Both/And: Reflections on Recent Anglo/Western Early Childhood Curriculum Statements

This paper identifies innovative work on curriculum undertaken in New Zealand in the 1990s as inspirational for recently developed frameworks in Australia and parts of Canada. The paper argues that the form this incorporation has taken in Canada and Australia is, almost literally, an opening of ‘some space’—as the more established modernist and technical understandings have not been entirely transformed or removed, but rather joined by perspectives that emphasize the need for a more complex, reflective and contextually situated practitioner—creating a contemporary policy landscape of ‘both/and’. The paper describes and analyzes in detail the processes and dynamics of the ‘both/and’ landscape achieved in British Columbia, Canada.
Monocytes in Aging and Exercise

Monocytes are cells of the innate immune system which perform important functions, including inflammatory cytokine production and phagocytosis, and participate in pathogen defense. In aging and disease, monocyte functions are dysregulated, and this dysregulation has been conclusively linked to the pathogenesis of a variety of age-related chronic diseases. In this review, we summarize the current literature supporting exercise training as a potential therapy for improving monocyte function, with particular attention to studies in the aging population. Substantial evidence suggests exercise is an effective therapy for modulating monocyte biology, although mechanisms by which this occurs are still not well understood.

<P><I>N</I><SUP>2</SUP>,3-Ethenoguanine (<I>N</I><SUP>2</SUP>,3-ϵG) is one of the exocyclic DNA adducts produced by endogenous processes (<I>e.g.</I> lipid peroxidation) and exposure to bioactivated vinyl monomers such as vinyl chloride, which is a known human carcinogen. Existing studies exploring the miscoding potential of this lesion are quite indirect because of the lability of the glycosidic bond. We utilized a 2′-fluoro isostere approach to stabilize this lesion and synthesized oligonucleotides containing 2′-fluoro-<I>N</I><SUP>2</SUP>,3-ϵ-2′-deoxyarabinoguanosine to investigate the miscoding potential of <I>N</I><SUP>2</SUP>,3-ϵG by Y-family human DNA polymerases (pols). In primer extension assays, pol η and pol κ replicated through <I>N</I><SUP>2</SUP>,3-ϵG, whereas pol ι and REV1 yielded only 1-base incorporation. Steady-state kinetics revealed that dCTP incorporation is preferred opposite <I>N</I><SUP>2</SUP>,3-ϵG with relative efficiencies in the order of pol κ > REV1 > pol η ≈ pol ι, and dTTP misincorporation is the major miscoding event by all four Y-family human DNA pols. Pol ι had the highest dTTP misincorporation frequency (0.71) followed by pol η (0.63). REV1 misincorporated dTTP and dGTP with much lower frequencies. Crystal structures of pol ι with <I>N</I><SUP>2</SUP>,3-ϵG paired to dCTP and dTTP revealed Hoogsteen-like base pairing mechanisms. Two hydrogen bonds were observed in the <I>N</I><SUP>2</SUP>,3-ϵG:dCTP base pair, whereas only one appears to be present in the case of the <I>N</I><SUP>2</SUP>,3-ϵG:dTTP pair. Base pairing mechanisms derived from the crystal structures explain the slightly favored dCTP insertion for pol ι in steady-state kinetic analysis. Taken together, these results provide a basis for the mutagenic potential of <I>N</I><SUP>2</SUP>,3-ϵG.</P>

Purpose: The current Classification of Periodontal and Peri-Implant Diseases and Conditions, published and disseminated in 2018, involves some difficulties and causes diagnostic conflicts due to its criteria, especially for inexperienced clinicians. The aim of this study was to design a decision system based on machine learning algorithms by using clinical measurements and radiographic images in order to determine and facilitate the staging and grading of periodontitis. Methods: In the first part of this study, machine learning models were created using the Python programming language based on clinical data from 144 individuals who presented to the Department of Periodontology, Faculty of Dentistry, Süleyman Demirel University. In the second part, panoramic radiographic images were processed and classification was carried out with deep learning algorithms. Results: Using clinical data, the accuracy of staging with the tree algorithm reached 97.2%, while the random forest and k-nearest neighbor algorithms reached 98.6% accuracy. The best staging accuracy for processing panoramic radiographic images was provided by a hybrid network model algorithm combining the proposed ResNet50 architecture and the support vector machine algorithm. For this, the images were preprocessed, and high success was obtained, with a classification accuracy of 88.2% for staging. However, in general, it was observed that the radiographic images provided a low level of success, in terms of accuracy, for modeling the grading of periodontitis. Conclusions: The machine learning-based decision system presented herein can facilitate periodontal diagnoses despite its current limitations. Further studies are planned to optimize the algorithm and improve the results.
Duplex-Specific Nuclease-Amplified Detection of MicroRNA Using Compact Quantum Dot-DNA Conjugates

<P>Advances in nanotechnology have provided new opportunities for the design of next-generation nucleic acid biosensors and diagnostics. Indeed, combining advances in functional nanoparticles, DNA nanotechnology, and nuclease-enzyme-based amplification can give rise to new assays with advantageous properties. In this work, we developed a microRNA (miRNA) assay using bright fluorescent quantum dots (QDs), simple DNA probes, and the enzyme duplex-specific nuclease. We employed an isothermal target-recycling mechanism, where a single miRNA target triggers the cleavage of many DNA signal probes. The incorporation of DNA-functionalized QDs enabled a quantitative fluorescent readout, mediated by Förster resonance energy transfer (FRET)-based interaction with the DNA signal probes. Our approach splits the reaction in two, performing the enzyme-mediated amplification and QD-based detection steps separately such that each reaction could be optimized for performance of the active components. Target recycling gave ca. 3 orders of magnitude amplification, yielding highly sensitive detection with a limit of 42 fM (or 1.2 amol) of miR-148, with excellent selectivity versus mismatched sequences and other miRNAs. Furthermore, we used an alternative target (miR-21) and FRET pair for direct and absolute quantification of miR-21 in RNA extracts from human cancer and normal cell lines.</P> [FIG OMISSION]</BR>

In her introduction to this special issue of Cross-Currents, Lorraine Paterson sets the scene for the featured articles and provides the context from which they emerged. This endnote essay works to a reverse formula, seeking if not to unweave then to unsettle the notion of bounded coherence and to spotlight instead the inchoate nature of any project to document the past. Rather than braiding together threads of our contributors' articles to signal the thematic coherence of this suite of papers on “mediating Chineseness,” I work instead from key themes presented in this collection to gesture towards loose ends and unstitched seams. Mirroring the genre of Willmott’s memoir essay, I invite reflection on these in-between spaces and the possibilities they represent for developing new approaches to the articulation, study, and mediation of “Chineseness” in Cambodia.
