      • Systematic Review of Single Large and/or Multinodular Hepatocellular Carcinoma: Surgical Resection Improves Survival

        Background: The role of surgical resection for patients with single large (${\geq}5cm$) and/or multinodular (${\geq}2$) hepatocellular carcinoma (HCC) is still controversial. This systematic review was performed to evaluate the safety and efficacy of resection for patients with single large and/or multinodular HCC. Materials and Methods: Databases (the PubMed, Web of Science, Embase, and Cochrane databases) were systematically searched to identify relevant studies exploring the safety and efficacy of resection for single large and/or multinodular HCC, published between January 2000 and December 2014. Perioperative morbidity and mortality, overall survival, and disease-free survival of the resection group were calculated. In addition, these outcome variables were also calculated for the control group in the included studies. Results: One randomized controlled trial and 42 nonrandomized studies involving 9,580 patients were eligible for analysis. Eight (1,594 patients) of the 43 studies also reported the outcomes of transarterial chemoembolization (TACE). Although 51.4% of patients featured cirrhosis, 90.7% of them demonstrated Child-Pugh A liver function in the resection group. The median rates of morbidity (24.5%) and mortality (2.5%) after resection were significantly higher than that of TACE (11.0%, P<0.001; 1.9%, P<0.001). However, patients who underwent resection had significantly higher median one-, three-, and five-year overall survival (76.1%, 51.7%, and 37.4%) than those who underwent TACE (68.3%, 31.5%, and 17.5%, all P<0.001). The median 1-, 3-, and 5-year DFS rates after resection were 58.3%, 34.6%, and 24.0%, respectively. Conclusions: Although tumor recurrence after resection for patients with single large and/ or multinodular HCC continues to be a major problem, resection should be considered as a strategy to achieve long-term survival.

      • Meta-analysis of Gene Expression Data Identifies Causal Genes for Prostate Cancer

        Prostate cancer is a leading cause of death in male populations across the globe. With the advent of gene expression arrays, many microarray studies have been conducted in prostate cancer, but the results have varied across different studies. To better understand the genetic and biologic mechanisms of prostate cancer, we conducted a meta-analysis of two studies on prostate cancer. Eight key genes were identified to be differentially expressed with progression. After gene co-expression analysis based on data from the GEO database, we obtained a co-expressed gene list which included 725 genes. Gene Ontology analysis revealed that these genes are involved in actin filament-based processes, locomotion and cell morphogenesis. Further analysis of the gene list should provide important clues for developing new prognostic markers and therapeutic targets.

        Isovitexin Is a Direct Inhibitor of Staphylococcus aureus Coagulase

        Staphylococcus aureus (S. aureus) is a major pathogen that causes human pneumonia, leading to significant morbidity and mortality. S. aureus coagulase (Coa) triggers the polymerization of fibrin by activating host prothrombin, which then converts fibrinogen to fibrin and contributes to S. aureus pathogenesis and persistent infection. In our research, we demonstrate that isovitexin, an active traditional Chinese medicine component, can inhibit the coagulase activity of Coa but does not interfere with the growth of S. aureus. Furthermore, we show through thermal shift and fluorescence quenching assays that isovitexin directly binds to Coa. Dynamic simulation and structure-activity relationship analyses suggest that V191 and P268 are key amino acid residues responsible for the binding of isovitexin to Coa. Taken together, these data indicate that isovitexin is a direct Coa inhibitor and a promising candidate for drug development against S. aureus infection.

      • Expression of Ang-2/Tie-2 and PI3K/AKT in Colorectal Cancer

        Purpose: To study the expression of angiogenin-2 (Ang-2) and its receptor Tie-2 in colorectal cancer and discuss the possible mechanisms behind this process. Materials and Methods: Using the streptavidin-peroxidase (SP) immunohistochemical method, paraffin sections from 100 colorectal cancer samples and 10 samples from tumor-adjacent normal tissue (> 2 cm from the edge of the gross tumor) were tested for protein expression of Ang-2, Tie-2, PI3K, and AKT. Reverse transcription-polymerase chain reaction and Western blots were further used to measure expression of the 4 genes and proteins in 20 freshly-resected colorectal cancer samples and tumor-adjacent normal tissues. Results: In colorectal cancer tissues, the expression of the Ang-2, Tie-2, PI3K, and AKT genes and their proteins was significantly higher than in tumor-adjacent normal tissues. Protein expression in poorly-differentiated adenocarcinoma was higher than that in well and moderately differentiated adenocarcinoma. According to Duke's classification, the protein expression in Stages C and D was significantly higher than that in Stages A and B. In the group with lymphatic metastasis, the protein expression was higher than that without lymphatic metastasis. Conclusions: In colorectal cancer, the expression of the Ang-2, Tie-2, PI3K, and AKT genes and their proteins is markedly higher than those in tumor-adjacent normal tissues. No correlation was observed between protein expression and gender, location, or histologic type. Correlations did exist between protein expression and differentiation level, stage of Duke's classification, and lymphatic metastasis; in colorectal cancer tissues with lower differentiation levels, higher stages of Duke's classification, and lymphatic metastasis, the expression of all 4 proteins was higher. The study of their expression patterns and relationships with aggression and metastasis will provide a valuable experimental foundation for assessing prognosis and targeted therapy of colorectal cancer.

