Fungi belonging to the Paecilomyces spp. have recently been used as food and herbal medicines in Korea and are greatly popular as commercially available powdered supplement or dried fruiting body. Despite this acceptance and its use, little is known of the genes related to its reactive agents. Presently, We have constructed an olig-d(T) primed directional cDNA library from the silkworm Dongchunghacho, an entomopathogenic fungus, of which species is belonging to Paecilomyces spp. based on the previous identification of ITS1 and ITS2 at the molecular level and collected from Jocheon Miryang, Korea. To isolate and screen genes in the fungus, 626 expressed sequence tags(ESTs) were generated by a partial sequencing from the cDNA library. cDNA encoding the glyceraldehyde-3-phosphate dehydrogenase(Pt-GAPDH) of Paecilomyces tenuipes- Jocheon was cloned from the above cDNA library. The complete cDNA sequence of Pt-GAPDH is comprised of 1,014bp encoding 338 amino acid residues. The deduced protein sequence of Pt-GAPDH showed higher homology with Beauberia bassiana-GAPDH(93% amino acid identity). Hydropathy analysis revealed that Pt-GAPDH protein is hydrophilic. The major three amino acids in its composition of amino acid residues were alanine(11.54%), valine(9.47%) and glycine(8.88%). The Pt-GAPDH gene of Paecilomyces tenuipes entomopathogenic fungus consisted of three exons and two introns coding for 338 amino acid residues, and the genomic DNA length of the gene spans 1302bp. The accession number of the gene in GenBank are GU997099 for Pt-GAPDH cDNA and GU997102 for Pt-GAPDH genomic DNA. More investigation works including gene expression, immunological analysis etc. will be carried continuously without hesitation after this presentation.

The accession number of the gene in GenBank are GU997099 for Pt-GAPDH cDNA and GU997102 for Pt-GAPDH genomic DNA.
Focus Types and the Presupposition Function of Korean -To

This paper aims to provide the semantics of Korean -to for its main usage as an additive focus particle, by considering its constraints on the immediate discourse. Previous studies on the particle have mostly focused on its syntactic and semantic characteristics on sentential levels. The traditional treatment of -to as a trigger of existential presupposition does not account for a wider variety of its uses, especially when the focus part is not an argument, but a predicate or even the whole sentence. I propose a proposition function specifying the relation between the sentence containing the particle and the sentence in the context which satisfies its presupposition.
Self-Compensation of PZT Errors in White Light Scanning Interferometry

One of main error sources in white light scanning interferometry is the inaccuracy of scanning mechanisms in that PZT(piezoelectric transducer) micro-actuators are preferably used. We propose a new calibration method that is capable of identifying actual scanning errors directly by analyzing the spectral distribution of sampled interferograms. This calibration provides an effective means of self-compensation for the non-linearity errors caused by PZT hysteresis, enhancing the measurement uncertainty to a level of 5 nanometers over an entire measuring range of 100 ${\mu}{\textrm}{m}$.

Protein O-phosphorylation can occur reciprocal with O-GlcNAc modification and represents a regulatory principle for proteins. O-phosphorylation of serine by GSK-3β on Snail1, a transcriptional repressor of E-cadherin and a key regulator of the epithelial-mesenchymal transition (EMT) program, results in its proteasomal degradation. We show that Snail1 carries O-GlcNAc at serine112 that stabilizes it by suppressing O-phosphorylation-mediated degradation. Stabilization by O-GlcNAc of Snail1 results in attenuation of E-cadherin mRNA expression. Enhanced O-GlcNAc modification occurred under hyperglycemic conditions and initiated EMT by transcriptional suppression of E-cadherin through Snail1. Thus, a molecular link exists between cellular glucose metabolism and the control of EMT by dynamic reciprocal O-phosphorylation and O-GlcNAc modification of Snail1.
Effects of Gentiana scabra var. buergeri Extract on Toxoplasmastic Activity of Macrophages

Gentiana scabra var. buergeri (G. scabra) is a herb known to have therapeutic effect in infection diseases. We studied cellular activation and antitoxoplasmosis in macrophages after G. scabra stimulation. Macrophage activation was detected by nitrite production. Macrophages were treated with G. scabra extracted with water or methanol. Maximal nitrite production was detected in macrophages after stimulation of G. scabra extract 0.1 ㎎/㎖. Maximal nitrite concentration was 23.22 0.003 uM/L in macrophages after water extract of G. scabra and was 24.07 1.41 uM/L after methanol extract of G. scabra. Effect of G. scabra in the phagocytic capacity of macrophages was monitored by using PI (percentage of macrophage infected by T. gondiii method. The minimum PI (42.5 2.31) was detected in macrophages treated by water extract of G. scabra 0.1 ㎎/㎖ before infection of T. gondii. We also examined toxoplasmastatic capacity of macrophage using FI (fold increase) method. The minimum FI (4.46 1.16) was shown in macrophages after water extract of G. scabra 0.1 ㎎/㎖ pretreatment before infection. Under electron microscope, proliferation of T. gondii was inhibited by extract of G. scabra treatment in macrophages and the mitochondrion and lysosomal vacuoles within cells were increased. Taken together, G. scabra extract activates macrophages and induces toxopalsmastatic activity after T. gondii infection. It is suggested that G. scabra may be used as a therapeutic drug against toxoplasmosis.
