        Immunohistochemical study on the gastro-entero-pancreatic(GEP) endocrine cells of the blue fox, Alopex lagopus

        북극여우의 위장췌 내분비세포의 부위별 분포, 출현빈도 및 세포의 종류를 밝히고자 면역조직화학적으로 관찰하였던 바 위장관에서는 6종, 췌장에서는 4종의 면역반응세포가 동정되었다. 5-HT 및 somatostatin 면역반응세포는 전위방관에 다수 분포하였으나 이중 somatostatin 면역반응세포는 장관에서 소수로 관찰되었다. Gas/CCK 면역반응세포는 주로 유문부와 십이지장에 국한하여 다수 분포하였다. Glucagon 면역반응세포는 분문부와 위저부에서 다수 그리고 장관에서 소수 관찰되었다. BPP 면역반응세포는 분문부와 위저부에 중등도, 대장에 소수분포하였다. 다수의 porcine CG 면역반응세포가 분문부와 위저부에서만 동정되었다. 한편 췌장에서는 somatostatin, glucagon, BPP 및 insulin 등 4종의 면역반응세포가 췌도 및 외분비에서 관찰되었다. 이상의 결과는 위췌장 내분비세포의 부위별 분포와 출현빈도가 동물종에 따라 매우 상이하다는 사실을 시사한다. The regional distribution and the relative frequencies of endocrine cells were studied in nine portions of the blue fox GI tract, and the distribution pattern and cell types of the pancreativc endocrine cells were also studied in the pancreas by immunohistochemical method. Six kinds of immunoreactive cells were identified in the GI tract, and four kinds of immunoreactive cells were also identified in the pancreas. Although numerous 5-HT- and somatostatin-immunoreactive cells were seen throughout the GI tract, somatostatin- immunoreactive cells were a few in the intestine. Very numerous Gas/CCK-immunoreactive cells were restricted generally in the pyloric region and duodenum. Numerous glucagon-immunoreactive cells were found in the stomach except the pyloric region, and generally a few in the intestine. Moderate number of BPP-immunoreactive cells were found in the stomach except the pyloric region, and a few in the large intestine. Numerous porcine CG-immunoreactive cells were restricted to the cardiac and fundic region. In the pancreas, four types of pancreatic endocrine cells- somatostatin-, glucagon-, BPP- and insulin-immunoreactive- were identified in the pancreatic islet and exocrine portion. These results suggest that the regional distribution, the relative frequencies and cell types of the GEP endocrine cells in the GI tract and pancreas varies considerably among the species.

        남생이 위장관 및 췌장 내분비세포에 관한 면역조직화학적 연구

        The regional distribution and relative frequency of gastrointestinal endocrine cells were studied immunohistochemically in the gastrointestinal mucosa and pancreas of the fresh water turtle. Ten kinds of endocrine cells were identified in the gastrointestinal tract. Cholecystokinin-8-, bovine pancreatic polypetide-and glucagon-immunoreactive cells were seen throughout the gastrointestinal tract, also among them cholecystokinin-8-immunoreactive cells were most predominant in segment III. Although gastrin- and gastrin/cholecystokinin-immunoreactive cells were found from segment III to VI and X, respectively, they were numerous in segment III. Somatostatin-immunoreactive cells were observed from segment I to VII. 5-hydroxytryptamine- immunoreactive cells were detected in segment I, III, VIII, IX and X. Human pancreatic polypeptide-immunoreactive cells were demonstrated in segment V, VI, VIII, IX and X. Insulin-immunoreactive cells were found from segment III to X except for segment VIII, but rare in segment VII. Neurotensin-immunoreactive cells were found to be restricted to segment VIII, IX and X. No porcine chromogranin-, substance P- and bombesin-immunoreactive cells were detected throughout the gastrointestinal tract of the fresh water turtle. Although typical mammalian pancreatic islets encapsulated by connective tissue were not present in this species, five kinds of endocrine cells-glucagon, insulin, somatostatin, bovine pancreatic polypeptide and 5-hydroxytryptamine-were found in forming small or large groups and scattered in the exocrine gland region. However porcine chromogranin- and motilin-immunoreactive cells could not be demonstrated in the pancreas.

