Radioprotective and Antioxidant Activity of Fractionated Extracts of Berries of Hippophae rhamnoides

        Rakesh Kumar Sharma,Raman Chawla,Rajesh Arora,Shikha Singh,Ravinder Kumar Sagar,Raj Kumar,Ashok Sharma,Manju L. Gupta,Surender Singh,Jagdish Prasad,Haider A. Khan,Anand Swaroop,A.K. Sinha,A.K. Gupta,R 한국식품영양과학회 2007 Journal of medicinal food Vol.10 No.1

Plants are an abundant source of medicinal compounds, some of which are useful in combating free radical-1002 (flavonoid-poor fraction) of Hippophae rhamnoideswere screened on the basis of their reducing power in the aqueousphase. REC-1001 was selected for further study, since it exhibited 27.38 times higher antioxidant activity than REC-1002.REC-1001 also showed significant (P. .05) membrane protection potential at 50 .g/mL, which was attributed to its abilityto scavenge peroxyl radicals (64.82. 1.25% scavenging within 1,440 min). A significant (P. .05) difference of 67.02% infree radical scavenging activity at 1,000 ng/mL between REC-1001 and vitamin E demonstrated the extract fraction’s worthc-tion. Further, REC-1001 was found to be nontoxic up to 200 mg/kg of body weight. This research suggests that the REC-1001 fraction of H. rhamnoidesextract is a safe and effective antioxidant nutraceutical product.

Role of Cytocentrifugation Combined with Nuclear Fast Picroindigocarmine Staining in Detecting Cryptozoospermia in Men Diagnosed with Azoospermia

        Sharma Rakesh Kumar,Gupta Sajal,Agarwal Ashok,Finelli Renata,Kuroda Shinnosuke,Saleh Ramadan,Boitrelle Florence,Kavoussi Parviz,Gül Murat,Tadros Nicholas,Ko Edmund,Farkouh Ala’a,Henkel Ralf,Arafa Moha 대한남성과학회 2022 The World Journal of Men's Health Vol.40 No.4

Purpose: Azoospermia is defined as the absence of spermatozoa in the pellet of a centrifuged semen sample. In fact, when a basic semen analysis fails to detect sperm in the ejaculate, there is still the possibility of detecting rare sperm after centrifugation of the sample and examination of the pellet. In this study, we assessed the role of Cytospin centrifugation in combination with the nuclear fast picroindigocarmine (NF-PIC) staining in identifying sperm in azoospermic samples. Materials and Methods: Semen samples of 251 men diagnosed as having azoospermia after standard examination were further analyzed by Cytospin centrifugation in combination with NF-PIC staining. Results: Sperm were detected in 60 men (23.9%), thus changing their diagnosis to cryptozoospermia. Conclusions: By identifying sperm in the semen of men who were thought to have total azoospermia, the Cytospin NF-PIC test can alter the diagnosis and further treatment of these men.

Can a brief period of double J stenting improve the outcome of extracorporeal shock wave lithotripsy for renal calculi sized 1 to 2 cm?

        Rakesh Sharma,Arpan Choudhary,Ranjit Kumar Das,Supriya Basu,Ranjan Kumar Dey,Rupesh Gupta,Partha Pratim Deb 대한비뇨의학회 2017 Investigative and Clinical Urology Vol.58 No.2

Purpose: Extracorporeal shock wave lithotripsy (ESWL) is an established modality for renal calculi. Its role for large stones is being questioned. A novel model of temporary double J (DJ) stenting followed by ESWL was devised and outcomes were assessed. Materials and Methods: The study included 95 patients with renal calculi sized 1 to 2 cm. Patients were randomized into 3 groups. Group 1 received ESWL only, whereas group 2 underwent stenting followed by ESWL. In group 3, a distinct model was applied in which the stent was kept for 1 week and then removed, followed by ESWL. Procedural details, analgesic requirements, and outcome were analyzed. Results: Eighty-eight patients (male, 47; female, 41) were available for analysis. The patients' mean age was 37.9±10.9 years. Stone profile was similar among groups. Group 3 received fewer shocks (mean, 3,155) than did group 1 (mean, 3,859; p=0.05) or group 2 (mean, 3,872; p=0.04). The fragmentation rate was similar in group 3 (96.7%) and groups 1 (81.5%, p=0.12) and 2 (87.1%, p=0.16). Overall clearance in group 3 was significantly improved (83.3%) compared with that in groups 1 (63.0%, p=0.02) and 2 (64.5%, p=0.02) and was maintained even in lower pole stones. The percentage successful outcome in groups 1, 2, and 3 was 66.7%, 64.5%, and 83.3%, respectively (p=0.21). The analgesic requirement in group 2 was higher than in the other groups (p=0.00). Group 2 patients also had more grade IIIa (2/3) and IIIB (1/2) complications. Conclusions: Stenting adversely affects stone clearance and also makes the later course uncomfortable. Our model of brief stenting followed by ESWL provided better clearance, comfort, and a modest improvement in outcome with fewer sittings and steinstrasse in selected patients with large renal calculi.

