A Fibrinolytic Enzyme from the Medicinal Mushroom Cordyceps militaris

In this study we purified a fibrinolytic enzyme from Cordyceps militaris using a combination of ion-exchange chromatography on a DEAE Sephadex A-50 column, gel filtration chromatography on a Sephadex G-75 column, and FPLC on a HiLoad 16/60 Superdex 75 column. This purification protocol resulted in a 191.8-fold purification of the enzyme and a final yield of 12.9 %. The molecular mass of the purified enzyme was estimated to be 52 kDa by SDS-PAGE, fibrin-zymography, and gel filtration chromatography. The first 19 amino acid residues of the N-terminal sequence were ALTTQSNV THGLATISLRQ, which is similar to the subtilisin-like serine protease PR1J from Metarhizium anisopliae var. anisopliase. This enzyme is a neutral protease with an optimal reaction pH and temperature of 7.4 and $37^{\circ}C$, respectively. Results for the fibrinolysis pattern showed that the enzyme rapidly hydrolyzed the fibrin $\alpha$-chain followed by the $\gamma$-$\gamma$ chains. It also hydrolyzed the $\beta$-chain, but more slowly. The A$\alpha$, B$\beta$, and $\gamma$ chains of fibrinogen were also cleaved very rapidly. We found that enzyme activity was inhibited by $Cu^{2+}$ and $Co^{2+}$, but enhanced by the additions of $Ca^{2+}$ and $Mg^{2+}$ ions. Furthermore, fibrinolytic enzyme activity was potently inhibited by PMSF and APMSF. This enzyme exhibited a high specificity for the chymotrypsin substrate S-2586 indicating it's a chymotrypsin-like serine protease. The data we present suggest that the fibrinolytic enzyme derived from the edible and medicinal mushroom Cordyceps militaris has fibrin binding activity, which allows for the local activation of the fibrin degradation pathway.

본 연구는 단열지질조사를 통한 대상지역의 지하수 단열 모델을 규명하여 지하수를 개발한 사례이다. 경기도 문산 동쪽에 위치하고 있는 산업시설물에서는 시설물 유지에 필요한 용수 수급용 지하수개발이 요구되었다. 지하수 개발 대상지역은 0.15 Km2 면적으로 제한되어 있었을 뿐만 아니라, 지하 매설물, 건물 등의 시설물을 피하여 선정, 개발해야 되는 매우 어려운 지리적 조건을 가지고 있었다. 지하수 개발 대상지역은 지하 매설물이 설치되어 있어 지구물리탐사가 불가한 곳으로 지질조사에만 의존하여 지하수개발을 수행하였다. 편마암의 엽리와 암상조사로 엽리 궤적도를 작성하여 조사대상지역에 발달하고 있는 남북방향 습곡축의 향사형 습곡이 인지되었다. 동서방향의 지질단면 분석에서는 아이스크림 스푼 모양의 F2 향사형 중첩습곡이 해석된다. 광역조사에서 인지된 N20oE, N30oW, NS 주향의 단층연장 상에 위치하는 평평한 조사대상지역에 수반성 인장단열들의 존재 가능성을 예측하였다. 우수향 운동감각을 갖고 있는 N20oE 단층은 F2 향사형 습곡에 발달하고 있는 두 조의 요곡성 공액형인 P-전단단층으로 해석된다. NE 주향 지질단면상에서, N20oE 단층과 엽리 간의 교각으로 약 100 m 심도에서 지하수저장이 가능한 삼각 프리즘형태의 공간형성이 가능함을 알수 있어, 중첩습곡의 서쪽 날개 쪽이 지하수개발 가능지역으로 예측하였다. 또한 N40oE 단층도 지하수공급원이 될 수 있다. 단열지질조사와 단면해석을 기초로 하여 N20oE와 N40oE 단층을 따라서 지하수 개발가능 위치를 선정하였으며, N20oE 단층선 상에 위치한 한 지점을 선정한 결과 이곳에서 145 ton/day의 지하수를 개발하였다. 결론적으로 개발에 성공한 지하수는 중첩습곡에 수반된 요곡단층과 엽리 교차로 형성된 지하수 저장고의 불투수 공간으로부터 공급된 것으로 해석되었다. This study is the case of groundwater development based on the geometrical fracture model of target area established only through geological fracture mapping technique. A fracture mapping of 9 km2, eastern Munsan, has been conducted to determine geological and hydrological factors for new water well placement in the Gyeonggi gneiss complex. Geophysical exploration was not applicable because of small restricted area and dense underground utilities at the site. Form line mapping on the basis of foliation orientation and rock type revealed a synform of NS fold axis bearing to the south. An EW geological cross-section passed through the site area shows a F2 synform as a double-wall ice cream spoon shape. Three regional faults of N20oE, N30oW and NS have been dragged into the site to help understand extensional fault paths. The N20oE fault with dextral sense is geometrically interpreted as a western fault of two flexural conjugate type-P shear faults in the F2 synformal fold. The NE cross-section reveals that a possible groundwater belt in the western limb of superposed fold area is formed as a trigonal prism within 100 m depth of the intersectional space between the N20oE fault plane the weakly sheared plane of transposed foliation. Another possible fault for water resource strikes N40oE. Recommended sites for new water well placement are along the N20oE and N40oE faults. As a result of fracture mapping, 145ton/day of water can be produced at one well along the N20oE fault line. Exploration of groundwater in the area is succeeded only using with geological fracture mapping and interpretation of geological cross-section, without any geophysical survey. Intersection of fault generated with the F2 synformal fold and foliation supply space of groundwater reservoir.
Is Routine Repeated Head CT Necessary for All Pediatric Traumatic Brain Injury?

