      • Selective sorting of semiconducting single-walled carbon nanotubes using thienylenevinylene-based conjugated polymers with high alkyl side-chain density

        <P>The demand for high-purity semiconducting single-walled carbon nanotubes (sc-SWNTs) has increased considerably with the aim of utilizing their superior properties in a range of future applications. Among post-sorting methods, conjugated polymers have been regarded as one of the candidates to selectively isolate sc-SWNTs with uniform electrical properties. Herein, we demonstrate the ability to selectively sort two types of SWNTs by the two polymers PCTV18T and PC12TV18T, which have different alkyl side-chain densities. PC12TV18T, with a high alkyl chain density, shows great sorting ability for both high-pressure carbon monoxide and plasma-torch-grown SWNTs with a weight ratio of almost 1:1 in toluene solution. In addition, it is found that PC12TV18T selectively sorts sc-SWNTs with the high purity. The chirality and diameters of the enriched sc-SWNTs are further confirmed by Raman spectroscopy and photoluminescence excitation/emission mapping. Finally, we fabricate bottom gate/bottom contact thin-film transistors using the enriched sc-SWNTs as semiconductors to verify the electrical performance. Devices with well-percolated sc-SWNT networks displayed p-dominant properties with average charge-carrier mobilities of 2.05 cm(2) V-1 S-1 and 9.87 cm(2) V-1 S-1 and on/off current ratios of approximately 10(4) and 10(5) for high-pressure carbon monoxide and plasma-torch-grown SWNTs, respectively.         Apoptotic Effects of Resveratrol via mTOR and COX-2 Signal Pathways in MCF-7 Breast Cancer Cells

        식물에서 추출한 파이토케미컬은 암세포의 여러 신호전달 기작에 관여함으로써 apoptosis를 유도한다. 본 연구에서는 파이토케미컬의 한 종류인 레스베라트롤을 MCF-7 세포에 처리함으로써 암세포의 증식 억제와 apoptosis 유도 효과를 알아보았고, 이러한 효과가 암세포의 성장과 증식에 관여하는 단백질인 mTOR와 COX-2의 발현 양상에 어떠한 영향을 미치는지 알아보고자 하였다. 그 결과 MCF-7 세포에 레스베라트롤을 처리했을 때 농도가 증가함에 따라 암세포의 생존률이 감소하였고, Hoechst 33342를 이용한 chromatin 염색과 Annexin V-propodium iodide staning을 통하여 암세포의 세포증식 효과 apoptosis에 의해 유도된 것임을 알 수 있었다. MCF-7 세포에 레스베라트롤을 처리했을 때 mTOR 및 COX-2의 발현 양상을 확인하기 위해 Western blotting을 실시한 결과, 레스베라트롤의 농도가 높아짐에 따라 mTOR 및 COX-2의 발현이 감소함을 확인 하였다. 이와 같은 결과는 MCF-7 유방암 세포에서 레스베라트롤에 의한 암세포의 증식 억제 및 apoptosis 유도가 mTOR 신호경로 저해를 통한 COX-2의 발현을 감소시킴으로써 나타나는 것으로 보인다. Resveratrol, a kind of phytochemical, is presented in grape skins. Resveratorl exerts antiproliferative, anti-cancer and pro-apoptotic activities in cancer cells. Mammalian target of rapamycin (mTOR) is a critical regulator of cellular growth and proliferation, and it is known to be a strategic target for anti-cancer therapeutic uses. mTOR is a major downstream of the PI3K/Akt pathway, which is activated in various cancer cells. It also plays an important role in the survival, proliferation and angiogenesis of cells. Cyclooxygenase-2 (COX-2) is an important protein that mediates inflammatory processes. It plays an important role in various tumors by affecting cell proliferation, mitosis, apoptosis and angiogenesis. In this study, we have investigated the effects of resveratrol on apoptosis through mTOR and COX-2 expression in MCF-7 breast cancer cells. The treatment of resveratrol with different concentrations inhibited proliferation of MCF-7. The data showed that resveratrol induced apoptotic cell death of cancer cells and decreased mTOR and COX-2 expression. These results suggest that resveratrol induces apoptosis of MCF-7 breast cancer cells by inhibiting mTOR and COX-2 expression.

