        Characterization in Terms of the NUX Rule of G - inserted Mutant Hammerhead Ribozymes with High Level of Catalytic Power

        Attempts using in vitro and in vivo selection procedures have been made to search for hammerhead ribozymes that have higher activities than the wild-type ribozyme and also to determine whether other sequences might be possible in the catalytic core of the hammerhead ribozyme. Active sequences selected in the past conformed broadly to the consensus core sequence except at A9, and no sequences were associated with higher activity than that of the hammerhead with the consensus core, an indication that the consensus sequence derived from viruses and virusoids is probably the optimal sequence [Vaish et al. (1997) Biochemistry 36, 6495-6501]. Recently, during construction of ribozyme expression vectors, we isolated a mutant hammerhead ribozyme, with an insertion of G between A9 and 610.1, that appeared to show significant activity [Kawasaki et at (1996) Nucleic Acids Res. 24, 3010-3016; Kawasaki et al. (1998) Nature 393, 284-289]. We, therefore, characterized kinetic properties of the G-inserted mutant ribozymes in terms of the NUX rule. We demonstrate that the NUX rule is basically applicable to the G-inserted ribozymes and, more importantly, one type of G-inserted ribozyme was very active with k_(cat), value of 6.4 min^(-1) in 50 mM Tris-HCl (pH 8.0) and 10 m1VI MgCl₂ at 37℃.

      • Recent progress of mutation breeding in modern genome science era -Ion-beam mutagenesis-

        The world is facing a serious food and energy crisis. Plant mutation breeding has played an important role in overcoming this crisis and maintaining world stability. New techniques are required to achieve faster and more effective breeding. At RIKEN, we have developed a unique technology for mutation induction by using heavy-ion beams from particle accelerators at the RI Beam Factory. This development was achieved through an efficient synergistic link between agricultural science and accelerator physics. The use of ion beams for mutagenesis has a number of advantages: the approach has low exposure levels and high survival rates with high mutation rates, and it creates a wide variety of different mutations. Because heavy-ion beams provide a very high amount of energy, even a single ion is enough to significantly damage a gene – in fact, the beams have enough energy to break the double strand of the DNA. The technique is also very useful in producing mutants that lack just a single gene; multiple propagation technology can be used to convert these mutants into new cultivars. Examples of such breeds include ‘Safinia Rose’ (petunia), ‘Temari Bright Pink’ (vervena) and ‘Olivia Pure White’ (dianthus). The development period for producing new varieties is only 2 years. Over last decade, molecular biology has made great advancements through technological innovation. We use high-throughput DNA sequencing techniques such as next-generation sequence instruments and microarray technologies for analysis of gene mutations. Mutants have become more and more useful and important in modern genetic studies, enabling the discovery of genes that control important traits, and revealing the functions and mechanisms underlying their operations. The discovery of genes using mutants may lead to the emergence of a new field in biology, ‘Mutagenomics’.

        Self-Regulated Learning Processes Outside the Classroom : Insights from a Case Study of Japanese EFL Students

        This study attempted to delve into out-of-class self-regulated learning processes of students with different English proficiency levels, focusing on their goal-setting and self-evaluation processes as well as their motivational beliefs. Four Japanese university students, two higher and two lower proficiency learners engaged in English self-study outside the classroom for 15 weeks. Their selfregulated learning processes were examined through a weekly English language learning journal kept by the participants. Goal-setting and self-reflection sheets were collected as supplemental data to gain insights into the descriptions in the journal. The textual data were analyzed using systematic content analysis procedures. Students’ English proficiency levels and learning progress were measured by the TOEIC Listening and Reading test. Their motivational beliefs (i.e., goal orientations) were investigated through their statements reported on the goal setting sheet. The analyses revealed that the two higher proficiency students, who had both instrumental and international orientations, were more metacognitively aware of their self-regulated learning processes and more actively engaged in selfregulatory processes than the two lower proficiency students, who had only instrumental orientations. In particular, the higher proficiency student showing the most learning progress exhibited effective self-regulatory cycles. Finally, pedagogical implications are discussed based on these findings.

      • Comparison of the implementation status of weight control and physical activities between South Korean and Japanese university students