        고슴도치 위점막의 내분비세포에 관한 면역조직화학적 연구

        한국산 고슴도치의 위점막에 분포하는 내분비세포에 대해 면역조직화학적으로 관찰하였던바, gastrin, somatostatin, 5-HT, glucagon, BPP, motilin, GIP 등 7종의 면역반응세포들이 동정되었으며, 이들 세포들의 부위별 분포와 출현빈도의 특정은 다음과 같다. Gastrin 면역반웅세포는 유문부에서만 아주 다수로 출현하였으며, somatostatin 면역반응세포는 유문부에서 다수, 분문부와 위저부에서는 중등도의 출현올 볼 수 있었다. 5-HT 면역반응세포는 분문부와 유문부에서 다수, 위저부에서는 중등도로 출현하였다. glucagon 면역반응세포는 위저부에서 소수로 그리고 유문부에서 극히 소수로 출현하였고, BPP 면역반응세포는 유문부에 다수로, 위저부에 중등도로 분문부에 소수로 분포하였다. motilin 면역반응세포는 유문부와 위저부에서 소수 또는 극소수로 출현하였으며, GIP 면역반응세포는 유문부에서만 소수의 분포를 볼 수 있었다. The gastric endocrine cells of the Korean hedgehog, Erinaceus korean us were studied immunohistochemically. Seven kinds of endocrine cells-, gastrin-, somatostatin-, 5-HT-, glucagon-, BPP-, motiIin-and GIP-immunoreactive cells- were identified in this study. The chracteristic findings of the regional distribution and relative frequency of them were examined. Gastrin-immunoreactive cells were very numerously detected only in the pyloric region. Somatostatin-immunoreactive cells were more numerous in the pyloric region than in the cardiac and fundic regions. 5-HT-immunoreactive cells were more numerous in the cardiac and pyloric regions than in the tundic one. Glucagon-immunoreactive cells were found few or rarely in the fundic and pyloric regions. BPP-imunoreactive cells were numerously distributed in the pyloric region, moderately in the fundic region and few in the cardiac region. Motilin-immunoreactive cells were found rarely or few in the fundic and pyloric regions. GIP-immnuoreactive Cells were detected onIy in the pyloric region.

        Serranidae (Coreoperca herzi) 췌장 내분비세포에 대한 면역조직화학적 연구

        The regional distribution and relative frequency of some endocrine cells in the pancreas of the Korean aucha perch, Coreoperca herzi Herzenstein belonging to the family Serranidae in order Perciformis, were observed using specific mammalian antisera against serotonin, insulin, glucagon, somatostatin and human pancreatic polypeptide (hPP) by peroxidase antiperoxidase (PAP) method. The pancreas was divided into four portions (principal and secondary islets, exocrine and pancreatic duct regions). In addition, the pancreatic islet regions were further subdivided into three regions (central, mantle and peripheral regions). Spherical to spindle or occasionally round to oval immunoreactive (IR) cells were demonstrated in the pancreatic islets and exoccrine portions, but no cells were detected in the pancreatic duct portions. In the principal islets, serotonin-IR cells were not detected but most of insulin-IR cells were located in the central regions and they were also demonstrated in the mantle and peripheral regions in moderate and rare frequencies, respectively. Glucagon- and hPP-IR cells were mainly situated in the mantle regions but the cells were also demonstrated in the peripheral regions in relatively lower frequency. Somatostatin-IR cells were evenly distributed in the central and mantle regions in a few frequency and cells were also demonstrated in the peripheral regions in rare frequency. Cell clusters were consisted of hPP-IR cells that were situated in the peripheral to mantle regions. In the secondary islet portions, serotonin-IR cells were randomly distributed throughout the whole pancreatic islet regions but lower frequency was detected in the peripheral regions compared to that in central and mantle regions where cells were detected in a few frequency, respectively. Insulin-IR cells were restricted to the central regions in numerous frequency and glucagon-IR cells were evenly distributed in the mantle and peripheral regions in moderate frequencies, respectively. Somatostatin-IR cells were observed in the central and mantle regions in moderate and a few frequencies, respectively. In addition, hPP-IR cells showed similar distributional patterns to those of glucagon-IR cells except cells were also located in the central regions in rare frequency. In the exocrine portions, only glucagon- and hPP-IR cells were demonstrated in rare and a few frequencies, respectively. In conclusion, the regional distribution and relative frequency of pancreatic endocrine cells of the Korean aucha perch showed general patterns, which were observed in other teleost. However, some species-dependent different distributional patterns and/or relative frequencies were also demonstrated especially to serotonin-IR cells. In pancreas of the Korean aucha perch, insulin-IR cells were the most predominant cell type followed by glucagon-, somatostatin-, hPP- and serotonin-IR cells.

