Association analysis of vitamin E and phytosterols content in dehulled rice

Vitamin E and phytosterols are both valuable nutrients that act as antioxidants in human bodies. Understanding the genetic basis of these traits is necessary for the improvement of nutritional quality by breeding. In this study, 119 rice accessions of diverse origin were genotyped using 232 SSR markers to identify marker–trait associations with Vitamin E and phytosterols in rice. Analysis of population structure revealed four subgroups in the population. Linkage disequilibrium (LD) patterns and distributions are of fundamental importance for genome-wide mapping associations. The mean r2 value for all intra-chromosomal loci pairs was 0.3361. LD between linked markers decreased with distance. Marker–trait associations were investigated using the unified mixed-model approach, considering both population structure (Q) and kinship (K). In total, 81 marker–trait associations were identified using 232 different SSR markers covering 12 chromosomes. The results suggest that association mapping in rice is a viable alternative to quantitative trait loci mapping. The results from this association mapping study will be the basis for improving rice nutritional quality.

The APETALA2/ethylene-responsive elementbinding factors (AP2/ERF) play central roles in the stressresponse in plants. In this study, we identified and isolateda novel salt stress-related gene, LcERF080, that encodes anAP2/ERF protein in Lotus corniculatus cultivar Leo. LcERF080 was classified into the B-4 group of the ERFsubfamily based on multiple sequence alignment andphylogenetic characterization. Expression of LcERF080was strongly induced by salt, abscisic acid, 1-aminocyclopropane-1-carboxylic acid, methyl jasmonate, and salicylicacid stresses. Subcellular localization assay confirmedthat LcERF080 is a nuclear protein. LcERF080 overexpressionin Arabidopsis resulted in pleiotropic phenotypeswith a higher seed germination rate and transgenic plantswith enhanced tolerance to salt stress. Further, under stressconditions, the transgenic lines exhibited elevated levels ofsoluble sugars and proline as well as relative moisturecontents but a lower malondialdehyde content than incontrol plants. The expression levels of hyperosmoticsalinity response genes COR15A, RD22, and P5CS1 werefound to be elevated in the LcERF080-overexpressingArabidopsis plants compared to the wild-type plants. Theseresults reveal that LcERF080 is involved in the responsesof plants to salt stress.

Although the overall structure of the chloroplast genome is generally conserved, a number of sequence variations have been identified, which are valuable for plant population and evolutionary studies. Here, we constructed a chloroplast variation map of 30 landrace rice strains of Korean origin, using the Oryza rufipogon chloroplast genome (Genbank: NC_017835) as a reference. Differential distribution of single nucleotide polymorphisms (SNPs) and indels across the rice chloroplast genome is suggestive of a region-specific variation. Population structure clustering revealed the existence of two clear subgroups (indica and japonica) and an admixture group (aus). Phylogenetic analysis of the 30 landrace rice strains and six rice chloroplast references suggested and supported independent evolution of O. sativa indica and japonica. Interestingly, two “aus” type accessions, which were thought to be indica type, shared a closer relationship with the japonica type. One hypothesis is that “Korean aus” was intentionally introduced and may have obtained japonica chloroplasts during cultivation. We also calculated the nucleotide diversity of 30 accessions and compared to six rice chloroplast references, which shown a higher diversity in the indica and aus groups than in the japonica group in lower level substitution diversity.

As one of the most important crop, rice is not only a staple food of half world’s population but a wonderful model plant, which has been leading the evolution and functional genomics study. The next-generation sequencing technology are expediting rice genomic study, by providing a simple but powerful way. In this study, we re-sequenced a core collection of 137 rice accessions from all over the world along with 158 Korean breeding varieties. Finally, 6.3G uniquely mapped reads were obtained, and about 10 million SNPs and ~1.2 million InDels were identified with average sequencing depth of 7.5X. These will help us to maximize our germplasm utilization and assists all the deep research in population dynamics and functional studies. Here, we’d like to show the approaches applied to resequencing data mining and on-going activities.

