        Perinatal Immunization With Vaccine-Grade Listeria monocytogenes Provides Protection Against Murine Th2 Airway Inflammation

        Purpose: Asthma is a chronic respiratory disorder that leads to inflammation and narrowing of the airways. Its global prevalence has attained epidemiclevels and treatment options that reach beyond temporary relief of symptoms are urgently needed. Since the processes leading to clinicallysymptomatic asthma start early in life, we set out to systematically evaluate a neonatal immunotherapeutic based on Listeria monocytogenes (Lm)for the control of allergic sensitization. Methods: We modified Lm to express the model allergen, ovalbumin (OVA), and tested the ability of neonatalimmunization with this strain to control allergic sensitization in a mouse model of OVA-induced asthma. Mice were immunized as newborns withlive or heat killed LmOVA or live Lm, followed 6 weeks later by allergic sensitization with OVA. In order to determine whether the TH1-polarizing effectof this vaccine vector inadvertently may exacerbate development of certain TH1-driven allergic diseases, mice immunized as newborns were assessedin a model of adult hypersensitivity pneumonitis (HP). Results: Both LmOVA and Lm-control vaccines were highly effective in providinglong-lasting protection from airway inflammation after only one immunization given perinatally. Serum antibody levels and lung cytokine productionsuggest that this prophylactic strategy is associated with an allergen specific TH1-dominated response. Specifically, LmOVA vaccinated mice displayedsignificantly elevated OVA-specific serum IgG2a, but no difference in anti-OVA IgE antibodies and only slightly decreased anti-OVA IgG1 antibodies. Importantly, Lm-based neonatal vaccination did not exacerbate Th1/Th17 driven HP, arguing against broad spectrum immune skewing. Conclusions:Our findings highlight the promise of early life Lm-based immunomodulatory interventions as a prophylactic strategy for allergic asthma.

      • Down-regulated MYH11 Expression Correlates with Poor Prognosis in Stage II and III Colorectal Cancer

        The MYH11 gene may be related to cell migration and adhesion, intracellular transport, and signal transduction. However, its relationship with prognosis is still uncertain. The aim of this study was to investigate correlations between MYH11 gene expression and prognosis in 58 patients with stage II and III colorectal cancer. Quantitative real-time polymerase chain reaction was performed in fresh CRC tissues to examine mRNA expression, and immunohistochemistry was performed with paraffin-embedded specimens for protein expression. On univariate analysis, MYH11 expression at both mRNA and protein levels, perineural invasion and lymphovascular invasion were related to disease-free survival (p<0.05; log-rank test). Cancers with lower MYH11 expression were more likely to have a poor prognosis. Otherwise, MYH11 expression was unrelated to patient clinicopathological features. On multivariate analysis, low MYH11 expression proved to be an independent adverse prognosticator (p<0.05). These findings show that MYH11 can contribute to predicting prognosis in stage II and III colorectal cancers.

        Glutamine-Supplemented Parenteral Nutrition and Probiotics in Four Adult Autoimmune Enteropathy Patients

        To evaluate the effects of glutamine-supplemented parenteral nutrition (PN) and probiotics in adult autoimmune enteropathy (AIE) patients. Four adult AIE patients were identified from April 2006 to January 2012. Clinical and nutritional data were obtained from the patients’ medical records. Glutamine-supplemented PN started immediately when the AIE diagnosis was confirmed. The total PN duration was 351 days. According to the PN prescription, the average caloric intake ranged from 20 to 25 kcal/kg/day, and the protein intake ranged from 1.2 to 1.5 g/kg/day. Alanyl-glutamine (20 g/day) was administered to AIE patients for 4 weeks followed by a 2-week break, and this treatment schedule was repeated when PN lasted for more than 6 weeks. Body weight gain and an increased serum albumin level were achieved after PN, and defecation frequency and quality also improved. Each patient received oral supplements, 250 mL of Ensure and two probiotics capsules (each capsule containing 0.5×108 colonies) three times a day when enteral nutrition started. Three AIE patients were successfully weaned off PN, and one patient died of pneumonia. Glutamine-supplemented PN and probiotics show promise in managing patients with AIE and related malnutrition.

