Adding ring-stiff eners is an eff ective strategy for strengthening CHS tubular joints. However, research on the capacity of ring-stiff ened CHS tube-gusset joints remains limited. In this study, we performed experimental and numerical analyses to derive static strength equations for this joint. For convenience, we simplifi ed the complex joint model into a ring plate to CHS joint, which consisted of a CHS tube and a ring-stiff ener. First, we tested fi ve specimens to obtain failure mode and ultimate strength. Second, a fi nite element model was established and validated by the experimental results. Then, we used the verifi ed numerical method to simulate the behavior of the ring plate to CHS joint. A wide range of geometric parameters was considered, and the relevant design guide was assessed. Finally, nonlinear analysis was performed, and a design formula was proposed. Good agreement was achieved between the proposed design formula and the results of experimental and numerical analyses.

In this letter, we consider a cooperation system with multipleuntrusted relays (URs). To keep the transmitted informationconfidential, we obtain joint channel characteristics (JCCs)through combining the channels from the source to thedestination. Then, in the null space of the JCCs, jammersconstruct artificial noise to confuse URs when the source nodebroadcasts its data. Through a distributed implementationalgorithm, the weight of each node can be obtained from itsown channel state information. Simulation results show thathigh-level security of the system can be achieved when internaland external eavesdroppers coexist.

Nanofiber mats have received considerable attention from researchers in the field of particulate protection. In thisstudy, polyvinylidene fluoride (PVDF) nanofiber mats were prepared on a large scale by a solution blowing process, and thencrosslinked by ultraviolet (UV)-radiation using trimethylolpropane tri-acrylate (TMPTA) as a crosslinker. The structure, morphology, mechanical and thermal properties, and protective performance of the nanofiber mats were evaluated. Resultsshow that the addition of TMPTA and UV treatment decreased the diameter of the nanofibers and enhanced the mechanicalproperties, air permeability, and moisture permeability. Nanofiber mats show higher filtration and lower filtration resistancecompared with a commercial product. Furthermore, the nanofiber mats were bonded with Nylon 6 fabric, which showedgood performance as a novel functional layer in protective clothing.

Recently, there are many studies, that considering green wireless cellular networks, have taken the energy consumption of the base station (BS) into consideration. In this work, we first introduce an energy consumption model of multi-mode sharing BS powered by multiple energy sources including renewable energy, local storage and power grid. Then communication load requests of the BS are transformed to energy demand queues, and battery energy level and worst-case delay constraints are considered into the virtual queue to ensure the network QoS when our objective is to minimize the long term electricity cost of BSs. Lyapunov optimization method is applied to work out the optimization objective without knowing the future information of the communication load, real-time electricity market price and renewable energy availability. Finally, linear programming is used, and the corresponding energy efficient scheduling policy is obtained. The performance analysis of our proposed online algorithm based on real-world traces demonstrates that it can greatly reduce one day’s electricity cost of individual BS.

In this study, an energetic material was prepared by depositing nano-Al on CuO arrays via electrophoretic deposition (EPD), which offers a feasible route for nano-Al integration. The morphology and structure of the CuO arrays and Al/CuO composites were characterized by scanning electron microscopy and X-ray diffraction. The CuO arrays were homogenously salix leaf-like structure with a width of ~150 to 200 nm. The energy density of Al/CuO composites was approximate to 1454.5 J/g by integrating the differential scanning calorimetry (DSC) plot and the combustion performance was recorded by a high-speed camera. Moreover, the combustion flames were violent and the whole reaction process only lasted 72.2 ms, indicating that the energy of the Al/CuO nanothermite can be released effectively.

Pulmonary vascular remodeling due to excessive proliferation and resistance to apoptosis of pulmonary artery smooth muscle cells (PASMCs) is the hallmark feature of pulmonary arterial hypertension (PAH). Recent evidence suggests that miR-125a-5p plays a role in a rat model of monocrotaline-induced PAH (MCT-PAH); however, the underlying mechanism is currently unknown. Here, we examined the expression profile of miR-125a-5p in MCT-PAH rats and investigated the putative therapeutic effect of miR-125a-5p using the miR-125a-5p agomir. In addition, the miR-125a- 5p agomir or antagomir was transfected into rat PASMCs, and proliferation and apoptosis were measured. Activity of the miR-125a-5p target STAT3 was measured using a luciferase reporter assay, and the expression of downstream molecules was measured using RT–qPCR and/or western blot analysis. Importantly, inducing miR-125a-5p expression in vivo slowed the progression of MCT-PAH by reducing systolic pulmonary arterial pressure, the Fulton index, and pulmonary vascular remodeling. Moreover, overexpressing miR-125a-5p inhibited the proliferation and promoted the apoptosis of PASMCs. In addition, stimulating PASMCs with TGF-β1 or IL-6 upregulated miR-125a-5p expression, whereas overexpressing miR-125a-5p reduced TGF-β1 and IL-6 production, as well as the expression of their downstream targets STAT3 and Smad2/3; in contrast, downregulating miR-125a-5p increased TGF-β1 and IL-6 production. Finally, a dual-luciferase reporter assay revealed that miR-125a-5p targets the 3′-UTR of STAT3, suppressing the downstream molecules PCNA, Bcl-2, and Survivin. Taken together, these findings suggest that miR-125a-5p ameliorates MCT-PAH in rats, has a negative feedback regulation with TGF-β1 and IL-6, and regulates the proliferation and apoptosis of PASMCs by directly targeting STAT3.

