Multilocus Sequence Typing and Virulence Factors Analysis of Escherichia coli O157 Strains in China

Escherichia coli O157:H7, an important food-borne pathogen, has become a major public health concern worldwide. The aim of this study was to investigate the molecular epidemiologic feature of E. coli O157:H7strains in China. 105 E. coli O157:H7 isolates were collected from various hosts and places over 9 years. A multilocus sequence typing scheme (MLST) was applied for bacteria genotyping and polymerase chain reaction (PCR) was used for virulence factor identification. Seven new MLST sequence types (STs), namely ST836, ST837, ST838, ST839, ST840, ST841, and ST842 were identified, which grouped into two lineages. Phylogenetic analysis suggested that the most two frequent STs in China, ST837 and ST836, may be the derivatives of E. coli O157:H7 Sakai or E. coli O157:H7 EDL933. Geographical diversity and host variety of E. coli O157:H7 were observed in China. In addition, the different distribution of tccp was detected. The data presented herein provide new insights into the molecular epidemiologic feature of E. coli O157:H7, and aid in the investigation of the transmission regularity and evolutionary mechanism of E. coli O157:H7.
The Magnetic Filtering Vacuum Arc Film Deposition System and Its Applications

A cathodic arc with beam filter is employed for the deposition of metallic and hydrogen-free amorphous carbon films. A solenoid filter is used to prevent macropaticles and nonionized atoms from reaching the substrate. The detail transport characters of the filter are presented in the paper. With an optmum filter arrangement we are able to obtain a filter output of 18.4% of the total number of ions produced by the vacuum arc discharge. The deposited amorphous cabon thin film contains no hydrogen and a high fraction of $sp^3$ is determined by XPS. A dense Ti film deposited on H13 steel improves the corrosion resistance of the H13 steel and significant improvements of corrosion resistance were observed by implanting Ti, C in the film.

<P>We present an analysis of microlensing event OGLE-2016-BLG-0693, based on the survey-only microlensing observations by the OGLE and KMTNet groups. In order to analyze the light curve, we consider the effects of parallax, orbital motion, and baseline slope, and also refine the result using a Galactic model prior. From the microlensing analysis, we find that the event is a binary composed of a low-mass brown dwarf (49(-18)(+20) Mj) companion and a K- or G-dwarf host, which lies at a distance of 5.0 +/- 0.6 kpc toward the Galactic bulge. The projected separation between the brown dwarf and its host star is less than similar to 5 au, thus it is likely that the brown dwarf companion is located in the brown dwarf desert.</P>

<P>A novel double-templates technique was adopted for solid-phase extraction packing agent, and the obtained hybrid molecularly imprinted polymers with double-templates (theophylline and chlorogenic acid) were characterized by fourier transform infrared and field emission scanning electron microscope. The molecular recognition ability and binding capability for theophylline and chlorogenic acid of polymers was evaluated by static absorption and dynamic adsorption curves. A rapid and accurate approach was established for simultaneous purification of theophylline and chlorogenic acid in green tea by coupling hybrid molecularly imprinted solid-phase extraction with high performance liquid chromatography. With optimization of SPE procedure, a reliable analytical method was developed for highly recognition towards theophylline and chlorogenic acid in green tea with satisfactory extraction recoveries (theophylline: 96.7% and chlorogenic acid: 95.8%). The limit of detection and limit of quantity of the method were 0.01 mu g/mL and 0.03 mu g/mL for theophylline, 0.05 mu g/mL and 0.17 mu g/mL for chlorogenic acid, respectively. The recoveries of proposed method at three spiked levels analysis were 98.7-100.8% and 983-100.2%, respectively, with the relative standard deviation less than 1.9%. Hybrid molecularly imprinted polymers with double-templates showed good performance for two kinds of targets, and the proposed approach with high affinity of hybrid molecularly imprinted polymers might offer a novel method for the purification of complex samples. A porous Nd-doped TiO<SUB>2</SUB> monolith was successfully synthesized with polystyrene spheres as a template using a sol-gel method followed by calcination at high temperature. The porous Nd-doped TiO<SUB>2</SUB> monolith was characterized by X-ray diffractometry, scanning electron microscopy and Brunauer-Emmett-Teller measurements. Nd doping can increase the TiO<SUB>2</SUB> surface area by hindering the growth of large TiO<SUB>2</SUB> particles, and suppressing the recombination of photo-produced hole/electron (h<SUP>+</SUP>/e<SUP>-</SUP>) pairs. Moreover, its porous structure can provide a large surface area, facilitating enhanced adsorption and fast transfer of pollutants. To evaluate the photocatalytic activity of the porous Nd-doped TiO<SUB>2</SUB> monolith, the photo-degradation for methyl orange was investigated under UV-Vis irradiation. The porous 0.5% Nd-doped TiO<SUB>2</SUB> monolith showed better behavior than the other as-prepared samples and Degussa P25 due to its narrow band gap, high efficiency for h<SUP>+</SUP>/e<SUP>-</SUP> pair separation, and large surface area. The synergistic combination of Nd doping and the porous structure is a promising material design strategy for use in the degradation of organic pollutants.

