        In Vitro Screening for Compounds Derived from Traditional Chinese Medicines with Antiviral Activities Against Porcine Reproductive and Respiratory Syndrome Virus

        Seventeen compounds derived from traditional Chinese medicines (TCMs) were tested for their antiviral activity against porcine reproductive and respiratory syndrome virus (PRRSV) in vitro. Visualization with the cytopathologic effect (CPE) assay and the 3-(4, 5-dimethyithiazol- 2-yl)-2,5-diphenyltetrazolium bromide test were used to determine the 50% cytotoxic concentration (CC50) and 50% effective concentration (EC50) in cultured Marc-145 cells. Among the tested compounds, chlorogenic acid and scutellarin showed potential anti-PRRSV activity. The EC50 values were 270.8 ± 14.6 μg/ml and 28.21 ± 26.0 μg/ml and the selectivity indexes were >5.54 and 35.5, respectively. The time-of-addition and virucidal assay indicated that the anti-PRRSV activity of the two compounds could be due to their inhibiting the early stage of virus replication and/or inactivating the virus directly. The inhibition of the virus attachment was not observed in the adsorption inhibition assay. The inhibition ratios of chlorogenic acid and scutellarin were, respectively, 90.8% and 61.1% at the maximum non-cytotoxic concentrations. The results have provided a basis for further exploration of their antiviral properties and mechanisms in vivo. We believe that the chlorogenic acid and scutellarin have a great potential to be developed as new anti-PRRSV drugs for clinical application.

        A new recombined porcine reproductive and respiratory syndrome virus virulent strain in China

        Porcine reproductive and respiratory syndrome (PRRS) is one of the most important swine diseases worldwide. In the present study, a new virulent strain of PRRS virus (PRRSV), GDsg, was isolated in Guangdong province, China, and caused high fever, high morbidity, and high mortality in sows and piglets. The genome of this new strain was 15,413 nucleotides (nt) long, and comparative analysis revealed that GDsg shared 82.4% to 94% identity with type 2 PRRSV strains, but only 61.5% identity with type 1 PRRSV Lelystad virus strain. Phylogenetic analysis indicated that type 2 PRRSV isolates include five subgenotypes (I, II, III, IV, and V), which are represented by NADC30, VR-2332, GM2, CH-1a, and HuN4, respectively. Moreover, GDsg belongs to a newly emerging type 2 PRRSV subgenotype III. More interestingly, the newly isolated GDsg strain has multiple discontinuous nt deletions, 131 (19 + 18 + 94) at position 1404–1540 and a 107 nt insertion in the NSP2 region. Most importantly, the GDsg strain was identified as a virus recombined between low pathogenic field strain QYYZ and vaccine strain JXA1-P80. In conclusion, a new independent subgenotype and recombinant PRRSV strain has emerged in China and could be a new threat to the swine industry of China.

      • Maximum power extraction for wind turbines through a novel yaw control solution using predicted wind directions

        <P><B>Abstract</B></P> <P>For modern horizontal axis wind turbines (WTs), a yaw drive mechanism is utilized to adjust the nacelle position to face the wind direction. Depending on historical signals from wind direction sensors, conventional yaw control methods could not provide sufficient performance in tracking winds, and thus result in a reduction of wind power extraction. This issue needs to be tackled using advanced control solutions. Taking advantage of predicted wind directions, a novel control solution is proposed in this study. Specifically, the proposed solution refers to a novel control structure that consists of a wind direction predictive model and a novel yaw control method. Under the proposed control structure, a hybrid autoregressive integrated moving average method-based Kalman filter (ARIMA-KF) model is used to predict the wind direction, and two novel yaw control methods are proposed: one created by using the predicted wind direction as the tracking reference, and the other based on a model predictive control (MPC) using a finite control set. To demonstrate the feasibility and the superiority of the proposed solution, two novel yaw controllers are developed and tested through some simulation tests using industrial data. Their performance is compared to the one of two industrial yaw controllers. Comparison results show that the two novel yaw controllers are capable of reducing yaw error, and thus increase wind power extraction for the WTs. Meanwhile, it is noticeable that the MPC-based controller has an advantage in the aspect of reducing yaw actuator usage.</P> <P><B>Highlights</B></P> <P> <UL> <LI> A novel yaw control solution using predicted wind direction is proposed. </LI> <LI> An ARIMA-KF based model is used to predict the wind direction. </LI> <LI> One novel yaw control method uses the predicted wind direction as the tracking reference. </LI> <LI> The other one is based on a model predictive control using a finite control set. </LI> <LI> The novel yaw control solution extracts 1% more wind power. </LI> </UL> </P>

