Aroma development in rice has been reported due to the lack of function of betaine aldehyde dehydrogenase gene (badh2) on rice chromosome 8. A lot of functional markers have been designed based on the InDels, such as 7bp deletion in exon 2, 803bp deletion in exon 4 and 5, 8bp deletion in exon 7, and 3bp insertion in exon 13. Although there were a lot of functional SNPs, other InDels have not been detected by a PCR-based marker. Here we developed a simple, co-dominant, functional cleaved amplified polymorphic sequence (CAPS) marker for fragrance trait based on 1bp insertion in exon 14. The developed marker showed a high efficiency in discriminating that special aromatic rice variety, and displayed perfect co-segregation with the trait of fragrance in the F2 population. This new marker developed in the present study would be useful in molecular breeding of fragrant rice varieties.
Aging impairs insulin-stimulated glucose uptake in rat skeletalmuscle via suppressing AMPKα

Insufficient intracellular fat oxidation is an important contributor to aging-related insulin resistance, while the precise mechanism underlying is unclear. AMP-activated protein kinase (AMPK) is an important regulator of intracellular fat oxidation and was evidenced to play a key role in high-glucose and high-fat induced glucose intolerance. In the present study, we investigated whether altered AMPK expression or activity was also involved in aging-related insulin resistance. Insulin sensitivity of rats’ skeletal muscles was evaluated using in-vitro glucose uptake assay. Activity of α subunit of AMPK (AMPKα ) was evaluated by measuring the phosphorylation of both AMPKα (P- AMPKα ) and acetyl-CoA carboxylase (P-ACC), while expression of AMPKα was assessed by determining the mRNA levels of AMPKα 1 and AMPKα 2, and protein contents of AMPKα . Compared with 4-month old rats, 24-month old rats exhibited obviously impaired insulin sensitivity. At the same time, AMPKα activity significantly decreased, while AMPKα expression did not alter during aging. Glucose transporter 4 expression also decreased in old rats. Compared with 24-month old rats, administration of the specific activator of AMPK, 5-aminoimidazole-4-carboxamide riboside (AICAR), significantly elevated AMPKα activity and GluT4 expression. Also, aging-related insulin resistance was significantly ameliorated by AICAR treatment. In conclusion, aging-related insulin resistance is associated with impaired AMPKα activity and could be ameliorated by AICAR, thus indicating a possible role of AMPK in aging-induced insulin resistance.
Prefrontal cortex miR-29b-3p plays a key role in the antidepressant-like effect of ketamine in rats

Ketamine has a rapid, obvious, and persistent antidepressant effect, but its underlying molecular mechanisms remain unknown. Recently, microRNAs (miRNAs) have emerged as important modulators of ketamine’s antidepressant effect. We investigated the alteration in miR-29b-3p in the brain of rats subjected to ketamine administration and chronic unpredictable mild stress (CUMS), and a sucrose preference test and forced swimming test were used to evaluate the rats’ depressive-like state. We used recombination adeno-associated virus (rAAV) or lentivirus-expressing miR-29b-3p to observe the change in metabotropic glutamate receptor 4 (GRM4). Cell culture and electrophysiological recordings were used to evaluate the function of miR-29b-3p. Ketamine dramatically increased miR-29b-3p expression in the prefrontal cortex of the normal rats. The dual luciferase reporter test confirmed that GRM4 was the target of miR-29b- 3p. The miR-29b-3p levels were downregulated, while the GRM4 levels were upregulated in the prefrontal cortex of the depressive-like rats. The ketamine treatment increased miR-29b-3p expression and decreased GRM4 expression in the prefrontal cortex of the depressive-like rats and primary neurons. By overexpressing and silencing miR-29b-3p, we further validated that miR-29b-3p could negatively regulate GRM4. The silencing of miR-29b-3p suppressed the Ca2+ influx in the prefrontal cortex neurons. The miR-29b-3p overexpression contributed to cell survival, cytodendrite growth, increases in extracellular glutamate concentration, and cell apoptosis inhibition. The overexpression of miR- 29b-3p by rAAV resulted in a noticeable relief of the depressive behaviors of the CUMS rats and a lower expression of GRM4. The miR-29b-3p/GRM4 pathway acts as a critical mediator of ketamine’s antidepressant effect in depressive-like rats and could be considered a potential therapeutic target for treating major depression disorder.
Genome-wide association studies approach and post-GWAS study in rice

