        Essential oil of Citrus limon Osbeck: An insecticidal natural product against Cryptolestes pusillus SchÖnherr

        This study investigated the insecticidal activity and mechanism of action of essential oil from Citrus limon Osbeck (C. limon oil). The fumigation method was used to determine the insecticidal activity of different concentrations of C. limon oil against adult Cryptolestes pusillus (C. pusillus). In addition, the effect of C. limon oil on the organs of adult C. pusillus was studied. The results showed that when the concentration of C. limon oil reached 0.6 μL/cm 3 , following fumigation for 4, 8, 12, 16, 20, and 24 h, the corrected mortality rates of adult C. pusillus were 13 %, 30 %, 47 %, 95 %, 97 %, and 100 %, respectively. Furthermore, C. limon oil caused vacuolization in the digestive tract and adipose cells, thus, as a plant-based insecticide, it has strong research and application value.

        Component Prototyping for the China Spallation Neutron Source Project

        The China Spallation Neutron Source (CSNS) complex consists of an H- linear accelerator, a rapid cycling synchrotron accelerating the beam to 1.6 GeV, a solid tungsten target station and instruments for spallation neutron applications. The facility operates at a 25-Hz repetition rate with an initial design beam power of 120 kW and is upgradeable to 500 kW. The primary challenge is to build a robust and reliable user-friendly facility with upgrade potential at a fraction of the \world standard" cost. Success of the project relies on the results of prototyping research & development (R&D) of key technical systems and components. This paper discusses the prototyping experiences of the past two and a half years.         Effects of moderate pressure on premeability and viability of Saccharomyces cerevisiae cells

        With CO2 and N2 as the pressure media, the effects of the moderate pressure (0.1-1.0MPa) and the holding time on the conductivities of the cell suspension of Saccharomyces cerevisiae CICC1447 and Saccharomyces cerevisiae CICC1339, as well as the absorbances of the supernatant (after centrifuged) at 280 nm (A280) and 260 nm (A260) were determined. The membrane permeability of Saccharomyces cerevisiae CICC1447 increased significantly and the cell leakage was aggravated with the pressure increase. For Saccharomyces cerevisiae CICC1339, the conductivity of the cell suspension, A280 and A260 of the supernatant fluctuated with the pressure increase; as a whole, they increased with pressure. Different from high pressure, a moderate pressure not only remarkably improved the permeability of the yeast cell membrane, but also kept yeast cell viability; moreover, the integrity of the yeast cell membrane could be maintained.

      • A Cluster-Based Countermeasure against MAC Layer Attacks in IEEE 802.11 MANETs

        <P>MANETs become a considerable interest nowadays. However, these types of networks are vulnerable to a large group of attacks, e.g., MAC layer attacks. Most of the previous researches related to security issues focus on detection of misbehaviors after it occurred. We propose a new way of thinking to evade occurrence of misbehaviors. A novel clustering algorithm is presented using analytic hierarchy process to elect clusterhead. Afterwards, the clusterhead is required to assign backoff values to originators in its transmission range, which are also the members in the corresponding cluster, rather than permitting the originators to choose backoff values. Meanwhile, the elected clusterhead also monitors whether originators obey the backoff values. With the proposed scheme, the misuse of the backoff in MAC layer in 802.11 DCF can be prevented.</P>

        Headspace volatiles influenced by infusion matrix and their release persistence: a case study of oolong tea

        The perceived aroma of oolong tea is primarilyand directly affected by its infusion matrix, and the releasepersistence of headspace volatiles can better illustrate thepersistent aroma. Headspace solid phase microextractioncoupled with gas chromatography–mass spectrometry wasperformed to analyze the headspace constituents of oolongtea. The presence of the infusion matrix seemed to preventthe headspace release of certain odorants. The release ofindole, nerolidol, and a-farnesene was also remarkablyenhanced or depressed (2.70, 1.56, and 0.69-fold in teainfusion versus in dry leaves). Moreover, the amount ofvolatile species gradually decreased with increased waterratio. Eight odorants were determined to be stable andpersistent during continuous infusion, whereas six weredetermined to be less persistent (gradually decreased orstopped releasing). The volatile dilution test further confirmedthe persistent release of nerolidol.

        Five new sesquiterpene lactones from Inula hupehensis

        Four new pseudoguaianolides (1–4), one newguaianolide (5), together with ten known compounds(6–15) were isolated from the aerial parts of Inula hupehensis. Their structures were elucidated mainly on the basisof 1D and 2D spectroscopic methods and circular dichroismanalysis. In addition, compounds 1–10 and 13 weretested for their inhibitory effects against LPS-induced NOproduction in RAW264.7 macrophages. Compounds 2, 6, 8and 9 exhibited significant inhibitory activities with IC50values in the range of 0.6–6.6 lM.

        The Expression of MicroRNA-155 in Plasma and Tissue Is Matched in Human Laryngeal Squamous Cell Carcinoma

        Purpose: Tumor-associated microRNAs have been detected in cancer, though whether plasma microRNA-155 (miR-155) could be a potential biomarker for laryngeal squamous cell carcinoma (LSCC) prognosis is unclear. We aimed to determine how miR-155 can be used to predict the clinical characteristics of patients with LSCC and correctly diagnose them. Materials and Methods: We collected tissue samples and peripheral blood samples before and after treatment from 280 LSCC cases and 560 controls. Real-time quantitative reverse transcription PCR was employed in this study to compare the relative expressionof miR-155. Results: A total of 280 LSCC patients and 560 age- and sex-matched controls were included in the study. The miR-155 level was more up-regulated in LSCC tissue than in the non-tumor tissues (13.6±2.4 vs. 3.1±0.80, p<0.001). Additionally, a significantly higher miR-155 level in plasma samples from LSCC patients than in those of the controls (8.9±1.25 vs. 1.8±0.8, p<0.001) was reported. Tissue miR-155 showed an area under the curve (AUC) of 0.933, with a sensitivity of 82.6% and a specificity of 89.2%. The AUC for plasma miR-155 was 0.757, with a sensitivity of 58.4% and a specificity of 69.5%. When early LSCC in TNM I stage was considered, tissue miR-155 showed an area under the curve of 0.804, with a sensitivity of 85.2% and a specificity of 87.3%. Conclusion: The expression of tissue and plasma miR-155 were significantly up-regulated in patients with LSCC. Our work will serve as a basis for further investigation, preferably large-scale validation in clinical trials.