The marine brown algal genus Colpomenia has a worldwide distribution, with five species reported in Korea and Japan. However, no studies to date attempted to identify the number of species and geographical distribution of Colpomenia along Chinese coast. To fill the biodiversity knowledge gap, we analyzed 63 mitochondrial cox3 and 62 mitochondrial atp6 sequences of Colpomenia specimens collected from 30 localities along the Chinese coast. Maximum likelihood and Bayesian inference trees suggest the presence of at least three Colpomenia species (i.e., C. peregrina, C. claytoniae, and C. sinuosa) in China. C. peregrina and C. claytoniae are documented for the first time. C. sinuosa was only found in the South China Sea and its distribution didn’t overlap with that of C. peregrina which was found in the Yellow-Bohai Sea and the East China Sea. C. claytoniae appears to be confined to three isolated islands in the East and the South China Sea, where it occurs in sympatry with, respectively, C. peregrina and C. sinuosa. Future study can focus on comparing ecophysiological differences of Colpomenia species in response to environmental variables and exploring possible genetic hybridization / introgression at inter-specific contact zones.

In this paper, we prove strong convergence theorems of Halpern iteration for relatively asymptotically nonexpansive map- pings in the framework of Banach spaces. Model-Deviation Compensation Control for Uncertain Systems with Time Delay

A novel model-deviation compensation control strategy is developed for a class of uncertain systems with time delay in this paper. Taking an identified second-order plus dead-time (SOPDT) process for example, the whole design methodology is proposed, which integrates the prediction model, the sliding function design, the equivalent control term and the model-deviation compensation control law. Firstly the delay-ahead predictor with the feedback correction is constructed to weaken the affection of time delay in the system. Then by improving the classical sliding mode control, after obtaining the equivalent control term, the model-deviation compensation term is derived from the actual regulated response and the deviation to be eliminated, so that the modeling uncertainties and external disturbances can be restrained. Finally the establishment and feasible solution of LMI guarantee that the system output reaches the desired value. The simulation results verify the effectiveness of the proposed strategy.
ANXA2 Regulates the Behavior of SGC-7901 Cells

ANXA2, a member of the annexin family, is overexpressed and plays important roles in tumor development. However, the significance of ANXA2 expression in gastric carcinoma has not been clarified.To elucidate its roles in growth of gastric cancer, ANXA2 expression in SGC-7901 cells was inhibited with a designated siRNA, then cell proliferation, cell cycling, apoptosis and motility were determined by MTT assay, flow cytometry, Hoechst 33342 staining and wound healing assay, respectively. To further assess the behavior of ANXA2 deleted SGC-7901 cells, changes of microstructures were observed under fluorescence microscopy, laser scanning confocal microscopy and electron microscopy. We found that inhibition of ANXA2 expression caused cell proliferation to decrease significantly with G1 arrest, motility to be reduced with changes in pseudopodia/filopodia structure and F-actin and ${\beta}$-tubulin expression, and apoptosis to be enhanced albeit without significance. At the same time, ANXA2 deletion resulted in fewer pseudopodia/filopodia, non-stained areas were increased, contact inhibition among cells reappeared, and expression of F-actin and ${\beta}$-tubulin was decreased, with induction of polymerized disassembled forms. Taken together, these data suggest that ANXA2 overexpression is important to maintain the malignancy of cancer cells, and this member of the annexin family has potential to be considered as a target for the gene therapy of gastric carcinoma.

Objective The study investigated cognitive performance and brain function between treatment-resistant depression (TRD) and non- TRD patients to find potential neurobiological markers associated with refractoriness in depression patients.Methods Fourteen TRD patients, 26 non-TRD patients and 23 healthy controls (HC) were included in the present study. The neural function of prefrontal cortex (PFC) and cognitive performance among the three group were examined using near-infrared spectroscopy (NIRS) during verbal fluency task (VFT).Results Both TRD and non-TRD groups exhibited significantly worse VFT performance and lower activation of oxygenated hemoglobin (oxy-Hb) changes in the bilateral dorsolateral PFC (DLPFC) compared to the HC group. Within the TRD and non-TRD groups, VFT performance was no significant difference, but activation of oxy-Hb changes in dorsomedial PFC (DMPFC) in TRD patients was significantly lower than non-TRD patients. In addition, activation of oxy-Hb changes in right DLPFC were negatively correlated with the severity of depressive symptoms in depression patients.Conclusion Both TRD patients and non-TRD patients exhibited lower oxy-Hb activation in DLPFC. TRD patients exhibit lower oxy- Hb activation in DMPFC than non-TRD patients. fNIRS maybe a useful tool for predict depressive patients with or without treatment resistant.
