      • Clinical Comparison between Paclitaxel Liposome (Lipusu<sup>®</sup>) and Paclitaxel for Treatment of Patients with Metastatic Gastric Cancer

        Aim: To compare the efficacy and safety of paclitaxel liposome (Lipusu$^{(R)}$) with paclitaxel in combination with tegafur and oxaliplatin in treating patients with advanced gastric cancer. Materials and Methods: Patients with advanced gastric cancer receiving chemotherapy were retrospectively collected, and divided into two groups. Patients in group A received paclitaxel liposomes at a dose of 135 $mg/m^2$ on day 1 of each cycle, and patients in group B were given paclitaxel at the same dose with the same timing. All patients received tegafur at a dose of 500 $mg/m^2$ on days 1-5, and oxaliplatin at a dose of 80-100 $mg/m^2$ on day 1 for 2 cycles (each cycle was 21 d in total). Results: Fifty-eight patients could be evaluated for efficacy. The overall response rate was 47% in group A (14/30), and 46% in group B (13/28). Disease control rate was 73% in group A (22/30), and 71% in group B (20/28) (P>0.05). No significant differences were detected in hematologic and neurologic toxicities between the two groups (P>0.05). However, nausea, vomiting and hypersensitive reactions were significantly lower in group A than in group B (P<0.05). Conclusion: Paclitaxel liposomes are as effective as paclitaxel when combined with tegafur and oxaliplation in treating patients with advanced gastric cancer, but adverse reactions with paclitaxel liposomes are less common.

        RNAi suppression of nuclear receptor genes results in increased susceptibility to sulfoxaflor in brown planthopper, Nilaparvata lugens

        Nuclear receptors (NRs), which belong to a superfamily of ligand-dependent transcription factors, play essential roles in gene regulation to affect numerous physiological pathways. Twenty NR genes were identified in Nilaparvata lugens by using genomic, transcriptomic and GenBank databases, and categorized into NR0-NR6 subfamilies according to standard nomenclature. Among them, three NR geneswere classed into NR0, four into NR1, eight into NR2, one into NR3, one into NR4, two into NR5, and one into NR6, respectively. A phylogenetic tree of NRs from N. lugens and other representative species was constructed, which provided evolutionary insight into genetic distance. In order to investigate the NRs,whichwere induced by sulfoxaflor, time- and tissue-specific expression profiles of NR genes in fourth-instar nymphs were determined following LD50 sulfoxaflor treatment (0.28 ng/insect) and compared with control samples. NlUSP, NlE78, NlTLL, NlHR51, NlHR83, NlPNR and NlFTZ-F1 were highly expressed following 12–48 h of sulfoxaflor treatment. NlHR39 and NlDSF transcripts were detected in the head and the thorax, respectively. High-level and constitutive expression of NlHR3, NlTLL, NlHR83, NlFTZF1 and NlHR4 were found in the abdomen. Feeding of dsRNAs reduced the expression of NlHR3, NlUSP, NlTLL, NlHR83, NlPNR, NlFTZ-F1 and NlHR4 (35.48–49.77%) and caused significant nymph mortality (69.21–81.45%). These NRs, considered as insecticide targets, may play important function in sulfoxaflor detoxification.

        Signal processing method of bubble detection in sodium flow based on inverse Fourier transform to calculate energy ratio

        Electromagnetic vortex flowmeter is a new type of instrument for detecting leakage of steam generator, and the signal processing method based on the envelope to calculate energy ratio can effectively detect bubbles in sodium flow. The signal processing method is not affected by changes in the amplitude of the sensor output signal, which is caused by changes in magnetic field strength and other factors. However, the detection sensitivity of the electromagnetic vortex flowmeter is reduced. To this end, a signal processing method based on inverse Fourier transform to calculate energy ratio is proposed. According to the difference between the frequency band of the bubble noise signal and the flow signal, only the amplitude in the frequency band of the flow signal is retained in the frequency domain, and then the flow signal is obtained by the inverse Fourier transform method, thereby calculating the energy ratio. Using this method to process the experimental data, the results show that it can detect 0.1 g/s leak rate of water in the steam generator, and its performance is significantly better than that of the signal processing method based on the envelope to calculate energy ratio.

