      • Hepatitis B Virus DNA Negativity Acts as a Favorable Prognostic Factor in Hepatocellular Carcinoma Patients

        Background: This retrospective study was aimed to investigate the efficacy of prophylactic agents in hepatocellular carcinoma (HCC) patients receiving TACE and compare the difference between lamivudine and entecavir. Materials and Methods: A consecutive series of 203 HBV-related HCC patients receiving TACE were analyzed including 91 patients given prophylactic agents. Virologic events, defined as an increase in serum HBV DNA level to more than 1 log10 IU/ml higher than the nadir level, hepatitis flares due to HBV reactivation and progression free survival (PFS) were the main endpoints. Results: Some 48 (69.6%) reached virologic response. Prophylaxis significantly reduced virologic events (8.8% vs 58.0%, p=0.000) and hepatitis flares (1.1% vs 13.4%, p=0.001). Patients presenting undetectable HBV DNA levels displayed a significantly improved PFS as compared to those who never achieved undetectable HBV DNA. Prophylaxis and e-antigen positivity were the only significant variables associated with virologic events. In addition, prophylaxis was the only independent protective factor for hepatitis flares. Liver cirrhosis, more cycles of TACE, HBV DNA negativity, a lower Cancer of the Liver Italian Program score, non-metastasis and no hepatitis flares were protective factors for PFS. Prophylactic lamivudine demonstrated similar efficacy as entecavir. Conclusions: Prophylactic agents are efficacious for prevention of HBV reactivation in HCC patients receiving TACE. Achievement of undetectable HBV DNA levels displayed a significant capability in improving PFS. Moreover, persistent tumor residual lesions, positive HBV DNA and hepatitis B flares might be causes of tumor progression in these patients.

      • Efficacy of Prophylactic Entecavir for Hepatitis B Virus-Related Hepatocellular Carcinoma Receiving Transcatheter Arterial Chemoembolization

        Background and Aims: Hepatitis B virus (HBV) reactivation was reported to be induced by transcatheter arterial chemoembolization (TACE) in HBV-related hepatocellular carcinonma (HCC) patients with a high incidence. The effective strategy to reduce hepatitis flares due to HBV reactivation in this specific group of patients was limited to lamivudine. This retrospective study was aimed to investigate the efficacy of prophylactic entecavir in HCC patients receiving TACE. Methods: A consecutive series of 191 HBV-related HCC patients receiving TACE were analyzed including 44 patients received prophylactic entecavir. Virologic events, defined as an increase in serum HBV DNA level to more than 1 log10 copies/ml higher than nadir the level, and hepatitis flares due to HBV reactivation were the main endpoints. Results: Patients with or without prophylactic were similar in host factors and the majorities of characteristics regarding to tumor factors, HBV status, liver function and LMR. Notably, cycles of TACE were parallel between the groups. Ten (22.7%) patients receiving prophylactic entecavir reached virologic response. The patients receiving prophylactic entecavir presented significantly reduced virologic events (6.8% vs 54.4%, p=0.000) and hepatitis flares due to HBV reactivation (0.0% vs 11.6%, p=0.039) compared with patients without prophylaxis. Kaplan-Meier analysis illustrated that the patients in the entecavir group presented significantly improved virologic events free survival (p=0.000) and hepatitis flare free survival (p=0.017). Female and Eastern Cooperative Oncology Group (ECOG) performance status 2 was the only significant predictors for virological events in patients without prophylactic antiviral. Rescue antiviral therapy did not reduce the incidence of hepatitis flares due to HBV reactivation. Conclusion: Prophylactic entecavir presented promising efficacy in HBV-related cancer patients receiving TACE. Lower performance status and female gender might be the predictors for HBV reactivation in these patients.

      • Expression and Clinical Significance of MicroRNA-376a in Colorectal Cancer

        The incidence of colorectal cancer (CRC) is increasing in many Asian countries and microRNAs have already been proven to be associated with tumorigenesis. Currently, microRNA-376a (miR-376a) expression and association with clinical factors in CRC remains unclear. In this study, real-time quantitative reverse transcriptase-polymerase chain reaction (qRT-PCR) was carried out on 53 matched pairs of CRC and adjacent normal mucosa to investigate the expression levels of miR-376a. According to the high or low expression of miR-376a, patients were divided into two groups. The relationship between miR-376a expression and clinicopathological factors of 53 patients was evaluated. Survival analysis of 53 CRC patients was performed with clinical follow-up information and survival curves were assessed by the Kaplan-Meier method. Immunohistochemistry (IHC) staining was performed on sections of paraffin-embedded tissue to investigate the vascular endothelial growth factor (VEGF) expression. MiR-376a showed low expression in cancer tissues compared to the adjacent normal tissues and altered high miR-376a expression tended to be positively correlated with advanced lymph node metastasis and shorter patient survival. VEGF IHC positivity was significantly more common in patients with high expression levels of miR-376a.Those results demonstrated that miR-376a may be a meaningful prognostic biomarker and potential therapeutic target in colorectal cancer.