      • Hiwi Knockdown Inhibits the Growth of Lung Cancer in Nude Mice

        Hiwi, a human homologue of the Piwi family, plays an important role in stem cell self-renewal and is overexpressed in various human tumors. This study aimed to determine whether an RNA interference-based strategy to suppress Hiwi expression could inhibit tumor growth in a xenograft mouse model. A rare population of $SSC^{lo}\;Alde^{br}$ cells was isolated and identified as lung cancer stem cells in our previous study. Plasmids containing U6 promoter-driven shRNAs against Hiwi or control plasmids were successfully established. The xenograft tumor model was generated by subcutaneously inoculating with lung cancer stem cell $SSC^{lo}\;Alde^{br}$ cells. After the tumor size reached about 8 mm in diameter, shRNA plasmids were injected into the mice via the tail vein three times a week for two weeks, then xenograft tumor growth was assessed. In nude mice, intravenously delivery of Hiwi shRNA plasmids significantly inhibited tumor growth compared to treatment with control scrambled shRNA plasmids or the vehicle PBS. No mice died during the experiment and no adverse events were observed in mice administered the plasmids. Moreover, delivery of Hiwi shRNA plasmids resulted in a significant suppressed expression of Hiwi and ALDH-1 in xenograft tumor samples, based on immunohistochemical analysis. Thus, shRNA-mediated Hiwi gene silencing in lung cancer stem cells by an effective in vivo gene delivery strategy appeared to be an effective therapeutic approach for lung cancer, and may provide some useful clues for RNAi gene therapy in solid cancers.

        Long Noncoding RNA HOXA11-AS Modulates the Resistance of Nasopharyngeal Carcinoma Cells to Cisplatin via miR-454-3p/c-Met

        To elucidate the mechanism of action of HOXA11-AS in modulating the cisplatin resistance of nasopharyngeal carcinoma (NPC) cells. HOXA11-AS and miR-454-3p expression in NPC tissue and cisplatin-resistant NPC cells were measured via quantitative reverse transcriptase polymerase chain reaction. NPC parental cells (C666-1 and HNE1) and cisplatin-resistant cells (C666-1/DDP and HNE1/DDP) were transfected and divided into different groups, after which the MTT method was used to determine the inhibitory concentration 50 (IC50) of cells treated with different concentrations of cisplatin. Additionally, a clone formation assay, flow cytometry and Western blotting were used to detect DDP-induced changes. Thereafter, xenograft mouse models were constructed to verify the in vitro results. Obviously elevated HOXA11-AS and reduced miR-454-3p were found in NPC tissue and cisplatin-resistant NPC cells. Compared to the control cells, cells in the si-HOXA11-AS group showed sharp decreases in cell viability and IC50, and these results were reversed in the miR-454-3p inhibitor group. Furthermore, HOXA11-AS targeted miR-454-3p, which further targeted c-Met. In comparison with cells in the control group, HNE1/DDP and C666-1/DDP cells in the si-HOXA11-AS group demonstrated fewer colonies, with an increase in the apoptotic rate, while the expression levels of c-Met, p-Akt/Akt and p-mTOR/mTOR decreased. Moreover, the si-HOXA11-AS-induced enhancement in sensitivity to cisplatin was abolished by miR-454-3p inhibitor transfection. The in vivo experiment showed that DDP in combination with si-HOXA11-AS treatment could inhibit the growth of xenograft tumors. Silencing HOXA11-AS can inhibit the c-Met/AKT/mTOR pathway by specifically upregulating miR-454-3p, thus promoting cell apoptosis and enhancing the sensitivity of cisplatin-resistant NPC cells to cisplatin.

      • Use of an Artificial Neural Network to Predict Risk Factors of Nosocomial Infection in Lung Cancer Patients

        Statistical methods to analyze and predict the related risk factors of nosocomial infection in lung cancer patients are various, but the results are inconsistent. A total of 609 patients with lung cancer were enrolled to allow factor comparison using Student's t-test or the Mann-Whitney test or the Chi-square test. Variables that were significantly related to the presence of nosocomial infection were selected as candidates for input into the final ANN model. The area under the receiver operating characteristic (ROC) curve (AUC) was used to evaluate the performance of the artificial neural network (ANN) model and logistic regression (LR) model. The prevalence of nosocomial infection from lung cancer in this entire study population was 20.1% (165/609), nosocomial infections occurring in sputum specimens (85.5%), followed by blood (6.73%), urine (6.0%) and pleural effusions (1.82%). It was shown that long term hospitalization (${\geq}22days$, P= 0.000), poor clinical stage (IIIb and IV stage, P=0.002), older age (${\geq}61days$ old, P=0.023), and use the hormones were linked to nosocomial infection and the ANN model consisted of these four factors. The artificial neural network model with variables consisting of age, clinical stage, time of hospitalization, and use of hormones should be useful for predicting nosocomial infection in lung cancer cases.