        생진양혈탕가미방이 Streptozotocin으로 유발된 당뇨 생쥐의 췌장에 미치는 영향

        Streptozotocin(STZ)을 투여하여 고혈당을 유발시킨 생쥐의 이자섬에 생진양혈탕가미방이 미치는 영향을 규명하고자, 전방액 (0.4ml/day)을 21일간 투여하여 혈당 및 BUN의 변화와 인슐린 면역조직화학적 검색 및 전자 현미경을 이용한 $\beta$세포의 미세구조 변화를 관찰하였다. STZ을 투여한 대조군 이자섬의 면역조직화학적 검사 결과 insulin에 양성반응을 보이는 세포가 거의 관찰되지 않았으나, 생진양혈탕가미방을 투여한 실험군의 $\beta$세포들은 대조군에 비하여 강한 양성반응을 보였다. 전자현미경 관찰에서, 대조군 생쥐의 $\beta$세포는 핵의 핵막이 정상군에 비하여 뚜렷하지 않았으며 세포질소기관의 발달도 미약하였다. 세포질내에 함유되어 있는 분비과립의 수가 현저히 감소하였으며 전자밀도가 높은 중심부분의 크기도 정상군에 비하여 현저히 작았다. 실험군의 $\beta$세포에서는 insulin분비소포의 수는 정상군에 비하여 다소 적게 관찰되었으나 대조준에 비해서는 증가하였으며, 전자밀도가 높은 중심부분의 크기도 대조군에 비하여 다소 증가하였다. 혈당은 대조군($365.33{\pm}91.01$, mg/dl)에비하여 실험군($179.56{\pm}73.36$, mg/dl)에서 유의성있게 감소하였다. 위와 같은 결과로 보아 생진양혈탕가미방이 STZ로 유발된 당뇨 생쥐의 치료에 효능이 있음을 확인할 수 있었다. This study was carried out to understand the effects of Saengchinyanghyoltang-gamibang (SGT) on pancreatic islets of the mice induced with streptozotocin (STZ). In the control group, two times injected with 50 mg/kg 572 at 24-hour intervals, a few number of insulin immunoreactive-cells are observed at the pancreatic islets of the mice. In the experimental group which administered with extract of SGT during 21-day, a number of immunoreactive-cells are observed at the pancreatic islets. According to the electron microscopic observation, $\beta-cells$ of the control group were contained a few of secretory granules, but also these granules were contained electro-lucent materials. In the experimental group, a lot o( secretory granules and well developed cell organelles are observed at the $\beta-cells$. The level of glucose was significantly decreased in the experimental group compared with control group, but the level of BUN was similar in these two groups. These results suggest that administration of SGT to the mice improved the damage of $\beta-cells$ from injected with STZ.

