        Impact of Ambient Temperature and Dietary Crude Protein in Wethers: Nitrogen Metabolism and Feed Efficiency

        Young lambs (Suffolk wethers, n=18), which were 22 to 26 kg average BW, were chronically exposed to temperatures of +1 to +$4^{\circ}C$ (cold) or +21 to +$24^{\circ}C$ (warm) during 10 wk experimental periods. The sheep were closely shorn and were housed in individual metabolism crates in controlled environment rooms. Sheep consumed pelleted diets ad libitum, which consisted of mainly barley grain and brome grass, and contained 7, 11, or 14% CP. The experimental design consisted of a $2{\times}3$ factorial with a single crossover of environment treatment. Feed intake, BW, feces, and urine excretion were measured. Apparent digestibilities were not affected by diet CP concentration or temperature treatments; however, voluntary intake per kg BW was increased (p<0.05) by diet CP content in both environments. Growing lambs gained weight slightly faster in a cold environment when N intake was above 27 g/d. Nitrogen excretion and N balance were positively related (p<0.01) with diet CP content, and fecal N excretion was significantly increased (p<0.05) in the cold environment. Therefore, dietary CP content strongly influenced N metabolism; however, cold exposure did alter only fecal N excretion. The higher DM intake per kg BW at 11% CP diet in the cold environment permitted ADG comparable to 14% CP diet in the warm environment. The results of this study do support the hypothesis that lambs are better able to utilize a moderate reduction in the CP content of the diet in a cold environment.

        Polymorphic Diversity of UBX Domain D from cDNA Isolated from Pectoral Muscle of Korean Native Chicken

        The objectives of this study are to identify specific functional genes which are related with growth and protein structure of the pectoral muscle of Korean native chicken. Pectoral muscle was isolated from three Korean native chickens (KNC, red brown, 12 months old, 2.41 ± 0.24 kg) and three Cornish chickens (16 month old, 2.76 ± 3.0 kg). The subtraction cDNA library was prepared in PCR4 Blunt-TOPO vector. The DNA sequence homology was compared with other breeds and species in GenBank. A clone NDS-81 was found to be unique for the DNA sequence homology with UBX family. Their partial sequence has high homology (98%) with chicken UBX domain D. Chicken UBX domain has chicken (93%), cattle (68%), dog (67%), mouse (64%) and, human (63%) nucleotide sequence homology. Several regions were mutated from T in chicken to C or G in the NDS-81 clone. The first site is LAD in chicken, but it was expressed as (L)RM in clone NDS-81. In this site, amino acids were changed from Ala to Arg, and from Asp to Met. The second site was changed from ER (Arg) in chicken to ED (Asp) in clone NDS-81. They are both containing functional side chains and play an important role in binding other proteins. Therefore, the clone NDS-81 could be a different candidate gene for the UBX family gene and could related with pectoral muscle structure of Korean native chicken.

        This report provides a study of effect on new interconnection in Dangjin area in terms of transient stability. KEPCO is planning to build a HVDC and HVAC line in Dangjin area in 2015. The case study used the Korean power system in summer peak in 2011. The results have been compared with the present status and test scenario followed the guidelines for analysis of transient stability in KEPCO system.

        Electroacupuncture induces Fos expression in the nucleus tractus solitarius via cholecystokinin A receptor signaling in rats.

        <P>OBJECTIVES: Cholecystokinin, a satiety hormone, acts on cholecystokinin A receptor on vagal afferent neurons that project to the nucleus tractus solitarius, resulting in inhibition of feeding. Cholecystokinin is known to be released by electroacupuncture stimulation at certain body sites which elicits profound psychophysiological responses. Our previous study has revealed the involvement of cholecystokinin and cholecystokinin A receptor in the electroacupuncture stimulation-induced modulation of feeding. The aim of the present study was to examine whether electroacupuncture stimulation at the acupuncture point ST36 (Joksamni) activates the nucleus tractus solitarius neurons and whether such effect is mediated by cholecystokinin A receptor. METHODS: Using an immunofluorescent analysis of Fos, a neuronal activation marker, we compared the Fos immunoreactivity of the nucleus tractus solitarius among three groups of Sprague-Dawley rats: (1) control (48 hour fasting + saline pre-treatment + no electroacupuncture stimulation); (2) SalEA (48 hour fasting + saline pre-treatment + ST36 electroacupuncture stimulation); (3) LorEA (48 hour fasting + pre-treatment of cholecystokinin A receptor antagonist, lorglumide + ST36 electroacupuncture stimulation). RESULTS: ST36 electroacupuncture stimulation significantly reduced 30 minute food intake (p<0.05, SalEA versus control) and increased Fos expression in the nucleus tractus solitarius (p<0.01, SalEA versus control). The effects of electroacupuncture on food intake and Fos were blocked by a lorglumide pre-treatment (p>0.05, LorEA versus control). DISCUSSION: Our finding suggests that ST36 electroacupuncture stimulation activates the nucleus tractus solitarius neurons via cholecystokinin A receptor signaling pathway, which may be the underlying central mechanism of electroacupuncture-induced satiety effect.</P>

