Optimal Protocol for Mass Propagation of Aloe vera

We established an advanced protocol for in vitro propagation of Aloe vera via comparison of basal media, sucrose contents, growth hormone combinations, and additional supplementation with various polyamines. The maximal number and growth of shoots after 5 weeks was obtained using MS media including $30g\;L^{-1}$ sucrose supplemented with $1.0mg\;L^{-1}$ BA and $0.1mg\;L^{-1}$ NAA. To improve shoot production, various concentrations of putrescine, spermidine, and spermine were added under optimal growth hormone conditions (MS media supplemented with $30g\;L^{-1}$ sucrose, $1.0mg\;L^{-1}$ BA, and $0.1mg\;L^{-1}$ NAA). Maximal shoot number and growth after 5 weeks were achieved with supplementation of $50mg\;L^{-1}$ spermidine. Regenerated plants were successfully acclimatized in soil with 100% efficiency. Cytogenetic inspection revealed that the regenerated plants maintained intact chromosomes identical to those of plants grown in field conditions. This protocol provides a valuable alternative for mass production of elite Aloe vera.
Novel 2,4-dianilino-5-fluoropyrimidine derivatives possessing ALK inhibitory activities

A new series of 2,4-dianilino-5-fluoropyrimidine derivatives were designed and synthesized and their anaplastic lymphoma kinase (ALK) inhibitory activities were evaluated by biochemical and cell-based assays in order to discover a new ALK inhibitor. Most compounds synthesized showed good inhibitory activities against ALK and good cytotoxic activities in H3122 cell line. The best compound 6f showed good activity against wild-type ALK along with crizotinib-resistant mutant ALK, and it showed 6 times better activity in cell-based assay than crizotinib. Some SAR studies were performed by the comparisons of the activities between 6 and the designed-synthesized compounds.

Lysimachia capillipes, a traditional herbal medicine, has been used to treat cold and arthritis. The aim of this study was to investigate the anti-inflammatory activity of L. capillipes extract in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. Treatments with L. capillipes extract at concentrations of 3.75,7.5,15, and 25 μg/mL had no cytotoxicity in LPS-stimulated RAW264.7 cells. L. capillipes extract dose-dependently suppressed the production of nitric oxide (NO) and the expression of pro-inflammatory cytokines including interleukin (IL)-1β, IL-6, and tumor necrosis factor (TNF)-α in LPS-stimulated RAW264.7 cells. These were associated with the decrease of the mRNA expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) by L. capillipes extract (3.75-25 μg/mL). Moreover, L. capillipes extract also significantly inhibited the phosphorylation of MAPKs including p38, JNK and ERK1/2 as well as the expression of phosphorylated NF-κB-p65 in a dose-dependent manner. These results suggest that L. capillipes extract has anti-inflammatory properties, which activates through the inhibition of the NF-κB and MAPKs signaling pathway.

In this paper, we investigated the carbonization characteristics of lignin hydrochar prepared by hydrothermal carbonization and established a model for predicting the carbonization degree using near-infrared spectroscopy and partial least squares regression. The carbon content of the hydrothermally carbonized lignin at the temperature of 200 ℃ was higher by approximately 3 wt% than that of the untreated sample, and the carbon content tended to gradually increase as the heating time increased. Hydrothermal carbonization made lignin more carbon-intensive and more homogeneous by eliminating the microparticles. The discriminant and predictive models using near-infrared spectroscopy and partial least squares regression approppriately determined whether hydrothermal carbonization has been applied and predicted the carbon content of hydrothermal carbonized lignin with high accuracy. In this study, we confirmed that we can quickly and nondestructively predict the carbonization characteristics of lignin hydrochar manufactured by hydrothermal carbonization using a partial least squares regression model combined with near-infrared spectroscopy. 본 논문에서는 열수 탄화(hydrothermal carbonization)에 의해 제조된 리그닌 하이드로차의 탄화 특성을 조사하였고, 근적외선 분광법과 부분 최소 제곱(partial least squares) 회귀를 이용하여 탄화 거동을 예측하기 위한 모델을 수립하였다. 온도 200℃에서 열수 탄화된 리그닌의 탄소 함량은 무처리 시료 보다 약 3 wt% 높았으며 가열 시간이 증가할수록 탄소 함량도 서서히 증가하는 경향이 나타났다. 열수 탄화는 리그닌을 더욱 탄소 집약적으로 변화시키고 마이크로 파티클을 제거하여 더욱 균질한 특성을 부여하였다. 근적외선 분광법과 부분 최소 제곱 회귀를 이용한 판별 및 예측 모델은 수열 탄화의 적용 여부를 완벽히 구분했으며 높은 정확도로 열수 탄화 리그닌의 탄소 함량을 예측하였다. 본 연구로부터 근적외선 분광법과 결합된 부분 최소 제곱 회귀 모델을 이용하여 열수 탄화에 의해 제조된 리그닌 하이드로차의 탄화 특성을 빠르고 비파괴적으로 예측할 수 있다는 것이 확인되었다.