        Radioprotective effects of fucoidan on bone marrow cells: improvement of the cell survival and immunoreactivity

        Fucoidan is a sulfated polysaccharide purified from brown algae including Fucus vesiculosus and has a variety of biological effects including mobilization of hematopoietic progenitor cells. Recently, we demonstrated that fucoidan stimulates the antigen-presenting functions of dendritic cells. In this study, we investigated the radioprotective effects of fucoidan on bone marrow cells (BMCs), which are the main cellular reservoir for the hematopoietic and immune system. To evaluate the effects of fucoidan, we assayed cell viability and immune responses. In a viability assay, fucoidan significantly increased the viability of BMCs. Based on the results of flow cytometric analysis, the increased viability of fucoidan-treated BMCs was attributed to the inhibition of radiation-induced apoptosis. Furthermore, fucoidan altered the production of immune-related cytokines from BMCs and increased the capability of BMCs to induce proliferation of allogeneic splenocytes. Taken together, our study demonstrated that fucoidan has radioprotective effects on BMCs with respect to cell viability and immunoreactivity. These results may provide valuable information, useful in the field of radiotherapy.

        자라 위장관 내분비세포에 관한 면역조직화학적 연구

        The gastrointestinal endocrine cells of the Pond tortoise, Amyda sinensis were studied immunohistochemically, and somatostatin-, gastrin/cholecystokinin(GAS/CCK)-, glucagon-, 5-hydroxytryptamine(5-HT)-, insulin- and chromogranin-immunoreactive cells were revealed. The characteristic findings of the regional distribution and relative frequency of these immunoreative cells in the gastrointestinal tract of the Pond tortoise were as follows; A few somatostatin-immunoreactive cells were distributed from the segment I to the segment V. GAS/CCK-immunoreactive cells were found from the segment III to the segment VII. These cells were numerous in the segment III and a few in the other segments. A few glucagon-immunoreactive cells were found in the segment I and rare in the segment II. 5-HT-immunoreactive cells were found throughout the gastrointestinal tract. Numerous numbers of them were found in the segment I, while moderate or a few in the other segments. Insulin-immunoreactive cells were distributed from the segment II to the segment IX. Moderate numbers of them were found in the segment VIII and IX, while a few in the other segments. Chromogranin-immunoreactive cells were found from the segment III to the segment VI. Moderate numbers of these cells were found in the segment IV and V, while a few in the other segments. BPP-immunoreactive cells were not observed throughout the gastrointestinal tract of the Pond tortoise, Amyda sinensis.

        Immunohistochemical study of the gastrointestinal endocrine cells in the Mongolian Gerbils, Meriones unguiculatus

        몽골리안 저빌(Meriones unguiculatus)의 위장관을 분문부, 기저부, 유문부, 십이지장, 공장, 회장, 맹장, 결장 및 직장의 9부분으로 구분하고 이들 부위에서 cholecystokinin(CCK)-8, gastrin, secretin, pancreatic polypeptide(PP), somatostatin, serotonin, glucagon 및 insulin 등 8종의 항혈청에 대한 내분비 세포의 부위별 분포 및 출현빈도를 면역조직화화적으로 관찰하였던 바, CCK-8, gastrin, somatostatin 및 serotonin 면역반응세포들이 관찰되었다. 이들 면역반응세포들은 위 부위에서는 원형에서 타원형으로 주로 위샘(gastric gland) 또는 주세포와 벽세포 사이에서 관찰되었고, 장 부위에서는 원형 또는 타원형의 면역반응세포들이 공장의 장샘(intestinal gland)에서 관찰되었으며, 타원형에서 방추형의 세포들이 장상피세포 사이에서 관찰되었다. CCK-8 면역반응세포들은 유문부와 십이지장에 국한되어 다수 또는 소수 관찰되었으며, gastrin 면역반응세포들은 유문부에 국한되어 다수 관찰되었다. Somatostatin 면역반응세포들 역시 gastrin 변역반응세포들과 유사하게 유문부에 국한되어 중등도의 출현빈도를 나타내었고, serotonin 면역반응세포들은 분문부와 맹장을 제외한 위장관 전부위에 걸쳐 중등도 또는 다수 관찰되었으나, secretin, PP, glucagon 및 insulin 면역반응세포들은 관찰되지 않았다. 이상에서 몽골리안 저빌의 위장관에 존재하는 내분비 세포들의 종류 및 출현빈도는 다른 설치류에 비해 매우 적거나 낮은 것으로 관찰되었으며, 이들 내분비 세포들의 출현부위 역시 다른 설치류에서와는 달리 일부 부분에 국한되는 것으로 관찰되었다. In order to study the regional distribution and relative frequency of the immunoreactive endocrine cells in the gastrointestinal tract of the Mongolian gerbil, Meriones unguiculatus, the gastrointestinal tract was divided into 9 portions (cardia, fundus, pylorus, duodenum, jejunum, ileum, cecum, colon and rectum) and immunostained by immunohistochemical (PAP) method using 8 types of specific antisera against cholecystokinin (CCK)-8, gastrin, secretin, pancreatic polypeptide(PP), somatostatin, serotonin, glucagon and insulin. CCK-8-, gastrin-, somatostatin- and serotonin-immunoreactive cells were demonstrated in this study. These immunoreactive cells were found in the gastric gland regions of the pylorus or between parietal and chief cells of the fundus with round to spherical shaped, and in the interepithelial regions of the intestinal tract with spherical to spindle shaped except for jejunum where some of immunoreactive cells were also observed in the intestinal glands with round to spherical shaped. CCK-8-immunoreactive cells were restricted to the pylorus and duodenum with numerous and a few frequency, respectively. Gastrin-immunoreactive cells were restricted to the pylorus with numerous frequency. Similar to those of gastrin-immunoreactive cells, somatostatin-immunoreactive cells were restricted to pylorus with moderate frequency. Serotonin-immunoreactive cells were detected throughout whole gastrointestinal tract except for cardia and cecum with moderate or numerous frequency. However, no secretin-, PP-, glucagon- and insulin-immunoreactive cells were observed in this study. From these results, the appearance type, regional distribution and relative frequency of immunoreactive endocrine cells in the gastrointestinal tract of the Mongolian gerbils were somewhat lowered or restricted compared to those of other mammals and these differences were might be caused by feeding habits and species specification.

