The purpose of this standard is to locate relevant existing guideline and test methods and to provide guidance for interim use of materials for which a standard does not exist. The standard may be use as guideline in tissue engineered research.

We provided standard practice for direct contact cell culture evaluation of materials for medical devices in this report. This practice introduced a reference method of direct contact cell culture testing. This method of direct contact cell culture may be used in the construction of medical materials and devices for test the cytotoxic potential of materials. Researcher for medical devices could use this practice to compare other cytotoxicity tests or directly to evaluate materials. Cell or materials could be sometimes modified depending on use in offered extend of this practice.

Matrix metalloproteinases (MMPs), a family of zinc-containing endopeptidases, participate in many normal processes such as embryonic development and wound repair, and in many pathological situations such as cancer, atherosclerosis, and arthritis. Peptidomimetic MMP inhibitors were designed and synthesized with N-formylhydroxylamine (retrohydroxamate) as a zinc-binding group and various side chains on the ${\alpha}$, P1', and P2' positions. Using in vitro MMP assays with purified MMPs (MMP-1, MMP-2, MMP-3, MMP-9, and MMP-14) and fluorogenic peptide substrates, it was found that compounds 2d and 2g selectively inhibit gelatinases (MMP-2 and MMP-9) and interstitial collagenase (MMP-1). They also inhibited the chemo-invasion of fibrosarcoma HT-1080 cells and tube formation of human umbilical vascular endothelial cells in a dose-dependent manner. Our results suggest that retrohydroxamate-based MMP inhibitors, especially compounds 2d and 2g, have the potential to be used as therapeutic drugs for cancer and other MMP-related diseases.

Toxicological studies have an object of detecting adverse effects of a chemical on an organism based on observed toxicity marker (i.e., serum biochemical markers and chemical-specific gene expression) or phenotypic outcome. To date, most toxicogenomic studies concentrated on hepatic toxicity. cDNA microarray analysis enable discrimination of the responses in animals exposed to different classes of hepatotoxicants. In an effort to further characterize the mechanisms of 2, 3, 7, 8,-Tetrachlorodibenzo-p-dioxin (TCDD or dioxin)-mediated toxicity, comprehensive temporal-responsive microarray analyses were performed on hepatic tissue from Sprague-Dawley rats treated with TCDD. Hepatic gene expression profiles were monitored using custom DNA chip containing 490 cDNA clones related with toxicology. Gene expression analysis identified 26 features which exhibited a significant change. In this study, we observed that the genes related with oxidative stress in rats exposed to Dioxin, such as CYPIIA3 and glutathione S-transferase, were up-regulated at 24hr after exposure. In this study, we carried out to discover novel evidence for previously unknown gene expression patterns related to mechanism of hepatic toxicity in rats exposed to dioxin, and to elucidate the effects of dioxin on the gene expression after exposure to dioxin.
Effects of Dioxin Exposed in Human by Using Radioactive cDNA Microarray

2, 3, 7, 8-Tetrachlorodibenzo-p-dioxin (TCDD) are well known as the most toxic environmental compound in these days. Many researches are reported that dioxin produces multiple toxic effects, such as endocrine toxicity, reproductive toxicity, immunotoxicity and cancer. In this study, we carried to discover novel evidence for previously unknown gene expression patterns in human exposed to dioxin by using radioactive cDNA microarray. 548 workers who were divided into experimental and control groups according to their urinary Naphthol levels were enrolled in our study. Blood mRNA in human was isolated, and the gene expression profiles were analyzed by cDNA microarray. Gene expression analysis identified 52 genes which exhibited a significant change. In our study, most notably, genes involved in cell cycle, cell proliferation, signal transduction and apoptosis in human exposed to dioxin, such as CCND3, TSHR, and EFRN5, were up-regulated. In the current study, we observed gene expression of people that are exposed to dioxin using radioactive cDNA microarray. Through these results, we suggest when objects are exposed to toxic compounds, such as dioxin, the radioactive cDNA microarray may be using in sensitively detecting of cancerous change.

Molecular weight distribution of copolyacrylonitrile, which was obtained from precipitation copolymerization without and with using dispersants in mixed solution, is studied. The contribution ratio of liquid phase polymerization and solid phase polymerization under different polymerization conditions could be worked out through the formula,which has been deduced in literature. From the calculated results, common points of each reaction system are, i) contribution ratio (r) of solid phase to liquid phase decreases with the increase of water content; thus the solid phase polymerization is gradually strengthened, which is apt to form chain of high molecular weight, ii) the higher temperature leads to higher compatibility between water and DMSO; thus the solid phase polymerization contribution would decrease,while the value of r is considerably larger. The limit molecular weight distribution of the system without dispersants in 100% water is approaching to 2; thus the corresponding r becomes larger, the molecular weight distribution ratio (Q) decreases in the system with dispersants.

This paper proposes quay crane (QC) scheduling algorithms that determine the working sequence of QCs over ship bays in a container vessel in automated container terminals. We propose two scheduling algorithms that examine the distribution of export containers in the stacking yard and determine the sequence of ship bays to balance the workload distribution among the yard blocks. One of the algorithms is a simple heuristic algorithm which dynamically selects the next ship bay based on the entropy of workloads among yard blocks whenever a QC finishes loading containers at a ship bay and the other uses genetic algorithm to search the optimal sequence of ship bays. To evaluate the fitness of each chromosome in the genetic algorithm, we have devised a method that is able to calculate an approximation of loading time of container vessels considering the workloads among yard blocks. Simulation experiments have been carried out to compare the efficiency of the proposed algorithms. The results show that our QC scheduling algorithms are efficient in reducing the turn-around time of container vessels.

Objective We investigated the impact of depressed mood (dysphoria) and loss of interest or pleasure (anhedonia)on the risk of dementia in cognitively-normal elderly individuals. Methods This study included 2,685 cognitively-normal elderly individuals who completed the baseline and 4-year follow-up assessments of the Korean Longitudinal Study on Cognitive Aging and Dementia. We ascertained the presence of dysphoria and anhedonia using the Mini International Neuropsychiatric Inventory. We defined subjective cognitive decline as the presence of subjective cognitive complaints without objective cognitive impairments. We analyzed the association of dysphoria and anhedonia with the risk of cognitive disorders using multinomial logistic regression analysis adjusted for age, sex, education, Cumulative Illness Rating Scale score, Apolipoprotein E genotype, and neuropsychological test performance. Results During the 4-year follow-up period, anhedonia was associated with an approximately twofold higher risk of mild cognitive impairment (OR=2.09, 95% CI=1.20–3.64, p=0.008) and fivefold higher risk of dementia (OR=5.07, 95% CI=1.44–17.92, p=0.012) but was not associated with the risk of subjective cognitive decline. In contrast, dysphoria was associated with an approximately twofold higher risk of subjective cognitive decline (OR=2.06, 95% CI=1.33–3.19, p=0.001) and 1.7-fold higher risk of mild cognitive impairment (OR=1.75, 95% CI=1.00–3.05, p=0.048) but was not associated with the risk of dementia. Conclusion Anhedonia, but not dysphoria, is a risk factor of dementia in cognitively-normal elderly individuals.
