Fermented cottonseed meal improves production performance and reduces fat deposition in broiler chickens

        Niu Jun Li,Wei Lian Qing,Luo Yuan Qing,Yang Wen Ting,Lu Qi Cheng,Zheng Xin Xia,Niu Yu Jie,Sheng Wen,Cheng Hong,Zhang Wen Ju,Nie Cun Xi 아세아·태평양축산학회 2021 Animal Bioscience Vol.34 No.4

Objective: This study was conducted to investigate the effect of fermented cottonseed meal (FCSM) on growth performance, carcass traits, and fat deposition in white-feather broiler chickens. Methods: A total of 480 male one-day-old white-feather broiler chickens were selected randomly and divided into four groups with six replicates of 20 chickens in each. The experimental chickens were fed diets including 3%, 6%, or 9% FCSM fermented by Candida tropicalis until 42 days old. In the experiment, the chickens of the control group were fed soybean meal. Results: FCSM supplementation linearly decreased the feed conversion ratio from d 15 to 21 and d 36 to 42, respectively (p<0.05). The percentage of carcass and semi-eviscerate increased in response to dietary FCSM supplementation at d 21 (p<0.05). The percentage of eviscerated and semi-eviscerate of 3FCSM was higher than that in other groups at d 35 (p<0.05). At the age of 42 d, the percentage of carcass increased in a quadratic way among increasing FCSM in diets (p<0.05). The subcutaneous fat thickness linearly decreased with the increasing levels of FCSM at d 21 (p<0.05). Gompertz and Logistic functions provided a better fit on abdominal fat and subcutaneous fat, respectively. The best fitted equation predicted that the maximum growth rate of abdominal fat weight and subcutaneous fat thickness occurred at d 28. FCSM had no significant effects on the shape of growth curve of abdominal fat weight and subcutaneous fat thickness, but reduced the height of the curve. Birds receiving the 6FCSM diet for 21 d had smaller adipocyte surface and lower serum glucose as well as triglyceride concentration. Conclusion: FCSM is beneficial for broiler chickens as it positively affects their growth and carcass in addition to altering their fat deposition. Optimization the extraction of anthocyanins from blueberry residue by dual-aqueous phase method and cell damage protection study

        Yuan Gao,Yubin Ji,Fuling Wang,Wenlan Li,Xiaomeng Zhang,Zhihui Niu,Zhuo Wang 한국식품과학회 2021 Food Science and Biotechnology Vol.30 No.13

Blueberry residue is usually discarded as waste, but has a high anthocyanins content. The extraction methodof anthocyanins from blueberry residue with ultrasonicassisted dual-aqueous phase system was optimized. Interms of the principle of central group and design (CCD)experimental design, three-factor and five-level responsesurface analysis was adopted to optimize the extractionconditions with the extraction rate of anthocyanins. Theoptimum extraction rate of anthocyanin was12.372 ± 0.078 mg/g. Anthocyanin extract could protectthe pBR322 DNA oxidative damage induced by Fentonreagent, increase the superoxide dismutase(SOD) and glutathioneperoxidase (GSH-Px) enzyme activities, anddecrease the H2O2-induced cell apoptosis of human normalliver cell (LO2 cell). The study indicates that the extractionrate of anthocyanin was increased by optimized ultrasonicassisted dual-aqueous phase system. The anthocyaninextract could protect DNA and LO2 cell from oxidativedamage.

Endophytic Fungi from Dalbergia odorifera T. Chen Producing Naringenin Inhibit the Growth of Staphylococcus aureus by Interfering with Cell Membrane, DNA, and Protein

        Yuan Gao,Yubin Ji,Wenlan Li,Fuling Wang,Fuling Wang,Xiaomeng Zhang,Zhihui Niu,Lulu Zhou,Lijun Yan 한국식품영양과학회 2021 Journal of medicinal food Vol.24 No.2