Background: Holothuria scabra is a costly gourmet and traditional Chinese tonic medicine. However, the lack of high-quality genome information hinders the genetic, phylogenetic, and bioactivator researches. Objective: To construct high-quality genomic data of H. scabra and conduct genome-wide phylogenetic analysis. Methods: The whole genome of a male H. scabra was sequenced based on Nanopore MinION platform, and the sequence was assembled by wtdbg2. Transcriptome sequencing was used to aid the gene annotation. Repeat sequences, non-coding RNA, pseudogene and gene functional annotation were analyzed. 750 single-copy gene families from ten species were applied to construct phylogenetic tree for evolutionary analysis by using the ML method. Results: The H. scabra genome of 1.18 Gb (N50 = 1557,492 bp) with 500.42 Mb of putative repetitive sequences was assembled from a male H. scabra individual, and 16,642 protein-coding genes, 951 pseudogenes, 1791 motifs and 45,400 domains from the generated assembly were identified. The divergence time between H. scabra and its ancestor was estimated approximately 192.6 million years ago. H. scabra and A. japonicas joined together while sea urchin and sea star diverged about 440 Mya ago. Some key genes involved in notochord and gill slits development, skeleton degeneration and nervous system, as well as homeobox genes differ between H. scabra and Apostichopus japonicas. Conclusion: We report the first whole genome of H. scabra with expectation that this will be a valuable resource for genetic, phylogenetic, breading, molecular biology and bioactivator studies of sea cucumbers and other invertebrates.

In this paper, weighted average consensus of second-order multi-agent systems (MASs) via sampleddata control is investigated. Specially, since the intelligence degree of each agent may be different in practical applications, the concept of intelligence level is introduced to characterize the intelligence degrees of agents in this paper. A novel distributed continuous-time control protocol with various intelligence levels is put forward based on sampled data in order to reduce the updating rates and computational burden of the controller. A necessary and sufficient condition is derived to achieve weighted average consensus under the proposed protocol. Moreover, according to the acquired result, the authors also give the choosing approaches of the feedback gains and sampling period. Finally, several numerical simulations are provided to illustrate the reliability of the theoretical results.
STAT3 promotes NLRP3 inflammasome activation by mediating NLRP3 mitochondrial translocation
Luo Ling,Wang Fupeng,Xu Xueming,Ma Mingliang,Kuang Guangyan,Zhang Yening,Wang Dan,Li Wei,Zhang Ningjie,Zhao Kai 생화학분자생물학회 2024 Experimental and molecular medicine Vol.56 No.-
Recognition of the translocation of NLRP3 to various organelles has provided new insights for understanding how the NLRP3 inflammasome is activated by different stimuli. Mitochondria have already been demonstrated to be the site of NLRP3 inflammasome activation, and the latest research suggests that NLRP3 is first recruited to mitochondria, then disassociated, and subsequently recruited to the Golgi network. Although some mitochondrial factors have been found to contribute to the recruitment of NLRP3 to mitochondria, the detailed process of NLRP3 mitochondrial translocation remains unclear. Here, we identify a previously unknown role for Signal transducer and activator of transcription-3 (STAT3) in facilitating the translocation of NLRP3 to mitochondria. STAT3 interacts with NLRP3 and undergoes phosphorylation at Ser727 in response to several NLRP3 agonists, enabling the translocation of STAT3 and thus the bound NLRP3 to mitochondria. Disruption of the interaction between STAT3 and NLRP3 impairs the mitochondrial localization of NLRP3, specifically suppressing NLRP3 inflammasome activation both in vitro and in vivo. In summary, we demonstrate that STAT3 acts as a transporter for mitochondrial translocation of NLRP3 and provide new insight into the spatial regulation of NLRP3.
