        RhoGDI2 induced malignant phenotypes of pancreatic cancer cells via regulating Snail expression

        Background: Rho GDP dissociation inhibitor 2 (RhoGDI2) has been shown to contribute to the aggressive phenotypes of human cancers, such as tumor metastasis and chemoresistance. Objective: This study aimed to assess the effects of RhoGDI2 on tumor progression and chemoresistance in pancreatic cancer cells. Methods: The expression of RhoGDI2 in pancreatic cancer cells was detected by Western blot analysis. Gain-of-function and loss-of-function approaches were done to examine the malignant phenotypes of the RhoGDI2-expressing or RhoGDI2-depleting cells. The correlation between RhoGDI2 and Snail was also analyzed. Results: Differential expression of RhoGDI2 protein in pancreatic cancer cell lines was identified. Gain-of-function and loss-of-function experiments showed that RhoGDI2 induced the malignant phenotypes of pancreatic cancer cells, including proliferation, migration, invasion, and gemcitabine (GEM) chemoresistance. The upregulation of RhoGDI2 stimulated the expression of Snail, resulting in the altered expression of epithelial marker E-cadherin and mesenchymal marker Vimentin, which were characteristics of the tumorigenic activity of epithelial-mesenchymal transition. The expression of RhoGDI2 and Snail was upregulated in clinical tumor samples, and higher expression of RhoGDI2 or Snail was significantly associated with poor patient survival in pancreatic ductal adenocarcinoma (PDAC). Conclusion: The findings indicated that RhoGDI2 promoted GEM resistance and tumor progression in pancreatic cancer and that RhoGDI2 might be a potential therapeutic target in patients with PDAC.

        Management strategy for Spodoptera frugiperda (Lepidoptera:Noctuidae) in mountain maize planting areas in China

        The fall armyworm, Spodoptera frugiperda (Lepidoptera: Noctuidae), is a major maize pest worldwide and has caused huge economic loss. To explore the control tactics of S. frugiperda in mountain maize planting fields, studies were performed in China from 2020 to 2021 for screening the S. frugiperda-resistant maize variety, seed coating agents, chemical insecticides, and biological agents. The results showed that 75 g/hm 2 Emamectin benzoate had better controlling efficacy (with a control efficacy of 91.05% at 1d) than other chemicals, so it was recommended in the high incidence period of pests. Compared with other seed coating treatments, 50% Lumi ngwei TM625FS (with a control efficacy of 76.19% at 10d) was the best seed coating against S. frugiperda. The biological control “Telenomus remus + Picromerus lewisi” and “T. remus + Eocanthecona furcellata” had good control efficacy on S. frugiperda with 76.01%, 64.10% at 20d, respectively. Additionally, there were lower numbers of S. frugiperda in maize variety GuiDan6, BaoYu9, and YanBaiYu8. In summary, this research rec ommended four effective pest management strategies (GuiDan6-resistant maize variety, 50% Lumingwei TM625FS-seed coating agents, 5% emamectin benzoate-chemical pesticide, and “T. remus + P. lewisi”-biological control) in mountain maize planting fields.         A Survey of the Geographic Distribution of Ophiocordyceps sinensis

        Ophiocordyceps sinensis is one of the best known fungi in Traditional Chinese Medicine. Many efforts have been devoted to locating the production areas of this species resulting in various reports; however, its geographic distribution remains incompletely understood. Distribution of O. sinensis at the county level is clarified in this work based on both a literature search and fieldwork. More than 3600 publications related to O. sinensis were investigated, including scientific papers, books, and online information. Herbarium specimens of O. sinensis and field collections made by this research group during the years 2000-2010 were examined to verify the distribution sites. A total of 203 localities for O. sinensis have been found, of which 106 are considered as confirmed distribution sites, 65 as possible distribution sites, 29 as excluded distribution sites and three as suspicious distribution sites. The results show that O. sinensis is confined to the Tibetan Plateau and its surrounding regions, including Tibet, Gansu, Qinghai, Sichuan, and Yunnan provinces in China and in certain areas of the southern flank of the Himalayas, in the countries of Bhutan, India and Nepal, with 3,000 m as the lowest altitude for the distribution. The fungus is distributed from the southernmost site in Yulong Naxi Autonomous County in northwestern Yunnan Province to the northernmost site in the Qilian Mountains in Qilian County, Qinghai Province, and from the east edge of the Tibetan Plateau in Wudu County, Gansu Province to the westernmost site in Uttarakhand, India. The clarification of the geographic distribution of O. sinensis will lay the foundation for conservation and sustainable use of the species.

