Alzheimer``s disease severely compromises cognitive function. One of the mechanisms to explain the pathology of Alzheimer’sdisease has been the hypotheses of amyloid-pore/channel formationby complex Aβ-aggregates. Clinical studies suggestedthe moderate alcohol consumption can reduces probability developingneurodegenerative pathologies. A recent report exploredthe ability of ethanol to disrupt the generation of complexAβ in vitro and reduce the toxicity in two cell lines. Molecular dynamics simulations were applied to understandhow ethanol blocks the aggregation of amyloid. On the otherhand, the in silico modeling showed ethanol effect over thedynamics assembling for complex Aβ-aggregates mediated bybreak the hydrosaline bridges between Asp 23 and Lys 28, wasare key element for amyloid dimerization. The amyloid pore/channel hypothesis has been explored only in neuronal models,however recently experiments suggested the frog oocytessuch an excellent model to explore the mechanism of the amyloidpore/channel hypothesis. So, the used of frog oocytes toexplored the mechanism of amyloid aggregates is new, mainlyfor amyloid/pore hypothesis. Therefore, this experimentalmodel is a powerful tool to explore the mechanism implicatesin the Alzheimer’s disease pathology and also suggests a modelto prevent the Alzheimer’s disease pathology.

The area under a receiver operating characteristic (ROC) curve (AUC) is a popular measure of pure diagnostic accuracy that is independent from the proportion of diseased subjects in the analysed sample. However, its actual usefulness in the clinical context has been questioned, because it does not seem to be directly related to the actual performance of a diagnostic marker in identifying diseased and non-diseased subjects in real clinical settings. This study evaluates the relationship between the AUC and the proportion of correct classifications (global diagnostic accuracy, GDA) in relation to the shape of the corresponding ROC curves. We demonstrate that AUC represents an upward-biased measure of GDA at an optimal accuracy cut-off for balanced groups. The magnitude of bias depends on the shape of the ROC plot and on the proportion of diseased and non-diseased subjects. In proper curves, the bias is independent from the diseased/non-diseased ratio and can be easily estimated and removed. Moreover, a comparison between 2 partial AUCs can be replaced by a more powerful test for the corresponding whole AUCs. Applications to 3 real datasets are provided: a marker for a hormone deficit in children, 2 tumour markers for malignant mesothelioma, and 2 gene expression profiles in ovarian cancer patients. In conclusion, the AUC is a measure of accuracy with potential clinical relevance for the evaluation of disease markers. The clinical meaning of ROC parameters should always be evaluated with an analysis of the shape of the corresponding ROC curve.
Functional and Structural Effects of Amyloid-beta Aggregate on Xenopus laevis Oocytes

Xenopus laevis oocytes exposed to amyloid- aggregate generated oscillatory electric activity (blips) that was recorded by two-microelectrode voltage-clamp. The cells exhibited a series of “spontaneous” blips ranging in amplitude from 3.8 0.9 nA at the beginning of the recordings to 6.8 1.7 nA after 15 min of exposure to 1 M aggregate. These blips were similar in amplitude to those induced by the channel-forming antimicrobial agents amphotericin B (7.8 1.2 nA) and gramicidin (6.3 1.1 nA). The amyloid aggregate-induced currents were abolished when extracellular Ca2+ was removed from the bathing solution, suggesting a central role for this cation in generating the spontaneous electric activity. The amyloid aggregate also affected the Ca2+-dependent Cl- currents of oocytes, as shown by increased amplitude of the transient-outward chloride current (Tout) and the serum-activated, oscillatory Cl- currents. Electron microcopy revealed that amyloid aggregate induced the dissociation of the follicular cells that surround the oocyte, thus leading to a failure in the electro-chemical communication between these cells. This was also evidenced by the suppression of the oscillatory Ca2+-dependent ATP-currents, which require proper coupling between oocytes and the follicular cell layer. These observations, made using the X. laevis oocytes as a versatile experimental model, may help to understand the effects of amyloid aggregate on cellular communication.

The occurrence of Laminarionema elsbetiae (Ectocarpaceae, Phaeophyceae), as epi-endophyte of Rhodymenia pseudopalmata (Rhodymeniales, Rhodophyta), described from Santa Isabel, Rawson, Argentina. L. elsbetiae grows in the host tissues forming epi-endophytic relationship in the epidermal, cortical and medullar layers. Epiphytic thalli of L. elsbetiae were unbranched filaments emerging from hostis surface. Reproductive structures of L. elsbetiae on the host were absent. On the contrary, free cultured individuals formed different reproductive structures. Macrozoosporangia containing a single large motile zoospore originated from vegetative cells, they were conical to cylindrical in shape, 30-50 ${\mu}m$ in length and 18-20 ${\mu}m$ in wide. Uniseriate plurilocular zoosporangia were cylindrical shape, 40 ${\mu}m$ in length and 10-13 ${\mu}m$ in wide. Sexual fusion was not seen. In mixed cultures of L. elsbetiae with R. pseudopalmata fronds, L. elsbetiae infected the host, grew as in natural host and, formed macrosporangia between host subcortical cells. Gametophytes of L. elsbetiae were filaments with diffuse growth, branched with a branch pattern alternate or opposite. Gametangia were plurilocular, uni or biseriate and lateral. When mature they contained 2 to 6 isogametes. The presence L. elsbetiae on R. pseudopalmata could be defined as an epi-endophytic relationship. The percentage of infection of R. pseudopalmata thalli by L. elsbetiae was 34%.A25% of the infected thalli presented a low, non-symptomatic level infection, whereas a 62% and a 13% of them exhibited respectively moderate and high indexes of infection.
Acute urticarial rash after COVID-19 vaccination containing Polysorbate 80

We present the case of a 48-year-old Caucasian woman, who developed an acute urticarial rash after the second dose of coronavirus disease 2019 (COVID-19) vaccination with Oxford-AstraZeneca. Though the most common cutaneous adverse reactions to vaccines are non-allergic, we believe the rash may represent an immediate hypersensitivity type I reaction against the vaccine excipient Polysorbate 80 (Pol80), configuring an acute allergic urticaria. Skin prick test with Pol80, were performed and resulted positive, confirming the role of Pol80 in eliciting immediate hypersensitivity in our patient. Of note, sensitizing excipients contained in COVID-19 vaccines are commonly used in everyday products and preexisting sensitizations may cause allergic reactions to vaccines, highlighting the need to undergo allergy consultation upon vaccine administration.

We present the case of a 24-year-old Caucasian man, who developed a scaly erythematous skin rash after the second dose of coronavirus disease 2019 (COVID-19) vaccination with Comirnaty (BNT162b2, BioNTech/Pfizer; Pfizer, New York, NY, USA) and proved positive for Epstein-Barr virus (EBV) reactivation. The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) mRNA vaccines have been associated with an up-regulated T helper type 1-cell response, possibly favoring an immune system imbalance. Also, EBV reactivation has been postulated after COVID-19 vaccination, but only in the immunosuppressed. Noteworthy we report the first case of EBV viral reactivation associated with cutaneous manifestations in an immunocompetent patient after the COVID-19 vaccine.
