        Review : The Role of Residues 103, 104, and 278 in the Activity of SMG1 Lipase from Malassezia globosa: A Site-Directed Mutagenesis Study

        The SMG1 lipase from Malassezia globosa is a newly found mono- and diacylglycerol (DAG) lipase that has a unique lid in the loop conformation that differs from the common alpha-helix lid. In the present study, we characterized the contribution of three residues, L103 and F104 in the lid and F278 in the rim of the binding site groove, on the function of SMG1 lipase. Sitedirected mutagenesis was conducted at these sites, and each of the mutants was expressed in the yeast Pichia pastoris, purified, and characterized for their activity toward DAG and pnitrophenol (pNP) ester. Compared with wild-type SMG1, F278A retained approximately 78% of its activity toward DAG, but only 11% activity toward pNP octanoate (pNP-C8). L103G increased its activity on pNP-C8 by approximately 2-fold, whereas F104G showed an approximate 40% decrease in pNP-C8 activity, and they both showed decreased activity on the DAG emulsion. The deletion of 103-104 retained approximately 30% of its activity toward the DAG emulsion, with an almost complete loss of pNP-C8 activity. The deletion of 103-104 showed a weaker penetration ability to a soybean phosphocholine monolayer than wild-type SMG1. Based on the modulation of the specificity and activity observed, a pNP-C8 binding model for the ester (pNP-C8, N102, and F278 form a flexible bridge) and a specific lipidanchoring mechanism for DAG (L103 and F104 serve as “anchors” to the lipid interface) were proposed.

        Design of 39-GHz Up- and Down-Conversion Mixers for 5G mmWave TDD Applications

        This article presents fully integrated 39-GHz bidirectional up- and down-conversion mixers for 5G millimeter-wave (mmWave) applications. Fabricated in a 65-nm CMOS process with a 1.2-V supply voltage, the up- and down-conversion mixers consume 39 and 43 mW, respectively. For 5G time-division duplexing (TDD) operation, intermediate-frequency (IF)/local-oscillator (LO)/radio-frequency (RF) T/R switches are introduced. For better isolation and low insertion loss between the up- and down-conversion mixer, a series-shunt singlepole double-throw (SPDT) structure and an equivalent lumped λ/4 transmission line are proposed for IF and RF T/R switches, respectively. To realize compact area and wide bandwidth, a transformer-based matching network is adopted in this design. Targeting multi-channel phased array applications, the measurement result shows that the up-conversion mixer achieves a 2.5-dB peak conversion gain with 6.5 GHz 3-dB bandwidth. Including the insertion loss of the switch and IF signal routing, at the maximum gain of 36.5 GHz, the up-conversion mixer achieves an output 1-dB gain compression point (OP1dB) of 2.5 dBm. Furthermore, the down-conversion mixer achieves a 5-dB peak conversion gain with a 9.7-GHz 3-dB bandwidth.

      • A piezoelectric linear ultrasonic motor with the structure of a circular cylindrical stator and slider

        <P>A piezoelectric linear ultrasonic motor is proposed, with a cylindrical stator and slider structure. The length and diameter of the motor are about 10 and 1.5 mm, respectively. The stator consists of two piezoelectric ceramic (PZT) tubes connected by a thin film metallic glass (TFMG) pipe. The stator is designed based on theoretical analyses and finite element method (FEM) simulation. The traveling wave propagation is obtained in the FEM simulation under the proper geometrical sizes, suitable boundary conditions and driving voltage signals. The trajectories of particles on the TFMG pipe are elliptical motion. In the experiment, a 25  µm thick TFMG pipe is fabricated using the rotating magnetron sputtering technique and the vibration characteristics of the stator are measured by a laser Doppler vibrometer (LDV) system. Bidirectional motion of the slider is observed around 600 kHz, the maximum velocity is near to 40  mm s<SUP> − 1</SUP> at 50 Vp–p for the loose slider and the maximum output force is 6 mN at 70 Vp–p for the tight slider. </P>

        Temperature distribution analysis of steel box-girder based on long-term monitoring data

        Temperature may have more significant influences on structural responses than operational loads or structural damage. Therefore, a comprehensive understanding of temperature distributions has great significance for proper design and maintenance of bridges. In this study, the temperature distribution of the steel box girder is systematically investigated based on the structural health monitoring system (SHMS) of the Sutong Cable-stayed Bridge. Specifically, the characteristics of the temperature and temperature difference between different measurement points are studied based on field temperature measurements. Accordingly, the probability density distributions of the temperature and temperature difference are calculated statistically, which are further described by the general formulas. The results indicate that: (1) the temperature and temperature difference exhibit distinct seasonal characteristics and strong periodicity, and the temperature and temperature difference among different measurement points are strongly correlated, respectively; (2) the probability density of the temperature difference distribution presents strong non-Gaussian characteristics; (3) the probability density function of temperature can be described by the weighted sum of four Normal distributions. Meanwhile, the temperature difference can be described by the weighted sum of Weibull distribution and Normal distribution.

