      • System Identification with Mixed Binary-valued and Precise Measurements

        This paper considers system identification based on mixed binary-valued and precise measurements with noises. The optimal Quasi-Convex Combination Estimator (QCCE) is constructed by combining the identification results of binary-valued and precise subsystems. The convergence properties are theoretically analysed in terms of strong consistency and asymptotical efficiency. Compared with the maximum likelihood (ML) estimator based on the whole system, the optimal QCCE has a smaller asymptotical variance that can achieve the cram´er-Rao (CR) lower bound. Extensive numerical simulations validate the superiority of the optimal QCCE.

        Mycobacterium vaccae induces a strong Th1 response that subsequently declines in C57BL/6 mice

        Mycobacterium (M.) vaccae is a fast-growing species of saprophytic bacteria that is widely distributed. To understand the host immuneresponses induced by M. vaccae isolated from bovine submaxillary lymph nodes, C57BL/6 mice were infected with reference strain M. bovisBacillus Calmette-Guérin (BCG) and isolated M. vaccae using intraperitoneal injections. Comparison of the bacterial replication and organpathology between M. vaccae and M. bovis BCG revealed that M. vaccae was more malignant than M. bovis in mice. We also demonstratedthat serum from the M. vaccae-infected mice contained a higher expression level of gamma-interferon (IFN-), tumor necrosis factor alpha,monocyte chemoattractant protein-1, interleukin (IL)-4, IL-12, IL-10 and transforming growth factor beta than did the other groups, especiallyafter week 4. Furthermore, when the numbers of CD3+CD4+IFN-+ and CD3+CD4+IL4+ cells in the infected mice were observed by flowcytometry, we found that a powerful T helper 1 (Th1) response was induced by M. vaccae infection, which was associated with the emergenceof CD3+CD4+IFN-+ cells. However, the Th1 response declined over time, which was associated with appearance of the CD4+CD25+FoxP3+and CD4+CD25+CD152+Treg cell reaction. In addition, a strong Th2 response was found. Finally, we found that M. vaccae infection increasedthe production of type I IFNs, which was associated with a reduced Th1 response.

      • The Investigation of Internal Coating for Natural Gas Pipeline

        Internal coating applied on natural gas pipeline not only can prevent the pipelines fiom internal corrosion, but also can greatly decrease the roughness of internal surface of the pipeline so that the gas flowing resistance would be reduced and the gas transmission efficiency could be improved largely. In order to meet with the requirement of western gas pipeline projects, a internal coating material special for reducing transmission resistance of gas pipeline has been developed by my Institute. A stimulating test equipment showthat transmission efficiency of pipe with special internal coating can be 15.3% more than that of steel pipe without coating.

        Analytical Solution of Loosening Pressure Model for Shallow Tunnel Based on Pile-beam-arch Method

        Loosening of earth pressure by tunnels is an important parameter for the structural design of subway stations, this study analyzes the difference between the loosening pressures of each span of a shallow multi-span tunnel station using the pile-beam-arch (PBA) method. By conducting a similarity model test of the PBA method, we simulate the complete excavation process. We study the sliding failure mode of the overlying soil of the shallow-buried station and the law of change of the loosening pressure of each span. A simplified calculation model for the loosening pressure of shallow buried multi-span tunnels by the cavity pile method was developed, and the applicability of the formulae and sensitivity analysis of the parameters were carried out. The results reveal that the loosening pressure of the mid-span is transferred to the stress of the side-span during tunnel excavation. This finding contradicts Terzaghi’s and Bierbaumer’s methods, which do not consider the influence of tunnel structure on the loosening pressure of the overlying soil. The side span relaxation pressure is only related to the soil capacity and the depth of burial of the tunnel This research can provide an important theoretical reference for the design of the overburden load and lateral confining pressure of the PBA structure design.

