      • Prognostic Role of Circulating Tumor Cells in Patients with Pancreatic Cancer: a Meta-analysis

        Background: Isolation and characterization of circulating tumor cells (CTCs) in patients suffering from a variety of different cancers have become hot biomarker topics. In this study, we evaluated the prognostic value of CTCs in pancreatic cancer. Materials and Methods: Initial literature was identified using Medline and EMBASE. The primary data were hazard ratios (HRs) with 95% confidence intervals (CIs) of survival outcomes, including overall survival (OS) and progression free survival/recurrence free survival (PFS/RFS). Results: A total of 9 eligible studies were included in this meta-analysis, published between 2002 and 2013. The estimated pooled HR and 95%CI for OS for all studies was 1.64 (95%CI 1.39-1.94, p<0.00001) and the pooled HR and 95%CI for RFS/DFS was 2.36 (95%CI 1.41-3.96, p<0.00001). The HRs and 95%CIs for OS and RFS/DFS in patients before treatment were 1.93 (95%CI 1.26-2.96, p=0.003) and 1.82 (95%CI 1.22-2.72, p=0.003), respectively. In patients receiving treatment, the HRs and 95%CI for OS and RFS/DFS were 1.37 (95%CI 1.00-1.86, p=0.05) and 1.89 (95%CI 1.01-3.51, p=0.05), respectively. Moreover, the pooled HR and 95%CI for OS in the post-treatment group was 2.20 (95%CI 0.80-6.02, p=0.13) and the pooled HR for RFS/DFS was 8.36 (95%CI 3.22-21.67, p<0.0001). Conclusions: The meta-analysis provided strong evidence supporting the proposition that CTCs detected in peripheral blood have a fine predictive role in pancreatic patients especially on the time point of post-treatment.

        Bayesian forecasting approach for structure response prediction and load effect separation of a revolving auditorium

        A Bayesian dynamic linear model (BDLM) is presented for a data-driven analysis for response prediction and load effect separation of a revolving auditorium structure, where the main loads are self-weight and dead loads, temperature load, and audience load. Analyses are carried out based on the long-term monitoring data for static strains on several key members of the structure. Three improvements are introduced to the ordinary regression BDLM, which are a classificatory regression term to address the temporary audience load effect, improved inference for the variance of observation noise to be updated continuously, and component discount factors for effective load effect separation. The effects of those improvements are evaluated regarding the root mean square errors, standard deviations, and 95% confidence intervals of the predictions. Bayes factors are used for evaluating the probability distributions of the predictions, which are essential to structural condition assessments, such as outlier identification and reliability analysis. The performance of the present BDLM has been successfully verified based on the simulated data and the real data obtained from the structural health monitoring system installed on the revolving structure.

      • FNC, a Novel Nucleoside Analogue, Blocks Invasion of Aggressive Non-Hodgkin Lymphoma Cell Lines Via Inhibition of the Wnt/β-Catenin Signaling Pathway

        Chemotherapy is the primary therapy for malignant lymphoma (ML). However, the clinical outcome is still far from satisfactory. Consequently, an understanding of the mechanism of modulating cancer cell invasion, migration and metastasis is important for the development of more effective chemotherapeutic agents. FNC, 2'-deoxy-2'-${\beta}$-fluoro-4'-azidocytidine, a novel cytidine analogue, has demonstrated significantly inhibitory effects on proliferation of several non-Hodgkin lymphoma (NHL) cell lines. A previous study indicated that FNC effectively inhibited the growth of Raji and JeKo-1 cells in dose-time dependent effects with $IC_{50}$ values of $0.2{\mu}M$ and $0.097{\mu}M$, respectively. This study was focused on investigating the anti-invasive properties of FNC on NHL cells and its potential mechanisms of action. Cell adhesion and transwell chamber assays were utilized to investigate the anti-invasive effects of FNC on Raji and JeKo-1 cells. Real-time PCR and Western blotting were employed to qualify the expression of ${\beta}$-catenin, the glycogen synthase kinase-3 beta (GSK-$3{\beta}$), E-cadherin vascular endothelial growth factor (VEGF), matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9). The results revealed that FNC remarkably inhibited the adhesion, migration and invasion of two human aggressive non-Hodgkin lymphoma cell lines in a dose dependent manner. Furthermore, ${\beta}$-catenin, MMP-2, MMP-9, VEGF mRNA and protein levels were decreased after FNC treatment, while GSK-$3{\beta}$ and E-cadherin increased. Our studies thus provide evidence and a rationale that FNC may offer an effective chemotherapeutic agent by regulating the invasion and metastasis of aggressive non-Hodgkin lymphoma via inhibition of the Wnt/${\beta}$-catenin signaling pathway.

