Recently, poly-γ-glutamic acid synthetase A (pgsA) has been applied to display exogenous proteins on the surface of Lactobacillus casei or Lactococcus lactis, which results in a surfacedisplayed component of bacteria. However, the ability of carrying genes encoded by plasmids and the expression efficiency of recombinant bacteria can be somewhat affected by the longer gene length of pgsA (1,143 bp); therefore, a truncated gene, pgsA, was generated based on the characteristics of pgsA by computational analysis. Using murine IL-10 as an exogenous gene, recombinant Lactobacillus plantarum was constructed and the capacity of the surface-displayed protein and functional differences between exogenous proteins expressed by these strains were evaluated. Surface expression of IL-10 on both recombinant bacteria with anchorins and the higher expression levels in L. plantarum-pgsA’-IL-10 were confirmed by western blot assay. Most importantly, up-regulation of IL-1β, IL-6, TNF-α, IFN-γ, and the nuclear transcription factor NF-κB p65 in RAW264.7 cells after stimulation with Poly(I:C) or LPS was exacerbated after co-culture with L. plantarum-pgsA. By contrast, IL-10 expressed by these recombinant strains could reduce these factors, and the expression of these factors was associated with recombinant strains that expressed anchorin (especially in L. plantarum-pgsA’-IL-10) and was significantly lower compared with the anchorin-free strains. These findings indicated that exogenous proteins could be successfully displayed on the surface of L. plantarum by pgsA or pgsA’, and the expression of recombinant bacteria with pgsA’ was superior compared with bacteria with pgsA.
Structure Analysis and Laxative Effects of Oligosaccharides Isolated from Bananas

Banana oligosaccharides (BOS) were extracted with water, and then separated and purified using column chromatography. Gel penetration chromatography was used to determine the molecular weights. Thin layer chromatogram and capillary electrophoresis were employed to analyze the monosaccharide composition. The indican bond and structure of the BOS molecule were determined using Fourier transform infrared spectroscopy and nuclear magnetic resonance. Results showed that BOS were probably composed of eight β-D-pyran glucose units linked with 1→6 indican bonds. The laxative effects of BOS were investigated in mice using the method described in “Handbook of Technical Standards for Testing and Assessment of Health Food in China.” The length of the small intestine over which a carbon suspension solution advanced in mice treated with low-, middle-, and high-dose BOS was significantly greater than that in the model group, suggesting that BOS are effective in accelerating the movement of the small intestine.
Geographic Distribution and Epidemiology of Lung Cancer During 2011 in Zhejiang Province of China

Background: To explore etiology for providing scientific clues for the prevention of lung cancer. Materials and Methods: Data for lung cancer incidence and meteorological geographic factors from 25 counties in Zhejiang province of China during 2011 were studied. Stepwise multiple regression and correlation analysis were performed to analyze the geographic distribution and epidemiology of lung cancer. Results: 8,291 new cases (5,998 in males and 2,293 females) of lung cancer during 2011 in Zhejiang province were reported in the 25 studied counties. Reported and standardized incidence rates for lung cancer were 58.0 and 47.0 per 100,000 population, respectively. The incidence of lung cancer increased with age. Geographic distribution analysis shows that the standardized incidence rates of lung cancer in northeastern Zhejiang province were higher than in the southwestern part, such as in Nanhu, Fuyang, Wuxing and Yuyao counties, where the rates were more than 50 per 100,000 population. In the southwestern Zhejiang province, for instance, in Yueqing, Xianju and Jiande counties, the standardized incidence rates of lung cancer were lower than 37 per 100,000 population. Spearman correlation tests showed that forest coverage rate, air quality index (AQI), and annual precipitation level are associated with the incidence of lung cancer. Conclusions: Lung cancer in Zhejiang province shows obvious regional differences. High incidence appears associated with low forest coverage rate, poor air quality and low annual precipitation. Therefore, increasing the forest coverage rate and controlling air pollution may play an important role in lung cancer prevention.

Novel small-molecule polysaccharide fractions,named POP II and POP III, were purified from Portulacaoleracea L. with average Mw values of 9.25 and 8.03 kDa,respectively. Monosaccharide analysis revealed that POP IIwas composed of Rha, Ara, Man, Glc, and Gal with molarratios of 1: 1.42: 0.44: 0.88: 1.59. POP III was composedof Rha, Ara, Glc, and Gal with molar ratios of 1: 1.16:0.23: 0.59. The antioxidant activities of the fractions wereevaluated using cell-free and cell-mediated radical generatingsystems. POPII and POP III possessed strong antioxidantactivities in both systems. The 2 novel polysaccharidefractions extracted from P. oleracea L. can be developed asnatural antioxidants for treatment of free radical-relateddiseases.

The aim of this study is to evaluate the antiinflammatory and protective eff ects of L -theanine in infl ammatorybowel disease (IBD) and to identify the underlyingmolecular mechanisms. Rats were pre-treated with L -theanineat 0, 50, 200, or 800 mg/kg/day. IBD was induced inrats using dextran sulfate sodium (DSS). Histopathologicalanalysis suggests that L -theanine can suppress DSS-inducedIBD with signifi cant inhibition of infl ammation in large andsmall intestinal tissues. Moreover, the 200 mg/kg/day L -theanine-treated DSS group had higher body and small intestineweights, a lower disease activity index and expression ofinfl ammatory factors than the DSS group without pre-treatment. In RNA sequencing and tandem mass tag labelinganalyses, large number of mRNAs and proteins expressionlevel diff ered when compared with the DSS-induced ratswith and without 200 mg/kg/day L -theanine pre-treatment. Moreover, Kyoto Encyclopedia of Genes and Genomes pathwayanalysis indicates the anti-infl ammatory activities ofL -theanine in DSS-induced IBD, with a high representationof genes in “Cholesterol metabolism” and “Retinol metabolism”pathways. Analysis of protein–protein interaction networksfurther indicates the involvement of these two pathways. These studies suggest that medium-dose L -theaninepre-treatment could ameliorate DSS-induced IBD throughmolecular mechanisms involving cholesterol and retinolmetabolism.

Aims: The aim of this study was to investigate the effects of adjuvant chemotherapy cycles on the prognosis of patients with post-operative stomach cancer through retrospective analysis. Methods: A total of 128 patients with gastric cancer who underwent gastrectomy, followed by adjuvant chemotherapy consisting of epirubicin, cisplatin or oxaliplatin, leucovorin, and 5-fluorouracil, according to a defined schedule, were divided into three groups according to the number of chemotherapy cycles: Group I (<6 cycles); Group II (6 cycles); and Group III (>6 cycles). Results: The 5-year overall survival (OS) was 20.8% in Group I, 45.0% in Group II, and 42.9% in Group III, with a median follow-up of 43 months. The 5-year relapse-free survival (RFS) was 15.1% in Group I, 40% in Group II, and 40% in Group III. The OS and RFS in Groups II and III were significantly better than in Group I (OS, p = 0.002 and p=0.003; RFS, P<0.001 and P=0.002). There was no difference in OS (p = 0.970) or in RFS (p = 0.722) between Groups II and III. Multivariate Cox hazard analysis determined that the number of adjuvant chemotherapy cycles was an independent factor that influenced OS and RFS. Conclusion: Six cycles of adjuvant chemotherapy gave encouraging outcomes in patients with resectable gastric cancer. Further prospective randomized controlled investigations are warranted in a multi-center setting.
