        Amelogenin 유전좌를 이용한 성별검색의 유용성에 관한 연구

        The aim of this study was to validate the sex typing based on amplification of X-Y homologous Amelogenin locus in Korean including mutation rate in this locus. It was found that there was no case with reported mutation that may hinder the exact sex typing among 240 Koreans, and the sex typing was successful even with subnanogram quantities of male and female DNA. There was no difference in the sensitivity of reaction among male and female. Differential amplification between X and Y amelogenin bands in some samples was noted, and dilution study revealed that this phenomenon was more frequent when the quantity of sample was low, usually less than 10 ng. That phenomenon was variable between amplification reactions, and was also dependent on different Taq enzyme used for the amplification. When there was differential amplification, the intensity ratio (Y band/X band) ranged about 0.68-0.87.

        위치민감형 광다이오드 검출기의 신호처리회로 개발과 적용

        본 연구는 위치 민감형 광 증폭 다이오드로부터 나오는 신호를 증폭 및 파형 변화 후 신호의 크기를 검출하여 일정시간 동안 유지시키는 뒷단 회로 개발에 관한 연구이다. 신호발생기에서 발생한 소신호를 증폭 소자를 통한 안정적인 증폭 후 미분회로를 통하여 신호 파형을 검출하기 수월한 형태로 변형 하고, peak/hold 회로에서 피크의 최대점을 일정시간 유지하여 신호의 수집을 원활하게 하였다. 본 회로에 대한 독립적인 성능 평가를 위하여 상용 장비로부터의 검사신호를 입력으로 사용하였다. The aim of this study was to develop a signal process circuit for a position sensitive avalanche photodiode detector. The circuit parts consisted of amplification, differential and peak/hold circuit. This research was the baseline to develop highly compact radiation detector. The signal was amplified by an amplification chip and its shape was changed in a differential circuit to minimize the pulse tailing. The peak/hold circuit detect the peak of the signal from the differential circuit and hold the amplitude of the peak for data acquisition. In order to test the intrinsic function of the circuit, the input signal was transmitted from a commercial pulse generator.

      • Identification of a Novel PGE<sub>2</sub> Regulated Gene in SNU1 Gastric Cancer Cell

        Prostaglandin $E_2$ ($PGE_2$) plays an important role in the regulation of various gastric functions, and the growth-inhibitory activities on tumor cells are studied in vitro and in vivo. Although the mechanisms have attracted many researchers in the past decade, the molecular mechanisms of cell cycle arrest, or induction of apoptosis by $PGE_2$, is unclear. We investigated the effects of $PGE_2$ on the growth of the human gastric carcinoma cell line SNU1 and genes that are regulated by $PGE_2$ and isolated them using differential display RT-PCR (DD RT-PCR). FACS analysis suggested that SNU1 cells were arrested at the G1 phase by $PGE_2$ treatment. This growth inhibitory effect was in a time- and dose-dependent manner. Treatment of SNU1 cells with $10\;{\mu}g/ml$ $PGE_2$, followed by DD RT-PCR analysis, revealed differently expressed bands patterns from the control. Among the differently expressed clones, we found an unidentified cDNA clone (HGP-27) overexpressed in $PGE_2$-treated cells. The full-length cDNA of HGP-27 was isolated using RACE, which consisted of a 30-nt 5'-noncoding region, a 891-nt ORF encoding the 296 amino acid protein, and a 738-nt 3'-noncoding region including a poly(a) signal. This gene was localized on the short arm of chromosome number 11. Using the Motif Finder program, a myb-DNA binding repeat signature was detected on the ORF region. The COOH-terminal half was shown to have similarity with the $NH_3$-terminal domain of thioredoxin (Trx). This relation between HGP-27 and Trx implied a potential role for HGP-27 in modulating the DNA binding function of a transcription factor, myb.

        Angle Sensors Based on Oblique Giant Magneto Impedance Devices

        The measurement of external magnetic field orientation using Giant Magneto Impedance (GMI) sensors has been performed. A soft magnetic alloy of Co30Fe34Ni36 was electroplated on a Si wafer with a CoFeNi seed layer. V-shaped microwire patterns were formed using a conventional photolithography process. An external magnetic field was generated by a rectangular AlNiCo permanent magnet. The reference direction was defined as the external magnetic field direction oriented in the middle of 2 GMI devices. As the orientation of the magnetic field deviated from the reference direction, variation in the field component along each device introduced voltage changes. It was found that, by taking the voltage difference between the left and right arms of the Vshaped device, the nonlinearity of each device could be significantly reduced. The fabricated angle sensor had a linear range of approximately 70° and an overall sensitivity of approximately 10 ㎷.
