      • Innovative Research on Auditory Experience Design Using Binaural Technology in Tourist Interactive Guide Desk

        In recent years, the tourism industry has witnessed multifaceted development trends, including digitalization, intelligence, and sustainable practices. Faced with the transformative shifts in the tourism landscape, epitomized by the emergence of smart tourism, nations globally have launched initiatives to leverage advanced information technology for enhancing the tourism experience and improving service efficiency. In order to present tourists with a more authentic and expressive urban landscape,this article employed research methodologies such as As-Is Analysis and Affinity Diagramming to analyze the travel needs of ten tourists, utilized Binaural, the core technology for achieving immersive audio effects, and innovatively designed the auditory experience in the interactive guide desk. We aim to create a more immersive and unique touring experience, offering a novel technological path for the South Korean tourism industry, thus contributing pioneering concepts to the advancement of smart tourism.

        A general structural design methodology for multi-hole orifices and its experimental application

        Compared to single-hole orifices (SOs), multi-hole orifices (MOs) have smaller orifice sizes and various patterns of orifice distribution. The geometric description of MOs is more complex, increasing the difficulty of MO structural design. Therefore, it is worthwhile to investigate the key factors affecting MO throttle or flow control characteristics and to develop a general MO design method. This work presents a practical geometric design methodology for MOs and applies this procedure in throttle experiments. To describe the MO geometry in detail, the methodology first introduces a comprehensive set of geometric architectures involving orifice arrangement criteria and geometric parameters such as the total orifice number, n; the orifice distribution density, Dd; and the equivalent diameter ratio, EDR. Then, a series of throttle tests in water flow are conducted to investigate the effect of various geometric features on the pressure loss characteristics of MOs. Finally, a simple model to calculate the pressure loss coefficient of MOs is presented.

        Improvement of FK506 Production by Selection of 4-Aminobutyric Acid-tolerant Mutant and Optimization of its Fermentation Using Response Surface Methodology

        Tacrolimus (FK506), a 23-membered polyketidemacrolide with immunosuppressant activity, can be produced byStreptomyces tsukubaensis. We studied a variety of mutant strainsof S. tsukubaensis for the microbial production of FK506. Thebest strain (CZ-19) was obtained from the parent strain LLZ-1 bynitrosoguanidine mutation and 4-Aminobutyric acid (FK506precursor structure analogs) adaption. In the shake-flask experiments,titer of FK506 by CZ-19 was 532.44 mg/L, increased by 65.13%compared to that of the parent strain. Through single factorexperiments and response surface methodology, we furtheroptimized the medium for improved FK506 production by CZ-19in shake flask culture. The optimal medium for enhanced FK506production was as follows: 17.19 g/L corn starch, 21.78 g/Lglucose, 8.06 g/L peptone, and 18.98 mg/L 4-aminobutyric acid. The predicted FK506 titer was 906.49 mg/L, and the experimentaldata confirmed the validity of the model. The present studydemonstrates that S. tsukubaensis CZ-19 is a promising strain forindustrial production of FK506.

        Mitochondrial dysfunction is underlying fluoroquinolone toxicity: an integrated mitochondrial toxicity assessment

        Background Antibiotics bear an inherent risk of mitochondrial toxicity due to the structural similarities between mitochondria and bacteria. Drug-induced mitochondrial dysfunction can contribute to organ toxicity and has resulted in a considerable number of market withdrawals. Fluoroquinolones were alerted for hepatotoxicity liability and were recently given black box warnings by FDA for the potential disabling side eff ects including tendonitis, pain in extremities and joints, neuropathies associated with paraesthesia, depression, impairment of memory, hearing, vision, etc. Objective In this study, the potential involvement of mitochondrial impairment in the toxicity of four fl uoroquinolones was investigated with a panel of mechanistic endpoints including ATP, mitochondrial respiration, as well as mitochondrial DNA and protein levels. Results Data revealed mitochondrial toxicity to diff erent extents, induced by the tested fl uoroquinolones and suggested mitochondrial protein synthesis as potential mechanism of action. Conclusion This study exemplifi es an integrated/holistic approach with a selected battery/panel of in vitro assays/endpoints serving to identify mitochondrial toxicity early on in the development of drugs, in particular, antibiotics.

      • Two Designs of Space-Time Block Codes Achieving Full Diversity With Partial Interference Cancellation Group Decoding

        <P>A partial interference cancellation (PIC) group decoding based space-time block code (STBC) design criterion was recently proposed by Guo and Xia, where the decoding complexity and the code rate traeoff is dealt when the full diversity is achieved. In this paper, two designs of STBC are proposed for any number of transmit antennas that can obtain full diversity when a PIC group decoding (with a particular grouping scheme) is applied at receiver. With the PIC group decoding and an appropriate grouping scheme for the decoding, the proposed STBC are shown to obtain the same diversity gain as the ML decoding, but have a low decoding complexity. The first proposed STBC is designed with multiple diagonal layers and it can obtain the full diversity for two-layer design with the PIC group decoding and the rate is up to 2 symbols per channel use. With PIC-SIC group decoding, the first proposed STBC can obtain full diversity for any number of layers and the rate can be full. The second proposed STBC can obtain full diversity and a rate up to 9/4 with the PIC group decoding. Some code design examples are given and simulation results show that the newly proposed STBC can well address the rate-performance-complexity tradeoff of the MIMO systems.</P>

