        Erythritol production by Yarrowia lipolytica mutant strain M53 generated through atmospheric and room temperature plasma mutagenesis

        Mutants of Yarrowia lipolytica with high erythritol production were generated through an atmospheric and room temperature plasma (ARTP) mutation system. Among these mutants, Y. lipolytica M53 exhibited the highest erythritol yield. In a batch culture, M53 produced 64.8 g/L erythritol from 100 g/L glycerol. The yields of byproducts (e.g. mannitol, arabitol, and a-ketoglutaric acid) were low, and the mechanisms underlying these changes were examined by measuring enzyme activities in the pentose phosphate pathway. Up to 145.2 g/L erythritol was produced by M53 from 200 g/L of glycerol, and erythritol accumulation was promoted by 3.7 mg/L of Cu2?, 10.15 mg/L of Mn2?, and 30.37 g/L of NaCl. Fed-batch cultivation of M53 in a 5-L fermentor produced 169.3 g/L erythritol with low levels of byproducts within 168 h. This finding confirmed the potential of M53 as an erythritol producer on a commercial scale.

      • Free vibration analysis of FG porous spherical cap reinforced by graphene platelet resting on Winkler foundation

        In this study, free vibration analysis of FG porous spherical cap reinforced by graphene platelets resting on Winkler-type elastic foundation has been surveyed for the first time. Three different types of porosity patterns are considered for the spherical cap whose two types of porosity patterns in the metal matrix are symmetric and the other one is uniform. Besides, five GPL patterns are assumed for dispersing of GPLs in the metal matrix. Tsai-Halpin and extended rule of the mixture are used to determine the Young modulus and mass density of the shell, respectively. Employing 3D FEM elasticity in conjunction with Hamilton's Principle, the governing motion equations of the structure are obtained and solved. The impact of various parameters including porosity coefficient, various porosity distributions in conjunction with different GPL patterns, the weight fraction of graphene Nano fillers, polar angles and stiffness coefficient of elastic foundation on natural frequencies of FG porous spherical cap reinforced by GPLs have been reported for the first time.

        Distortion-Free Intelligent Sampling of Sparse Surfaces Via Locally Refined T-Spline Metamodelling

        Automatic design of the number of sample points and sample locations when measuring surfaces with different geometries is of critical importance to enable autonomous manufacturing. Uniform sampling has been widely used for simple geometry measurement, e.g. planes and spheres. However, there is a lack of appropriate sampling techniques that can be applied to complex freeform surfaces, especially those with sparse topographical features, e.g. cutting edges and other high-curvature features. In this paper, a distortion-free intelligent sampling and reconstruction method with improved efficiency for sparse surfaces is proposed. In this method, a locally-refined T-spline approximation is firstly applied which maps a surface to a simplified T-spline space; then a shift-invariant space sampling method and corresponding reconstruction are applied for the surface measurement. This sampling strategy provides a cost-effective sampling design and guarantees the surface reconstruction without information loss in a T-spline space. Theoretical demonstrations and case studies show that this sampling strategy can provide up to an order of magnitude improvement in accuracy or efficiency over state-of-the-art methods, for the measurement of sparse surfaces, from macro- to nano-scales.

        Application of Near-Infrared Fluorescence Imaging with Indocyanine Green in Totally Laparoscopic Distal Gastrectomy

        aPurpose: Recently, totally laparoscopic gastrectomy has been gradually accepted by surgeons worldwide for gastric cancer treatment. Complete dissection of the lymph nodes and the establishment of the surgical margin are the most important considerations for curative gastric cancer surgery. Previous studies have demonstrated that indocyanine green (ICG)-traced laparoscopic gastrectomy significantly improves the completeness of lymph node dissection. However, it remains difficult to identify the tumor location intraoperatively for gastric cancers that are staged ≤T3. Here, we investigated the feasibility of ICG fluorescence for lymph node mapping and tumor localization during totally laparoscopic distal gastrectomy. Materials and Methods: Preoperative and perioperative data from consecutive patients with gastric cancer who underwent a totally laparoscopic distal gastrectomy were collected and analyzed. The patients were categorized into the ICG (n=61) or the non-ICG (n=75) group based on whether preoperative endoscopic mucosal ICG injection was performed. Results: The ICG group had a shorter operation time and less intraoperative blood loss. Moreover, significantly more lymph nodes were harvested in the ICG group than the non-ICG group. No pathologically positive margin was found and there was no significant difference in either the proximal or distal surgical margins between the 2 groups. Conclusions: Near-infrared fluorescence imaging with ICG can be successfully used in totally laparoscopic distal gastrectomy, and it contributes to both the completeness of D2 lymph node dissection and confirmation of the gastric transection line. Well-designed prospective randomized studies are needed in the future to fully validate our findings.

        Expression and Regulation of Transcription Factor FoxA2 in Chronic Rhinosinusitis With and Without Nasal Polyps

        Purpose: Chronic rhinosinusitis (CRS) is characterized by the excessive production of mucus. However, the molecular mechanism underlying mucin overproduction in CRS with or without nasal polyps (CRSwNP and CRSsNP, respectively) is poorly understood. This study was conducted to assess the importance of the transcription factor FoxA2 in mucin production and to investigate the targeting of FoxA2 as a potential therapeutic strategy for mucus hypersecretion in CRS patients. Methods: We enrolled 15 CRSwNP patients, 15 CRSsNP patients, and 10 normal controls in this study. The expression levels of FoxA2, MUC5AC, and MUC5B in inflamed and healthy nasal tissues were examined via immunohistochemistry and quantitative reverse transcription-polymerase chain reaction, and the levels of several proinflammatory cytokines in nasal secretions were measured via FlowCytomix analysis. In addition, the expression of MUC5AC and FoxA2 was determined in polyp-derived epithelial cells and NCI-H292 cells after in vitro stimulation. Results: FoxA2 was significantly down-regulated, and MUC5AC and MUC5B were significantly up-regulated in both the CRSwNP and CRSsNP patients compared to the controls (P<0.05), and the protein level of FoxA2 was negatively associated with the IL-6 level in the CRS patients (P<0.05). IL-6 significantly increased MUC5AC expression but inhibited FoxA2 expression in vitro (P<0.05). Transfection with a FoxA2 expression plasmid significantly decreased MUC5AC promoter activity (P<0.05) and inhibited IL-6-induced MUC5AC production (P<0.05). In addition, clarithromycin significantly alleviated IL-6-induced FoxA2 suppression and decreased MUC5AC expression in vitro (P<0.05). Conclusions: Our findings suggest that FoxA2 may be considered a therapeutic target for the modulation of mucus hypersecretion in CRS patients.