AGenome-wide association studies (GWAS) have proven a useful technique for identifying genetic loci responsible for natural variation in rice. With the fast developed next-generation sequencing technology, it is possible for people to carry out GWAS by phenotyping different traits. However, how to make full use of huge data, abandon unnecessary data, and solve the problem of data application effectively seems still an obstacle for many researchers. Taking the case of whole-genome resequencing of Korean authentic rice core set, here we present a general technological path of GWAS including: 1) a schematic view of sequencing-based GWAS in rice; 2) a user-friendly and interactive web application for GWAS in rice by the aid of experience from Arabidopsis; 3) Haplotype and association analysis of candidate genes in a certain mechanism pathway, giving 10 starch synthesis genes as example; and 4) functional validation by Trans- and Mata-Omics analysis.

The diagnosis of malignant mesothelioma (MM) remains a clinical challenge and the fluorescence in situ hybridization (FISH) assay has been reported to be one promising tool. The present meta-analysis aimed to establish the overall diagnostic accuracy of FISH for diagnosing MM. After a systematic review of English language studies, the sensitivity, specificity and other measures of accuracy of FISH in the diagnosis of MM were pooled using random-effects models. Summary receiver operating characteristic curves were applied to summarize overall test performance. Nine studies met our inclusion criteria, the pooled sensitivity and specificity for FISH for diagnosing MM being 0.72 (95% CI 0.67-0.76) and 1.00 (95% CI 0.98-1.00), respectively. The positive likelihood ratio was 34.5 (95% CI 14.5-82.10), the negative likelihood ratio was 0.24 (95% CI 0.16-0.36), and the diagnostic odds ratio was 204.9 (95% CI 76.8-546.6), the area under the curve being 0.99. Our data suggest that the FISH assay is likely to be a useful diagnostic tool for confirming MM. However, considering the limited studies and patients included, further large scale studies are needed to confirm these findings.
Recognition of Radar Emitter Signals Based on SVD and AF Main Ridge Slice

Recognition of radar emitter signals is one of core elements in radar reconnaissance systems. A novel method based on singular value decomposition (SVD) and themain ridge slice of ambiguity function (AF) is presented for attaining a higher correct recognition rate of radar emitter signals in case of low signal-tonoise ratio. This method calculates the AF of the sorted signal and ascertains the main ridge slice envelope. To improve the recognition performance, SVD is employed to eliminate the influence of noise on the main ridge slice envelope. The rotation angle and symmetric Holder coefficients of the main ridge slice envelope are extracted as the elements of the feature vector. And kernel fuzzy cmeans clustering is adopted to analyze the feature vector and classify different types of radar signals. Simulation results indicate that the feature vector extracted by the proposed method has satisfactory aggregation within class, separability between classes, and stability. Compared to existing methods, the proposed feature recognition method can achieve a higher correct recognition rate.
Multi-mode Radar Signal Sorting by Means of Spatial Data Mining

For multi-mode radar signals in complex electromagnetic environment, different modes of one emitter tend to be deinterleaved into several emitters, called as "extension", when processing received signals by use of existing sorting methods. The "extension" problem inevitably deteriorates the sorting performance of multi-mode radar signals. In this paper, a novel method based on spatial data mining is presented to address above challenge. Based on theories of data field, we describe the distribution information of feature parameters using potential field, and makes partition clustering of parameter samples according to revealed distribution features. Additionally, an evaluation criterion based on cloud model membership is established to measure the relevance between different cluster-classes, which provides important spatial knowledge for the solution of the "extension" problem. It is shown through numerical simulations that the proposed method is effective on solving the "extension" problem in multi-mode radar signal sorting, and can achieve higher correct sorting rate.
