        Cytological, genetic, and proteomic analysis of a sesame (Sesamum indicum L.) mutant Siyl‑1 with yellow–green leaf color

        Background Both photosynthetic pigments and chloroplasts in plant leaf cells play an important role in deciding on the photosynthetic capacity and efficiency in plants. Systematical investigating the regulatory mechanism of chloroplast development and chlorophyll (Chl) content variation is necessary for clarifying the photosynthesis mechanism for crops. Objective This study aims to explore the critical regulatory mechanism of leaf color mutation in a yellow–green leaf sesame mutant Siyl-1. Methods We performed the genetic analysis of the yellow-green leaf color mutation using the F2 population of the mutant Siyl-1. We compared the morphological structure of the chloroplasts, chlorophyll content of the three genotypes of the mutant F2 progeny. We performed the two-dimensional gel electrophoresis (2-DE) and compared the protein expression variation between the mutant progeny and the wild type. Results Genetic analysis indicated that there were 3 phenotypes of the F2 population of the mutant Siyl-1, i.e., YY type with light-yellow leaf color (lethal); Yy type with yellow-green leaf color, and yy type with normal green leaf color. The yellowgreen mutation was controlled by an incompletely dominant nuclear gene, Siyl-1. Compared with the wild genotype, the chloroplast number and the morphological structure in YY and Yy mutant lines varied evidently. The chlorophyll content also significantly decreased (P < 0.05). The 2-DE comparison showed that there were 98 differentially expressed proteins (DEPs) among YY, Yy, and yy lines. All the 98 DEPs were classified into 5 functional groups. Of which 82.7% DEPs proteins belonged to the photosynthesis and energy metabolism group. Conclusion The results revealed the genetic character of yellow-green leaf color mutant Siyl-1. 98 DEPs were found in YY and Yy mutant compared with the wild genotype. The regulation pathway related with the yellow leaf trait mutation in sesame was analyzed for the first time. The findings supplied the basic theoretical and gene basis for leaf color and chloroplast development mechanism in sesame.

        From the results of this research as to the exemplary model of Un-Yong Kim's question of sports and power deviation were following conclusions by investigating the real identification of Un-Yong Kim as a person who is regarded as a self-made man in the world sport industry, and by coping with those problems in the P.E. and sport coming up right after his retirement and the problem of how we can turn out sport leaders who can be spokesmen for Korea P.E. and sports and are capable and competitive in the world. 1. For a short-term period, it is necessary that the Ministry of Culture & Tourism should form a sport human resource pool for international committees in the Korea Olympic Committee as a short-term measure and that it should devise a mid and long term measure to take a proper step for training sport professionals for the diplomatic service and thus that a sufficient government budget is required for all these projects. 2. Through its influence and the mass media, the national government needs to support Korea sport leaders and athletes who are presently working in foreign countries, and also it should establish an extended policy for supporting civil sector sport exchanges with overseas countries. 3. It is necessary to improve the disposition of leaders for the image reform in the sport industry of the country, and the people themselves in the P.E. and sport need to secure the capability of attaining the influential power for the systemic and bureaucratic movements. 4. The national government and responsible institutions related with the P.E. and sport are to collect public opinions in a nation-broad dimension. 5. The continuance of authoritarianism regimes in our society have reinforced authoritative implicitness assertively for public interest, and it is true that it caused a tendency of regarding even the public good and interests as antagonism and suppression against the society. Accordingly, it is necessary to rediscovering the supplementary relation of personal interest with that of the public, and to seek for models that can produce moral leaders in the P.E. and sport, pursuing the ideals for the public and the personal harmoniously.

        남성중심의 경쟁적 직장문화에서 여성의 프로페셔널 리더십 : 광고인 최인아에 대한 사례연구

        본 연구는 남성중심의 직장문화를 극복하고 고위직 전문가로 진출한 여성 광고인의 사례연구를 통해 프로레셔널 리더십의 특서을 귀납적으로 규명하였다. 연구 대상은 공채출신 여성으로는 최초로 S그룹 임원으로 승진한 광고인 최인아이다. 최인아는 '남성중심의 직장문화'에서 합리적인 행동과 수범(垂範)사례를 통해 선례를 구축하고 직장문화를 변화시키는 단계별 전략을 취하였다. 그리고 프로로서 '성과로 승부'하겠다는 장기 전략에 입각하여 목표를 달성하기 위해 끊임없이 노력하였다. 이 과정에서 최인아는 부하들과 성과를 중시하는 과업 지향적인 행도을 취하면서도, 대화와 설득을 통해 신뢰관계를 형성하는 비권위주의적인 행동을 보이고 있다. 부하들과의 관계와 조직행동을 종합적으로 분석해 볼 때 최인아의 리더십은 성과를 최우선으로 하는 여성적 프로페셔널리즘으로 평가할 수 있다. This research is designed to draw a certain special type of leadership through the case study with inductive reasoning on the female worker who successfully overcame the male-oriented corporate culture and advanced into the high level of career position. The respondent of this research is In-A Choi, Executive Director, who has advanced into the first-ever female executive at so young age of 40 years as a publicly selected worker in the S Corporate. As a research turns out, In-A Choi has taken a gradational strategy to change the corporate culture by conducting reasonable ways and setting example cases by herself in such a 'Male-oriented corporate culture'. She has also shwoed the systematic behavior in which she has set up the long-term plan of being appreciated by her accomplishment as a professional. In the process of carrying out her own strategy, she has taken the horizontal and open-hearted actions which has been task-oriented and reflected the female traits. Moreover, her relationship with subordinates or systematic abhavior has stemmed from the 'Professionalsim' based on the preferential criteria of the accomplishment.