      • Prognostic Value of Matrix Metalloproteinase 9 Expression in Breast Cancer Patients: A Meta-analysis

        Background: Matrix metalloproteinase 9 (MMP-9) is related to tumor invasion and metastasis. However, the role of MMP-9 expression in breast cancer survival remains controversial. The purpose of this study was to accomplish a more accurate estimation of the association between MMP-9 expression and survival results in breast cancer patients through meta-analysis. Methods: A meta-analysis of published studies investigating the effects of positive MMP-9 expression on both relapse free survival (RFS) and overall survival (OS) was performed. Relevant literature was confirmed by searching electronic databases including PubMed, Ovid, EMBASE and China National Knowledge Infrastructure (CNKI) before November 1, 2012. Individual hazard ratios (HRs) and 95% confidence intervals (CIs) were extracted and pooled HRs with 95% CIs were used to evaluate the strength of the association between positive MMP-9 expression and survival results of breast cancer patients. Funnel plot and Egger's regression tests were used to evaluate publication bias. Heterogeneity and sensitivity analysis was also conducted. All the work was completed using STATA. Results: A total of 2,344 patients from 15 evaluative studies were finally included. Pooled HRs and 95% CIs suggested that MMP-9 overexpression had an unfavorable impact on both OS (HR: 1.70, 95% CI: 1.41-2.04) and RFS (HR: 1.54, 95% CI: 1.17-2.01) in breast cancer patients. There was no significant heterogeneity observed in the studies reported for OS (P=0.360, $I^2$=8.8%), but not RFS (P=0.002, $I^2$=67%). Publication bias was absent among the studies both in OS and RFS cases (t=-0.54, P=0.605 and t=1.71, P=0.131, respectively). Omission of any single study had little effect on the combined risk estimates on sensitivity analysis. Conclusion: The results of this meta-analysis suggest that positive MMP-9 expression confers a higher risk of relapse and a worse survival in patients with breast cancer. Larger prospective studies are now needed to evaluate the clinical utility of MMP-9 expression.

      • miR-181b as a Potential Molecular Target for Anticancer Therapy of Gastric Neoplasms

        Objective: MicroRNAs (miRNAs) play important roles in carcinogenesis. The aim of the present study was to explore the effects of miR-181b on gastric cancer. Methods: The expression level of miR-181b was quantified by qRT-PCR. MTT, flow cytometry and matrigel invasion assays were used to test proliferation, apoptosis and invasion of miR-181b stable transfected gastric cancer cells. Results: miR-181b was aberrantly overexpressed in gastric cancer cells and primary gastric cancer tissues. Further experiments demonstrated inducible expression of miR-181b by Helicobacter pylori treatment. Cell proliferation, migration and invasion in the gastric cancer cells were significantly increased after miR-181b transfection and apoptotic cells were also increased. Furthermore, overexpression of miR-181b downregulated the protein level of tissue inhibitor of metalloproteinase 3 (TIMP3). Conclusion: The upregulation of miR-181b may play an important role in the progress of gastric cancer and miR-181b maybe a potential molecular target for anticancer therapeutics of gastric cancer.

        Theoretical analysis of simply supported channel girder bridges

        Channel girder bridges that consist of a deck slab and two side beams are good choices for railway bridges and urban rail transit bridges when the vertical clearance beneath the bridge is restricted. In this study, the behavior of simply supported channel girder bridges was theoretical studied based on the theory of elasticity. The accuracy of the theoretical solutions was verified by the finite element analysis. The global bending of the channel girder and the local bending of the deck slab are two contributors to the deformations and stresses of the channel girder. Because of the shear lag effect, the maximum deflection due to the global bending could be amplified by 1.0 to 1.2 times, and the effective width of the deck slab for determining the global bending stresses can be as small as 0.7 of the actual width depending on the width-tospan ratio of the channel girder. The maximum deflection and transversal stress due to the local bending are obtained at the girder ends. For the channel girders with open section side beams, the side beam twist has a negligible effect on the deflections and stresses of the channel girder. Simplified equations were also developed for calculating the maximum deformations and stresses.

      • Comparison of Primary Breast Cancer Size by Mammography and Sonography

        Purpose: To compare tumor size by mammography and sonography and align with pathological results in primary breast cancer cases. Materials and Methods: We retrospectively reviewed 95 primary breast cancer patients who underwent mammography and sonography from January 2011 to June 2012. The largest tumor diameter was chosen as sizing reference for each imaging modality. The measurements of mammography and sonography were considered concordant if they were within the measurement of pathological results ${\pm}0.5cm$. Pearson's correlation coefficient was calculated for imaging results. Results: The range of the maximum diameter was 0.6cm-10.5cm and mean value was $3.81{\pm}2.04cm$ by pathological results, 0.7cm-12.4 cm and $3.99{\pm}2.19cm$ by mammography, and 0.9cm-11.0cm and $3.63{\pm}2.01cm$ by sonography, respectively. Sonography (R: 0.754), underestimated tumor size, but had a better correlation with pathological tumor size compared to mammography (R: 0.676), which overestimated tumor size. Conclusions: Sonography is superior to mammography in assessment of primary breast cancer.

