        Biological Characteristics of Recombinant Arthrobotrys oligospora Chitinase AO-801

        Chitinase AO-801 is a hydrolase secreted by Arthrobotrys oligospora during nematode feeding, while its role remained elusive. This study analyzed the molecular characteristics of recombinant chitinase of Arthrobotrys oligospora (reAO-801). AO-801 belongs to the typical glycoside hydrolase 18 family with conserved chitinase sequence and tertiary structure of (α/β)<sub>8</sub> triose-phosphate isomerase (TIM) barrel. The molecular weight of reAO-801 was 42 kDa. reAO-801 effectively degraded colloidal and powdered chitin, egg lysate, and stage I larval lysate of Caenorhabditis elegans. The activity of reAO-801 reached its peak at 40℃ and pH values between 4-7. Enzyme activity was inhibited by Zn2<sup>+</sup>, Ca2<sup>+</sup>, and Fe3<sup>+</sup>, whereas Mg2<sup>+</sup> and K<sup>+</sup> potentiated its activity. In addition, urea, sodium dodecyl sulfate, and 2-mercaptoethanol significantly inhibited enzyme activity. reAO-801 showed complete nematicidal activity against C. elegans stage I larvae. reAO-801 broke down the C. elegans egg shells, causing them to die or die prematurely by hatching the eggs. It also invoked degradation of Haemonchus contortus eggs, resulting in apparent changes in the morphological structure. This study demonstrated the cytotoxic effect of reAO-801, which laid the foundation for further dissecting the mechanism of nematode infestation by A. oligospora.

      • Research and Application of GEP Algorithm Based on Cloud Model

        Aiming at the traditional GEP algorithm adopted fixed rate of mutation and crossover rate in the process of evolution, and ignored the dynamic change of individual fitness, which leaded to the presence of premature convergence and local optimization problem. By using the cloud adaptive strategy and cloud cross strategy of cloud model, a genetic algorithm based on cloud model (Cloud Model Gene Expression Programming, CMGEP) was proposed. The algorithm adjusted the mutation rate and crossover rate in evolution through the cloud adaptation strategy according to the change of dynamic, and timely calculated population similarity to achieve cloud cross to increase the diversity of population and jump out of the premature convergence. It was applied to the field of railway engineering and its results were compared with those obtained by traditional GEP Algorithm and CMGEP Algorithm. Experiments show that the algorithm can improve the adaptability and the prediction accuracy, it has better convergence.

      • Catalytic topological insulator Bi<sub>2</sub>Se<sub>3</sub> nanoparticles for <i>in</i> <i>vivo</i> protection against ionizing radiation

        <P><B>Abstract</B></P> <P>Bi<SUB>2</SUB>Se<SUB>3</SUB> nanoparticles (NPs) have attracted wide interests in biological and medical applications. Layer-like Bi<SUB>2</SUB>Se<SUB>3</SUB> with high active surface area is promising for free radical scavenging. Here, we extended the medical applications of Bi<SUB>2</SUB>Se<SUB>3</SUB> NPs further to <I>in vivo</I> protection against ionizing radiation based on their superior antioxidant activities and electrocatalytic properties. It was found that Bi<SUB>2</SUB>Se<SUB>3</SUB> NPs can significantly increase the surviving fraction of mice after exposure of high-energy radiation of gamma ray. Additionally, the Bi<SUB>2</SUB>Se<SUB>3</SUB> NPs can help to recover radiation-lowered red blood cell counts, white blood cell counts and platelet levels. Further investigations revealed that Bi<SUB>2</SUB>Se<SUB>3</SUB> NPs behaved as functional free radical scavengers and significantly decreased the level of methylenedioxyamphetamine. <I>In vivo</I> toxicity studies showed that Bi<SUB>2</SUB>Se<SUB>3</SUB> NPs did not cause significant side effects in panels of blood chemistry, clinical biochemistry and pathology.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Bi<SUB>2</SUB>Se<SUB>3</SUB> NPs can significantly increase the surviving fraction of mice up to 70% after Gamma radiation. </LI> <LI> Bi<SUB>2</SUB>Se<SUB>3</SUB> NPs can help to recover radiation-lowered red blood cell counts, white blood cell counts and platelet levels. </LI> <LI> Bi<SUB>2</SUB>Se<SUB>3</SUB> NPs behaved as functional free radical scavengers and significantly decreased the level of methylenedioxyamphetamine. </LI> </UL> </P> <P><B>Graphical Abstract</B></P> <P>Layer-like Bi<SUB>2</SUB>Se<SUB>3</SUB> with high active surface area can protect mice against ionizing radiation based on their superior antioxidant activities and electrocatalytic properties.         Map-based cloning and functional analysis of the chromogen gene C in rice (Oryza sativa L.)

        The chromogen gene C is critical for anthocyanin regulation in rice, and apiculus color is an important agronomic trait in selective breeding and variety purification. Mapbased cloning and in-depth functional analysis of the C gene will be useful for understanding the molecular mechanism of anthocyanin biosynthesis and for rice breeding. Japonica landrace Lijiangxintuanheigu (LTH) has red apiculi and purple stigmas. Genetic analysis showed that red apiculus and purple stigma in LTH co-segregated indicating control by a single dominant gene, or by two completely linked genes. Using 1,851 recessive individuals from two F2 populations, the target gene OsC was delimited to a 70.8 kb interval on chromosome 6 that contains the rice homologue of the maize anthocyanin regulatory gene C1. When the entire OsC gene and its full-length cDNA cloned from LTH were transformed into japonica cultivar Kitaake with colorless apiculi and stigmas all positive transformants had red apiculi but non-colored stigmas, validating that OsC alone was responsible for the apiculus color and represented the functional C gene. OsC was constitutively expressed in all tissues examined, with strongest expression in leaf blades. These results set a foundation to clarify the regulatory mechanisms of OsC in the anthocyanin biosynthetic pathway.

