      • Carotenoids 식이와 알코올이 혈액학적 성분과 간조직에 미치는 영향에 관한 연구

        Alcohol is well known agent which can damage the human tissues such as liver via stimulating lipid peroxidation and storage, denaturation of macromolecules, and inhibiting protein metabolism. On the other hand, carotenoids in addition to vitamins A, C, E and lipoic acid, play important roles in protecting these oxidative damages as well as preventing the production of free radicals. This study was carried out to elucidate the precise effects of alcohol administration into rats on the antioxidative functions of dietary carotenoids and isolated β-carotene, and to find out any parameters to uncover more detailed biochemical mechanisms of these agents. For these purposes, two different approaches were performed: 1) examination of the changes in hematological parameters (e.g., total proteins, A/G ratio, ALT/AST et. al) and statistical correlations among inter- and intragroups, 2) examination of the histopathological changes by an electron microscope. The results were analyzed and summarized as following; (1) Carrot diet for one week caused a slight increase in albumin. A/G ratio and AST levels, and a slight decrease in ALT, ALP, BUN and uric acid levels. Two-week alcohol administration following carrot diet increased total proteins, albumin A/G ratio and BUN levels. However, both groups were shown to have little significant changes in cholesterol concentrations. (2) Cheese diet for one week caused a significant increase in total proteins, albumin, AST, ALP, uric acid and total cholesterol levels, and a significant decrease in A/G ratio and glucose concentrations. However, two-week β-carotene diet following cheese intake increased total proteins albumin, A/G ratio, BUN and total cholesterol levels, whereas AST, ALT, ALP and uric acid levels were decreased. (3) Alcohol administration for one week caused an increase in AST activities and a decrease in total proteins, albumin, ALT, ALP and BUN levels. A successive β-carotene diet following alcohol administration increased total proteins, albumin, ALT, ALP and BUN levels, whereas decreased AST and uric acid levels. However, there were not significant changes in A/G ratio, glucose and total cholesterol levels in both groups. (4) β-carotene diet for one week caused a slight increase in albumin, glucose, AST and uric acid levels, and a slight decrease in ALP and BUN levels. Two-weeks alcohol administration following β-carotene diet increased albumin, glucose, BUN and total cholesterol levels, whereas decreased AST, ALT, ALP and uric acid levels. (5) Although the pathological investigation on the liver did not reveal significant changes, cheese diet group (CH-BC/1-CH) was shown to have some lipid deposits. Some results were unexpected and different from typical hematological changes shown by other researchers. Nonetheless, these results strongly suggest that the liver damage or hepatism caused by alcohol intake affects many kinds of biochemical metabolisms, which results in significant changes in many hematological parameters. In addition, dietary carotenoid and isolated β-carotene were shown to have protective roles against the biochemical changes by alcohol intake.

      • Using Real-Time Electron Microscopy To Explore the Effects of Transition-Metal Composition on the Local Thermal Stability in Charged Li<sub><i>x</i></sub>Ni<sub><i>y</i></sub>Mn<sub><i>z</i></sub>Co<sub>1–<i>y</i>–<i>z</i></sub>O<sub>2</sub

        <P>In this work, we use in situ transmission electron microscopy (TEM) to investigate the thermal decomposition that occurs at the surface of charged LixNiyMnzCo1-y-zO2 (NMC) cathode materials of different composition (with y, z = 0.8, 0.1, and 0.6, 0.2, and 0.4,and 0.3), after they have been charged to their practical upper limit voltage (4.3 V). By heating these materials inside the TEM, we are able to directly characterize near surface changes in both their electronic structure (using electron energy loss spectroscopy) and crystal structure and morphology (using electron diffraction and bright-field imaging). The most Ni-rich material (y, z = 0.8, 0.1) is found to be thermally unstable at significantly lower temperatures than the other compositionsthis is manifested by changes in both the electronic structure and the onset of phase transitions at temperatures as low as 100 degrees C. Electron energy loss spectroscopy indicates that (i) the thermally induced reduction of Ni ions drives these changes, and (ii) this is exacerbated by the presence of an additional redox reaction that occurs at 4.2 V in the y, z = 0.8, 0.1 material. Exploration of individual particles shows that there are substantial variations in the onset temperatures and overall extent of these changes. Of the compositions studied, the composition of y, z = 0.6, 0.2 has the optimal combination of high energy density and reasonable thermal stability. The observations herein demonstrate that real-time electron microscopy provide direct insight into the changes that occur in cathode materials with temperature, allowing optimization of different alloy concentrations to maximize overall performance.</P>