New polymorphism of major histocompatibility complex B-G genes was investigated by amplification and digestion of a 401bp fragment including intron 1 and exon 2 using polymerase chain reaction-restriction fragment length polymorphism (PCRRFLP) technique with two restriction enzymes of Msp I and Tas I in eight breeds of Chinese indigenous chickens and one exotic breed. In the fragment region of the gene, three novel single nucleotide polymorphisms (SNPs) were detected at the two restriction sites. We found the transition of two nucleotides of A294G and T295C occurred at Tas I restriction site, and consequently led to a nonsynonymous substitution of asparagine into serine at position 54 within the deduced amino acid sequence of immunoglobulin variableregion- like domain encoded by the exon 2 of B-G gene. It was observed at rare frequency that a single mutation of A294G occurring at the site, also caused an identical substitution of amino acid, asparagine 54-to-serine, to that we described previously. And the transversion of G319C at Msp I site led to a non-synonymous substitution, glutamine 62-to-histidine. The new alleles and allele frequencies identified by the PCR-RFLP method with the two enzymes were characterized, of which the allele A and B frequencies at Msp I and Tas I loci were given disequilibrium distribution either in the eight Chinese local breeds or in the exotic breed. By comparison, allele A at Msp I locus tended to be dominant, while, the allele B at Tas I locus tended to be dominant in all of the breeds analyzed. In Tibetan chickens, the preliminary association analysis revealed that no significant difference was observed between the different genotypes identified at the Msp I and Tas I loci and the laying performance traits, respectively.

Catastrophic brittle failure prevents the application of bulk metallic glasses (BMGs).The brittle La69Co17Al14BMG showslarge compressive plasticity when compressed in constrained conditions. Here the compressive plasticity of this BMG wassystematically studied by decreasing aspect ratio (height/diameter) of the samples in uniaxial compression and the brittleductiletransition occurs at an aspect ratio of 0.36. With the increase of the plasticity (decrease of the aspect ratio), the hardnessdecreases firstly and then increases, however, the relaxation enthalpy indicating its structure increases monotonically. Our findings provide a powerful experimental support for the application of brittle La-based BMGs.

<P>We analyze an ensemble of microlensing events from the 2015 Spitzer microlensing campaign, all of which were densely monitored by ground-based high-cadence survey teams. The simultaneous observations from Spitzer and the ground yield measurements of the microlensing parallax vector pi(E), from which compact constraints on the microlens properties are derived, including less than or similar to 25% uncertainties on the lens mass and distance. With the current sample, we demonstrate that the majority of microlenses are indeed in the mass range of M dwarfs. The planet sensitivities of all 41 events in the sample are calculated, from which we provide constraints on the planet distribution function. In particular, assuming a planet distribution function that is uniform in log q, where q is the planet-to-star mass ratio, we find a 95% upper limit on the fraction of stars that host typical microlensing planets of 49%, which is consistent with previous studies. Based on this planet-free sample, we develop the methodology to statistically study the Galactic distribution of planets using microlensing parallax measurements. Under the assumption that the planet distributions are the same in the bulge as in the disk, we predict that similar to 1/3 of all planet detections from the microlensing campaigns with Spitzer should be in the bulge. This prediction will be tested with a much larger sample, and deviations from it can be used to constrain the abundance of planets in the bulge relative to the disk.</P>
KMT-2016-BLG-0212: First KMTNet-Only Discovery of a Substellar Companion

We present the analysis of KMT-2016-BLG-0212, a low flux-variation $(I_{\rm flux-var}\sim 20$ mag) microlensing event, which is in a high-cadence ($\Gamma = 4 {\rm hr}^{-1}$) field of the three-telescope Korea Microlensing Telescope Network (KMTNet) survey. The event shows a short anomaly that is incompletely covered due to the brief visibility intervals that characterize the early microlensing season when the anomaly occurred. We show that the data are consistent with two classes of solutions, characterized respectively by low-mass brown-dwarf $(q=0.037)$ and sub-Neptune $(q<10^{-4})$ companions. Future high-resolution imaging should easily distinguish between these solutions.
