        Inhibition of MicroRNA-15a/16 Expression Alleviates Neuropathic Pain Development through Upregulation of G Protein-Coupled Receptor Kinase 2

        There is accumulating evidence that microRNAs are emerging as pivotal regulators in the development and progression of neuropathic pain. MicroRNA-15a/16 (miR-15a/16) have been reported to play an important role in various diseases and inflammation response processes. However, whether miR-15a/16 participates in the regulation of neuroinflammation and neuropathic pain development remains unknown. In this study, we established a mouse model of neuropathic pain by chronic constriction injury (CCI) of the sciatic nerves. Our results showed that both miR-15a and miR-16 expression was significantly upregulated in the spinal cord of CCI rats. Downregulation of the expression of miR-15a and miR-16 by intrathecal injection of a specific inhibitor significantly attenuated the mechanical allodynia and thermal hyperalgesia of CCI rats. Furthermore, inhibition of miR-15a and miR-16 downregulated the expression of interleukin-1β and tumor-necrosis factor-αin the spinal cord of CCI rats. Bioinformatic analysis predicted that G protein-coupled receptor kinase 2 (GRK2), an important regulator in neuropathic pain and inflammation, was a potential target gene of miR-15a and miR-16. Inhibition of miR-15a and miR-16 markedly increased the expression of GRK2 while downregulating the activation of p38 mitogen-activated protein kinase and NF-κB in CCI rats. Notably, the silencing of GRK2 significantly reversed the inhibitory effects of miR-15a/16 inhibition in neuropathic pain. In conclusion, our results suggest that inhibition of miR-15a/16 expression alleviates neuropathic pain development by targeting GRK2. These findings provide novel insights into the molecular pathogenesis of neuropathic pain and suggest potential therapeutic targets for preventing neuropathic pain development.

        Synthesis of polyethylenimine-modified magnetic hydrogel nanocomposite absorbents for heavy metals removal

        Polyethylenimine(PEI)-modified magnetic hydrogel nanocomposite absorbents (PEI-mHNAs) were fabricated based on poly(acrylamide-co-acrylic acid) and PEI-modified magnetic nanoparticles by radical copolymerization. FTIR and XRD results preliminarily confirmed the target structure of PEI-mHNAs without destroying the structure of magnetic nanoparticles during modification and radical copolymerization. PEI-mHNAs with many rough porous and interstitial structure had high adsorption capacity of Cu(II) (217mg/g), Cd(II) (232mg/g) and Pb(II) (459mg/g). PEI-mHNAs had the best absorption capacity for heavy metal ions in the synthesis condition of acrylic acid/acrylamide mass ratio of 60 : 40, 0.8% initiator, AA neutralization degree of 70%, 0.75% crosslinker, and 15% PEImodified magnetic nanoparticles. Moreover, PEI-mHNAs had good magnetic responsiveness, high thermal stability and reusability, which make it a potential application in removing heavy metal ions from the contaminated wastewater.

        Influence of crosslinker amount on swelling and gel properties of hectorite/poly(acrylamide/itaconic acid) nanocomposite hydrogels

        A new hectorite/poly(acrylamide/itaconic acid) nanocomposite hydrogel was synthesized by inverse microemulsion polymerization. The effects of crosslinker amount on water absorbency and salt absorbency, swellability, pHsensitivity, gel strength, temperature-and shear-resistance were investigated. Water and salt absorbencies, pH-sensitivity and swellability decreased, while gel strength and temperature-and shear-resistance increased with increasing crosslinker amount. Nanocomposite hydrogels had good thermal stability with onset decomposing temperature of 252 oC. The as-synthesized hydrogel particles were regular and spherical-like and had an average particle size of 43 nm in the range of 30-65 nm. Hectorite clay platelets were exfoliated and transformed into amorphous structure.

      • Cryptanalysis of a Biometric-based Multi-Server Authentication Scheme

        Authentication and key agreement protocol becomes an important security issue for multi-server architecture. Combining biometrics with password enhances the level of security. Recently, Baruah et al. analyzed that Mishra et al.’s protocol has several drawbacks and proposed an improved biometric based multi-server authentication scheme. They claimed that their scheme satisfies all the required security attributes for a secure authentication. In this paper, we indicate that their scheme is not secure against key reveal attack, replay attack, and smart card forgery attack. Any registered user can retrieve the session key or launch the replay attack by eavesdropping on the communication channel. In addition, registered user can forge smart card when colluding with registered server.

        Caveolin-1 is a scaffold protein on the cell membrane. As the main component of caveolae, caveolin-1 is involved in many biological processes that include substance uptake and transmembrane signaling. Many of these processes and thus caveolin-1 contribute to cell transformation, tumorigenesis, and metastasis. Of particular interest are the dual rolesof tumor suppressor and oncogene that caveolin-1 appear to play in different malignancies, including pancreatic cancer. Therefore, analyzing caveolin-1 regulators and understanding their mechanisms of actionis key to identifying novel diagnostic and therapeutic tools for pancreatic cancer. This review details the mechanisms of action of caveolin-1 regulators and the potential significance for pancreatic cancer treatment.

