<P>Interferon-tau (IFNT), a type I interferon (IFN), is known as pregnancy recognition signaling molecule secreted from the ruminant conceptus during the preimplantation period. Type I IFNs, such as IFN-alpha and IFN-beta, are known to activate cell-death pathways as well as induce apoptosis. In cows, induction of apoptosis with DNA fragmentation is induced by IFNT in cultured bovine endometrial epithelial cells. However, the status of cell-death pathways in the bovine endometrium during the preimplantation period still remains unclear. In the present study, we investigated the different cell-death pathways, including apoptosis, pyroptosis, and autophagy, in uterine tissue obtained from pregnant cows and <I>in vitro</I> cultured endometrial epithelial cells with IFNT stimulation. The expression of <I>CASP7</I>, <I>8,</I> and <I>FADD</I> (apoptosis-related genes) was significantly higher in pregnant day 18 uterine tissue in comparison to non-pregnant day 18 tissue. The expression of <I>CASP4</I>, <I>11,</I> and <I>NLRP3</I> (pyroptosis-related genes) was significantly higher in the pregnant uterus in comparison to non-pregnant uterus. In contrast, autophagy-related genes were not affected by pregnancy. We also investigated the effect of IFNT on the expression of cell-death pathway-related genes, as well as DNA fragmentation in cultured endometrial epithelial cells. Similar to its effects in pregnant uterine tissue, IFNT affected the increase of apoptosis-related (<I>CASP8</I>) and pyroptosis-related genes (<I>CASP11</I>), but did not affect autophagy-related gene expression. IFNT also increased γH2AX-positive cells, which is a marker of DNA fragmentation. These results suggest that apoptosis- and pyroptosis-related genes are induced by IFNT in the pregnant bovine endometrial epithelial cells.</P>

Study Design: This is a single-center retrospective cohort study with a university hospital setting.Purpose: This study aims to evaluate the short-term course of physical function and walking ability after intramedullary spinal cord tumor (ISCT) resection and predict walking independence 1 year after surgery.Overview of Literature: Although several reports have shown the postoperative functional prognosis of spinal intramedullary tumors with long-term follow-up, no reports have identified the predictors associated with the functional outcome at an early stage. Methods: A total of 79 individuals who underwent ISCT resection at our institute between 2014 and 2019 were enrolled in the study, whose preoperative walking state was independent ambulator regardless of cane support with the Functional Independence Measure Locomotor Scale (FIM-L) score of ≥6. The FIM-L, the American Spinal Injury Association (ASIA) motor and sensory scores in the lower extremities, and the Walking Index for Spinal Cord Injury II (WISCI II) were assessed for walking independence, lower-limb function, and walking ability, respectively. These evaluations were performed at 4 time points: preoperatively, 1 week (1W), 2 weeks (2W), and 1 year after surgery.Results: In the early phase after surgery, 71% and 43% of the participants were nonindependent ambulators at 1W and 2W, respectively. Histopathology indicated that patients with solid tumors (ependymoma, astrocytoma, or lipoma) showed significantly lower indices at 1W and 2W than those with vascular tumors (hemangioblastoma or cavernous hemangioma). Regarding tumor location, thoracic cases exhibited poorer lower-limb function at 1W and 2W and poorer walking ability at 2W than cervical cases. According to the receiver operating characteristic (ROC) analysis, 2 WISCI II points at 2W had the highest sensitivity (100%) and specificity (92.2%) in predicting the level of walking independence at 1 year postoperatively (the area under the ROC curve was 0.99 (95% confidence interval, 0.93–1.00).Conclusions: The higher the lower-limb function scores in the early phase, the better the improvement in walking ability is predicted 1 year after ISCT resection.

The geographical distribution of the toxic blue-lined octopus (commonly known as the blue-ringed octopus), Hapalochlaena spp., around the East/Japan Sea was investigated. Observation records of the octopus were gathered using commercial search engines on the Internet. A questionnaire to complement and enhance the base data was conducted that targeted fishermen from areas where the octopus was most likely to occur, i.e., the southeast coast and islands of Korea in the East/Japan Sea. Overall, 32 observational records of the blue-lined octopus were found from Korea and Japan. In Korea, only one record, from 2003, was found on a website; none of the 240 fishermen who participated in the questionnaire reported seeing blue-lined octopus. However, a total of 31 observations of the blue-lined octopus from 2004 to July 2010 were found from 17 different regions in the East/Japan Sea and neighboring waters in Japan. Twenty-two cases were from coastal Honshu Island, and nine were from the west coast of Kyushu Island, Japan. The northern distributional boundary of the blue-lined octopus on the Japanese coast was off Fukui Prefecture around latitude 36º10' N. Our results indicate that the blue-lined octopus is distributed extensively along the Japanese coast, at a low frequency, in the East/Japan Sea.

The quality of harvested perishable products greatly depends on temperatures during transport and storage or market display. In this study, we evaluated the influence of low temperature and duration of storage on the storage quality in iceberg lettuce that were kept at 1±0.2℃ for 0 (control), 1 and 2 weeks and subsequently exposed to 20±2℃ for 10 days. Lettuce heads with prior storage at 1℃ for 1 or 2 weeks showed higher visual quality and lower level of wilting as compared to the control sample until 4 days of storage at 20℃. However, higher butt discoloration ratings were observed for 1 or 2-week stored samples. Higher hue angle value was observed in the 1-week sample until 4 days of storage as compared to the control or 2-week samples. No significant difference in textural quality was found between the control and 1℃ stored samples. In general, 1-week sample exhibited slightly higher puncture force and breaking energy value as compared with the control and 2-week samples. At the end of the storage, soluble sugar content declined by 52, 45 and 54% in the outer leaves and 70, 62 and 65% in the inner leaves of the control, 1 and 2-week stored samples, respectively. The heads with prior storage at 1℃ for 1 week showed a little higher sugar contents than those with 2-week or the control samples. In general, the outer leaves contained higher amount of sucrose, whereas the inner leaves contained higher amount of glucose and fructose. Although citric acid was found in trace amount, malic acid was the main component in organic acid fractions. Results suggest that iceberg lettuce could be stored at 1℃ for 2 weeks without affecting quality and that a better quality could be maintained at 20℃ with prior storage at 1℃ than continuous storage at 20℃.

The changes in ammonia content as well as activity and gene expression of glutamate dehydrogenase (GDH; EC 1.4.1.2) were investigated in lettuce during storage. GDH amination activity increased with the increases in ammonia content in the outer leaf portion after 24 h of storage. GDH amination activity was substantially higher than deamination activity. The isolated partial cDNA clone referred to as LsGDH (Lactuca sativa glutamate dehydrogenase; AB334207) consisted of 757 nucleotides and was highly homologous with the GDH genes of other plants. Although the transcript of LsGDH was found in both the outer and inner leaves, the level of transcript gradually increased in the outer leaves with the progress of storage, but was only expressed in the inner leaves when higher enzyme activity was observed. Results suggest that GDH expression in lettuce is controlled by tissue specific manner and/or multiple levels of regulations.