      • Luciferase Assay to Screen Tumour-specific Promoters in Lung Cancer

        Objective: Specific promoters could improve efficiency and ensure the safety of gene therapy. The aim of our study was to screen examples for lung cancer. Methods: The firefly luciferase gene was used as a reporter, and promoters based on serum markers of lung cancer were cloned. The activity and specificity of seven promoters, comprising CEACAM5 (carcinoembryonic antigen, CEA), GRP (Gastrin-Releasing Peptide), KRT19 (cytokeratin 19, KRT), SFTPB (surfactant protein B, SP-B), SERPINB3 (Squamous Cell Carcinoma Antigen, SCCA), SELP (Selectin P, Granule Membrane Protein 140kDa, Antigen CD62, GMP) and DKK1 (Dickkopf-1) promoters were compared in lung cancer cells to obtain cancer-specific examples with strong activity. Results: The CEACAM5, DKK1, GRP, SELP, KRT19, SERPINB3 and SFTPB promoters were cloned. Furthermore, we successfully constructed recombinant vector pGL-CEACAM5 (DKK1, GRP, SELP, KRT19, SERPINB3 and SFTPB) contained the target gene. After cells were transfectedwith recombinant plasmids, we found that the order of promoter activity from high to low was SERPINB3, DKK1, SFTPB, KRT19, CEACAM5, SELP and GRP and the order for promoters regarding specificity and high potential were SERPINB3, DKK1, SELP, SFTPB, CEACAM5, KRT19 and GRP. Conclusion: The approach adopted is feasible to screen for new tumour specific promoters with biomarkers. In addition, the screened lung-specific promoters might have potential for use in lung cancer targeted gene therapy research.

        Identification and expression patterns of UDP-glycosyltransferase (UGT) genes from insect pest Athetis lepigone (Lepidoptera: Noctuidae)

        UDP-glucuronosyltransferase (UGT) genes,which belong to an ancient gene family and play very important roles in all organisms, encode extracellularly secreted proteins that are involved in the transfer of glycosyl residues fromactivated nucleotide sugars to acceptor hydrophobicmolecules (aglycones). Athetis lepigone is an important polyphagous pest worldwide, and since 2011 it has become one of the major maize pests in North China. However, there have been no studies on pesticides for the effective control of this pest. In this study,we identified 23 putative UGT genes in A. lepigone by analysing previous antennal transcriptomic data. Phylogenetic analysis showed that all of the AlUGTs are distributed within 11 of 14 insect UGT sub-families. Tissue expression analysis revealed that N70% of AlUGTs were primarily expressed in adult antennae, of which three (AlUGT33AD1, AlUGT40F6 and AlUGT40L4) and four (AlUGT33B18, AlUGT33F10, AlUGT40Q3 and AlUGT41D3) displayed male-biased and female-biased expression, respectively. SomeAlUGTs, however, had higher expression levels in non-antennal tissues. Our study is the first to identify UGT genes in A. lepigone,which will help us to elucidate the diverse functions of these genes, and ultimately provide potential targets that will facilitate the development of efficient and environmentally friendly pesticides against A. lepigone.

        Remarkable impact of steam temperature on ginsenosides transformation from fresh ginseng to red ginseng

        Background: Temperature is an essential condition in red ginseng processing. The pharmacological activities of red ginseng under different steam temperatures are significantly different. Methods: In this study, an ultrahigh-performance liquid chromatography quadrupole time-of-flight tandem mass spectrometry was developed to distinguish the red ginseng products that were steamed at high and low temperatures. Multivariate statistical analyses such as principal component analysis and supervised orthogonal partial least squared discrimination analysis were used to determine the influential components of the different samples. Results: The results showed that different steamed red ginseng samples can be identified, and the characteristic components were 20-gluco-ginsenoside Rf, ginsenoside Re, ginsenoside Rg1, and malonyl-ginsenoside Rb1 in red ginseng steamed at low temperature. Meanwhile, the characteristic components in red ginseng steamed at high temperature were 20R-ginsenoside Rs3 and ginsenoside Rs4. Polar ginsenosides were abundant in red ginseng steamed at low temperature, whereas higher levels of less polar ginsenosides were detected in red ginseng steamed at high temperature. Conclusion: This study makes the first time that differences between red ginseng steamed under different temperatures and their ginsenosides transformation have been observed systematically at the chemistry level. The results suggested that the identified chemical markers can be used to illustrate the transformation of ginsenosides in red ginseng processing.