      • Life-Style Habits in a High-Risk Area for Upper Gastrointestinal Cancers: a Population-Based Study from Shanxi, China

        Background: Cancer is a burden on humanity and ranks as a leading cause of morbidity and mortality in China. Shanxi province has its unique cancer patterns and the burden is increasing. In this study, we aimed to assess the pattern of dietary habits and life-style in Shanxi, a high-risk area for upper gastrointestinal cancers in China and further evaluate the trends in cancer incidence and mortality based on registered data. Materials and Methods: Data on lifestyle, diet, physical activity were obtained from the household health survey at Zhongyang from 2013 to 2015. Cancer diagnoses were reported to Shanxi Center for Disease Control and Prevention (SCDCP). Population-based cancer incidence data and mortality data of 2012 were collected from the SCDCP. All incidence and death rates were expressed per 100,000 populations. Univariate analysis was performed using the Chi-squared test or Fisher's exact test. Results: Overall, deficiencies in fresh fruits and vegetable food, and intake of hot food, salted food, or pickled food are serious problems in Shanxi, especially in rural areas. Upper gastrointestinal cancers were the most commonly diagnosed cancers, and the incidence in rural areas is higher than those in urban areas. Cervical cancer is the most common cancer for females. Moreover, the agespecific incidence exhibited an increased trend before 40 years old. Consistent with the previous literature, our epidemiological investigation results suggest that lifestyle, nutrition deficient, and infections were major risk factors for upper gastrointestinal cancers or cervical cancer in Shanxi. Facing a serious situation, we further explored defensible recommendations for the general public in order to promote changes in environments that support healthful eating and physical activity habits, to reduce cancer risk. Conclusions: Our results present the current cancer trends in Shanxi and its related etiologic risk factors and provide a theoretical basis to guide public health efforts to prevent and control cancers in the province.

      • Lentivirus-mediated shRNA Interference Targeting SLUG Inhibits Lung Cancer Growth and Metastasis

        Objective: Lung cancer is a deadly cancer, whose kills more people worldwide than any other malignancy. SLUG (SNAI2, Snail2) is involved in the epithelial mesenchymal transition in physiological and in pathological contexts and is implicated in the development and progression of lung cancer. Methods: We constructed a lentivirus vector with SLUG shRNA (LV-shSLUG). LV-shSLUG and a control lentivirus were infected into the non-small cell lung cancer cell A549 and real-time PCR, Western blot and IHC were applied to assess expression of the SLUG gene. Cell proliferation and migration were detected using MTT and clony formation methods. Results: Real-time PCR, Western Blot and IHC results confirmed down-regulation of SLUG expression by its shRNA by about 80%~90% at both the mRNA and protein levels. Knockdown of SLUG significantly suppressed lung cancer cell proliferation. Furthermore, knockdown of SLUG significantly inhibited lung cancer cell invasion and metastasis. Finally, knockdown of SLUG induced the down-regulation of Bcl-2 and up-regulation of E-cadherin. Conclusion: These results indicate that SLUG is a newly identified gene associated with lung cancer growth and metastasis. SLUG may serve as a new therapeutic target for the treatment of lung cancer in the future.

        인터넷 쇼핑이 새로운 유통 채널이 되었고, 왕성히 발전하기 때문에 연구할 가치가 있다. 이 논문은 인터넷 쇼핑의 편리성, 저렴한 가격, 만족도, 고객의 재구매 의도.가 연구되었다. 이 연구를 위하여 샘플링으로 240부 설문 조사지를 발부하였다. 그 다음에 spss20.0으로 연구 분석하였다. 연구 방법은 성과 분석, 신뢰도 분석, 공동 분석, 회귀 분석으로 인터넷 쇼핑의 편리성, 저렴한 가격, 만족도, 고객의 재구매 의도를 확인하였다. 이 연구의 중요성은 고객의 구매력의 영향을 미치는 4 가지 요소를 분석을 하는 것이다. 연구결과는 인터넷 쇼핑의 큰 잠재력을 보여주었다. 결론으로, 인터넷쇼핑이 고객만족의 구매의도에 많은 영향을 미치는 것으로 조사되었다. Internet shopping is worth studying since it has become a new distribution channel and its booming development. Through research described in this paper: the convenience of internet shopping, low price, pleasure and diversity impact to the customers`re-purchase intent. This study examined a random sample of 240 copies of questionnaires. And use spss20.0 research and analysis. By using the following study methods: result analysis, reliability analysis, mutual analysis and regression analysis, proved that, the convenience of internet shopping, low price, pleasure and diversity produced positive impact to the customers` re-purchase intent. The significance of this study is analyzing the impact the customers` purchasing power by the four factors of internet shopping motivation. And the result shows great potential of internet shopping. In conclusion of this study will contribute to growth in the network market, so that flourish the emerging distribution channels.