        The influence of the extent of lymph node metastasis on the prognosis for patients with intrahepatic cholangiocarcinoma

        Purpose: Reports showed that some of intrahepatic cholangiocarcinoma (ICC) patients with lymph node metastasis (LNM) may also gain survival benefit undergone resection. However, the effect of the extent of LNM on prognosis and surgical indication is barely discussed. Methods: From September 1994 to November 2018, primary ICC patients undergone initial curable surgery were enrolled. Based on the extent of LNM, we divided these patients into 4 groups, including patients with no LNM (group N0), LNM to hepatoduodenal ligament or common hepatic artery (region A, group A), LNM to gastrohepatic lymph nodes for left liver ICC and periduodenal and peripancreatic lymph node for right liver ICC (region B, group B), or LNM beyond these regions (region C, group C). Multivariable Cox regression analysis was performed to identify the prognostic factors for recurrence- free survival (RFS) and overall survival (OS) in all groups. Results: A total of 133 patients were enrolled. There were 56, 21, 17, and 39 patients in groups N0, A, B, and C, respectively. There was significant difference between groups N0 and C in RFS (P < 0.001) and OS (P = 0.002). When we compared group N0 + A + B with group C, we also found that RFS (P < 0.001) and OS (P = 0.007) were significantly different. In multivariable analysis, the extent of LNM was an independent risk factor for RFS (P < 0.050). Conclusion: ICC patients with the LNM to regions A and B could still achieve good prognosis with resection. Surgery should be carefully considered when LNM to region C.

        Resonance Analysis of Medium Voltage Multi-Microgrids Considering the Interaction of Controllable Series Compensator and Grid-Connected Inverters

        The interaction of a controlled series compensator (CSC) with other power electronics and basic power components in a multi-microgrid (MMG) maybe lead to complex resonance problems. In this paper, the frequency domain analysis method and the mode analysis method are combined to analyze the resonance characteristics of the medium-voltage microgrid cluster system under the action of two controllable series compensators, transformer-coupled series compensator (TCSC) and capacitive-coupled series compensator (CCSC). Firstly, based on the equivalent output impedance models of TCSC and CCSC, the equivalent output impedance characteristics of TCSC and CCSC are explored by frequency domain analysis. Secondly, combined with the impedance network model of cluster system, the resonance characteristics of the MMG after separate and together access to these two controllable series compensators at diferent locations are explored by modal analysis method. The results show that the equivalent output impedance of the two couple-type compensators in a certain frequency band is capacitive, and CCSC has a wider capacitive frequency band. Changes in controller parameter selection can afect band range and capacitance degree. The CSC control parameters afect the system resonance frequency and resonance center, and when the grid-connected inverter circuit parameters are diferent, the mode of system resonance will also change. The combination of frequency domain analysis and modal analysis is an efective means to study the resonance stability of microgrid cluster systems.

        Extended Equal Service and Differentiated Service Models for Peer-to-Peer File Sharing

        Peer-to-peer (P2P) systems have proved the most effective and popular file sharing applications in recent years. Previous studies mainly focused on equal service and differentiated service strategies when peers have no initial data before their downloads. For an upload-constrained P2P file sharing system, we model both the equal service process and the differentiated service process when the initial data distribution of peers satisfies some special conditions. Moreover, we show how to minimize the time required to distribute the file to any number of peers. The proposed fluid-based models can reveal the intrinsic relations among the initial data amount, the peer set size, and the minimum last finish time. The closed-form expressions derived from the extended models can closely approximate chunk-based models and systems, especially for relatively large files. As an application of the extended models, we show how to provide differentiated service efficiently to multiple peer sets. Since no limits are imposed on the upload bandwidth of peers or the size of each peer set, we believe that our analytic process and the results achieved can provide not only fundamental insights into bandwidth allocation and data scheduling but also a helpful reference for both improving system performance and building an effective incentive mechanism for P2P file sharing systems.

        With the reform and development of modern education and the continuous development of the five-education plan, the state’s emphasis on the mental health of young people and college students is increasing, the construction principle and basic requirement of school psychological education system should not only attach importance to the cultivation of students’ positive psychological quality from the source, but also promote the professional level of school psychological consultation and Crisis Prevention and intervention. At present, the construction of psychological education system in colleges and universities is faced with the realistic difficult that the professional ability of counselors needs to be improved urgently. To sum up the practical problems they are facing, put forward the improvement path of college psychological education system based on the improvement of counselors’ professional ability, and give full play to their cooperative education mechanism and professional functions, in order to enrich and improve the current school psychological education system construction and development. 随着现代教育的改革发展与五育并举计划持续推进,国家对青少年及大学生心理健康成长的重视度不断提升,明确提出学校心理育人体系的构建原则与基本要求,既需要从源头重视学生积极心理品质培养,也需要提升学校心理咨询和危机预防与干预的专业水平。当前高校在心理育人体系建设中面临着辅导员专业能力亟需提升的现实困难,本文即以此为研究基础,通过分析高校心理育人体系建设的背景和要求,总结其面临的现实问题,提出基于辅导员专业能力提升的高校心理育人体系改进路径,发挥其协同育人机制和专业功能,以此充实并完善现阶段学校心理育人体系的建设与发展。
