        Ad libitum Pasture Feeding in Late Pregnancy Does Not Improve the Performance of Twin-bearing Ewes and Their Lambs

        The present study evaluated the effect of controlled ryegrass-white clover herbage availability from day 128 until day 142 of pregnancy in comparison to unrestricted availability, on the performance of twin-bearing ewes of varying body condition score (BCS; 2.0, 2.5, or 3.0) and their lambs. It was hypothesised that under conditions of controlled herbage availability, the performance of lambs born to ewes with a greater BCS would be greater than those born to ewes with a lower BCS. During the period that the nutritional regimens were imposed, the pre- and post-grazing herbage masses of the Control regimen (1,070±69 and 801±30 kg dry matter [DM]/ha) were lower than the ad libitum regimen (1,784±69 and 1,333±33 kg DM/ha; p<0.05). The average herbage masses during lactation were 1,410±31 kg DM/ha. Nutritional regimen had no effect on ewe live weight, BCS and back fat depth or on lamb live weight, indices of colostrum uptake, maximal heat production, total litter weight weaned or survival to weaning (p>0.05). The difference in ewe BCSs and back fats observed among body condition groups was maintained throughout pregnancy (p<0.05). At weaning, ewes from the BCS2.0 group had lower BCS and live weight (2.4±0.2, 74.3±2.6 kg) than both the BCS2.5 (2.6±0.2, 78.6±2.4 kg) and BCS3.0 ewes (2.7±0.2, 79.0±2.6 kg; p<0.05), which did not differ (p>0.05). Ewe BCS group had no effect on lamb live weight at birth or weaning or on maximal heat production (p>0.05). Serum gamma glutamyl transferase concentrations of lambs born to BCS3.0 ewes were higher within 36 hours of birth than lambs born to BCS2.0 ewes and BCS2.5 ewes (51.8±1.9 vs 46.5±1.9 and 45.6±1.9 IU/mL, respectively [p<0.05]). There was, however, no effect of ewe body condition on lamb plasma glucose concentration (p>0.05). Lamb survival was the only lamb parameter that showed an interaction between ewe nutritional regimen and ewe BCS whereby survival of lambs born to BCS2.5 and BCS3.0 ewes differed but only within the Control nutritional regimen ewes (p<0.05). These results indicate farmers can provide twin-bearing ewes with pre- and post-grazing ryegrass-white clover herbage covers of approximately 1,100 and 800 kg DM/ha in late pregnancy, provided that herbage covers are 1400 in lactation, without affecting lamb performance to weaning. The present results also indicate that under these grazing conditions, there is little difference in ewe performance within the BCS range of 2.0 to 3.0 and therefore they do not need to be managed separately.

      • An experimental and simulation study of novel channel designs for open-cathode high-temperature polymer electrolyte membrane fuel cells

        <P>A minimum balance of plant (BOP) is desired for an open-cathode high temperature polymer electrolyte membrane (HTPEM) fuel cell to ensure low parasitic losses and a compact design. The advantage of an open-cathode system is the elimination of the coolant plate and incorporation of a blower for oxidant and coolant supply, which reduces the overall size of the stack, power losses, and results in a lower system volume. In the present study, we present unique designs for an open-cathode system which offers uniform temperature distribution with a minimum temperature gradient and a uniform flow distribution through each cell. Design studies were carried out to increase power density. An experimental and simulation approach was carried out to design the novel open-cathode system. Two unique parallel serpentine flow designs were developed to yield a low pressure drop and uniform flow distribution, one without pins and another with pins. A five-cell stack was fabricated in the lab based on the new design. Performance and flow distribution studies revealed better performance, uniform flow distribution, and a reduced temperature gradient across the stack; improving overall system efficiency.       • Genome-wide association study in a Chinese population identifies a susceptibility locus for type 2 diabetes at 7q32 near <i>PAX4</i>