        Vp28 of Shrimp White Spot Syndrome Virus Is Involved in the Attachment and Penetration into Shrimp Cells

        White spot disease (WSD) is caused by the white spot syndrome virus (WSSV), which results in devastating losses to the shrimp farming industry around the world. However, the mechanism of virus entry and spread into the shrimp cells is unknown. A binding assay in vitro demonstrated VP28-EGFP (envelope protein VP28 fused with enhanced green fluorescence protein) binding to shrimp cells. This provides direct evidence that VP28-EGFP can bind to shrimp cells at pH 6.0 within OS It However, the protein was observed to enter the cytoplasm 3 h post-adsorption. Meanwhile, the plaque inhibition test showed that the polyclonal antibody against VP28 (a major envelope protein of WSSV) could neutralize the WSSV and block an infection with the virus. The result of competition EI,ISA further confirmed that the envelope protein VP28 could compete with WSSV to bind to shrimp cells. Overall, VP28 of the WSSV can bind to shrimp cells as an attachment protein, and can help the virus enter the cytoplasm.

        RON and MET Co-overexpression Are Significant Pathological Characteristics of Poor Survival and Therapeutic Targets of Tyrosine Kinase Inhibitors in Triple-Negative Breast Cancer

        Purpose Triple-negative breast cancer (TNBC) is highly malignant and has poor prognosis and a high mortality rate. The lack of effective therapy has spurred our investigation of new targets for treating this malignant cancer. Here, we identified RON (macrophage-stimulating 1 receptor) and MET (MET proto-oncogene, receptor tyrosine kinase) as a prognostic biomarker and therapeutic targets for potential TNBC treatment. Materials and Methods We analyzed RON and MET expression in 187 primary TNBC clinical samples with immunohistochemistry. We validated the targeted therapeutic effects of RON and MET in TNBC using three tyrosine kinase inhibitors (TKIs): BMS-777607, INCB28060, and tivantinib. The preclinical therapeutic efficacy of the TKIs was mainly estimated using a TNBC xenograft model. Results Patients with TNBC had widespread, abnormal expression of RON and MET. There was RON overexpression, MET overexpression, and RON and MET co-overexpression in 63 (33.7%), 63 (33.7%), and 43 cases (23.0%), respectively, which had poor prognosis and short survival. In vivo, the TKI targeting RON ant MET inhibited the activation of the downstream signaling molecules, inhibited TNBC cell migration and proliferation, and increased TNBC cell apoptosis; in the xenograft model, they significantly inhibited tumor growth and shrank tumor volumes. The TKI targeting RON and Met, such as BMS-777607 and tivantinib, yielded stronger anti-tumor effects than INCB28060. Conclusion RON and MET co-overexpression can be significant pathological characteristics in TNBC for poor prognosis. TKIs targeting RON and MET have stronger drug development potential for treating TNBC.

        Crosstalk evaluation in multiview autostereoscopic three-dimensional displays with an optimized diaphragm applied

        The crosstalk evaluation of multiview autostereoscopic three-dimensional (3D) displays is discussed, with both the human and technical factors investigated via image quality assessment. In the imaging performance measurements and analysis for a multiview autostereoscopic display prototype equipment, it was inferred that crosstalk would have both a positive and a negative effect on the imaging performance of the equipment. The importance of the attached diaphragm in the crosstalk evaluation was proposed and then experimentally verified, using the developed prototype equipment. The luminance distribution and crosstalk situation were given, with two different diaphragm arrays applied. The analysis results showed that the imaging performance of this 3D display system can be improved with minimum changes to the system structure.

        Semiglobal Robust Backstepping Output Tracking for Strict-feedback Form Systems with Nonlinear Uncertainty

        Output tracking controller design problem is dealt with for a class of nonlinear systems in strict-feedback form in the presence of time-varying nonlinear uncertainties and unmodeled dynamics with multi-operation points. A new method based on signal compensation is proposed to design a robust controller, which consists of a nominal controller and a robust compensator. It is shown that semiglobal robust tracking property can be achieved. A new feature of our results is that the controller is a linear and time-invariant one and “explosion of complexity” problem is avoided.