        Development of a Gas Monitor Detector for the PAL-XFEL

        This paper presents the configuration and responses of a transmissive gas monitor detector (GMD) developed by the Pohang Accelerator Laboratory for its X-ray free-electron laser. The GMD is installed on the hard X-ray beamline to monitor the intensity of the X-ray pulses. The GMD exploits the photo-ionization of Xe at ~10-8 - 10-5 Torr by interactions with intense X-ray pulses and uses two-stage microchannel plates (MCPs) to detect the photo-ions and photo-electrons that are generated. The nominal gain of the MCP is 8.47 × 105 at an operation voltage of 1.5 kV. The results of the experiment show that the combined gain and detection efficiency of the MCP was approximately one-third of its reference value. However, readings obtained by using the developed GMD showed a strong correlation pulse by pulse with other diagnostic detectors, and the arrival time obtained from the time-of-flight spectrum of the photo-ions was consistent with our expectation and numerical results.

        Gene Set Analyses of Genome-Wide Association Studies on 49 Quantitative Traits Measured in a Single Genetic Epidemiology Dataset

        Gene set analysis is a powerful tool for interpreting a genome-wide association study result and is gaining popularity these days. Comparison of the gene sets obtained for a variety of traits measured from a single genetic epidemiology dataset may give insights into the biological mechanisms underlying these traits. Based on the previously published single nucleotide polymorphism (SNP) genotype data on 8,842 individuals enrolled in the Korea Association Resource project, we performed a series of systematic genome-wide association analyses for 49 quantitative traits of basic epidemiological, anthropometric, or blood chemistry parameters. Each analysis result was subjected to subsequent gene set analyses based on Gene Ontology (GO) terms using gene set analysis software, GSA-SNP, identifying a set of GO terms significantly associated to each trait ($p_{corr}$ < 0.05). Pairwise comparison of the traits in terms of the semantic similarity in their GO sets revealed surprising cases where phenotypically uncorrelated traits showed high similarity in terms of biological pathways. For example, the pH level was related to 7 other traits that showed low phenotypic correlations with it. A literature survey implies that these traits may be regulated partly by common pathways that involve neuronal or nerve systems.

      • <i>ChARM</i> : Discovery of combinatorial chromatin modification patterns in hepatitis B virus X-transformed mouse liver cancer using association rule mining

        <P><B>Background</B></P><P>Various chromatin modifications, identified in large-scale epigenomic analyses, are associated with distinct phenotypes of different cells and disease phases. To improve our understanding of these variations, many computational methods have been developed to discover novel sites and cell-specific chromatin modifications. Despite the availability of existing methods, there is still room for further improvement when they are applied to resolve the histone code hypothesis. Hence, we aim to investigate the development of a computational method to provide new insights into <I>de novo</I> combinatorial pattern discovery of chromatin modifications to characterize epigenetic variations in distinct phenotypes of different cells.</P><P><B>Results</B></P><P>We report a new computational approach, <I>ChARM</I> (<I>C</I>ombinatorial C<I>h</I>romatin Modification Patterns using <I>A</I>ssociation <I>R</I>ule <I>M</I>ining), that can be employed for the discovery of <I>de novo</I> combinatorial patterns of differential chromatin modifications. We used <I>ChARM</I> to analyse chromatin modification data from the livers of normal (non-cancerous) mice and hepatitis B virus X (HBx)-transgenic mice with hepatocellular carcinoma, and discovered 2,409 association rules representing combinatorial chromatin modification patterns. Among these, the combination of three histone modifications, a loss of H3K4Me3 and gains of H3K27Me3 and H3K36Me3, was the most striking pattern associated with the cancer. This pattern was enriched in functional elements of the mouse genome such as promoters, coding exons and 5′UTR with high CpG content, and CpG islands. It also showed strong correlations with polymerase activity at promoters and DNA methylation levels at gene bodies. We found that 30 % of the genes associated with the pattern were differentially expressed in the HBx compared to the normal, and 78.9 % of these genes were down-regulated. The significant canonical pathways (Wnt/ß-catenin, cAMP, Ras, and Notch signalling) that were enriched in the pattern could account for the pathogenesis of HBx.</P><P><B>Conclusions</B></P><P><I>ChARM</I>, an unsupervised method for discovering combinatorial chromatin modification patterns, can identify histone modifications that occur globally. <I>ChARM</I> provides a scalable framework that can easily be applied to find various levels of combination patterns, which should reflect a range of globally common to locally rare chromatin modifications.</P><P><B>Electronic supplementary material</B></P><P>The online version of this article (doi:10.1186/s12859-016-1307-z) contains supplementary material, which is available to authorized users.</P>