        Risk Assessment Mmodel of Karst Tunnel Flood Based on Distance Discriminant Weighting and Set Pair Cloud

        Flood has become the main hidden threat to the safety of the karst tunnel. Scientific and reasonable flood classification assessment and prevention measures have also become the main problems to be solved urgently in the construction and operation of tunnels. Aiming at the problems of multiple influencing factors, complex index systems, and linear processing of the connection degree of set pair analysis, a set pair cloud model based on cloud theory eigenvalues is proposed to optimize the connection degree of set pair analysis. Regarding the uncertainty of the evaluation system and the ambiguity of the evaluation grade boundary, firstly, the ridge-type membership function is used to determine the basic probability distribution of each measured index. Secondly, the distance discrimination weighting theory based on the index distance is also established, so that each measured index can assign the dynamic weight of the evaluation system. It can comprehensively determine the weight of the cloud connection degree for the system, and draw the cloud level diagram to determine the tunnel flood risk level. Finally, the proposed model is verified with tunnel flood. The results show that the evaluation results are consistent with other evaluation models and actual tunnel conditions. This model can provide guidance for flood prediction and prevention in karst tunnels.

        Immunomodulatory Properties of Lactobacillus plantarum NC8 Expressing an Anti-CD11c Single-Chain Fv Fragment

        The lactic acid bacteria species Lactobacillus plantarum (L. plantarum) has been used extensively for vaccine delivery. Considering to the critical role of dendritic cells in stimulating host immune response, in this study, we constructed a novel CD11c-targeting L. plantarum strain with surface-displayed variable fragments of anti-CD11c, single-chain antibody (scFv-CD11c). The newly designed L. plantarum strain, named 409-aCD11c, could adhere and invade more efficiently to bone marrow-derived DCs (BMDCs) in vitro due to the specific interaction between scFv-CD11c and CD11c located on the surface of BMDCs. After incubation with BMDCs, the 409-aCD11c strain harboring a eukaryotic vector pValac-GFP could lead to more efficient expression of GFP compared with wild-type strains shown by flow cytometry analysis, indicating the enhanced translocation of pValac-GFP from L. plantarum to BMDCs. Similar results were also observed in an in vivo study, which showed that oral administration resulted in efficient expression of GFP in both Peyer’s patches (PP) and mesenteric lymph nodes (MLNs) within 7 days after the last administration. In addition, the CD11c-targeting strain significantly promoted the differentiation and maturation of DCs, the differentiation of IL-4+ and IL-17A+ T helper (Th) cells in MLNs, as well as production of B220+ IgA+ B cells in the PP. In conclusion, this study developed a novel DC-targeting L. plantarum strain which could increase the ability to deliver eukaryotic expression plasmid to host cells, indicating a promising approach for vaccine study.

        Influence of δ' Phase with GP‑I Zones Fillings on Slip Behavior and Cold Rolling Texture in AA2099

        The cold rolling texture of AA2099 sheets was strongly influenced by the prior heat treatment. Two typical heat treatments,including solid solution (T4) and underaging (T6) were utilized to control slip behavior by different precipitates during coldrolling. A much weaker texture with an orientation scatter towards higher Φ angles was found in the as-rolled T6 specimen(T6R). Slip behavior was analyzed using geometry necessary boundaries distribution. Results show that the T6R specimenexperienced a strong co-planar slip compared to the as-rolled T4 specimen. Prosperity of co-planar slips and limitation ofco-direction slips were main reasons for the weakening of Copper texture component in the T6R specimen. The distinguishslip behavior was caused by existence of δ' phase with GP-I zones fillings in the T6 specimen. This phase could be easilysheared by dislocations and then promoted co-planar slips in further deformation.

        Challenges for QTL Analysis in Crops

        Quantitative trait loci, a genetic concept introduced in the 1940s, for explaining the inheritance of non-Mendelian traits, have been realized as particular fragments of chromosomes even unique genes in most crops in the 21st century. However, only very a small portion of QTL has been screened out by geneticists comparing to a great number of genes underneath the quantitative traits. These identified QTL even have been seldom used into breeding program because crop breeders may not find the QTL in their breeding populations in their field station. Several key points will be proposed to meet the challenges of QTL analysis today: a fine mapping population and the related reference genetic map, QTL evaluation in multiple environments, recognizing real QTL with small genetic effect, map integration.
