Background/Aims Platelet-derived growth factor receptor alpha-positive (PDGFRα+) cells function in the purinergic regulation of gastrointestinal motility, and purines are reportedly inhibitory neurotransmitters in the enteric nervous system. We explore the distribution and function of PDGFRα+ cells related to purinergic inhibitory neurotransmission in human right and left colons. Methods Human colonic segments were prepared with mucosa and submucosa intact, and the circular muscle tension and longitudinal muscle tension were recorded. Purinergic neurotransmitters were administered after recording the regular contractions. Immunohistochemistry was performed on the circular muscle layers. Intracellular recording was performed on the colonic muscular layer. SK3, P2RY1, and PDGFR-α mRNA expression was tested by quantitative real-time polymerase chain reaction (qPCR). Results Adenosine triphosphate (ATP) treatment significantly decreased the frequency and area under the curve (AUC) of the segmental contraction in right and left colons. Beta-nicotinamide adenine dinucleotide (β-NAD) decreased the frequency in the right colon and the amplitude, frequency and AUC in the left colon. Apamin significantly increased frequency and AUC in the left colon, and after apamin pretreatment, ATP and β-NAD did not change segmental contractility. Through intracellular recordings, a resting membrane potential decrease occurred after ATP administration; however, the degree of decrease between the right and left colon was not different. PDGFRα+ cells were distributed evenly in the circular muscle layers of right and left colons. SK3, P2RY1, and PDGFRα expression was not different between the right and left colon. Conclusion Purines reduce right and left colon contractility similarly, and purinergic inhibitory neurotransmission can be regulated by PDGFRα+ cells in the human colon.

In several lysosomal storage disorders, including Niemann-Pick disease Type C (NP-C), sphingolipids, including glycosphingolipids, particularly gangliosides, are the predominant storage materials in the brain, raising the possibility that accumulation of these lipids may be involved in the NP-C neurodegenerative process. However, correlation of these accumulations and NP-C neuropathology has not been fully characterized. Here we derived NP-C mice with complete and partial deletion of the Siat9 (encoding GM3 synthase) gene in order to investigate the role of ganglioside in NP-C pathogenesis. According to our results, NP-C mice with homozygotic deletion of GM3 synthase exhibited an enhanced neuropathological phenotype and died significantly earlier than NP-C mice. Notably, in contrast to complete depletion, NP-C mice with partial deletion of the GM3 synthase gene showed ameliorated NP-C neuropathology, including motor disability, demyelination, and abnormal accumulation of cholesterol and sphingolipids. These findings indicate the crucial role of GM3 synthesis in the NP-C phenotype and progression of CNS pathologic abnormality, suggesting that well-controlled inhibition of GM3 synthesis could be used as a therapeutic strategy.

사람 난소내 난포의 수적 감소와 배란 후 황체의 퇴축 과정은 apoptosis에 의해서 이루어진다. 그러나 이러한 생리적 과정에 어떤 인자들이 영향을 주는지는 아직 밝혀져 있지 않다. 따라서 저자들은 난소내 apoptosis와 이에 영향을 끼치는 인자들의 관계를 규명하기 위하여 Fas, Fas-ligand, Bcl-2 및 Bad 항체를 이용한 면역조직화학적 방법으로 사람 난소의 원시 난포, 일차 난포, 우성 난포에서의 발현을 관찰하였다. To investigate the effects of feeding restricted on growth, carcass characteristics and plasma profiles in an attempt for optimum responses, a total of 108 cross-bred finishing barrows [(Landrace${\times}$Yorkshire)${\times}$Duroc]weighing an average of $46.88{\pm}0.52kg$ were assigned in a randomized complete block (RCB) design to one of four treatments with three replicates and nine pigs per pen. Feeding regimens were, 1) ad libitum from 50 kg to market weight (Ad 3/3), 2) restricted feeding from 90 kg to market weight (Ad 2/3), 3) restricted feeding from 70 kg to market weight (Ad 1/3), and 4) restricted feeding from 50 kg to market weight (Ad 0/3). During the experimental period, average daily feed intake (ADFI) was decreased from 2.53 kg (AD 3/3) to 2.09 kg (AD 0/3) with increasing restricted feeding duration of (p<0.05). Average daily gain (ADG) of AD 3/3 (0.79 kg) was significantly higher (p<0.05) than those of AD 1/3 (0.74 kg) or AD 0/3 (0.72 kg). Feed efficiency was not influenced by restriction regimens. Blood IGF-I concentrations were increased from 74.14 to 134.25 (167.36-115.66) ng/ml as body weight increased. Blood leptin concentrations were affected by feed intake level and coincided with blood IGF-I concentrations. Most of carcass characteristics were not significantly affected by restricted feeding, however cooking losses in AD 1/3 and Ad 0/3 treatment diet were higher than those in Ad 3/3 and Ad 2/3. In addition, there was a trend that backfat thickness was lowered in proportional to decreasing feed intake (p>0.05). In conclusion, restricted feeding improved feed efficiency after 50 kg body weight without deteriorating the pork quality of barrows.

Conjugated linoleic acid (CLA), when incorporated into mouse liver microsomal membranes, selectively inhibits the mutagenesis of 2-amino-3-methylimidazo[4,5-f]quinoline (IQ). Nine-week old female ICR mice were given (p.o.) 0.1 mL olive oil alone (control), 0.1 mL olive oil plus 0.1 mL linoleic acid, or 0.1 mL olive oil plus 0.1 mL CLA, twice weekly for four weeks. The animals were then sacrificed and liver S-9 fractions were prepared. Activation of IQ for mutagenesis by the liver S-9 from CLA-treated mice was significantly reduced in comparison with liver S-9 from control or linoleic acid-treated mice. By contrast, the activation of 7,12-dimethylbenz[a]anthracene (DMBA) and benzo[a]pyrene (BP) was unaffected. Hence, CLA incorporated into phospholipids may selectively affect cytochrome P450 isozymes responsible for activating IQ, but not those which activate BP or DMBA. The addition of free CLA or the methyl esters of CLA, linoleic acid, or oleic acid, to control S-9 inhibited the activation of all three mutagens (IQ, BP, and DMBA).