Black Carbon Concentration during Spring Season at High Altitude Urban Center in Eastern Himalayan Region of India

        Khushboo Sharma,Rakesh Kumar Ranjan,Sargam Lohar,Jayant Sharma,Rajeev Rajak,Aparna Gupta,Amit Prakash,Alok Kumar Pandey 한국대기환경학회 2022 Asian Journal of Atmospheric Environment (AJAE) Vol.16 No.1

This study analyzed the BC associated with PM1 and the contribution of biomass burning to the BC using a portable seven-channel Dual spot Aethalometer in and around Gangtok, the capital city of Sikkim, India, during April 2021. Additionally, CO2 and meteorological parameters (Temperature, Pressure, and Relative Humidity) was measured. The minimum concentration of BC was found in rural areas where the contribution of biomass burning to the BC is highest. The observed spatial variability of BC over Gangtok Municipal Corporation (GMC) area is minimal. Five days back-trajectory analysis was done using the Hybrid Single Particle Lagrangian Integrated Trajectory (HYSPLIT) model to understand the regional influences of air masses at Gangtok. The air mass of the studied region is under influence of trans-regional transport from Indo-Gangetic Plains affecting the BC concentration over the studied region. The black carbon presence in the ambient air near the glacier heights in the Eastern Himalayan region may significantly cause localized warming, thereby enhancing glacier melts. The results have significant bearing for the policy-makers to take corrective steps in addressing the issue of rising BC concentration in high altitude regions. A further detailed study is needed to examine the effect of BC on radiative forcing and its large-scale effect on the East Asian summer monsoon using regional climate models.

      • Interactions between Oxidative Stress, Lipid Profile and Antioxidants in Breast Cancer: A Case Control Study

Gupta, Rakesh Kumar,Patel, Amit Kumar,Kumari, Rajni,Chugh, Seema,Shrivastav, Chitrangada,Mehra, Siddharth,Sharma, Ajay Narayan Asian Pacific Journal of Cancer Prevention 2012 Asian Pacific journal of cancer prevention Vol.13 No.12

Oxidant/antioxidant balance has been suggested as an important factor for initiation and progression of cancer. The objective of this study was to determine changes in the levels of malondialdehyde (MDA), nitric oxide (NO), total cholesterol, triglycerides, LDL-cholesterol, HDL-cholesterol, total antioxidant capacity (TAC), glutathione peroxidase (GSH-Px), and superoxide dismutase (SOD) activities in serum samples of breast cancer patients (n=30) and healthy subjects (n=100). MDA and NO levels were found to be increased in breast cancer patients compared to the healthy subject group (p<0.05). Total cholesterol and triglycerides were elevated; and HDL-cholesterol level was found to be decreased in the cancer patients as compared to the healthy subjects (p<0.05). Compared to the healthy group, both serum TAC levels (p<0.001) and activity of SOD and GSH-Px (p=0.05) were found to be decreased in the breast cancer patients as compared to the healthy controls. Considering the data presented in this study, we suggest that free radicals induce lipid eroxidation and peroxidation of unsaturated fatty acid with decreased activity of enzymatic antioxidants in breast cancer.