      • Thermoplasma acidophilum의 20S Proteasome에 의한 ATPase-활성적 단백질분해에 관한 연구

        The eukaryotic 26S proteasome is an ATP/ubiquitin-dependent proteolytic complex consisting of the 20S core particle and 19S ATPase complex. However, because of its complexity and unstable properties, this study was carried out to present more simple and stable model for the ATP-activated proteolytic complex in prokaryotes which can take the place of the eukaryotic 26S proteasome. For this purpose, recombinant Thermoplasma 20S proteasome (T20S) and Methanococcus MS4, a sequence homolog of one ATPase subunit in the 19S ATPase complex, were successfully isolated from Escherichia coli (E. coli). The α and β subunits of T20S expressed in E. coli could assemble for themselves, and showed the peptide-hydrolyzing activity. Whereas both T20S and R20S (the 20S complex from rabbit skeletal muscle) had the highest peptidase activity against Suc-LLVY-AMC, a good substrate for chymotrypsin-like peptidase activity, the specific activity of T20S was slightly lower than that of R20S. In addition, several reagents such as KCI, SDS, and ovalbumin were shown to have different effects on the peptidase activities between T20S and R20S. When the ATPase activity of the purified MS4 were assayed , the Km for ATP was about 0.5 mM, and casein could stimulate the activity more than 2 fold without the change in Km. This result implicates that protein-activated ATPase may induce the conformational change of casein, and therefore suggests that MS4 ATPase may activate the proteolytic activity of the 20S proteasome via accelerating the recognition and translocation of the protein substrates.

        천연 갈변 억제제 개발을 위한 양파,사과 및 감귤 과피 추출물의 항산화 및 갈변 저해 효과

        This study was carried out to investigate antioxidative properties of various extracts and antibrowning effects of extracts in apple slices were investigated by ΔE value and PPO relative activity. Apples were cut into 1.5 ㎝ thickness and they were dipped in 1% extract solutions(OW: water extracts of onion, OE: 80% EtOH extracts of onion, AW: water extracts of apple, AE: 80% EtOH extracts of apple, MW: water extracts of mandarin orange peel, ME: 80% EtOH extracts of mandarin orange peel) for 1 min. OW showed higher than the other treatments for total phenolic contents(94.35 ㎎/g), PPO inhibition(74.00%). And the highest DPPH free radical scavenging activity(40.27%) measured in ME. ΔE value of apple slices dipped in MW was 2.37 whereas ΔE value of apple slices dipped in AW was 12.12. These results suggest that onion and mandarin orange peel extracts should be a potential source for controlling browning during storage of apple slices.

        Original Article : Morphologic Characteristics of the Outer Retina in Cone Dystrophy on Spectral-domain Optical Coherence Tomography