        Phase transformation and roasting kinetics of cobalt-rich copper sulfide ore in oxygen atmosphere assisted by sodium sulfate

        The recovery rates of copper (99 %) and cobalt (93 %) could be significantly increased by adding sodiumsulfate in activated roasting of Cobalt-rich copper sulfide (CRCS). The mechanism and kinetic parametersof CRCS roasting with sodium sulfate addition were not clear. Thus, in this study, the thermal roastingkinetics of carrollite (the predominant cobalt occurrence mineral) in CRCS adding Na2SO4 with an airatmosphere was studied in detail by TG-DTG method. The apparent activation energy (E) and preexponentialfactor (A) were obtained via Flynn-Wall-Ozawa method (FWO) and Kissinger-Akahira-Sunose method (KAS) methods, where E values were 113.89 kJmol1 (KAS) and 120.87 kJmol1(FWO). The thermodynamic parameters of the roasting process were evaluated. The model function forthe carrollite reaction was determined via Malek analysis. The mechanism of sodium sulfate additionin CRCS roasting was studied. Results showed that the addition of sodium sulfate in activated roastingcan promote the transformation of carrollite mineral phase. The activation energy value of the oxidationprocess of carrollite decreases. From the kinetic point of view, the addition of sodium sulfate can promotethe activation roasting.

        Pneumatic Measuring Method for Inclination Angle of the Edge of Inner Ring Groove

        The machining quality of the functional edges of inner ring groove have great influence on the performance of electro-hydraulic servo valve. The inclination angle of the functional edge is one of the most important indicators to evaluate the processing quality of functional edges. Therefore, a pneumatic measuring method based on pneumatic measuring theory for measuring the inclination angle of the edge of inner ring groove is proposed. The principle of measuring method and calculation method are analyzed. The measuring system parameters such as air source pressure, inlet nozzle diameter and slit width, which influence the measurement results are compared and analyzed, respectively. The results showed that those three measurement parameters significantly affect the slope of the measuring pressure curve. When the air source pressure, the inlet nozzle diameter and slit width are chosen as 90 kPa, 0.3 mm and 3 mm, respectively, the measurement system can be kept in a "subsonic-subsonic" state and good curve slope can be obtained. Based on the analysis, the measuring air path and other mechanical structure are designed. The experimental results indicate that the inclination angle relative to the central axis is 0.0387°. The proposed pneumatic measuring method can effectively measure the inclination angle of edge of the inner ring groove, which provides a method for evaluating the performance of the valve sleeve.

        Positive Association of TEAD1 With Schizophrenia in a Northeast Chinese Han Population

        Objective Schizophrenia is a complex and devastating psychiatric disorder with a strong genetic background. However, much uncertainty still exists about the role of genetic susceptibility in the pathophysiology of schizophrenia. TEA domain transcription factor 1 (TEAD1) is a transcription factor associated with neurodevelopment and has modulating effects on various nervous system diseases. In the current study, we performed a case–control association study in a Northeast Chinese Han population to explore the characteristics of pathogenic <i>TEAD1</i> polymorphisms and potential association with schizophrenia.Methods We recruited a total of 721 schizophrenia patients and 1,195 healthy controls in this study. The 9 single nucleotide polymorphisms (SNPs) in the gene region of <i>TEAD1</i> were selected and genotyped.Results The genetic association analyses showed that five SNPs (rs12289262, rs6485989, rs4415740, rs7113256, and rs1866709) were significantly different between schizophrenia patients and healthy controls in allele or/and genotype frequencies. After Bonferroni correction, the association of three SNPs (rs4415740, rs7113256, and rs1866709) with schizophrenia were still evident. Haplotype analysis revealed that two strong linkage disequilibrium blocks (rs6485989-rs4415740-rs7113256 and rs16911710-rs12364619-rs1866709) were globally associated with schizophrenia. Four haplotypes (C-C-C and T-T-T, rs6485989-rs4415740-rs7113256; G-T-A and G-T-G, rs16911710-rs12364619-rs1866709) were significantly different between schizophrenia patients and healthy controls.Conclusion The current findings indicated that the human <i>TEAD1</i> gene has a genetic association with schizophrenia in the Chinese Han population and may act as a susceptibility gene for schizophrenia.