This study focused on the antibacterial effects of the endophytic fungi producing naringenin from Dalbergia odorifera T. Chen against Staphylococcus aureus. The antibacterial activity was measured by the inhibition diameters, minimum inhibitory concentration (MIC), and minimum bactericidal concentration (MBC). The time-killing curve was also used to evaluate its antibacterial efficacy. The results of antibacterial activity determinations showed that endophytic fungi secondary metabolites can inhibit the growth of five pathogenic bacteria (S. aureus, Escherichia coli, Salmonella enteritidis, Pseudomonas aeruginosa, and Bacillus subtilis) and the most sensitive strain was S. aureus that had the MIC and MBC values of 0.13 and 0.50 mg/mL, respectively. The membrane permeability study was measured by a DNA leakage assay and electrical conductivity assay. Furthermore, the whole-cell protein lysates and DNA fragmentation assay was evaluated. The morphology of S. aureus treated with the endophytic fungi products was observed by scanning electron microscopy (SEM). The probable antibacterial mechanism of endophytic fungi secondary metabolites was the increased membrane permeability that leads to leaks of nucleic acids and proteins. SEM results further confirmed that the extracts can interfere with the integrity of S. aureus cell membrane and further inhibit the growth of bacteria, resulting in the death of bacteria. This study provides a new perspective for the antibacterial functions of endophytic fungi secondary metabolites for biomedical applications.

      • Alternative Messenger RNA Splicing of Autophagic Gene Beclin 1 in Human B-cell Acute Lymphoblastic Leukemia Cells

        Niu, Yu-Na,Liu, Qing-Qing,Zhang, Su-Ping,Yuan, Na,Cao, Yan,Cai, Jin-Yang,Lin, Wei-Wei,Xu, Fei,Wang, Zhi-Jian,Chen, Bo,Wang, Jian-Rong Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.5

Beclin 1 is a key factor for initiation and regulation of autophagy, which is a cellular catabolic process involved in tumorigenesis. To investigate the role of alternative splicing of Beclin1 in the regulation of autophagy in leukemia cells, Beclin1 mRNA from 6 different types of cell lines and peripheral blood mononuclear cells from 2 healthy volunteers was reversely transcribed, subcloned, and screened for alternative splicing. New transcript variants were analyzed by DNA sequencing. A transcript variant of Beclin 1 gene carrying a deletion of exon 11, which encoded a C-terminal truncation of Beclin 1 isoform, was found. The alternative isoform was assessed by bioinformatics, immunoblotting and subcellular localization. The results showed that this variable transcript is generated by alternative 3' splicing, and its translational product displayed a reduced activity in induction of autophagy by starvation, indicating that the spliced isoform might function as a dominant negative modulator of autophagy. Our findings suggest that the alternative splicing of Beclin 1 might play important roles in leukemogenesis regulated by autophagy.

Implementation and verification of a four-probe motion error measurement system for a large-scale roll lathe used in hybrid manufacturing

        Chen, Yuan-Liu,Niu, Zengyuan,Matsuura, Daiki,Lee, Jung Chul,Shimizu, Yuki,Gao, Wei,Oh, Jeong Seok,Park, Chun Hong IOP Pub 2017 Measurement science & technology Vol.28 No.10

In this paper, a four-probe measurement system is implemented and verified for the carriage slide motion error measurement of a large-scale roll lathe used in hybrid manufacturing where a laser machining probe and a diamond cutting tool are placed on two sides of a roll workpiece for manufacturing. The motion error of the carriage slide of the roll lathe is composed of two straightness motion error components and two parallelism motion error components in the vertical and horizontal planes. Four displacement measurement probes, which are mounted on the carriage slide with respect to four opposing sides of the roll workpiece, are employed for the measurement. Firstly, based on the reversal technique, the four probes are moved by the carriage slide to scan the roll workpiece before and after a 180-degree rotation of the roll workpiece. Taking into consideration the fact that the machining accuracy of the lathe is influenced by not only the carriage slide motion error but also the gravity deformation of the large-scale roll workpiece due to its heavy weight, the vertical motion error is thus characterized relating to the deformed axis of the roll workpiece. The horizontal straightness motion error can also be synchronously obtained based on the reversal technique. In addition, based on an error separation algorithm, the vertical and horizontal parallelism motion error components are identified by scanning the rotating roll workpiece at the start and the end positions of the carriage slide, respectively. The feasibility and reliability of the proposed motion error measurement system are demonstrated by the experimental results and the measurement uncertainty analysis.