Effectiveness of various nonpharmacological analgesic methods in newborns

Kumar, Pancham,Sharma, Rakesh,Rathour, Sukhdev,Karol, Sunidhi,Karol, Mohit The Korean Pediatric Society 2020 Clinical and Experimental Pediatrics (CEP) Vol.63 No.1

Background: Pain during the developmental period may adversely affect developing neuronal pathways and result in adverse neurodevelopmental, cognitive, and behavioral effects in later life. Immunizations, e.g., hepatitis B vaccine (HBV), administered at birth are painful experiences to which neonates are universally subjected. Purpose: Here we aimed to study and compare the effectiveness of various nonpharmacological pain management methods in newborns to enable the development of safe and effective analgesic methods for newborns. Methods: This prospective study was conducted at a tertiary care hospital in the Himalayan region. Three hundred term healthy neonates were divided into 6 groups of 50 each. Groups 1-5 were intervention groups, patients of which received a nonpharmacological intervention (breastfeeding, nonnutritive sucking, rocking, 25% sucrose, or distilled water) before the intramuscular HBV, while patients in group 6 received no intervention. The pain response in each group after the HBV injection was assessed and compared using cry duration and Douleur Aigue Nveau-ne (DAN) score, a behavioral acute pain rating scale for newborns. Results: Cry duration was decreased in all intervention groups, significantly so in the sucrose (19.90 seconds), breastfeeding (31.57 seconds), and nonnutritive sucking (36.93 seconds) groups compared with controls (52.86 seconds). DAN scores decreased significantly (P<0.05) at one or more points i.e. 30, 60, or 120 seconds in the breastfeeding and 25% sucrose intervention groups compared with controls. Conclusion: Oral sucrose and nonnutritive sucking are simple yet underutilized nonpharmacological interventions that effectively reduce pain in newborns.

Image-Guided Nanoparticle-Based siRNA Delivery for Cancer Therapy.

Tekade, Rakesh Kumar,Maheshwari, Rahul G S,Sharma, Piyoosh A,Tekade, Muktika,Chauhan, Abhay Singh Bentham Science Publishers 2015 CURRENT PHARMACEUTICAL DESIGN Vol.21 No.31

With the discovery of RNA interference technology, small-interfering RNA (siRNA) has emerged as new powerful tool for gene therapy because of its high targeting specificity and selectivity. However, one of the limitations to successful gene therapy is the inability to monitor delivery of genes and therapeutic responses at the targeted site. Hence, a combinatorial approach of gene therapy with molecular imaging has been crucial in optimizing gene therapy. Recent advances in nanotechnology have made tremendous efforts to develop multifunctional nanoparticles that contain imaging and therapeutic agents together for image-guided therapy. The nanoparticles serve as contrast agents in imaging for disease detection with simultaneous delivery of therapeutics to cure the diseases. The therapy also helps to monitor the drug accumulation and assimilation in the body, thereby facilitating the evaluation of treatment effects. Here, we present an overview of polymer and lipid-based carriers for siRNA delivery, along with imaging agents as imageguided therapy, in the treatment of breast, lung, liver, ovarian, cervical, and prostate cancers.

Friction and wear properties of core-shell (CI is a core & GO is a shell) particles based magnetorheological fluid under steel on steel point contacts

Sanjay Kumar,Rakesh Sehgal,M.F. Wani,Mukund Dutt Sharma 한국공업화학회 2023 Journal of Industrial and Engineering Chemistry Vol.118 No.-

This work is concerned with the experimental investigation of tribological and rheological properties ofcore–shell (carbonyl iron is the core and graphene oxide is the shell) particle-based MR fluid. Carbonyliron (CI) particles were encapsulated with graphene oxide (GO) by a simple wet stirring process. The surfacemorphology, structural properties, and magnetic properties of graphene oxide encapsulated CI particleswere analyzed by Field Emission Spectroscopy (FE-SEM), Raman Spectroscopy, and VibrationalSample Magnetometer (VSM). The rheological properties of prepared samples were examined on a modularcompact rheometer. The CI@GO particle-based MR fluid exhibited lower yield stress compared tobare CI particle-based MR fluid. Tribological tests of developed MR fluid were conducted on a universaltribometer with and without the presence of a magnetic field to ascertain the friction and wear behaviour. Experimental results revealed that the tribological properties are significantly improved by usingCI@GO-based MR fluid in comparison to bare CI-based MR fluid and dry sliding conditions. Further, thewear analysis of worn surfaces was observed on FE-SEM, Energy-Dispersive X-ray Spectroscopy (EDS),and 3D-profilometer before and after each test. Three body abrasion wear mechanism was observed withsome ploughing and groove formation.