      • A hybrid artificial intelligence and IOT for investigation dynamic modeling of nano-system

        In the present study, a hybrid model of artificial neural network (ANN) and internet of things (IoT) is proposed to overcome the difficulties in deriving governing equations and numerical solutions of the dynamical behavior of the nano-systems. Nano-structures manifest size-dependent behavior in response to static and dynamic loadings. Nonlocal and length-scale parameters alongside with other geometrical, loading and material parameters are taken as input parameters of an ANN to observe the natural frequency and damping behavior of micro sensors made from nanocomposite material with piezoelectric layers. The behavior of a micro-beam is simulated using famous numerical methods in literature under base vibrations. The ANN was further trained to correlate the output vibrations to the base vibration. Afterwards, using IoT, the electrical potential conducted in the sensors are collected and converted to numerical data in an embedded mini-computer and transferred to a server for further calculations and decision by ANN. The ANN calculates the base vibration behavior with is crucial in mechanical systems. The speed and accuracy of the ANN in determining base excitation behavior are the strengths of this network which could be further employed by engineers and scientists.

        Adaptive Finite-time Consensus for Second-order Nonlinear Multi-agent Systems with Input Quantization

        In this paper, the adaptive finite-time consensus (FTC) control problem of second-order nonlinear multiagent systems (MASs) with input quantization and external disturbances is studied. With the help of finite time control technology, a novel distributed adaptive control protocol is constructed to achieve FTC performance for second-order nonlinear MASs by using the recursive method. The control input is quantized through a hysteresis quantizer, which reduces the communication rate of arbitrary two agents. The unknown functions are approximated by adopting the radial basis function neural networks. Under the consensus protocols and adaptive laws, it can be proved that velocity errors of arbitrary two agents reach a small region of zero in finite time as well as position errors. Finally, the effectiveness of the proposed method is illustrated via a simulation example.

        AN IMPROVED METHOD FOR TRANSFERRING GRAPHENE GROWN BY CHEMICAL VAPOR DEPOSITION

        In this paper, we report an improved transfer of graphene by directly picking up the graphene with target substrates and the comparison of such transferred graphene samples with other graphene samples transferred by the commonly used "PMMA-based transfer" method. Raman spectroscopy studies show that this "direct transfer method" does not degrade the graphene structure and field effect transistor measurements show that it does not introduce any extra doping in graphene; in contrast, PMMA-based transfer samples have strong n-type doping. We also find that graphene samples achieved by this direct transfer method show higher mobility than graphene obtained from the PMMA-based transfer method.

        Improved Function Augmented Sliding Mode Control of Uncertain Nonlinear Systems with Preassigned Settling Time

        This paper investigates the robust tracking control of second-order uncertain nonlinear systems by adopting the function augmented sliding mode control approach. An improved version of this approach is proposed such that the exact information of the initial tracking error is not required when generating the desired trajectory of the tracking error. This evidently enlarges the application scope of the function augmented sliding mode control approach. Performance functions are introduced to form the performance envelopes for the sliding mode variables. A robust sliding mode controller is constructed such that the sliding mode variables are confined within their performance envelopes. This further guarantees that the tracking error converges to the given neighbourhood of zero within the preassigned settling time provided that proper control parameters are selected. An application example on the rendezvous control of spacecraft is also employed to illustrate the effectiveness of the proposed control approach.

        Damage detection of nonlinear structures with analytical mode decomposition and Hilbert transform

        This paper proposes an analytical mode decomposition (AMD) and Hilbert transform method for structural nonlinearity quantification and damage detection under earthquake loads. The measured structural response is first decomposed into several intrinsic mode functions (IMF) using the proposed AMD method. Each IMF is an amplitude modulated-frequency modulated signal with narrow frequency bandwidth. Then, the instantaneous frequencies of the decomposed IMF can be defined with Hilbert transform. However, for a nonlinear structure, the defined instantaneous frequencies from the decomposed IMF are not equal to the instantaneous frequencies of the structure itself. The theoretical derivation in this paper indicates that the instantaneous frequency of the decomposed measured response includes a slowly-varying part which represents the instantaneous frequency of the structure and rapidly-varying part for a nonlinear structure subjected to earthquake excitations. To eliminate the rapidly-varying part effects, the instantaneous frequency is integrated over time duration. Then the degree of nonlinearity index, which represents the damage severity of structure, is defined based on the integrated instantaneous frequency in this paper. A one-story hysteretic nonlinear structure with various earthquake excitations are simulated as numerical examples and the degree of nonlinearity index is obtained. Finally, the degree of nonlinearity index is estimated from the experimental data of a seven-story building under four earthquake excitations. The index values for the building subjected to a low intensity earthquake excitation, two medium intensity earthquake excitations, and a large intensity earthquake excitation are calculated as 12.8%, 23.0%, 23.2%, and 39.5%, respectively.