        <P><B>Aims/hypothesis</B></P><P>Most genetic variants identified for type 2 diabetes have been discovered in European populations. We performed genome-wide association studies (GWAS) in a Chinese population with the aim of identifying novel variants for type 2 diabetes in Asians.</P><P><B>Methods</B></P><P>We performed a meta-analysis of three GWAS comprising 684 patients with type 2 diabetes and 955 controls of Southern Han Chinese descent. We followed up the top signals in two independent Southern Han Chinese cohorts (totalling 10,383 cases and 6,974 controls), and performed in silico replication in multiple populations.</P><P><B>Results</B></P><P>We identified <I>CDKN2A/B</I> and four novel type 2 diabetes association signals with <I>p</I> < 1 × 10<SUP>−5</SUP> from the meta-analysis. Thirteen variants within these four loci were followed up in two independent Chinese cohorts, and rs10229583 at 7q32 was found to be associated with type 2 diabetes in a combined analysis of 11,067 cases and 7,929 controls (<I>p</I><SUB>meta</SUB> = 2.6 × 10<SUP>−8</SUP>; OR [95% CI] 1.18 [1.11, 1.25]). In silico replication revealed consistent associations across multiethnic groups, including five East Asian populations (<I>p</I><SUB>meta</SUB> = 2.3 × 10<SUP>−10</SUP>) and a population of European descent (<I>p</I> = 8.6 × 10<SUP>−3</SUP>). The rs10229583 risk variant was associated with elevated fasting plasma glucose, impaired beta cell function in controls, and an earlier age at diagnosis for the cases. The novel variant lies within an islet-selective cluster of open regulatory elements. There was significant heterogeneity of effect between Han Chinese and individuals of European descent, Malaysians and Indians.</P><P><B>Conclusions/interpretation</B></P><P>Our study identifies rs10229583 near <I>PAX4</I> as a novel locus for type 2 diabetes in Chinese and other populations and provides new insights into the pathogenesis of type 2 diabetes.</P><P><B>Electronic supplementary material</B></P><P>The online version of this article (doi:10.1007/s00125-013-2874-4) contains peer-reviewed but unedited supplementary material, which is available to authorised users.</P>

        Linear Peptides Are the Major Products of a Biosynthetic Pathway That Encodes for Cyclic Depsipeptides

        <P>Three new dentigerumycin analogues are produced by Streptomyces sp. M41, a bacterium isolated from a South African termite, Macrotermes natalensis. The structures of the complex nonribosomal peptide synthetase polyketide synthase (NRPS/PKS) hybrid compounds were determined by 1D- and 2D-NMR spectroscopy, high-resolution mass spectrometry, and circular dichroism (CD) spectroscopy. Both cyclic and linear peptides are reported, and the genetic organization of the NRPS modules within the biosynthetic gene cluster accounts for the observed structural diversity.</P>

        Self-assembled silver nanoparticles in glass microstructured by poling for SERS application

        A simple technique to fabricate microchannels in glasses with self-assembled silver nanoparticles (NPs) in the channels is presented. It combines thermal-electric poling of silver-to-sodium ion-exchanged glass slides with a patterned anodic electrode, formation of the microchannels via selective etching off the unpoled slide regions, and hydrogen annealing. The annealing results in the growth of NPs only on the bottom of the channels. The studies performed allowed optimizing the channels’ depth and NPs surface density for Surface Enhanced Raman Scattering (SERS) based sensing and microfluidic applications. We have demonstrated that the formed NPs allow detection of 1/20 of BPE (1,2-Di(4-pyridyl)ethylene 97%) monolayer, the evaluated Raman enhancement factor being ~4·107. The proposed approach based on the glass poling allowed us the fabrication of ~1 μm deep channels and easy multiplication of the structures because the anodic electrodes used for the poling are capable of multiple usage.

        Non-uniform effect of COVID-19 lockdown on the air quality in different local climate zones of the urban region of Kochi, India

        Deterioration of ambient air quality associated with urbanisation is a serious concern in many parts of the world. In India, air pollution, primarily due to particulate matter, has increased exponentially in the last few decades due to rapid urbanization, industrialization and population growth. This study investigates the non-uniform influence of COVID-19 lockdown on the ambient air quality of three distinct local climate zones (LCZs) within the urban region of Kochi (Kerala, India). The analysis of the air pollutant data of the ambient air quality monitoring stations during the pre-lockdown (PRLD), lockdown (LD) and post-lockdown (PTLD) periods of 2021 implies the significance of lockdown measures in the improvement of urban air quality. The air quality index (AQI) exhibits characteristic variability in different LCZs and contrasting behaviour between the LD period of 2020 and 2021, primarily due to the differences in the lockdown restrictions and strategies as well as the influence of local climatic factors. This study highlights the need for multiple monitoring stations in the urban regions with respect to different LCZs to identify the urban air quality hot spots.

      • Organization of the Mammalian Metabolome according to Organ Function, Lineage Specialization, and Longevity

        Biological diversity among mammals is remarkable. Mammalian body weights range seven orders of magnitude and lifespans differ more than 100-fold among species. While genetic, dietary, and pharmacological interventions can be used to modulate these traits in model organisms, it is unknown how they are determined by natural selection. By profiling metabolites in brain, heart, kidney, and liver tissues of 26 mammalian species representing ten taxonomical orders, we report metabolite patterns characteristic of organs, lineages, and species longevity. Our data suggest different rates of metabolite divergence across organs and reveal patterns representing organ-specific functions and lineage-specific physiologies. We identified metabolites that correlated with species lifespan, some of which were previously implicated in longevity control. We also compared the results with metabolite changes in five long-lived mouse models and observed some similar patterns. Overall, this study describes adjustments of the mammalian metabolome according to lifespan, phylogeny, and organ and lineage specialization.
