        Extracellular products-mediated interspecific interaction between Pseudomonas aeruginosa and Escherichia coli

        The Gram-negative pathogen Pseudomonas aeruginosa adopts several elaborate strategies to colonize a wide range of natural or clinical niches and to overcome the neighboring bacterial competitors in polymicrobial communities. However, the relationship and interaction mechanism of P. aeruginosa with other bacterial pathogens remains largely unexplored. Here we explore the interaction dynamics of P. aeruginosa and Escherichia coli, which frequently coinfect the lungs of immunocompromised hosts, by using a series of on-plate proximity assays and RNA-sequencing. We show that the extracellular products of P. aeruginosa can inhibit the growth of neighboring E. coli and induce a large-scale of transcriptional reprogramming of E. coli, especially in terms of cellular respiration- related primary metabolisms and membrane components. In contrast, the presence of E. coli has no significant effect on the growth of P. aeruginosa in short-term culture, but causes a dysregulated expression of genes positively controlled by the quorum-sensing (QS) system of P. aeruginosa during subsequent pairwise culture. We further demonstrate that the divergent QS-regulation of P. aeruginosa may be related to the function of the transcriptional regulator PqsR, which can be enhanced by E. coli culture supernatant to increase the pyocyanin production by P. aeruginosa in the absence of the central las-QS system. Moreover, the extracellular products of E. coli promote the proliferation and lethality of P. aeruginosa in infecting the Caenorhabditis elegans model. The current study provides a general characterization of the extracellular products-mediated interactions between P. aeruginosa and E. coli, and may facilitate the understanding of polymicrobial infections.

        Preparation and Characterization of Sulfated TiO2/zeolite Composite Catalysts with Enhanced Photocatalytic Activity

        Sulfated TiO2 nanoparticles were successfully immobilized on zeolite through improving hydrolysis-deposition method. Microstructure, crystallization, surface state and surface area of composite catalysts were characterized by SEM, XRD, FTIR spectra, XPS and BET and the photocatalytic activity was evaluated by degradation of methyl orange under UV irradiation. We optimized these factors (SO4 -2) ions, calcination temperature and loading amount of sulfated TiO2) on photocatalytic activity and crystallization of composite photocatalysts. The results indicated that the SO4 -2 ) ions are successfully immobilized on the surface of TiO2, and sulfated TiO2/zeolite show the highest photocatalytic activity for methyl orange at the [SO4 -2]/[Ti4+] molar rate of 1:1, calcination temperature of 600 ℃ for 2 h, and sulfated TiO2 loading amount of 40%, respectively.

        Development of Integrated Multi-Station System to Precisely Detect and Mitigate Surface Damage on Fused Silica Optics

        In high power laser facility, irreversible damage on fused silica optics, induced by laser irradiation or processing, seriously affects the service life of optics. Therefore, the work of inhibiting damage growth has been carried out in various countries. In our work, an integrated multi-station system is designed to detect and mitigate surface damage on fused silica. The process of processing fused silica optics include UV laser conditioning, surface damage detection and surface damage mitigation with CO2 laser. UV laser conditioning pre-initiates surface damage on fused silica optics with the laser flux less than Laser-Induced Damage Threshold (LIDT). Images of surface damage acquired from camera are processed by improved global threshold segmentation algorithm to extract damage information. Finally, CO2 laser is applied to process the damage with specific morphology to enhance the laser damage resistance. This integrated multi-station system saves the repeated optics installation time between the workstations with the positioning accuracy of 20 μm. Furthermore, the damage with diameter of 10 μm is mitigated to prolong service life of processed fused silica optics. The efficient and accurate integrated multi-station system is of great significance for off -line detecting and mitigating surface damage of fused silica optics in high power laser facility.

      • High Expression of Bcl-2 Protein Predicts Favorable Outcome in Non-small Cell Lung Cancer: Evidence from a Systematic Review and Meta-analysis

        Background: The prognostic value of Bcl-2 protein expression in non-small cell lung cancer (NSCLC) is under debate. We therefore systematically reviewed the evidence for Bcl-2 protein effects on NSCLC survival to elucidate this issue. Materials and Methods: An electronic search in Pubmed and Embase complemented by manual searches in article references were conducted to identify eligible studies to evaluate the association between Bcl-2 protein expression and overall survival (OS) as well as disease free survival (DFS) of NSCLC patients. Combined hazard ratios (HRs) with corresponding 95% confidence intervals (95%CIs) were pooled using the random-effects model. Results: A total of 50 trials (including 52 cohorts) encompassing 7,765 patients were pooled in the meta-analysis regarding Bcl-2 expression and OS of NSCLC patients. High expression of Bcl-2 protein had a favorable impact (HR=0.76, 95%CI=0.67-0.86). In the group of Bcl-2 expression and DFS, 11 studies including 2,634 patients were included. The synthesized result indicated high expression of Bcl-2 protein might predict good DFS (HR=0.85, 95%CI=0.75-0.95). Conclusions: Our present meta-analysis demonstrated favorable prognostic values of Bcl-2 expression in patients with NSCLC. Further prospective trails are welcomed to validate the utility of assessing Bcl-2 in NSCLC patient management.