        1) Parallel operation : The parallel operation having a different capacity brings about a head loss due to the reverse flow of the unbalance in pressure between two pumps at some condition. So it should be operated in the careful consideration of frictional head loss and pump's head capacity to operate in parallel operation. But it can be obtained from this study that the capacity of small pump is equal to that of large pump and the performance of parallel operation is Tmproved. And therefore, it could be otained the optimum operation at Case Ⅶ (N_L = 2,000, N_s = 000) and Case Ⅷ(N_L = 2,000, N_s = 2,500) in parallel operation. 2) Series operation : The series operation having a different capacity brings about a head loss at some condition due to the flow limit of small pump. And it can be obtained from this study that the capacity of small pump is approachaed to large pump and the performance of series operation is Tmproved. And therefore it can be obtained the optimum operation at Cast I (N_L =3,000, N_s = 3,000), Case Ⅳ (N_L = 2,500, N_s = 3,000), Case Ⅶ(N_L = 2,000, N_s = 3,000) in series operation.

      • LDV를 이용한 저속 원심회전차내 유동 속도 측정에 관한 연구

        To understand the flow phenomena within a channel of impeller and estimate the performance of compressor or pump, the flow velocity was measured by LDV with the advantages of non-intrusion in a flow field. The object of this study is to present a measuring method of flow velocity by LDV and to improve the measuring techniques. In order to estimate the characteristics of flow field within a channel, the flow velocity has been measured with respect to variation of blade angle of exit and to increase the S/N ratio of LDV, the half angle of lens has been increased and the hub wall coated with the soot to absorb the diffused reflection light. For the blade angles of exit, flow losses and deviation angle were small at 54° and large at 122°, and energy transfer coefficient had a good performance at 90°.

        The distribution of pressure in an impeller of a centrifugal pump was investigated to operate under the optimum, conditions according to the variation of dynamic factors such as rotational speed and flowrate in an impeller. The interchange of energy actually are as follows. 1) The variations of pressure in the radial direction had almost the same patterns as the flowrate was changed and the outlet pressure decreased with the increase of the flowrate. 2) It showed that the pressure increased in the radial direction of impeller as the rotational speed increased, and the same phenomenon took place with the variation of flowrate. Otherwise there was little recovery of pressure in the region of low rotational speed and high flowrates(5.0, 5.5 lps) . 3) The head-flowrate of the characteristic curves increased simultaneously according to the increase of rotational speed, and the power gradient of the power-flowrate curve increased as the rotational speed increased and the efficiency-flowrate curve showed that the efficiency was the same in a low flowrate and higher in a high flowrate according to the increase of rotational speed.

        The Ⅰ-Ⅴ characteristics of chalcogenide glass switches(As_3 Ge_1 Te_6 system) are measured withthe multimode pulse technique. The poole Frenkel effect is observed in the preswitching region except in weak field. The activation energy of the switch is 0.36eV. The temparature rise in the conducting channel just before switching is estimated below several degrees in Celsius. Switching parameters such as threshold conditions, minimum holding conditions, and the On state characteristics are investigated with the programmed mode. Negative dynamic resistance is observed in the on state near the minimum holding region. When two successive pulses of which hight of the second pulse is lower than the first one, slow speed transition from the on state to the off state is observed. The influences of the on state on the second switching are as follows; (1)when the time gap between the first pulse and the second one is narrowed within 2 micro seconds, the second threshold voltage decreases remarkably, (2) When the duration of the on state is prolonged, the next threshold voltage increases in some region, (3) slow speed transition from the on state to the off state makes next threshold voltage increase and suppress the next firing.

        Bacterial Community Structure and Function Shift in Rhizosphere Soil of Tobacco Plants Infected by Meloidogyne incognita

        Root-knot nematode disease is a widespread and catastrophic disease of tobacco. However, little is known about the relationship between rhizosphere bacterial community and root-knot nematode disease. This study used 16S rRNA gene sequencing and PICRUSt to assess bacterial community structure and function changes in rhizosphere soil from Meloidogyne incognita-infected tobacco plants. We studied the rhizosphere bacterial community structure of M. incognita-infected and uninfected tobacco plants through a paired comparison design in two regions of tobacco planting area, Yuxi and Jiuxiang of Yunnan Province, southwest China. According to the findings, M. incognita infection can alter the bacterial population in the soil. Uninfested soil has more operational taxonomic unit numbers and richness than infested soil. Principal Coordinate Analysis revealed clear separations between bacterial communities from infested and uninfested soil, indicating that different infection conditions resulted in significantly different bacterial community structures in soils. Firmicutes was prevalent in infested soil, but Chloroflexi and Acidobacteria were prevalent in uninfested soil. Sphingomonas, Streptomyces, and Bradyrhizobium were the dominant bacteria genera, and their abundance were higher in infested soil. By PICRUSt analysis, some metabolism-related functions and signal transduction functions of the rhizosphere bacterial community in the M. incognita infection-tobacco plants had a higher relative abundance than those uninfected. As a result, rhizosphere soils from tobacco plants infected with M. incognita showed considerable bacterial community structure and function alterations.