        Prevalence, Risk Factors, and Psychological Effects of Overactive Bladder in Chinese University Students

        Purpose: The purpose of this study was to investigate the prevalence and risk factors of overactive bladder (OAB) in young adults and to explore the influence of OAB on mental health. Methods: Between October 2019 and January 2020, 14,010 anonymous questionnaires were distributed to freshmen at 2 universities in Henan, China. The students came from all over the country. The questionnaire included general items and information necessary to calculate the overactive bladder symptom score, the Chinese version of the Pittsburgh Sleep Quality Index (PSQI) score, Self-Esteem Scale (SES) score, and Self-Rating Depression Scale (SDS) score. The relationships between the prevalence of OAB and its risk factors were evaluated. Results: The overall prevalence of OAB was 6.0%, with 4.3% of participants characterized as having dry OAB and 1.7% as having wet OAB. The prevalence of mild OAB was 5.5%, and that of moderate OAB was 0.5%; no severe OAB was observed. Higher prevalence rates of OAB were found among women, respondents with constipation, and respondents with primary nocturnal enuresis (PNE) (P <0.05). Compared to healthy controls, the OAB group exhibited a higher mean SDS score (52.12±8.986 vs. 47.71±9.399, P<0.001) and mean PSQI score (5.28±2.486 vs. 4.27±2.431, P<0.001), but a lower mean SES score (27.78±3.599 vs. 29.57±4.109, P<0.001). Conclusions: OAB significantly affects the mental health of young adults. Female sex, constipation, and PNE are risk factors for OAB.

        <P>Rice blast, caused by the fungus Magnaporthe oryzae, is one of the most devastating diseases of rice. To understand the molecular basis of Pi5-mediated resistance to M. oryzae, we cloned the resistance (R) gene at this locus using a map-based cloning strategy. Genetic and phenotypic analyses of 2014 F2 progeny from a mapping population derived from a cross between IR50, a susceptible rice cultivar, and the RIL260 line carrying Pi5 enabled us to narrow down the Pi5 locus to a 130-kb interval. Sequence analysis of this genomic region identified two candidate genes, Pi5-1 and Pi5-2, which encode proteins carrying three motifs characteristic of R genes: an N-terminal coiled-coil (CC) motif, a nucleotide-binding (NB) domain, and a leucine-rich repeat (LRR) motif. In genetic transformation experiments of a susceptible rice cultivar, neither the Pi5-1 nor the Pi5-2 gene was found to confer resistance to M. oryzae. In contrast, transgenic rice plants expressing both of these genes, generated by crossing transgenic lines carrying each gene individually, conferred Pi5-mediated resistance to M. oryzae. Gene expression analysis revealed that Pi5-1 transcripts accumulate after pathogen challenge, whereas the Pi5-2 gene is constitutively expressed. These results indicate that the presence of these two genes is required for rice Pi5-mediated resistance to M. oryzae.</P>

        Bulk Synthesis of Fe3Al Intermetallic Compound Nanoparticles by Flow-Levitation Method

        Metallic nanopowders have an increasing application in magnetic materials, catalysts and chemical and metallic industries. In this research, a novel bulk synthesis method for preparing high pure intermetallic Fe3Al nanoparticles was developed by flow-levitation (FL) method. The Fe and Al vapors ascending from the high-temperature levitated droplet were condensed by cryogenic argon gas under atmospheric pressure. X-ray diffraction (XRD) and selected-area electron diffraction (SAED) were used to identify and characterize the prepared nanopowders exhibiting a Fe3Al phase. Measurement of transmission electron microscopy (TEM) indicated that the Fe3Al particles are nearly spherical, and the particle size of the compound ranges from 10 nm to 200 nm in diameter. The chemical composition of the nanoparticles were determined with energy dispersive spectrometer. The magnetic properties of the nanopowder indicate that Fe3Al intermetallic compound is a soft magnet at room temperature, with coercivity of 24.2 Oe and saturation magnetization of 173.2 emu/g. The production rate of Fe3Al nanoparticles was estimated to be about 4 g/h in a continuous manner, by using the FL method. This method as great potential in mass production of Fe3Al nanoparticles.