      • Combined Expression of Metastasis Related Markers Naa10p, SNCG and PRL-3 and its Prognostic Value in Breast Cancer Patients

        Combinations of multiple biomarkers representing distinct aspects of metastasis may have better prognostic value for breast cancer patients, especially those in late stages. In this study, we evaluated the protein levels of N-${\alpha}$-acetyltransferase 10 protein (Naa10p), synuclein-${\gamma}$ (SNCG), and phosphatase of regenerating liver-3 (PRL-3) in 365 patients with breast cancer by immunohistochemistry. Distinct prognostic subgroups of breast cancer were identified by combination of the three biomarkers. The Naa10p+SNCG-PRL-3-subgroup showed best prognosis with a median distant metastasis-free survival (DMFS) of 140 months, while the Naa10p-SNCG+PRL-3+subgroup had the worst prognosis with a median DMFS of 60.5 months. Multivariate analysis indicated Naa10p, SNCG, PRL-3, and the TNM classification were all independent prognostic factors for both DMFS and overall survival (OS). The three biomarker combination of Naa10p, SNCG and PRL-3 performed better in patients with lymph node metastasis, especially those with more advanced tumors than other subgroups. In conclusion, the combined expression profile of Naa10p, SNCG and PRL-3, alone or in combination with the TNM classification system, may provide a precise estimate of prognosis of breast cancer patients.

        Effects of Hydrogen Peroxide Accumulation, Lipid Peroxidation, and Polyphenol Oxidation during Superficial Scald Development in ‘Fuji’ Apples

        Hydrogen peroxide (H2O2) and malondialdehyde (MDA) levels and polyphenol oxidase (PPO) activity weremeasured in ‘Fuji’ apples with different susceptibilities to the development of superficial scald during storage at 0and 20°C. Both scald incidence and severity increased gradually in the cold and the symptoms became apparent inthe fruits with an early, middle, and late harvest after 12, 15, and 21 weeks storage, respectively. Early harvestfruits had the highest scald incidence and severity as well as H2O2 and MDA contents, while late harvest had thelowest. The H2O2 and MDA levels increased, while PPO activity remained constant during storage at 0°C. All fruitsshowed a rapid increase in scald incidence and severity after warming, especially at the first 4 days, when H2O2 andMDA levels and PPO activities also increased. The H2O2 and MDA levels in scald-free tissues in late harvest fruitswere lower initially but became higher in the scald tissues after warming for 4 days. The PPO activities were equalin both tissues. These results indicated that mature fruits were relatively resistant to scald, and that scald symptomdevelopment was highly dependent on the accumulations of H2O2 and MDA during cold storage, but PPO activitywas less important.

        The Analysis and Comparison of the Needle-puncture Property of Four Types of Silkworm Cocoons

        Silkworm cocoons display outstanding properties in many aspects, such as thermal protection, biocompatibilityand mechanical resistance and so on. In this paper, we focus on the puncture properties without detailed exploration of fourtypes of cocoons (three domesticated cocoon and Eri silkworm). Different sections, end and middle, as well as differentdirections, puncturing from inner and outer surface, of four types of aforementioned cocoons were systematically analyzedand compared. The results of present research show that the middle section exhibits a relatively greater needle-punctureresistance compare with end section for three B. mori silkworm cocoons. With regard to different puncture directions, fromexternal and internal surface, puncturing from internal surface exhibits a relatively higher needle-puncture load as well asfracture displacement tendencies compared with needle-puncture performance from external surface no matter in middle orend sections. Combining physical parameter assessment, statistical analysis and needle-puncture performance, this researchinspires a clear-cut frame to design a composite with high puncture resistance.

      • Prognostic Value of MGMT Promoter Methylation and TP53 Mutation in Glioblastomas Depends on IDH1 Mutation

        Several molecular markers have been proposed as predictors of outcome in patients with glioblastomas. We investigated the prognostic significance of $O^6$-methylguanine-DNA methyltransferase (MGMT) promoter methylation and TP53 mutation status dependent on isocitrate dehydrogenase 1 (IDH1) mutation in glioblastoma patients. A cohort of 78 patients with histologically confirmed glioblastomas treated with radiation therapy and chemotherapy were reviewed retrospectively. We evaluated the prognostic value of MGMT promoter methylation and TP53 mutation status with regard to progression-free survival (PFS) and overall survival (OS). It was revealed that mutations in IDH1, promoter methylation of MGMT, TP53 mutation, age, Karnofsky performance status (KFS), and extension of resection were independent prognostic factors. In patients with an IDH1 mutation, those with an MGMT methylation were associated with longer PFS (p=0.016) and OS (p=0.013). Nevertheless, the presence of TP53 mutation could stratify the PFS and OS of patients with IDH1 wild type (p=0.003 and 0.029 respectively, log-rank). The MGMT promoter methylation and TP53 mutation were associated with a favorable outcome of patients with and without mutant IDH1, respectively. The results indicate that glioblastomas with MGMT methylation or TP53 mutations have improved survival that may be influenced by IDH1 mutation status.

