        Manipulations of glucose/lipid metabolism and gut microbiota of resistant starch encapsulated Ganoderma lucidum spores in T2DM rats

        Our team previously demonstrated that Ganodermalucidum spores (GLS) and resistant starch (RS) hadhypoglycemic effects separately on type 2 diabetic mellitus(T2DM) rats. This work was to explore the effects ofadministering encapsulated GLS within RS (referred to asEGLS) in the T2DM rats, which were induced by streptozotocin(STZ). The EGLS was orally administered torats for 28 days. The parameters of glycometabolismand lipometabolism were evaluated, and fecal microbiotacomposition was investigated. The results showed thatEGLS significantly enhanced glycometabolism and lipometabolismparameters in T2DM rats, which might beassociate with the enhancement of the glucose and lipidmetabolism, insulin secretion, and glycogen synthesis andreduced lipogenesis. Furthermore, the intervention ofEGLS also reduced the Proteobacteria community andimproved dysfunctional gut microbiota. This study indicatedEGLS may be a potential candidate for dietaryintervention to modulate diabetes.

        Cell Membrane Hybrid Lipid Nanovesicles Enhance Innate Immunity for synergistic immunotherapy by promoting Immunogenic Cell Death and cGAS Activation

        Immunotherapy shows great therapeutic potential for long-term protection against tumor relapse and metastasis. Innate immune sensors, such as cyclic GMP-AMP synthase (cGAS) and stimulator of interferon genes (STING), dissolve DNA and induce type I interferon. Through activation of the cGAS/STING pathway, chemotherapy drugs and reversine (REV) may provide synergetic anti-tumor effects. Here, we prepared drug-loaded cell membrane hybrid lipid nanovesicles (LEVs) (designated LEV@DOX@REV) by fusion of cell membranes, phospholipids, doxorubicin (DOX), and REV, to realize accurate delivery to tumors and chemo-immunotherapy. The cell membranes of LEVs confer “homing” abilities. DOX can induce immunogenic cell death as a result of its specific immunomodulatory effects, which promotes the maturation of immune cells and improves the microenvironment of the immune system. REV is proven to efficiently activate cGAS/STING signaling, thereby enhancing the immune system. The antitumor efficacy of LEV@DOX@REV was evaluated in a 4T1 subcutaneous tumor xenograft model, a distant metastatic tumor model, and a liver metastatic tumor model. LEV@DOX@REV facilitated the infiltration of cytotoxic T lymphocytes within tumors, increased the secretion of proinflammatory cytokines, and modified the tumor microenvironment. In conclusion, LEV@DOX@REV displayed favorable antitumor effects and extended the survival of tumor-bearing mice. We therefore successfully developed nanoparticles capable of enhancing immune activation that have potential therapeutic applications for cancer immunotherapy.

      • Effect of Ligustilide Supplement on Physical Fitness of Athletes

        PURPOSE: In this study, high content ligustilide plant extract was supplemented to monitor the changes of physical fitness and physiological function of athletes, and to explore the effect of ligustilide plant extract on physical function and physical fitness of athletes. METHODS: Thirty five high-level basketball players from Zhengzhou University were selected and divided into two groups randomly: nutritional supplement group (S, n = 18) and control group (C, n=17). This study was approved by the Ethics Committee of Zhengzhou University. Informed consent was obtained from each subject. The intervention period was 8 weeks, six days per week. The training method adopted the small cycle plate training scheme. We measured changes of the scores of rating of perceived exertion (RPE), physical fitness test (15m running, 30m running, 400m running) and routine blood test (mainly including red blood cell count, hemoglobin concentration and average erythrocyte hemoglobin concentration, etc.) before and after training. RESULTS: In group S, the percentage of RPE (grade 6-11) and RPE (grade 12-18) before experiment was 37.5% and 62.5%. After 8 weeks of experiment, 62.5% of RPE (grade 6-11) and 37.5% of RPE (grade 12-18). In group C, 14% of RPE (grade 6-11), 86% of slight RPE (grade 12-18), and 100% of RPE (grade 12-18) after 8 weeks of experiment. It was found that after 8 weeks of training, the number of red blood cells, the concentration of hemoglobin and the average concentration of red blood cell hemoglobin in group S were significantly higher than those before the experiment (p <0.05). According to the comparison of the physical fitness test results of the athletes in group S, it was found that the performance of 400m after 4 weeks and 8 weeks was significantly higher compared with pre-experiment (p<0.05). The changes of 30m and 15m were not significant different (p>0.05). The 400-meter test performance of athletes in group C was significantly higher than that before the experiment only after 8 weeks of training (p<0.05). CONCLUSIONS: Ligustilide supplementation can increase the number of red blood cell and hemoglobin concentration of athletes and has a positive impact on the physical fitness of athletes and the recovery of fatigue.

        Pulsed-laser-diode Intermittently Pumped 2-μm Acousto-optic Q-switched Tm:LuAG Laser

        The heat distribution in crystals in a 2-µm acousto-optic Q-switched Tm:LuAG laser pumped by pulsed-laser-diode (pulsed-LD) intermittent-pumping technology was analyzed using COMSOL software. The thermal lensing effect of the Tm:LuAG crystal can be mitigated by pulsed-LD intermittent-pumping techniques. An experimental setup using this kind of approach achieved maximum output energy of 8.31 mJ, minimum pulse width of 101.9 ns, and highest peak power of 81.55 kW, reached at a Q-switched repetition rate of 200 Hz. It offers significant improvement of performance of the output laser beam, compared to pulsed-LD double-ended pumping technology at the same repetition rate.