        Homo-succinic acid production by metabolically engineered <i>Mannheimia succiniciproducens</i>

        <P><B>Abstract</B></P> <P>Succinic acid (SA) is a four carbon dicarboxylic acid of great industrial interest that can be produced by microbial fermentation. Here we report development of a high-yield homo-SA producing <I>Mannheimia succiniciproducens</I> strain by metabolic engineering. The PALFK strain (<I>ldhA</I> <SUP> <I>-</I> </SUP>, <I>pta</I> <SUP> <I>-</I> </SUP>, <I>ackA</I> <SUP> <I>-</I> </SUP> <I>, fruA</I> <SUP> <I>-</I> </SUP>) was developed based on optimization of carbon flux towards SA production while minimizing byproducts formation through the integrated application of <I>in silico</I> genome-scale metabolic flux analysis, omics analyses, and reconstruction of central carbon metabolism. Based on <I>in silico</I> simulation, utilization of sucrose would enhance the SA production and cell growth rates, while consumption of glycerol would reduce the byproduct formation rates. Thus, sucrose and glycerol were selected as dual carbon sources to improve the SA yield and productivity, while deregulation of catabolite-repression was also performed in engineered <I>M. succiniciproducens</I>. Fed-batch fermentations of PALFK with low- and medium-density (OD<SUB>600</SUB> of 0.4 and 9.0, respectively) inocula produced 69.2 and 78.4g/L of homo-SA with yields of 1.56 and 1.64mol/mol glucose equivalent and overall volumetric SA productivities of 2.50 and 6.02g/L/h, respectively, using sucrose and glycerol as dual carbon sources. The SA productivity could be further increased to 38.6g/L/h by employing a membrane cell recycle bioreactor system. The systems metabolic engineering strategies employed here for achieving homo-SA production with the highest overall performance indices reported to date will be generally applicable for developing superior industrial microorganisms and competitive processes for the bio-based production of other chemicals as well.</P> <P><B>Highlights</B></P> <P> <UL> <LI> <I>Mannheimia succiniciproducens</I> was engineered to produce homo-succinic acid. </LI> <LI> Different carbon sources were examined for optimal succinic acid production. </LI> <LI> Catabolic repression deregulation allowed co-fermentation of sucrose and glycerol. </LI> <LI> High productivity and yield of homo-succinic acid were achieved. </LI> <LI> Membrane cell recycling fermentation resulted in productivity of 38.6g/L/h. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        A case report of an unusual temporomandibular joint mass: Nodular fasciitis

        Nodular fasciitis (NF) is a benign myofibroblastic proliferation that grows very rapidly, mimicking a sarcoma on imaging. It is treated by local excision, and recurrence has been reported in only a few cases, even when excised incompletely. The most prevalent diagnoses of temporomandibular joint (TMJ) masses include synovial chondromato- sis, pigmented villonodular synovitis, and sarcomas. Cases of NF in the TMJ are extremely rare, and only 3 cases have been reported to date. Due to its destructive features and rarity, NF has often been misdiagnosed as a more aggressive lesion, which could expose patients to unnecessary and invasive treatment approaches beyond repair. This report presents a case of NF in the TMJ, focusing on various imaging features, along with a literature review aiming to determine the hallmark features of NF in the TMJ and highlight the diagnostic challenges

      • Construction of High-density Linkage Map and QTL Analysis of Korean Watermelon using SNP and InDel Markers