      • 중국인 고급 학습자를 위한 한국어 미각 고정표현 교육방안 연구

        본 논문은 중국인 고급 학습자를 대상으로 한국어 기본 미각어 `달다, 쓰다, 짜다, 시다, 맵다`류의 고빈도 미각 고정표현을 가르치고자 한다. 그리고 중국인 학습자의 특성에 맞게 효과적인 교육방안을 제안하고, 또한 이러한 교육 방안을 실제 한국어 교육 현장에 적용되면 효과가 있는지 검증하고자 한다. 이를 위하여 우선 한·중 양 언어의 미각어에 대한 기본적 논의를 살펴보았다. 또한 `달다/甛`류, `쓰다/苦`류, `시다/酸`류, `짜다/咸`류, `맵다/辣`류를 중심으로 한·중 미각어의 파생의미를 비교하였다. 그리고 그 파생의미로 구성된 한·중 미각 고정표현을 비교하여 서로 간의 대응관계도 밝혔다. 4장에서는 교재 분석을 통해 한국어 미가 고정표현의 교육 현황을 파악하고 문제점을 찾았으며, 고빈도 미각 고정표현의 학습 목록을 추출하였고, 또한 중국인 학습자들의 한국어 미각 고정표현에 대한 인지도를 조사하였다. 그 후에 한국어 미각 고정표현을 난이도별로 나누었다. 그 미각 고정표현을 효과적으로 가르치기 위하여 한·중 대조를 토대로 공기 관계 교육방법 및 맵을 활용한 교육방법을 제안하였다. 5장에서는 한국어 미각 고정표현의 교육방법이 실제 한국어 교육 현장에 적용되면 효과가 있는지 검증한 내용이다. 마지막으로 6장은 결론 부분으로, 본 논문의 논의를 요약하고 본 논문의 의미와 한계점을 밝혔다. 한국어 교육에서 고정표현에 대한 교육이 중요하게 강조된 것은 많은 선행연구에서 반영되고 있다. 그러나 현재 한국어 교육에서 파생의미 및 미각 고정표현에 대한 교육이 아직 이루어지지 않았다. 그리고 한·중 미각 고정표현 간의 차이점이 많이 존재하기 때문에 학습자가 어려움을 겪을 수밖에 없다. 따라서 중국인 학습자들이 한국어 미각 고정표현의 학습이 체계적으로 이루어져야 하다는 인식을 가지고 본 논문을 시작하여 연구하였다. 본 논문은 실용적 교육 방안을 모색했다는 데에 의미를 두고자 한다. 그러나 실제 다양한 한국어 미각어들을 모두 다루지 못한 점과 제안된 교육방안은 실제 한국어 교육현장에서 많이 사용하는 방법과 겹치는 부분이 있지만 중국인 학습자에게 한국어 미각 고정표현을 학습하는 데에 도움이 되었다고 검증하였다. 앞으로 미각 고정표현에서 외국인 학습자에게 재미있고 효율성이 높은 자료를 구축하고 학습자에게 쉽게 배울 있고 잘 기억할 수 있도록 돕는 교육방안을 많이 연구해야 할 것으로 바란다. The objective of this study was to find effective methods of teaching korean pragmatic conventional expressions of flavors for chinese advanced learners. An analysis is done in Chapter Ⅲ by centering about the extended meaning of five basic flavors "sweet", "bitter", "sour", "spicy", "salty" and focusing on contrastive analysis of pragmatic conventional expressions of five basic flavors in Korean and China. Chapter Ⅳ was the process of selecting the high frequency of korean pragmatic conventional expressions and about teaching methods. In ChapterⅤ, based on the observation done before, it does the search about education programs of korean pragmatic conventional expressions of flavors, which is subjected to Chinese leaners. In the real educational class, educational effect is measured through a test in which Chinese learner were educated with the method of korean pragmatic conventional expressions of flavors. Finally Chapter is the conclusion part, which concludes the argument of this research and points out the meaning and shortcomings of this research. This research realizes the difficulties that Chinese learners will meet while learning korean pragmatic conventional expressions of flavors in South Korean and tries to solve these problems in practical teaching. This research is dedicated to find out the ways suitable for South korean education scenes in the process of contrasting. However, the research has shortcomings such as the teaching method had been used in practical education, be limited to five basic flavors, etc. which are expected to be studied in the future.

      • Differential Diagnosis of Malignant Biliary Tract Cancer from Benign Tissues using Apparent Diffusion Coefficient Measurements with Diffusion Weighted Imaging in Asians

        Background: The aim of this meta-analysis was to assess the efficacy of the apparent diffusion coefficient (ADC) value of diffusion-weighted MRI (DWI) for differentiating biliary tract cancer (BTC) from benign biliary tract diseases in Asians. Materials and Methods: We systematically searched Embase and PubMed prior to December 2014. Eight studies conducted in Asians met our predetermined inclusion criteria. Results: Our meta-analysis results showed that ADC values in BTC tissues were significantly lower than in benign biliary tract tissues (SMD = -1.54, 95%CI: -1.75~-1.33, P<0.001). Subgroup analysis based on the MRI machine type showed that the ADC values were consistent, accurate and reliable in the diagnosis of BTC when comparing cancer tissue vs. benign tissue under the Siemens 1.5 T/3.0 T, Philips 1.5 T/3.0 T, GE 1.5 T, and Toshiba 1.5 T types, respectively (all P<0.05). Further, ADC values were still consistent and accurate in the differential diagnosis of BTC under the b value of 800 and $1000s/mm^2$ (all P<0.05). Conclusions: Our findings supported potential clinical applications of DWI ADC values in differentiating BTC from benign biliary tract diseases in Asians.
