        紅藤湯이 LPS 膣內 注入 생쥐에 미치는 抗炎症효과

        Purpose : There are various oriental medicine therapy of treating Pelvic Inflammatory Disease(PID) in clinics. We made the PID mice model by injection Lipopolysaccharide (LPS) in vagina, and investigate anti-inflammatory effects of Hongdeung-Tang among oral medication, retention enema therapy and herbal-acupuncture treatment. Method : The ICR(20-30g) mice(♀) were used. To examine the occurrence of inflammation, LPS in different concentration was injected into the vagina of the mice, and White Blood Cell(WBC) in blood was counted. To examine anti-inflammatory effects, 6 mice were assigned to each of the normal group, the control group and the sample group. Hongdeung-Tang was medicated in oral and rectal with 1.0g/㎏ low dose and 3.0g/㎏ high dose and by herbal acupucture for 5 days, 2 days before LPS injection. After 3days from LPS injection, blood was collected from retro-orbital plexus, and WBC, Interleukin-6(IL-6), Tumor Necrosis Factor-α(TNF-α) in the blood was counted. Result : After LPS injection with each dose, WBC count showed significant increase depending on LPS concentration from 100㎍/㎏, and it was maximized at 3 or 4 days after LPS injection. the Hongdeung-Tang treatment groups, The number of WBC was decreased significantly only in low dose and high dose oral medication, and IL-6 concentration showed significant decrease in oral and rectal medication as well as in herbal acupuncture treatment. TNF-α concentration was decreased significantly in oral and rectal medication of low dose and high dose. herbal-acupuncture treatment group datas showed reductive tendency. Conclusion : Based on above results, we made the PID mice model by injection LPS in vagina , and demonstrated anti-inflammatory effect of Hongdeung-Tang of oral medication, retention enema therapy and herbal-acupuncture treatment.

        강녕탕(康寧湯)의 LPS 질내 주입 생쥐에 미치는 항염증(抗炎症) 효과

        Objective: We made the genital organs inflammatory mice model by vaginal injection of Lipopolysaccharide(LPS), and we intended to make study about anti-inflammatory effects of Gangyeong-Tang, among oral and rectal medication and Herbal-Acupuncture treatment. Method: The female ICR(20-30g) mice were used, the temperature was controlled within $22{\pm}0.5^{\circ}C$ and water and food was not limited. The environment was manipulated to simulate 12 hours of day and 12 hours of night. After LPS injection into vagina, we confirmed on-set of inflammation. 2 days before LPS injection, we started to medicate Gangyeong-Tang in Oral and Rectal and Aqua-acupucture treatment. After 3days from LPS injection, we mesured the White Blood Cell(WBC), Interrleukin-6(IL-6), Tumor Necrosis Factor-${\alpha}$(TNF-${\alpha}$) in blood which was collected from the Retro-orbital Plexus. Results: 1. We made the genital organs inflammatory mice model by vaginal injection of LPS successfully. 2. The number of WBC was decreased significantly as we medicated Gangyeong- Tang in Oral 1g/kg dose, 3g/kg dose and Rectal 1g/kg dose, rectal 3g/kg dose. 3. The concentration of IL-6 was decreased significantly as we medicated Gangyeong-Tang in all group of the Oral, Rectal, Herbal-Acupuncture treatment. 4. The concentration of TNF-${\alpha}$ was decreased significantly as we medicated Gangyeong-Tang in Oral 3g/kg dose and Rectal 1g/kg dose, Rectal 3g/kg dose group. Herbal-Acupuncture treatment group datas showed reductive tendency. Conclusion: As a result of this experiment, we made the genital organs inflammatory mice model by vaginal injection of LPS successfully and demonstrated anti-inflammatory effect of Gangyeong-Tang.

        MiR-200c-3p inhibits LPS-induced M1 polarization of BV2 cells by targeting RIP2

        Background: Microglia are important immune cells, which can be induced by lipopolysaccharide (LPS) into M1 phenotype that express pro-inflammatory cytokines. Some studies have shown that microRNAs play critical roles in microglial activation. Objective: This study was designed to investigate the role of miR-200c-3p in regulating inflammatory responses of LPS-treated BV2 cells. Methods: The expression of miR-200c-3p in BV2 cells was detected by real-time PCR. Receptor-interacting protein 2 (RIP2) was predicted as a target gene of miR-200c-3p. Their relationship was verified by dual-luciferase reporter assay. The function of miR-200c-3p and RIP2 in microglial polarization and NF-κB signaling was further evaluated. Results: LPS treatment reduced miR-200c-3p expression in a dose-dependent and time-dependent manner in BV2 cells. LPS treatment increased the expression of M1 phenotype markers inducible nitric oxide synthase (iNOS) and major histocompatibility complex class (MHC)-II, promoted the release of pro-inflammatory cytokines interleukin (IL)-1β, IL-6 and tumor necrosis factor (TNF)-α, and enhanced the nuclear translocation and phosphorylation of nuclear factor-kappaB (NF-κB) p65. Reversely, miR-200c-3p mimics down-regulated the levels of these inflammatory factors. Furthermore, RIP2 was identified to be a direct target of miR-200c-3p. RIP2 knockdown had a similar effect to miR-200c-3p mimics. Overexpression of RIP2 eliminated the inhibitory effect of miR-200c-3p on LPS-induced M1 polarization and NF-κB activation in BV2 cells. Conclusions: MiR-200c-3p mimics suppressed LPS-induced microglial M1 polarization and NF-κB activation by targeting RIP2. MiR-200c-3p/RIP2 might be a potential therapeutic target for the treatment of neuroinflammation-associated diseases.

        Anti-inflammatory effects of trans-1,3-diphenyl-2,3-epoxypropane-1-one in zebrafish embryos in vivo model

        <P>In this study, trans-1,3-diphenyl-2,3-epoxypropane-1-one (DPEP) was evaluated for its anti-inflammatory activity in lipopolysaccharide (LPS)-stimulated zebrafish embryos. In the present study, DPEP exhibited potential protective effect in the zebrafish embryos as confirmed by survival rate. DPEP acts as an effective agent against reactive oxygen species (ROS) formation induced by LPS. Moreover, DPEP effectively inhibited LPS-induced nitric oxide (NO) production in zebrafish embryos. In addition, DPEP significantly reduced the expression of inducible NOS (iNOS) and cycloxygenase 2 (COX-2), which generate NO as a key mediator of inflammation in a concentration-dependent manner. According to these results, DPEP could be considered an effective anti-inflammatory agent, which might be further developed as a functional ingredient. This is the first report of the anti-inflammatory activity of DPEP in the LPS-stimulated zebrafish model. (C) 2016 Elsevier Ltd. All rights reserved.</P>