      • Association Between MDM2 SNP309 T>G and Risk of Gastric Cancer: A Meta-analysis

        Background: As a negative regulator of P53, MDM2 plays an important role in carcinogenesis; a polymorphism in its promoter region. SNP309 T>G, is known to increase the expression of MDM2, thus being considered related to higher susceptibility to neoplasia. However, no agreement has been achieved regarding its effects on gastric cancer. Methods: The present systematic meta-analysis was performed based on comprehensive literature search from Pubmed, Web of science and CBM databases. Results: It was suggested from 6 independent studies that the GG genotype is associated with a significantly increased risk of gastric cancer (Recessive: OR = 1.43, 95% CI = 1.08-1.91, P = 0.013), and subgroup analysis also confirmed the relationship (English publications-recessive model: OR = 1.45, 95% CI = 1.10-1.91, P = 0.009; Studies in China-recessive model: OR = 1.58, 95% CI = 1.08-2.30, P = 0.017). No publication bias was detected. Conclusion: The meta-analysis indicated a significant inverse association between GG genotype carriage and elevated risk of gastric cancer. However, more studies and detailed information are needed to fully address the topic.

      • Relative Risk of Metabolic Syndrome in Middle Aged Adults with Different Weight Living in Urban Beijing, China

        The purpose of this study is to compare the relative risk of metabolic syndrome (MS) in middle aged adults with different body weights. 155 subjects living in urban Beijing were recruited from 24 neighborhood committees of urban Beijing. They were divided into normal weight, overweight and obese groups according to their BMIs. The general information of the subjects was collected using an interview-administered questionnaire. Standard procedure was followed to measure subject's weight, height and waist. Biochemical parameters (total cholesterol (TC), low- and high­density lipoprotein cholesterol (LDL-C ; HDL-C), triglyceride (TG), and fasting glucose) and blood pressure were also determined. The results indicated that the systolic and diastolic blood pressure, HDL-C of obese group was lower than that of the normal weight group. Fasting glucose of obese males was significantly higher than that of normal weight males. No significant difference of fasting glucose was found among female groups. No significant difference of TG was found among male groups, while TG of overweight and obese females was both significantly higher than normal weight females. There was no significant difference of TC and LDL-C among normal weight, overweight and obese groups in both males and females. The MS rate of obese males was significantly higher than the normal weight and overweight males, as was the female. The relative risk of MS in obese group was about 11 times higher (OR=11.249, $95\%CI$ = 3.812 - 33.191) than the normal weight group after adjusting for age, gender, smoking, drinking, family economic level and education status. It is concluded that obesity contributed to lower HDL-C, hypertriglyceride, hypertension and MS after controlling the effects of age, gender, socioeconomic status, alcohol drinking and smoking. Obese individuals have a higher risk of having MS than their normal weight counterparts.

        Comparative transcriptome analysis of ATP-binding cassette (ABC) transporter genes in eri-silkworm, Samia cynthia ricini in response to 1- deoxynojirimycin

        1-Deoxynojirimycin (DNJ) as a kind of alkaloid has been confirmed that could modulate glycometabolism andhas toxicity for the eri-silkworm in our previous research. On the contrary, what is the potentially defensivemechanism when the DNJ enters the eri-silkworm. Based on comparative transcriptome sequencing, we foundthat ATP-binding cassette (ABC) transporter genes could be induced significantly by DNJ. In this study, a total of16 putative ABC transporter genes were identified, which can be classified into seven subfamilies, namely oneABCA, four ABCBs, three ABCCs, two ABCDs, one ABCE, three ABCFs, and two ABCGs. Phylogenetic analysisrevealed that ScABCs had strong conservation with Bombyx mori. Reverse transcription quantitative PCR (RTqPCR)suggested that 6 ABC transporters had a strong positive correlation between RT-qPCR and transcriptomedata. Additionally, S. c. ricini ABC transporter C-subfamily 4 (ScABCC4), S. c. ricini ABC transporter G-subfamily4 (ScABCG4), S. c. ricini ABC transporter A-subfamily 3 (ScABCA3) and S. c. ricini ABC transporter C-subfamily10 (ScABCC10) showed different expression pattern in two feed dose (1% and 2% DNJ) and three time points(6h, 12 h, 48 h). This study provides the first study on identification, characterization and expression patterns ofABC transporter genes in S. c. ricini response to DNJ, and lays a foundation for further understanding of theirphysiological roles response to the alkaloid.