        Purpose: Adolescent Japanese girls have a strong desire to be thin and, therefore, engage in many unhealthy weight-control behaviors such as dieting. In recent years, this drive for thinness has spread to include boys as well as girls. Meanwhile, the desire to be thin is also increasing among adolescents in South Korea. The present study aimed to investigate the correlation between weight-control (WC) behaviors and physical activity (PA) among university students in Japan and South Korea. Method: The subjects comprised 1,833 university students in Japan (male, 454; female, 456) and South Korea (male, 449; female, 474). All subjects were asked whether they engaged in WC behaviors and were queried about the intensity (mild, moderate, and vigorous), number of days per week, and duration of PA. The correlations between sex, WC behaviors, and PA were investigated by country using a chi-square test. Multiple correspondence analysis (MCA) was then applied to investigate overall correlations. Result: The rates of WC behaviors were higher among female students than male students in both countries (58.7% vs. 18.4% in Japan [χ2=151.9***] and 61.1% vs. 42.3% in South Korea [χ2=32.6***]). Regarding PA, male students tended to exercise more days per week than female students in South Korea for all PA intensities. The same tendency was observed in Japan in relation to moderate and vigorous PA, but not for mild PA which was the same for males and females. Two-dimensional (2D) plots based on the results of MCA showed a correlation between not engaging in WC behaviors and performing PA among male students and between engaging in WC behaviors and not performing PA among female students in both Japan and South Korea for mild PA (1D: α=.455, eigenvalue=1.519; 2D: α=.336, eigenvalue=1.337). This tendency became increasingly marked for moderate (1D: α=583, eigenvalue=1.776, 2D: α=.463, eigenvalue=1.531) and vigorous (1D: α=.656, eigenvalue=1.970, 2D: α=.470, eigenvalue=1.544) PA. Conclusion: Female university students in Japan characteristically engaged in WC behaviors without PA, and this tendency became more marked as exercise intensity increased. Female university students in South Korea also engaged in WC behaviors, but they also performed moderate PA; and WC behaviors were associated with PA. Male university students in South Korea engaged in WC behaviors, but these were associated with PA and the number of days per week; and durations of PA were greater than those of female university students in South Korea at all PA intensities. The rates of WC behaviors were low among male university students in Japan; however, the number of days per week and duration of PA were greater than those of male university students in South Korea at all PA intensities. The present findings show that the correlations between WC behaviors and PA observed among university students in South Korea are stronger in Japan.

        Effects of Chios mastic gum and exercise on physical characteristics, blood lipid markers, insulin resistance, and hepatic function in healthy Japanese men

        The effects of Chios mastic gum (Pistacia lentiscus var. Chia) and exercise on physical characteristics, blood lipid markers, insulin resistance, and hepatic function were investigated in healthy, non-smoking Japanese men aged C 40 years. Participants were allocated to control (C, 5 g/day placebo powder, n = 7), mastic (M, 5 g/day mastic powder, n = 7), or mastic plus physical activity groups (M ? PA, 5 g/day mastic powder and 30-min exercise three times/week, n = 7), and measurements were taken at baseline, 3 and 6 months. Serum triglycerides were significantly reduced at 3 months in M and M ? PA compared with C (P\0.05). Serum insulin and homeostatic model assessment of insulin resistance values were significantly reduced at 3 and 6 months in M ? PA, and at 6 months in M, compared with C (P\0.05). These results indicate that Chios mastic gum intake for 6 months reduced serum triglyceride and insulin concentrations, and additional exercise enhanced the effect on insulin.

      • CO-CREATION WITH FANS TO ENHANCE CUSTOMER ENGAGEMENT IN PRODUCT AND SERVICE DEVELOPMENT

        The concept of co-creation with consumers has been extensively studied in the literature documented in innovation management and marketing literatures. In this study, we contribute to the literature threefold. First, we develop a model to investigate how co-creation with fans enhance purchase intent and engagement of other customers. Second, we test the model using the data collected from consumers on four different product categories including beer, car, cosmetics and travel. By doing so, we examine the differences between product vs. service as well as different product categories. Third, we will compare the results with that of ordinary consumers to check if there are any differences in the case of co-creating with fans. Most of the extant studies have found a positive effect of co-creation on the outcome evaluation. However, engaging other consumers by co-creation with fans has not been studied. For bridging this research gap, we developed a conceptual model to investigate the antecedents and consequences of co-creation with fans. The hypotheses are as follows. H1: Product class involvement and domain specific knowledge affect perception on co-creation positively. H2: Perception on co-creation affects purchase intent and word-of-mouth positively. H3: The relationships of H1 and H2 differs depending on product categories. H4: The relationships of H1 and H2 differs if companies work with ordinary customers or fans. We tested the hypotheses with the data collected from consumers an online questionnaire survey. Data collection was conducted through a market research agency in 2016 for beer and cars, and in 2019 for cosmetics and travel. The data was collected from 240 consumers in their 20s, 30s, and 40s in Japan. Each segment has same number of males and females, 40 people each for six segments. The findings shed a new light on the co-creation literature and help companies to design better co-creation with fans in different product and service categories.

        Brown Tumor of the Patella Caused by Primary Hyperparathyroidism: A Case Report

        It has been reported that the common sites of brown tumors are the jaw, pelvis, ribs, femurs and clavicles. We report our experience in a case of brown tumor of the patella caused by primary hyperparathyroidism. An initial radiograph and CT showed an osteolytic lesion and MR images showed a mixed solid and multiloculated cystic tumor in the right patella. One month after the parathyroidectomy, rapid bone formation was observed on both radiographs and CT images.