        Dissimilar Resistance Spot Welding of DP 600/A5052/DP 600 Triple Sheets

        Triple sheets of DP 600/A5052 aluminum alloy/DP 600 steel were joined by means of resistance spot welding in this study. The experimental results showed that the microstructure of fusion zone was lath martensite for DP 600 and column crystals for A5052. Their grains coarsened with increasing distance away from the base metals. The interface zone between DP 600 and A5052 was composed of continuous Fe2Al5 intermetallic compound (IMC) layer and needle-like Fe4Al13 IMC. When welding time prolonged from 8 cycles to 17 cycles, the average length of Fe4Al13 phase adjacent to A5052 shortened from 2.3 μm to 0.89 μm, whereas the thickness of Fe2Al5 IMC layer near DP 600 widened from 1.59 μm to 2.59 μm. The tensile-shear load of DP 600/A5052/DP 600 joints reached a maximum value of 10.796 kN with 14 cycles welding time. Failure during tensile-shear testing occurred along DP 600/A5052 interfaces because Fe2Al5 and Fe4Al13 were brittle phases, but an amount of tearing area was found on the fracture surface.

        Determination of equivalent blasting load considering millisecond delay effect

        In the analysis of the effects of rock tunnel blasting vibration on adjacent existing buildings, the model of simplified equivalent load produces higher calculation result of vibration, due to the lack of consideration of the millisecond delay effect. This paper, based on the static force equivalence principle of blasting load, proposes a new determination method of equivalent load of blasting vibration. The proposed method, based on the elastic-static force equivalence principle of stress wave, equals the blasting loads of several single blastholes in the same section of millisecond blasting to the triangle blasting load curve of the exploded equivalent elastic boundary surface. According to the attenuation law of stress wave, the attenuated equivalent triangle blasting load curve of the equivalent elastic boundary is applied on the tunnel excavation contour surface, obtaining the final applied equivalent load. Taking the millisecond delay time of different sections into account, the time-history curve of equivalent load of the whole section applied on the tunnel excavation contour surface can be obtained. Based on Sailing Tunnel with small spacing on Sanmenxia-Xichuan Expressway, an analysis on the blasting vibration response of the later and early stages of the tunnel construction is carried out through numerical simulation using the proposed equivalent load model considering millisecond delay effect and the simplified equivalent triangle load curve model respectively. The analysis of the numerical results comparing with the field monitoring ones shows that the calculation results obtained from the proposed equivalent load model are closer to the measured ones and more feasible.

        Nugget Formation and Its Mechanism of Resistance Spot Welded Joints in DP600 Dual-Phase and DC54D Ultralow Carbon Steel

        Resistance spot welded joints in different configurations of DP600 and DC54D were investigated to elucidate thenugget formation process and mechanical properties of the resultant joints. Results show that, when the weldingtime was less than 4 cycles, the fusion zone (FZ) was not formed, but the heat-affected zone (HAZ) occurred with a“butterfly” shape. In 4 cycles, the FZ in dissimilar sheets occurred with an “abnormal butterfly” shape because of nuggetshift. When the welding time increased to 14 cycles, the FZ exhibited a “bread loaf” shape and the weld shifted to“dog bones.” The nugget can be divided into three regions, namely, FZ, HAZ1, and HAZ2, and the FZ consisted oflath martensite. The micro hardness of DP600 FZ was lower than that of HAZ because of the dilution of DC54D. The failure mode of B changed from interfacial failure to plug failure during the nugget formation process. The tensile-shear load of sound weld is 6.375, 6.016, and 19.131 kN.

        A novel active disturbance rejection-based control strategy for a gun control system

        To compensate for the nonlinearity and to achieve finely-tuned tracking accuracy of a gun control system driven by an AC machine,an improved active disturbance rejection control (IADRC) strategy with neural network embedding (NN-IADRC) is developed in this paper. The proposed IADRC, which has amnestic memory effects, can be regarded as an extension of the conventional ADRC (CADRC),making it a special case of the IADRC. To further attenuate the dependence on system models and enhance the disturbance rejection capacities of the IADRC strategy, an on-line NN-based optimum updating approach is also developed in this paper. Finally, a series of experiments are conducted on the semi-physical simulation platform to estimate the performance of the control system and the effects of the memory factor on the system. The experimental results confirm that the proposed NN-IADRC is highly robust. The results also confirm that it performs more excellently than the CADRC and that its fine tuning has attained tracking accuracy.