Objective : Repeated computed tomography (CT) follow up for traumatic brain injury (TBI) patients is often performed. But there is debate the indication for repeated CT scans, especially in pediatric patients. Purpose of our study is to find risk factors of progression on repeated CT and delayed surgical intervention based on the repeated head CT. Methods : Between March, 2007 and December, 2013, 269 pediatric patients (age 0-18 years) had admitted to our hospital for head trauma. Patients were classified into 8 subgroups according to mechanisms of injury. Types, amount of hemorrhage and amount changes on repeated CT were analyzed as well as initial Glasgow Coma Scale (GCS) scores. Results : Within our cohort of 269 patients, 174 patients received repeat CT. There were progression in the amount of hemorrhage in 48 (27.6%) patients. Among various hemorrhage types, epidural hemorrhage (EDH) more than 10 cc measured in initial CT was found to be at risk of delayed surgical intervention significantly after routine repeated CT with or without neurological deterioration than other types of hemorrhage. Based on initial GCS, severe head trauma group (GCS 3-8) was at risk of delayed surgical intervention after routine repeated CT without change of clinical neurologic status. Conclusion : We suggest that the patients with EDH more than 10 cc or GCS below 9 should receive repeated head CT even though absence of significant clinical deterioration.

Cortical malformation-associated epileptic seizures are resistant to conventional anticonvulsant drugs. Relatively little research has been conducted on the effects of antiepileptic drugs (AEDs) on seizure activity in a rat model of dysplasia. We have used rats exposed to methylazoxymethanol acetate (MAM) in utero, an animal model featuring nodular heterotopia, to investigate the effects of ethosuximide (ETX) in the dysplastic brain. Pilocarpine was used to induce acute seizure in MAM-exposed and age-matched vehicle-injected control animals. Field potential recordings were used to monitor the amplitude and number of population spikes, and paired pulse inhibition in response to stimulation of the commissural pathway. Pharmaco-resistance was tested by measuring seizure latencies after pilocarpine administration (320 mg/kg, Lp.) with and without pre-treatment with ETX. Pre-treatment with 300 mg of ETX significantly prolonged the latency to the status epilepticus (SE) in both control and MAM-treated groups. Pre-treatment with ETX 100mg and ETX 200 mg had little effect in MAMexposed rats. However, ETX 200 mg prolonged the latency to the SE in control groups. Spontaneous field potential and secondary after-discharges were higher for MAM-treated rat in comparison with control rats injects with ETX. The main findings of this study are that acute seizures initiated in MAM-exposed rats are relatively resistant to standard ETX assessed in vivo. These data suggest that ETX do not prolong seizure latencies in MAM-rats exposed to pilocarpine.

Aluminum nitride (AlN) powder was easily synthesized by the direct nitridation of Al melt containing ~20 wt.% Mg catalyst and the nitriding behavior was investigated by thermodynamic calculation and through observations of electron microscopy and X-ray diffraction. The addition of Mg catalyst decreased the nitriding temperature below $1,000^{\circ}C$, which is comparable to the high nitriding temperature of $1,400^{\circ}C$ required in carbothermal method. It was caused by a significant increase of the solubility of nitrogen gas due to the increase of Mg catalyst in Al melt. The dissolved nitrogen gas met Mg catalyst and was transformed into metastable $Mg_3N_2$. Finally the metastable phase reacted with Al to AlN.