        The mitochondrial genome of the Kentish Plover Charadrius alexandrinus (Charadriiformes: Charadriidae) and phylogenetic analysis of Charadrii

        The suborder Charadrii (Aves: Charadriiformes), one of the most species-rich radiations within shorebirds, which contains good source for studies of ecology, behaviour and evolution. The resources of mitogenome have rapidly accumulated in recent years due to the advanced genomic sequencing, while suborder Charadrii’s mitogenome has not been well studied. The primary objective of this study was to determine the complete mitogenome sequence of Charadrius alexandrinus, and investigated the evolutionary relationship within Charadrii. The mitogenome of C. alexandrinus were generated by amplification of overlapping Polymerase Chain Reaction (PCR) fragments. In this study, we determined the complete mitogenome sequence of the Kentish Plover Charadrius alexandrinus, and comparative analysed 11 species to illustrate mitogenomes structure and investigated their evolutionary relationship within Charadrii. The Charadrii mitogenomes displayed moderate size variation, the mean size was 16,944 bp (SD = 182, n = 11), and most of the size variation due to mutations in the control region (CR). Nucleotide composition was consistently biased towards AT rich, and the A+T content also varies for each protein-coding genes. The variation in ATP8 and COIII was the highest and lowest respectively. The GC skew was always negative, with the ATP8 had higher value than other regions. The average uncorrected pairwise distances revealed heterogeneity of evolutionary rate for each gene, the COIII, COI and COII have slow evolutionary rate, whereas the gene of ATP8 has the relative fast rate. The highest value of Ks and Ka were ND1 and ATP8, and the ratios of Ka/Ks are lower than 0.27, indicating that they were under purifying selection. Phylogenomic analysis based on the complete mitochondrial genomes strongly supported the monophyly of the suborder Charadrii. This study improves our understanding of mitogenome structure and evolution, and providing further insights into phylogeny and taxonomy in Charadrii. In future, sequencing more mitogenomes from various taxonomic levels will significantly improve our understanding of phylogenetic relationships within Charadrii.

      • Diagnostic Value of Fluorescence in Situ Hybridization Assay in Malignant Mesothelioma: A Meta-analysis

        The diagnosis of malignant mesothelioma (MM) remains a clinical challenge and the fluorescence in situ hybridization (FISH) assay has been reported to be one promising tool. The present meta-analysis aimed to establish the overall diagnostic accuracy of FISH for diagnosing MM. After a systematic review of English language studies, the sensitivity, specificity and other measures of accuracy of FISH in the diagnosis of MM were pooled using random-effects models. Summary receiver operating characteristic curves were applied to summarize overall test performance. Nine studies met our inclusion criteria, the pooled sensitivity and specificity for FISH for diagnosing MM being 0.72 (95% CI 0.67-0.76) and 1.00 (95% CI 0.98-1.00), respectively. The positive likelihood ratio was 34.5 (95% CI 14.5-82.10), the negative likelihood ratio was 0.24 (95% CI 0.16-0.36), and the diagnostic odds ratio was 204.9 (95% CI 76.8-546.6), the area under the curve being 0.99. Our data suggest that the FISH assay is likely to be a useful diagnostic tool for confirming MM. However, considering the limited studies and patients included, further large scale studies are needed to confirm these findings.

      • Cloning of Phospholipase D from Grape Berry and Its Expression under Heat Acclimation

        To investigate whether phospholipase D (PLD, EC 3.1.4.4) plays a role in adaptive response of post-harvest fruit to environment, a PLD gene was firstly cloned from grape berry (Vitis Vinifera L. cv. Chardonnay) using RT-PCR and 3'- and 5'-RACE. The deduced amino acid sequence (809 residues) showed 84.7% identity with that of PLD from Ricinus communis. The secondary structures of this protein showed the characteristic C2 domain and two active sites of a phospholipid-metabolizing enzyme. The PLD activity and its expression in response to heat acclimation were then assayed. The results indicated PLD was significantly activated at enzyme activity, as well as accumulation of PLD mRNA and synthesis of new PLD protein during the early of heat acclimation, primary suggesting that the grape berry PLD may be involved in the heat response in post-harvest grape berry. This work offers an important basis for further investigating the mechanism of post-harvest fruit adaptation to environmental stresses.

        A Method Based on Spectrum Superposition for Minimizing the Hazards of Blasting Vibration

        The selection of delay time will directly affect the effect of vibration reduction in millisecond delay blasting. The commonly used frequency spectrum control methods have some problems such as the difficulty of seed wave selection and the complexity of nonlinear superposition model. Based on the principle of waveform linear superposition, this study proposes a new spectrum superposition method for minimizing the vibration hazards in an open deep-hole bench blasting. This method, with no need to select the seed waves, mainly uses the ratio of the Fourier spectrum of the combined vibration waveform and the single hole vibration waveform to quantify the suppressed vibration. By defining the suppression ratio K, the suppression ratio with frequency range of 10 − 60 Hz under different delay times was calculated, and the theoretical optimal vibration reduction delay time value of 10 − 15 ms was given. A high precision digital electronic detonator was used to verify the proposed spectrum superposition method. The test results show that the best delay time of field vibration reduction is 5 − 10 ms, which is lower than the theoretical best delay time of 10 − 15 ms. It is the authors’ belief that this phenomenon is mainly caused by the joints and cracks in the rock mass, which will prolong the actual superposition time of blasting seismic waves between adjacent holes. In addition, it is found that there is a negative correlation between the delay time and the main vibration frequency, which is mainly caused by the low-pass filtering characteristic of rock medium. In order to minimize the probability of resonance failure, an optimal delay of 5 ms based on the main vibration frequency was given.