Fragment pathways of 3-alkyl and acyl-5-amino-l,3,4-thiadiazolin-2-ones were completely assigned by mass analyzed kinetic energy spectra (MIKE), collisional activated dissociation (CAD) spectra and high resolution mass spectra. [NH_(2)CS]^(+) and [S=C=O]^(+) ions were directly formed from molecular ion of the 3-alkyl compounds and from 5-amino-3H-l,3,4-thiadiazolin-2-one ion (1) which was produced by McLafferty rearrangement from molecular ion of 3-acyl compounds. A loss of neutral SCO was only detected from 3-alkyl substituted molecular ions.
Custom-Made ITE Type Hearing Protection Device Using a Small Acoustic Filter

Noise induced hearing loss (NIHS), the well-known occupational disease, is caused by continuous excessive noise. The prevention of NIHS is very important, because it is unrecoverable. There are some kinds of hearing protection device (HPD), and those are effective in preventing NIHS. But workers in noisy environment often resist to wearing them. Because they are ready - made products, so workers feel uncomfortable to wear. Also, they didn't maintain the conversation frequency range, so workers are hard to communicate in wearing them. To prevent hearing loss effectively, it is important that workers keep wearing HPD. Therefore, a HPD is needed to be comfortable to wear and be effective not only in hearing protection but also in preserving communication ability. So we proposed a custom - made hearing protection device in which a small acoustic filter is inserted. We designed several kinds of small acoustic filters and carried out some acoustic experiments for measuring characteristics of filters. We confirmed that acoustic transmission characteristic can be adjusted from experimental results using designed small acoustic filters. And we researched for the actual efficiency of a new developed custom - made hearing protection device using a small size acoustic filter. Also, we found out that workers are more satisfied with the new development than a former protection device from a workers' response.

Earth physics survey is raised not only to measure interiors and exteriors of nation's surface but also, through measuring physical property and property of material data on universe space, to utilize various fields such as land survey, map manufacture, earth physics expedition, geological feature resources developments and earthquake foresight and disasters prevention monitoring. Currently, though the intrinsic task, roles and the utilization field are changed little by little according to social environment variation and introductions of new survey technology, this is still holding large gravity in land survey fields and, recognized as one of the important infrastructure buildings in almost most nations, they are establishing utilization DB through systematic observation activity and data collections Specially, earth physics survey is playing very a vital role that provides survey and standard documents for map manufacture of a nation. This study will present a general improvement plan for earth physics survey activation including, relevant to earth physics survey, domestic and foreign present situation, quality elevation plans of observation data and budget estimate plan etc.

Epidemiologic interest in particulate matter (PM) is growing particularly because of its impact of respiratory health. It has been elucidated that PM evoked inflammatory signal in pulmonary epithelia. However, it has not been established $Ca^{2+}$ signaling mechanisms involved in acute PM-derived signaling in pulmonary fibroblasts. In the present study, we explored dust particles PM modulated intracellular $Ca^{2+}$ signaling and sought to provide a therapeutic strategy by antagonizing PM-induced intracellular $Ca^{2+}$ signaling in human lung fibroblasts MRC5 cells. We demonstrated that PM10, less than $10{\mu}m$, induced intracellular $Ca^{2+}$ signaling, which was mediated by extracellular $Ca^{2+}$. The PM10-mediated intracellular $Ca^{2+}$ signaling was attenuated by antioxidants, phospholipase blockers, polyADPR polymerase 1 inhibitor, and transient receptor potential melastatin 2 (TRPM2) inhibitors. In addition, PM-mediated increases in reactive oxygen species were attenuated by TRPM2 blockers, clotrimazole (CLZ) and N-(p-amylcinnamoyl) anthranilic acid (ACA). Our results showed that PM10 enhanced reactive oxygen species signal by measuring DCF fluorescence and the DCF signal attenuated by both TRPM2 blockers CLZ and ACA. Here, we suggest functional inhibition of TRPM2 channels as a potential therapeutic strategy for modulation of dust particle-mediated signaling and oxidative stress accompanying lung diseases.