      • Guided waves of porous FG nanoplates with four edges clamped

        Based on the nonlocal strain gradient (NSG) theory and considering the influence of moment of inertia, the governing equations of motion of porous functionally graded (FG) nanoplates with four edges clamped are established; The Galerkin method is applied to eliminate the spatial variables of the partial differential equation, and the partial differential governing equation is transformed into an ordinary differential equation with time variables. By satisfying the boundary conditions and solving the characteristic equation, the dispersion relations of the porous FG strain gradient nanoplates with four edges fixed are obtained. It is found that when the wave number is very small, the influences of nonlocal parameters and strain gradient parameters on the dispersion relation is very small. However, when the wave number is large, it has a great influence on the group velocity and phase velocity. The nonlocal parameter represents the effect of stiffness softening, and the strain gradient parameter represents the effect of stiffness strengthening. In addition, we also study the influence of power law index parameter and porosity on guided wave propagation.

      • Functional Investigation on Aromatase in Endometrial Hyperplasia in Polycystic Ovary Syndrome Cases

        Objective: To explore the possible significance of aromatase P450 in endometrial hyperplasia with a background of polycystic ovary syndrome (PCOS). Methods: Immunohistochemistry was used to determine the expression of aromatase P450 in endometrium of PCOS patients. Semiquantitative analysis of aromatase P450 expression of mRNA and protein level wasalso carried out by real-time quantitative RT-PCR method. After endometrial cells were stimulated by testosterone and letrozole in vitro, the estradiol ($E_2$) level was determined, and the expression of cell aromatase P450 mRNA was assessed. Results: The aromatase P450 mRNA level was increased in endometria of PCOS patients. When endometrial cells were cultured with $10^{-6}M$ testosterone, the $E_2$ level in the culture medium increased. An inhibitory effect on $E_2$ generation and expression of aromatase P450 mRNA was observed when the endometrial cells were treated with $10^{-5}M$ letrozole. Conclusions: There is an increased expression of aromatase P450 in PCOS patient endometrium. Androgen stimulation could enhance the synthesis of aromatase P450 mRNA and the production of $E_2$ in endometrial cells in vitro while letrozole could do the reverse.

        A dual-site modulated FRET-based two-photon ratiometric fluorescent probe for tracking lysosomal pH changes in living cells, tissues and zebrafish

        <P><B>Abstract</B></P> <P>Lysosomal pH plays an essential role in mediating various biological processes such as immunization, cell metabolism and enzyme activity. Herein, by utilizing the fluorescence resonance energy transfer (FRET) strategy, a two-photon (TP) ratiometric fluorescent probe (NpLys-pH) has been developed for tracking of lysosomal pH changes in living cells, tissues, and zebrafish. NpLys-pH was constructed by conjugating a pH turn-on TP fluorophore <B>1</B> (D-π-A-structured naphthalene derivative) with a pH turn-off naphthalimide fluorophore <B>2</B> <I>via</I> a non-conjugated linker. Meanwhile, NpLys-pH has two potential pH response sites that modulate the fluorescence signal by ICT and PET, respectively. The FRET process exists between the TP fluorophore <B>1</B> and naphthalimide fluorophore <B>2</B>. In addition, NpLys-pH respond to pH rapidly and reversibly with high selectivity and sensitivity and has been applied for tracking lysosomal pH changes in living cells, tissues and zebrafish. We expect that the new probe present here can prompt the development of a wide variety of TP ratiometric fluorescent probes which can find application in detecting other important analytes in biological systems.</P> <P><B>Highlights</B></P> <P> <UL> <LI> A FRET-based TP ratiometric probe for tracking of lysosome pH changes. </LI> <LI> It has a pH turn-on donor and a pH turn-off acceptor. </LI> <LI> NpLys-pH possesses two well-resolved emission peaks separated by 115 nm. </LI> <LI> It successfully tracked lysosome pH changes in live cells, tissues, and zebrafish. </LI> </UL> </P>

        CD44 Antibody-Conjugated Gold Nanostars as SERS Probes for Distinguishing Cancer Cells (A549 Cells, H1229 Cells) from Normal Cells (ATII Cells)

        In this paper, we report a novel antibody conjugated gold nanostars (AuNSs) as surface-enhanced Raman spectroscopy (SERS) probes for distinguishing cancer cells (A549 cells, H1229 cells) from normal cells (ATII cells). In such a probe, bovine serum albumin (BSA) was served as the protective agent and stabilizing agent, 4-mercaptobenzoic acid (4-MBA) was used as the Raman reporter to generate SERS signals as well as the conjugation agent for attaching CD44 antibody (anti-CD44) molecules to AuNSs, where anti-CD44 could effectively target to CD44 protein overexpressed cells. All the results of the experiments con¯rmed that more SERS probes have been targeted to cancer cells (A549 cells and H1229 cells) than that of normal cells (ATII cells) under the same condition. The anti-CD44 SERS probes hold a potential application in distinguishing cancer cells from normal cells with high sensitivity and good biocompatibility.

        Effect of Hollow Microspheres as Reinforcing Fillers on Natural Rubber Composites

        This study aims to investigate the effect of a new hollow microsphere (RiM01) as a reinforcing filler fornatural rubber (NR) instead of carbon black (CB) on rubber composites. The differences in vulcanization characteristicsand mechanical properties of the natural rubber composites are being investigated by carbon black and hollowmicrosphere ratios. The results show that the addition of hollow microspheres decreases the tensile strength, abrasionresistance, and hardness of the rubber composites. However, when the hollow microspheres completely replace carbonblack, the compression set is reduced from the initial 33% to 15%. In addition, the tensile and tear strengths of the rubbercomposites with 50 phr hollow microspheres are only reduced by 7.04% and 1.68% compared with those before aging;with 40 phr hollow microspheres, the 300% constant tensile stresses after aging are enhanced by 45% compared withthose before aging. The aging resistance is improved.