        An immunohistochemical study of the endocrine cells in gastrointestinal tract of the Korean native goat

        한국 재래산양의 위장관 내분비세포를 면역조직화학적으로 관찰하였던 바 5-HT, somatostatin, Gas/CCk, glucagon, chromogranin, PP 면역 반응세포들이 동정되었다. 한국 재래산양의 위장관에 있어서 이들 면역반응세포의 부위별 분포와 출현빈도의 특징은 다음과 같다. 5-HT 면역반응세포는 제 4 위 및 유문부에서 보다 대장과 소장에서 더 많이 출현하였다. Somatostatin면역반응세포는 대장과 소장에서 보다 제 4 위와 유문부에서 다수 출현하였다. Gas/CCk 면역반응세포는 유문부에서 가장 다수로 분포하였으며 기타 부위에서는 소수로 출현하였다. 대장과 소장에서는 중등도의 glucagon 면역반응세포가 관찰되었으나, 제 4위와 유문부에서는 소수로 관찰되었다. Chromogranin 면역반응세포는 전 위장관에서 골고루 그리고 아주 많이 출현하였다. PP면역반응세포는 대장에서 중등도로, 회장에서 소수로 분포하였다. Insulin 면역반응세포는 전 위장관에서 관찰할 수 없었다. The gastrointestinal endocrine cells of the Korean native goat were studied immunohistochemically, and 5-HT-, somatostatin-, Gas/CCK-, glucagon-, chromogranin- and PP- immunoreactive cells were revealed. The characteristic findings of the regional distribution and relative frequency of these immunoreactive cells in the gastrointestinal tract of the Korean native goat were as follows. 5-HT-immunoreactive cells were more numerous in the small and large intestine than in the abdomen stomach. Somatostatin-immunoreactive cell were more numerous in the abdomen stomach than in the small and large intestine. Gas/CCK-immunoreactive cells were concentrated very numerously in the pyloric region with a few in the other regions. Moderate numbers of glucagon-immunoreactive cells were found in the small and large intestine, but a few of them were found in the abdomen stomach. Very numerous chromogranin-immunoreactive cells were detected throughout the gastrointestinal tract. PP-immunoreactive cells were observed moderate numbers in the large intestine with few in the ileum. No insulin-immunoreactive cell was found in the gastrointestinal tract.