        Vertical Handoff Decision Algorithm combined Improved Entropy Weighting with GRA for Heterogeneous Wireless Networks

        Future network scenario will be a heterogeneous wireless network environment composed of multiple networks and multimode terminals (MMT). Seamless switching and optimal connectivity for MMT among different networks and different services become extremely important. Here, a vertical handoff algorithm combined an improved entropy weighting method based on grey relational analysis (GRA) is proposed. In which, the improved entropy weight method is used to obtain the objective weights of the network attributes, and GRA is done to rank the candidate networks in order to choose the best network. Through simulation and comparing the results with other vertical handoff decision algorithms, the number of handoffs and reversal phenomenon are reduced with the proposed algorithm, which shows a better performance.

        Different Characteristics of Psychological and Sleep Symptoms Across Social Media Addiction and Internet Gaming Disorder in Chinese Adolescents- A Network Analysis

        Objective Previous research has explored a variety of mental disorders associated with Internet Gaming Disoder (IGD) and Social Media Addiction (SMA). To date, few studies focused on the network characteristics and investigated mood and sleep symptoms across SMA and IGD of adolescence at a group-specific level. This study aims to identify different characteristics of IGD and SMA and further determine the group-specific psychopathology process among adolescents. Methods We conducted a cross-sectional study to recruit a cohort of 7,246 adolescents who were scored passing the cutoff point of Internet Gaming Disorder Scale-Short Form and Bergen Social Media Addiction Scale, as grouped in IGD and SMA, or otherwise into the control group. Patient Health Questionnaire-9, Generalized Anxiety Disorder 7-item, and Pittsburgh Sleep Quality Index were assessed for the current study, and all assessed items were investigated using network analysis. Results Based on the analytical procedure, the participants were divided into three groups, the IGD group (n=789), SMA group (n=713) and control group (n=5,744). The edge weight bootstrapping analysis shows that different groups of networks reach certain accuracy, and the network structures of the three groups are statistically different (pcontrol-IGD=0.004, pcontrol-SMA<0.001, pIGD-SMA<0.001). The core symptom of SMA is “feeling down, depressed, or hopeless”, while IGD is “feeling tired or having little energy”. Conclusion Although IGD and SMA are both subtypes of internet addiction, the psychopathology processes of IGD and SMA are different.         Effects of Pressure and Deposition Time on the Characteristics of In2Se3 Films Grown by Magnetron Sputtering

        Crystalline In2Se3 films were fabricated by magnetron sputtering from a sintered In2Se3-compound target and the effects of the deposition parameters, including the working pressure and deposition time, on the phase composition, structure, morphology, and optical properties were clarified. Single-phase κ-In2Se3 was prepared at 4.0 Pa, but γ-In2Se3 was recognized when the working pressure was lower than 4.0 Pa. The optical transmittance of the films decreased to 45% and the optical band gap varied from 2.9 to 2.0 eV with increasing film thickness from 80 to 967 nm. Metal-semiconductor-metal (MSM) photodetectors based on γ-In2Se3 thin films with various thicknesses were also fabricated. The result of photosensitivity research on such MSM photodetectors suggests that it may be impossible to fabricate wide-absorption-range MSM devices by just using a single material (γ-In2Se3) because of spatial potential fluctuations in the layers.

        Multi-Epitope Fusion Protein Eg mefAg-1 as a Serodiagnostic Candidate for Cystic Echinococcosis in Sheep

        Cystic echinococcosis (CE) in sheep is a hazardous zoonotic parasitic disease that is caused by Echinococcus granulosus (Eg). At present, serological test is an important diagnostic method for Eg infection in domestic animals. Here, a fusion protein Eg mefAg-1 harboring 8 dominant B-cell epitopes of Eg such as antigen B, tetraspanin 1, tetraspanin 6, reticulon and Eg95 was produced in E. coli and evaluated for CE in sheep by indirect ELISA. Eg mefAg-1 showed in ELI- SA a high sensitivity (93.41%) and specificity (99.31%), with a coincidence rate of 97.02%. Overall, it is suggested that the Eg mefAg-1 could be a potential antigen candidate for CE serodiagnosis in sheep.

        The effect of Glomus intraradices on the physiological properties of Panax ginseng and on rhizospheric microbial diversity

        Background: Glomus intraradices is a species of arbuscular mycorrhizal fungi that, as an obligate endomycorrhiza, can form mutually beneficial associations with plants. Panax ginseng is a popular traditional Chinese medicine; however, problems associated with ginseng planting, such as pesticide residues, reduce the ginseng quality. Methods: In this experiment, we studied the effect of inoculating G. intraradices on several physiological properties and microbial communities of ginseng. UV-Visible Spectrum method was used to detect physical properties. Denaturing gradient gel electrophoresis method was used to analyze microbial communities. Results: The results indicated that inoculation with G. intraradices can improve the colonization rate of lateral ginseng roots, increase the levels of monomeric and total ginsenosides, and improve root activity as well as polyphenol oxidase and catalase activities. We also studied the bacterial and fungal communities in ginseng rhizospheric soil. In our study, G. intraradices inoculation improved the abundance and Shannon diversity of bacteria, whereas fungi showed a reciprocal effect. Furthermore, we found that G. intraradices inoculation might increase some beneficial bacterial species and decreased pathogenic fungi in rhizospheric soil of ginseng. Conclusion: Our results showed that G. intraradices can benefit ginseng planting which may have some instructive and practical significance for planting ginseng in farmland.