        Construction of an Efficient Mutant Strain of Trichosporonoides oedocephalis with HOG1 Gene Deletion for Production of Erythritol

        The mitogen-activated protein kinase HOG1 (high-osmolarity glycerol response pathway) plays a crucial role in the response of yeast to hyperosmotic shock. Trichosporonoides oedocephalis produces large amounts of polyols (e.g., erythritol and glycerol) in a culture medium. However, the effects of HOG1 gene knockout and environmental stress on the production of these polyols have not yet been studied. In this study, a To-HOG1 null mutation was constructed in T. oedocephalis using the loxP-Kan-loxP/Cre system as replacement of the targeted genes, and the resultant mutants showed much smaller colonies than the wild-type controls. Interestingly, compared with the wild-type strains, the results of shake-flask culture showed that To-HOG1 null mutation increased erythritol production by 1.44-fold while decreasing glycerol production by 71.23%. In addition, this study investigated the effects of citric acid stress on the T. oedocephalis HOG1 null mutants and the wild-type strain. When the supplementation of citric acid in the fermentation medium was controlled at 0.3% (w/v), the concentration of erythritol produced from the wild-type and To-HOG1 knockout mutant strains improved by 18.21% and 21.65%, respectively.

        Hepatotoxic mechanism of diclofenac sodium on broiler chicken revealed by iTRAQ-based proteomics analysis

        Background: At the therapeutic doses, diclofenac sodium (DFS) has few toxic side effects on mammals. On the other hand, DFS exhibits potent toxicity against birds and the mechanisms remain ambiguous. Objectives: This paper was designed to probe the toxicity of DFS exposure on the hepatic proteome of broiler chickens. Methods: Twenty 30-day-old broiler chickens were randomized evenly into two groups (n = 10). DFS was administered orally at 10 mg/kg body weight in group A, while the chickens in group B were perfused with saline as a control. Histopathological observations, serum biochemical examinations, and quantitative real-time polymerase chain reaction were performed to assess the liver injury induced by DFS. Proteomics analysis of the liver samples was conducted using isobaric tags for relative and absolute quantification (iTRAQ) technology. Results: Ultimately, 201 differentially expressed proteins (DEPs) were obtained, of which 47 were up regulated, and 154 were down regulated. The Gene Ontology classification and Kyoto Encyclopedia of Genes and Genomes pathway analysis were conducted to screen target DEPs associated with DFS hepatotoxicity. The regulatory relationships between DEPs and signaling pathways were embodied via a protein-protein interaction network. The results showed that the DEPs enriched in multiple pathways, which might be related to the hepatotoxicity of DFS, were “protein processing in endoplasmic reticulum,” “retinol metabolism,” and “glycine, serine, and threonine metabolism.” Conclusions: The hepatotoxicity of DFS on broiler chickens might be achieved by inducing the apoptosis of hepatocytes and affecting the metabolism of retinol and purine. The present study could provide molecular insights into the hepatotoxicity of DFS on broiler chickens.

        Enterococcus faecium R0026 Combined with Bacillus subtilis R0179 Prevent Obesity-Associated Hyperlipidemia and Modulate Gut Microbiota in C57BL/6 Mice

        Bacillus subtilis and Enterococcus faecium are commonly used probiotics. This study aimed to identify the effect of live combined Bacillus subtilis R0179 and Enterococcus faecium R0026 (LCBE) on obesityassociated hyperlipidemia and gut microbiota in C57BL/6 mice. Forty male C57BL/6 mice were divided into four groups: normal group (N group), model group (M group), low-dose group (L group), and high-dose group (H group). Mice were gavaged with LCBE at 0.023 g/mice/day (L group) or 0.23 g/mice/day (H group) and fed with a high-fat diet for 8 weeks. In vitro E. faecium R0026 showed an ability to lower the low-concentration of cholesterol by 46%, and the ability to lower the highconcentration of cholesterol by 58%. LCBE significantly reduced the body weight gain, Lee index, brown fat index and body mass index of mice on a high-fat diet. Moreover, LCBE markedly improved serum lipids (including serum triglyceride, total cholesterol, low-density lipoprotein and highdensity lipoprotein) while also significantly reducing liver total cholesterol. Serum lipopolysaccharide and total bile acid in L and H groups decreased significantly compared with M group. PCR-DGGE analysis showed that the composition of gut microbiota in the treatment groups was improved. Akkermansia muciniphila was found in H group. The PCA result indicated a similar gut microbiota structure between LCBE treatment groups and normal group while the number of bands and Shannon diversity index increased significantly in the LCBE treatment groups. Finally, qPCR showed Bifidobacterium spp. increased significantly in H group compared with M group, LCBE alleviated liver steatosis and improved brown adipose tissue index.