        Purpose: To investigate the morphologic changes in the outer retina of patients with cone dystrophy, using spectral-domain optical coherence tomography (SD-OCT). Methods: The medical records of 15 cone dystrophy patients examined from January 2007 to January 2012 were reviewed retrospectively. All patients underwent ophthalmic evaluation including best-corrected visual acuity (BCVA), color vision testing, fundus examination, full-field standard electroretinography (ERG), multifocal (mf) ERG, and SD-OCT. Qualitative and quantitative SD-OCT data and ERG responses were analyzed and compared among the patient categories and the normal control group. Results: There were 4 major categories of SD-OCT findings, based on the status of the ellipsoid portion of the photoreceptor inner segment (ISe), outer segment (OS) contact cylinder, and retinal pigment epithelium (RPE) layer. Category 0 showed no structural abnormalities. Category 1 showed foveal ISe loss and obscurity of the border between the ISe band and the external limiting membrane (ELM). Category 2 showed foveal thinning and focal foveal ISe disruption with an intact ELM. Category 3 showed foveal thickening and perifoveal disruption of the ISe layer. Category 1 to 3 showed OS contact cylinder layer absence and RPE thickening. The patients in category 0 tended to be younger (mean, 10.0 years) than those in categories 1 to 3 (mean, 17.6 years), although this difference was not statistically significant. Category 1 to 3 patients exhibited statistically significant thinning of the central retina and outer nuclear layer and thickening of the RPE layer relative to the category 0 and normal control group. There was a significant correlation between the central foveal thickness and BCVA in the patients with cone dystrophy. ERG and mfERG responses did not differ significantly among the different cone dystrophy categories. Conclusions: The morphologic features of cone dystrophy as revealed by SD-OCT, could be categorized as either normal or 1 of 3 different types of outer retinal changes. The presence of normal retinal structures in young cone dystrophy patients with functional impairment (category 0) indicates that electrophysiologic studies are superior to current imaging modalities for the early diagnosis of cone dystrophy. The characteristic SDOCT findings in cone dystrophy patients may aid in differential diagnosis and be useful for future research on the pathology of cone dystrophy.

        Poly(vinylbenzyl chloride-glycidyl methacrylate)/Polyethylene Composite Anion Exchange Membranes for Vanadium Redox Battery Application

        Anion exchange membranes for a vanadium redox flow battery (VRB) were prepared by pore-filling on a PE substrate with the copolymerization of vinylbenzyl chloride (VBC) and glycidyl methacrylate (GMA). The ion exchange capacity, water uptake and weight gain ratio were increased with a similar tendency up to 65% of GMA content, indicating that the monomer improved the pore-filling degree and membrane properties. The vanadium ion permeability and open-circuit voltage were also investigated. The permeability of the VG65 membrane was only $1.23{\times}10^{-7}\;cm^2\;min^{-1}$ compared to $17.9{\times}10^{-7}\;cm^2\;min^{-1}$ for Nafion 117 and $1.8{\times}10^{-7}\;cm^2\;min^{-1}$ for AMV. Consequently, a VRB single cell using the prepared membrane showed higher energy efficiency (over 80%) of up to 100 cycles compared to the commercial membranes, Nafion 117 (ca. 58%) and AMV (ca. 70%).

        Concerns about global climate change and potential legislation that may impact the use of refrigerants with high global warming potential (GWP) have accelerated the development of new refrigerants with reduced impact on climate change. The most common refrigerant used globally in residential and light commercial air conditioning systems and heat pumps is R410A, with a GWP of 2088. Regulations are already in place to limit to GWP to below 750 in many regions globally, and there may be further reductions needed in the future. This paper discusses alternative fluids that have been developed to most nearly meet the most often cited requirements for environmental, physical, and performance properties for air conditioning and heat pump applications. R454B, with a GWP of 466 and both performance and properties similar to R410A is a potential alternative to meet these requirements while maintaining compatibility with systems similar to those of existing R410A designs to enable an orderly transition to lower GWP.

