methods: Data were collected from the Pubmed, Medline and EMBASE databases, with the last report up to 1 December, 2011. Crude ORs with 95% CIs were used to assess the strength of the association by (1) the additive, (2) the codominant, (3) the dominant, and (4) the recessive models. Results: A total of seven studies, including six studies on FAS -1377G>A polymorphism, five studies on FAS -670A>G polymorphism, and six studies on FASL -844T>C polymorphism, were identified in the current meta-analysis. Overall, an association of FAS -1377G>A (AA versus GG: OR = 1.313, 95% CI = 1.045-1.650, Ph = 0.347, $I^2$ = 10.8) and FASL -844T>C (CC versus TT: OR = 1.352, 95% CI = 1.043-1.752, Ph = 0.461, $I^2$ = 0.0) polymorphisms with gastric cancer was found in the codominant model. However, we did not detect any association between gastric cancer and the FAS -670A>G polymorphism. In the subgroup analysis by ethnicity, similar elevated risks were also observed in Asian population for FAS -1377G>A (AA versus GG: OR = 1.309, 95% CI = 1.041-1.646, Ph = 0.240, $I^2$ = 27.3) and FASL -844T>C (CC versus TT: OR = 1.420, 95% CI = 1.081-1.865, Ph = 0.524, $I^2$ = 0.0) polymorphisms. Conclusions: This meta-analysis indicated that FAS -1377G>A and FASL -844T>C polymorphisms might be associated with gastric cancer risk.

Aim: The significance of the mucinous adenocarcinoma in TNM staging and prognosis for colorectal tumor patients is still controversial. The aim was to provide a meta-analysis for TNM staging and prognostic features of colorectal tumors. Methods: 30 individual case-control studies were finally included into this meta-analysis, involving a total of 444,489 cancer cases and 45,050 mucinous adenocarcinomas, of relations with TNM staging and prognostic features. Results: Compared to non-mucinous adenocarcinoma patients, the TNM IV stage accounted for a larger percentage of mucinous adenocarcinomas (OR=1.48, 95%CI 1.28-1.71, POR<0.001) and the prognosis was significantly poor (HR=1.06, 95%CI 1.04-1.08, P<0.001). After heterogeneity testing, the results was similar to the holistic approach outcome (HR=1.48, 95%CI 1.35-1.62, P<0.001). Conclusion: Compared to patients with non-mucinous adenocarcinomas, mucinous adenocarcinoma patients with later TNM staging make up a big percentage, and mucinous adenocarcinoma is an independent risk factor for poor prognosis.

The traction motors in electric multiple unit (EMU) trains are powered by AC-DC-AC converters, and the DC link voltage is generated by single phase PWM converters, with a fluctuation component under twice the frequency of the input catenary AC grid, which causes fluctuations in the motor torque and current. Traditionally, heavy and low-efficiency hardware LC resonant filters parallel in the DC side are adopted to reduce the ripple effect. In this paper, an analytical model of the ripple phenomenon is derived and analyzed in the frequency domain, and a ripple control scheme compensating the slip frequency of rotor vector control systems without a hardware filter is applied to reduce the torque and current ripple amplitude. Then a relatively simple discretization method is chosen to discretize the algorithm with a high discrete accuracy. Simulation and experimental results validate the proposed ripple control strategy.

A new conjugated polymer with bulky substituents was synthesized by the polymerization of ethynylestradiol 3-methyl ether (EEDME) by such transition metal catalysts as PdCl2, RuCl3, and (NBD)PdCl2. The polymerization proceeded well in homogeneous manner to give a moderate yield of polymer. The chemical structure of poly(EEDME) was characterized to have the conjugated polymer backbone with the designed substituents. From the CV measurements, the HOMO energy level of the polymer was calculated to be 5.02 eV. The photoluminescence spectra of poly(EEDME) showed that the two photoluminescence peaks are located at 434 and 406 nm corresponding to the photon energy of 2.86 and 3.06 eV, respectively.