        Compensation-based Characteristic Modeling and Tracking Control for Electromechanical Servo Systems With Backlash and Torque Disturbance

        For two-inertia servo mechanisms, the high order, backlash nonlinearity and external disturbance make the precise modeling and control difficult to implement. This article provides an easily implemented modeling and control strategy to deal with this problem. A characteristic modeling framework of disturbed nonlinear systems is proposed. To restrain the modeling error, the backlash nonlinearity and torque disturbance are observed by constructing a finite-time extended state observer (FESO) based on homogeneity properties and then the compensation action can be taken. Based on the compensated system, the discrete-time characteristic model is established using the sampled input-output data, which degrades the modeling complicacy. To estimate the model parameters, an adaptation law with projection algorithm is proposed using the tracking error and the estimation error as the excitation signal. A discrete-time second-order fast terminal sliding-mode control (DSFTSC) is proposed based on the characteristic model to stabilize the whole system, where an improved reaching law is designed to enhance the rapidity and weaken the chattering and the utilization of the fast terminal switching surface also speeds up the regression rate and decreases the tracking error. Finally, the effectiveness of the characteristic modeling, the adaptive law and the control scheme is validated by simulations in Matlab and experiments in a practical test rig, respectively.

        Computational Prediction of Alzheimer's and Parkinson's Disease MicroRNAs in Domestic Animals

        As the most common neurodegenerative diseases, Alzheimer's disease (AD) and Parkinson's disease (PD) are two of the main health concerns for the elderly population. Recently, microRNAs (miRNAs) have been used as biomarkers of infectious, genetic, and metabolic diseases in humans but they have not been well studied in domestic animals. Here we describe a computational biology study in which human AD- and PD-associated miRNAs (ADM and PDM) were utilized to predict orthologous miRNAs in the following domestic animal species: dog, cow, pig, horse, and chicken. In this study, a total of 121 and 70 published human ADM and PDM were identified, respectively. Thirty-seven miRNAs were co-regulated in AD and PD. We identified a total of 105 unrepeated human ADM and PDM that had at least one 100% identical animal homolog, among which 81 and 54 showed 100% sequence identity with 241 and 161 domestic animal miRNAs, respectively. Over 20% of the total mature horse miRNAs (92) showed perfect matches to AD/PD-associated miRNAs. Pigs, dogs, and cows have similar numbers of AD/PD-associated miRNAs (63, 62, and 59). Chickens had the least number of perfect matches (34). Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analyses suggested that humans and dogs are relatively similar in the functional pathways of the five selected highly conserved miRNAs. Taken together, our study provides the first evidence for better understanding the miRNA-AD/PD associations in domestic animals, and provides guidance to generate domestic animal models of AD/PD to replace the current rodent models.

        Long non-coding RNA RP11-6O2.4 indicates poor prognosis and suppresses cell cycle progression through the p38-MAPK signaling pathway in gastric cancer

        Backgrounds: The role of long non-coding RNAs (lncRNA) in gastric cancer (GC) has been highlighted in studies conducted over the past decade. However, the potential clinical value and the mechanisms of action of RP11-6O2.4 in GC have not been thoroughly elucidated to date. The specific aim of the present study was to assess RP11-6O2.4 and to explore its role in human GC. Methods: Quantitative real-time polymerase chain reaction (qPCR) was performed to analyze the expression levels of RP11-6O2.4 in GC tissues, paired adjacent noncancerous tissues (ANTs) and GC cell lines. In addition, the correlation between RP11-6O2.4 expression and the clinical characteristics and prognosis of patients with GC was statistically analyzed. The effects of RP11- 6O2.4 on the GC cell cycle transformation through the p38-MAPK signaling pathway were explored by flow cytometry, qPCR and Western blot analysis after treatment with SB203580, a p38MAPK specific inhibitor, in vitro. Results: The expression levels of RP11-6O2.4 in GC tissues were significantly lower than the paired ANTs (P<0.05). In addition, RP11-6O2.4 expression was significantly lower in cases with older age, longer maximum tumor diameter, higher ASA grade and deeper invasive depth (P<0.05). RP11-6O2.4 expression was significantly higher in cases with well/middle differentiation than poor/no differentiation; higher in cases without lymph node metastasis than in lymph node metastasis; and higher in cases in stage Ⅰ/Ⅱ than in stage Ⅲ/Ⅳ. An in vitro assay showed that RP11-6O2.4 induced G0/ G1 phase cell cycle arrest, likely by regulating the p38- MAPK signaling pathway. Conclusion: The above mentioned data suggested that RP11-6O2.4 was a tumor-suppressor gene in GC. RP11- 6O2.4 might play an important role in the cell cycle transformation by regulating the p38-MAPK signaling pathway, thereby representing a specific biomarker and a potential molecular target for the treatment of GC.