      • 담배 박각시나방 Manduca sexta의 계획된 세포죽음동안의 Proteasome의 변화

        모든 진핵세포의 주요 단백질 분해요소인 proteasome은 변성되거나 상해된 불필요한 단백질을 제거할 뿐만 아니라, 세포주기를 비롯한 다양한 대사과정들의 주요 효소들을 필요 시 적절히 분해함으로써 항상성을 유지시키는 필수적인 효소로 잘 알려져 있다. 또한 proteasome은 특정 포유동물 조직의 계획된 세포죽음(programmed cell death 또는 apoptosis)와 밀접하게 관련되어 있다고 보고된 바, 본 연구에서는 담배 박각시나방(Manduca sexta)의 변태과정에서 일어나는 마디간 근육(intersegmental muscle)의 급격한 파괴과정동안의 proteasome의 변화양상을 발현 및 단백질 분해활성을 중점으로 조사하였다. 정상근육과 파괴직전의 근육으로부터 proteasome을 분석한 결과 단백질 분해활성이 최소한 6배 이상 증가하였으며, proteasome의 주요 구성성분들의 발현 변화를 각각의 항체(antibody)를 이용하여 조사한 결과 세포죽음 직전에 급격히 증가함을 보였다. 이러한 구성성분들의 발현증가가 proteasome 복합체 형성을 통해 활성의 증가를 일으키는지 조사한 결과, 세포내에서 실질적으로 단백질 분해활성을 담당한다고 여겨지는 26S proteasome(2,000 kDa)의 위치에서 커다란 활성의 변화와 구성성분의 증가를 볼 수 있었다. 따라서, 담배 박각시나방의 계획된 세포죽음 현상은 특정 호르몬의 신호에 의해 proteasome의 발현이 급격히 증가하여 26S proteasome의 복합체 형태로 세포 내 대부분의 단백질들을 분해함으로써 30시간이내 근육조직을 완전히 소멸시키는 것을 알 수 있었다. 한편, proteasome을 구성하는 특정 구성성분의 과다 발현이 세포의 단백질 분해현상에 어떠한 영향을 미치는 지 조사하기 위하여 쥐의 fibroblast cell에 26S proteasome의 ATPase 구성성분인 m56 유전자를 삽입한 후 세포의 추출물로부터 proteasome의 양과 활성의 변화를 관찰하였다. Proteasome을 초원심분리로 추출하여 관찰한 결과, 담배 박각시나방의 경우와는 달리 유전자 삽입 세포는 모든 실험에서 정상 세포와 전혀 차이를 보이지 않았다. 이 결과로부터 세포의 생명 유지에 필수적이고 매우 다양한 기능을 가진 proteasome은 40여개의 구성성분들의 발현이 매우 정교하게 조절되고 있으며, 특정 세포죽음 자극(apoptotic signal)이 왔을 때 일련의 구성 성분들의 발현과 26S proteasome 복합체 형성이 증가함으로써 그 기능을 수행하는 것을 알 수 있으며, 보다 자세한 연구는 비정상적 단백질 분해나 세포죽음을 동반하는 여러 질환들의 원인과 현상을 규명할 수 있으며, 또한 그 활성을 조절함으로써 궁극적인 치료의 방향도 제시 가능할 것으로 기대된다.

      • 지도자 유형이 태권도 수련생의 사회성 발달에 미치는 영향

        This study aims at exploring the effect of leadership type on Taekwondo trainees' development in sociality. in order to help as a reference for a Taekwondo training. The measuring instrument(Leadership Scale for Sport) used in the survey was developed by Chellandurai and Saleh(1980) and revised by Kim(1990). Items related with development in sociality argued by Ahn(1997) and Yeon(2001) were also used. Data were processed through SPSS. Frequency analysis was done to figure out background variables of the objects. Reliability analysis was done to verify the questionnaire reliability. Multiple regression was used to explore the effect of leadership behaviors on the trainees' development in sociality. The results were as follows Firstly, social supporting highly affected sub-factors of sociality development such as trust, justice. and patriotism. Secondly, positive rewarding behaviors highly affected sub-factors of trainees' sociality such as language use habits, responsibility. Thirdly, training and instructing behaviors by the trainers highly affected trainees' teamwork. When synthetically looking, it was found that trainees' development in sociality had various sub-factors as did the trainers' types. Accordingly, it was thought that proper custom-made training in developing sociality was important to trainees.