        Watermelon (Citrullus lanatus) is an important fruit that grows in temperate tropical climates around the world. In this study, quantitative trait loci (QTL) analysis was performed using the genotypes and phenotypes of 87 F2 populations derived from CBARES1 and CBARES2 from the Korean watermelon resources. A total of 37 quantitative traits related to leaves, stems, flowers, and fruits were investigated. The single nucleotide polymorphism (SNP) locus and the insertion or deletion (InDel) locus were found by comparing the nucleotide sequences of the parent generation, CBARES1 and CBARES2. SNP markers were genotyped through resequencing and InDel markers genotyped through PCR and electrophoresis. A linkage map with a total length of 3036.90 cM was constructed consisting of 1,539 SNP markers and 80 InDel markers. InDel data from the actual experimental results were compared with SNP data to confirm the linkage distance and location on the chromosome. As a result, from the 80 InDel markers, 56 were matched to the SNP location and 24 slightly deviated. QTL analysis with the inclusive composite interval mapping (ICIM) method revealed that 37 traits showed significant QTL results in 73 regions of the chromosome. The epistasis QTL analysis was performed to supplement the part that did not show a clear logarithm of the odds ratio (LOD) value due to the additive effect. The linkage map and QTL created in this study are important resources for gene discovery and improvement of watermelon quality.

      • Anti-angiogenic effects of resveratrol in combination with 5-fluorouracil on B16 murine melanoma cells.

        <P>5-Fluorouracil (5-FU) has been used as a chemotherapeutic drug for various types of cancer, although the development of resistance remains a major limitation for its use in clinical settings. In the present study, the anti-angiogenic effects of resveratrol and 5-FU either alone or in combination were examined in a B16 murine melanoma model. Co-treatment using resveratrol and 5-FU inhibited cell proliferation more efficiently compared with use of either drug alone and the antiproliferative effect coincided with changes in the expression levels of AMP-activated protein kinase (AMPK), cyclooxygenase-2, vasodilator-stimulated phosphoprotein (VASP) and vascular endothelial growth factor (VEGF). Furthermore, co-treatment with resveratrol and 5-FU reduced tumor growth compared with that in the control group and this growth-inhibitory effect was associated with changes in the expression levels of AMPK, VASP and VEGF. Immunohistochemical staining for angiogenesis demonstrated that co-treatment with resveratrol and 5-FU reduced the number of microvascular vessels compared with that in the control group. These results suggested that co-treatment with resveratrol and 5-FU suppressed cell growth and angiogenesis in B16 murine melanoma tumors.</P>

        GDNF secreted by pre-osteoclasts induces migration of bone marrow mesenchymal stem cells and stimulates osteogenesis

        Bone resorption is linked to bone formation via temporal and spatial coupling within the remodeling cycle. Several lines of evidence point to the critical role of coupling factors derived from pre-osteoclasts (POCs) during the regulation of bone marrowderived mesenchymal stem cells (BMMSCs). However, the role of glial cell-derived neurotrophic factor (GDNF) in BMMSCs is not completely understood. Herein, we demonstrate the role of POC-derived GDNF in regulating the migration and osteogenic differentiation of BMMSCs. RNA sequencing revealed GDNF upregulation in POCs compared with monocytes/macrophages. Specifically, BMMSC migration was inhibited by a neutralizing antibody against GDNF in pre-osteoclast-conditioned medium (POC-CM), whereas treatment with a recombinant GDNF enhanced migration and osteogenic differentiation. In addition, POC-CM derived from GDNF knock-downed bone marrow macrophages suppressed BMMSC migration and osteogenic differentiation. SPP86, a small molecule inhibitor, inhibits BMMSC migration and osteogenic differentiation by targeting the receptor tyrosine kinase RET, which is recruited by GDNF into the GFRa1 complex. Overall, this study highlights the role of POC-derived GDNF in BMMSC migration and osteogenic differentiation, suggesting that GDNF regulates bone metabolism. [BMB Reports 2020; 53(12): 646-651]