        Effects of different selenium sources and levels on antioxidant status in broiler breeders

        Objective: This study was conducted with the objectives to examine the impacts of inorganic selenium (Se) and different types and levels of organic selenium on the serum and tissues Se status and antioxidant capacity in broiler breeders. Methods: Five hundred and forty 48-wk-old Lingnan Yellow broiler breeders were randomly assigned to 6 dietary treatments, provided same basal diet (0.04 mg/kg of Se) with 0.15 mg/kg, or 0.30 mg/kg of Se from sodium selenite (SS) or from selenium-enriched yeast (SY) or from selenomethionine (SM). The broiler breeders were slaughtered after an 8-wk experiment. Results: The results showed that SM was better than SY and SS, 0.30 mg/kg level was better than 0.15 mg/kg level in Se deposition (p<0.05) in serum, liver, kidney, pancreas and muscle; in antioxidant status, organic selenium had better effects than SS in broiler breeders (p<0.05), but SM and SY had a similar result, and 0.15 mg/kg level was better than 0.30 mg/kg (p<0.05). Conclusion: The results demonstrated the evident advantage of supplementation of broiler breeders with 0.15 mg/kg SM, which improved tissue Se concentrations and antioxidant status, and can be considered as the best selenium source.

      • Efficacy and Tolerance of Pegaspargase-Based Chemotherapy in Patients with Nasal-Type Extranodal NK/T-Cell Lymphoma: a Pilot Study

        Nasal-type extranodal natural killer (NK)/T-cell lymphoma (ENKL) is a highly invasive cancer with a poor prognosis. More effective and safer treatment regimens for ENKL are needed. Pegaspargase (PEG-Asp) has a similar mechanism of action to L-asparaginase (L-Asp), but presents lower antigenicity. The aim of the present research was to evaluate the safety profile and the latent efficacy of a PEG-Asp-based treatment regimen in patients with ENKL. Data collected from 20 patients with histologically confirmed ENKL, admitted to the Third Affiliated Hospital of Sun Yat-Sen University from January 2009 to August 2013, were included in the study. All patients received $2500IU/m^2$/IM PEG-Asp on day 1 of every 21-day treatment cycle. Patients received combination chemotherapy with CHOP (n=5), EPOCH (n=7), GEMOX (n=7) or CHOP with bleomycin (n=1). After 2-5 treatment cycles (median, 4 cycles) of PEG-Asp-based chemotherapy, five patients (25%) showed a complete response (CR), and the overall response rate (ORR) was 60%. Grade 3/4 neutropenia occurred in fourteen patients (70%). Grade 3 alanine aminotransferase (ALT) elevation was observed in two. Grade 1-2 non-hematological toxicity consisted of activated partial thromboplastin time (APTT) elongation (n=9), hypofibrinogenemia (n=6), hypoproteinemia (n=17), hyperglycemia (n=3), and nausea (n=6). No allergic reactions were detected. No treatment related death was reported. Our results suggested that PEG-Asp-based chemotherapy presented an acceptable tolerance and a potential short-term outcome in patients with nasal-type ENKL.

        The attenuation of chlorogenic acid on oxidative stress for renal injury in streptozotocin-induced diabetic nephropathy rats

        The present study was undertaken to investigate whether chlorogenic acid (CGA) could protect kidney function against oxidative stress in the diabetic nephropathy (DN) rats. The treatment with CGA could decrease significantly the levels of blood glucose, blood urea nitrogen and serum creatinine in DN rats. Moreover, CGA significantly increased the activity of superoxide dismutase, glutathione peroxidase, and catalase. Moreover, the level of lipid peroxidation malondialdehyde was reduced markedly after CGA administration. Immunohistochemical analysis also showed that CGA downregulated significantly cyclooxygenase-2 protein expression in renal tissue, which is considered as one of the major pathogeneses of oxidative stress. Furthermore, we demonstrated that CGA could block the expression of activating transcription factor-6, C/EBP homology protein and the phosphorylation of eukaryotic initiation factor 2α and double stranded RNAactivated protein kinase-like endoplasmic reticulum kinase. In addition, we attempted to detect the presence of diabetic renal tissues apoptosis-related proteins. Our data provided evidence to support this fact that CGA attenuated oxidative stress in streptozocin-induced DN rats. Its molecular mechanism may inhibit the endoplasmic reticulum-stress response in DN.
